Stellantchasmus falcatus (Digenea: Heterophyidae) in Cambodia: Discovery of Metacercariae in Mullets and Recovery of Adult Flukes in an Experimental Hamster

  • Stellantchasmus falcatus (Digenea: Heterophyidae) is first reported from Cambodia through recovery of the metacercariae from mullet fish and adult flukes from an experimentally infected hamster. We purchased 7 mullets, Chelon macrolepis, in a local market of Phnom Penh, Cambodia, and each of them was examined by the artificial digestion method on May 2010. The metacercariae of S. falcatus were detected in all mullets (100%) examined, and their average density was 177 per fish. They were elliptical, $220{\times}168{\mu}m$ in average size. They were orally infected to an hamster to obtain adult flukes. Adults recovered at day 10 post infection were observed with a light microscope and a scanning electron microscope (SEM). They were small, $450{\times}237{\mu}m$ in average size, had a small oral sucker ($41{\times}50{\mu}m$), subglobular pharynx ($29{\times}21{\mu}m$), slender esophagus ($57{\mu}m$), long and thick-walled expulsor ($119{\times}32{\mu}m$), spherical ovary ($58{\times}69{\mu}m$), and 2 ovoid testes (right: $117{\times}74{\mu}m$; left: $114{\times}63{\mu}m$). Eggs were small, yellow, and $23{\times}12{\mu}m$ in average size. In SEM observations, tegumental spines were densely distributed on the whole tegument, and single small type I sensory papillae were distributed around the lip of oral sucker. The small ventral sucker was dextrally located and had 8 type I sensory papillae on the left margin. It has been first confirmed in the present study that the mullet, C. macrolepis, is playing the role of a second intermediate host of S. falcatus in Cambodia.

Secretory Differentiation of Hamster Tracheal Epithelial Cells Increases Activation of Matrix Metalloproteinase-2

  • In chronic airway inflammatory diseases such as asthma and chronic bronchitis, it has been suggested that matrix metalloproteinases secreted from infiltrating neutrophil contribute the pathogenesis of the disease and have been a focus of intense investigation. We report here that hamster tracheal surface epithelial goblet cells (HTSE cells) produce matrix metalloproteinase-2 (MMP-2) and tissue inhibitor of metalloproteinase-2 (TIMP-2). Matrix metalloproteinase activities were investigated using [$^3H$]collagen-digestion assay and gelatin zymography. The subtype of matrix metalloproteinases expressed from HTSE cells was MMP-2 (gelatinase A), which was determined by Western blot with various subtype selective anti-matrix metalloproteinase antibodies. The MMP-2 and TIMP-2 cDNAs from HTSE cells were partially cloned by RT-PCR and they reveal more than 90% of sequence homology with those from human, rat and mouse. The collagenolytic activity was increased with the secretory differentiation of the HTSE cell and it was found that zymogen activation was responsible for the increased MMP-2 activity in HTSE cells. The results from the present study suggest that the metaplastic secretory differentiation of airway goblet cells may affect chronic airway inflammatory process by augmenting the zymogen activation of MMP-2.

A Promising Serum Autoantibody Marker, Anti-Heat Shock Protein 90α, for Cholangiocarcinoma

  • The present study was designed to investigate cholangiocarcinoma (CCA) antibodies in hamster serum. Hamster CCA cell lines were processed using sodium dodecyl sulfate-polyacrylamide gel electrophoresis and matrix-assisted laser desorption/ionization time-of-flight mass spectrometry. A candidate biomarker was confirmed by immunoprecipitation and western blot, and was further analyzed using ELISA and sera from normal control hamsters, hamsters with opisthorchiasis and hamsters with various stages of CCA, as well as from CCA patients and healthy individuals. One candidate marker was identified as $HSP90{\alpha}$, as indicated by a high level of anti-$HSP90{\alpha}$ in hamster CCA sera. It was found that the levels of anti-$HSP90{\alpha}$ were specifically elevated in the sera of hamsters with CCA compared with other groups and progressively increased with the clinical stage. At the cut-off point of 0.4850 on the receiver operating characteristic curve, anti-$HSP90{\alpha}$ could discriminate CCA from healthy control groups with a sensitivity of 76.2%, specificity of 71.4% and total accuracy 75.5%. In the present study, we have shown that anti-$HSP90{\alpha}$ may be a potential useful serum biomarker to discriminate CCA cases from healthy persons.

Saffron Reduction of 7,12-Dimethylbenz[a]anthracene-induced Hamster Buccal Pouch Carcinogenesis

  • Our aim was to investigate the chemopreventive potential of saffron in DMBA-induced hamster buccal pouch carcinogenesis. Assessment was by monitoring the percentage of tumor bearing hamsters, tumor size as well as the status of detoxification agents, lipid peroxidation and antioxidants. Oral squamous cell carcinomas were induced in the buccal pouch of Syrian golden hamsters by painting them with 0.5% DMBA in liquid paraffin three times a week for 14 weeks. We observed 100% oral tumor formation with severe histopathological abnormalities in all the hamsters treated with DMBA alone, activities of phase I and phase II detoxification enzymes, lipid peroxidation and antioxidants being significantly altered. Though oral administration of saffron completely prevented the formation of tumors, we noticed severe hyperplasia and dysplasia in hamsters treated with DMBA, suggesting that tumors might eventually develop. Oral administration of saffron return detoxification enzymes, lipid peroxidation and antioxidants to normal ranges. The chemopreventive potential of saffron thus is likely due to antioxidant properties and modulating effects on detoxification in favour of the excretion of carcinogenic metabolites during DMBA-induced hamster buccal pouch carcinogenesis.

Overproduction of Recombinant Human VEGF (Vascular Endothelial Growth Factor) in Chinese Hamster Ovary Cells

  • Vascular endothelial growth factors (VEGFs) are a family of proteins that mediate angiogenesis. $VEGF_{165}$ is a VEGF-A isoform and has been extensively studied owing to its potential use in therapeutic angiogenesis. This study established Chinese hamster ovary (CHO) cells overexpressing recombinant human $VEGF_{165}$ $(rhVEGF_{165})$ protein. The production rate of the established CHO cells was over 80mg/l of $rhVEGF_{165}$ protein from a 7-day batch culture process using a 7.5-l bioreactor with a 5-l working volume and serum-free medium. The $rhVEGF_{165}$ protein was purified to homogeneity from the culture supernatant using a two-step chromatographic procedure that resulted in a 48% recovery rate. The purified $rhVEGF_{165}$ protein was a glycosylated homodimeric protein with a higher molecular weight (MW) than the protein expressed from insect cells, suggesting that the glycosylation of the $rhVEGF_{165}$ protein in CHO cells differed from that in insect cells. The purified $rhVEGF_{165}$ protein in this study was functionally active with a half-maximal effective concentration of 3.8ng/ml and specific activity of $2.5{\times}10^5U/mg$.

Morphological Changes in The Central Canal of the Hamster Spinal Cord after Treatment with 6-Aminonicotinamide (6-Aminonicotinamide 투여 후 햄스터 척수 중심관의 형태변화)

  • Hydrocephalus is induced experimentally in prenatal and suckling animals following an injection of 6-aminonicotinamide (6-AN). The most remarkable characteristic of these animals is aqueduct stenosis caused by swellings of the ependymal cells and subependymal cells in the periaqueductal gray matter and the central canal of the spinal cord. The present study was undertaken to investigate the morphological changes of the ependymal cells in the central canal of the spinal cord of 3.5 months old hamster after treatment with 6-AN. Intraperitoneal administrations of 6-AN (10 mg/kg body weight) every two days gave rise to partial central canal stenosis of the spinal cord after 27-29 days (13-l4th injection), but cilia and microvilli were located in the strictural area of the con#rat canal. The vacuolations in the ependymal cells were not observed and degenerating changes of intracellular organelles of the ependymal cells did not occur, so that the ependymal cells lining the central canal of the hamster spinal cord were not affected by 6-AN. But the present study demonstrate that 6-AN causes to create numerous vacuoles in the subependymal area of the central canal. Although the vacuoles were well developed in the neuroglial cells and the neuropils of the subependymal area, the neurons were not affected by 6-AN. These results strongly suggests that partial central canal stenosis occurred by 6-AN was due to vacuolations and swellings of the neuroglial cells and nueropils in the subependymal area.

The Study On The Effect Of Chungsang bangpungtang and Lonicerae Flos, Coicis Semen (CBTLC) On Acnes (淸上防風湯加味이 面포에 미치는 影響)

  • This study was carried out to prove the effects of CBTLC on the $5{\alpha}$-reductase inhibition, the sterilizing power for Propionibactrium acnes, the cytotoxicity of human monocyte, the inhibition for prosta-glandins($PGE_2$), Interleukins(IL-$1{\beta}$) and TNF-${\alpha}$ in inflammation, and the size of Hamster ear sebaceous gland concerned with Acnes. The result were obtained as follows : 1. On the $5{\alpha}$-reductase inhibition of CBTLC in vitro, An undiluted solution of CBTLC was $71{\%}$ inhibition on $5{\alpha}$-reductase and $\frac{1}{10}$ diluted solution of CBTLC was $20{\%}$ inhibition on $5{\alpha}$-reductase. 2. On the sterilizing power for Propionibactrium acnes concerned in Acnes, an undiluted solution and $\frac{1}{10}$ diluted solution of CBTLC formed 12mm clear zone diameters. 3. CBTLC was showed the cytotoxicity of human monocyte in $0.08{\%}$ and $0.12{\%}$ of CBTLC. 4. $0.01{\%}$ and $0.04{\%}$ of CBTLC inhibited the production of prostaglandins($PGE_2$) in the human monocyte stimulated with P. acnes LPS. 5. $0.08{\%}$ and downward of GMHBS inhibited the production of interleukins(lL-$1{\beta}$) in the human monocyte stimulated with P. acnes LPS. 6. $0.08{\%}$ and $0.12{\%}$ of GMHBS inhihited the production of TNF-${\alpha}$ in the human monocyte stimulated with P. acnes LPS. 7. As the antiandrogenic compound, CBTLC was used in hamster ears with topical application. CBTLC was diminished slightly on the size of hamster ears sebaceous gland.

Expression and Purification of Biologically Active Human Bone Morphogenetic Protein-4 in Recombinant Chinese Hamster Ovary Cells

  • Bone morphogenetic protein-4 (BMP-4) is considered to have therapeutic potential for various diseases, including cancers; however, the high expression of biologically active recombinant human BMP-4 (rhBMP-4) needed for its manufacture for therapeutic purposes has yet to be established. In the current study, we established a recombinant Chinese hamster ovary (rCHO) cell line overexpressing rhBMP-4 as well as a production process using 7.5-l bioreactor (5 L working volume). The expression of the mature rhBMP-4 was significantly enhanced by recombinant furin expression. The combination of a chemically defined medium and a nutrient supplement solution for high expression of rhBMP-4 was selected and used for bioreactor cultures. The 11-day fed-batch cultures of the established rhBMP-4-expressing rCHO cells in the 7.5-L bioreactor produced approximately 32 mg/l of rhBMP-4. The mature rhBMP-4 was purified to homogeneity from the culture supernatant using a two-step chromatographic procedure, resulting in a recovery rate of approximately 55% and a protein purity greater than 95%. The N-terminal amino acid sequences and N-linked glycosylation of the purified rhBMP-4 were confirmed by N-terminal sequencing and de-N-glycosylation analysis, respectively. The mature purified rhBMP-4 has been proved to be functionally active, with an effective dose concentration of $EC_{50}$ of 2.93 ng/ml.

Some Chromosome Alteranations in the Cultured Chinese Hamster Cells Treated by Steroids (Steroid 물질처리를 받은 Chinese hamster 세포에 있어서의 염색체 이상)

  • This study is concerned with alterations in chromosomes (numbers and morphology) when the culture of Chinese hamster cells (FAF-28 strain) was treated by steroids, testosterone and DOC. 1. In 200 cells of normal untreated cells as control population the chromosome of stemline was decided as which was contained in 158 cells ; that is , in 79 percent of the population. The average chromosome number in above 20 cells observed was calculated as 23.95 with minimum limit at 20 and maximum limit at 70. 2. Many different chromosome numbers, ranging from 19 to 352 were observed in the 200 cells treated by testosterone. The diploid number of 22 showed the peak of variation curve was counted in 71 cells (35.5%) and an average chromosome number of stemline was 22 which was counted in 74 cells (37%). While all of the chromosome number of stemline was 22 which was counted in 74 cells (37%). While all of the chromosome numbers in the 200 cells observed ranged from 20 to 181 , an average chromosome number was also found to be 30.09. 4. The chromosome component in the cultured normal FAF-28 cells with 22 diploid chromosomeswas as follows ; 9a) 2 paris were long and metacentric (LM), (b) 3 pairs were medium length and metacentric (MM), (c) 3 pairs were small and subtelocentric (SS) and (d) 3 pairs were small and metacentric (SM). 5. The twenty cells with 44 chromosomes were selected at random from each cell population treated with testosterone and DOC , so that chromosome idiogram and morphology could be studies. In the twenty cells of the testosterone treated population the average ratio of above four groups, LM ; MM;Ss:SM, was found to be 8.6 : 10.8:13.5:10.7. On the other hand, the average ratio in the same number of cells of the DOC treated one was 7.7 :11.4:12.5:12.7. 6. The five types of the altered chromosomes morphologically in the hundred cells selected at random from each cell population treated by testosterone and DOC were observed (Type I-V). The thirty-one altered chromosomes were found to be in the testosterone treated cell population and the sixteen in DOC treated.

Evaluation of Genotoxicity of CP Pharmacopuncture Using an In Vitro Chromosome Aberration Test in Chinese Hamster Lung Cell (Chinese Hamster Lung 세포를 이용한 염색체이상 시험을 이용한 CP약침의 유전독성평가)

  • This study was designed to assess the toxicity of capsaicin-containing (CP) pharmacopunture using an in vitro chromosomal aberrations in Chinese hamster lung (CHL/IU) cells. In order to determine the high dose level in the main study of this study, a dose range finding study was conducted first. The high dose was selected at 10.0% of CP pharmacopuncture extract, and then diluted sequentially to produce lower dose levels of 5.00, 2.50, 1.25, 0.625 and 0.313% by applying a geometric ratio of 2. As a result, the cytotoxicity and precipitation of the CP pharmacopuncture as a test substance were not evident at any dose level during short-time treatment with and without metabolic activation and continuous treatment without metabolic activation. Therefore, the dose levels for this study were chosen as 10.0, 5.0, and 2.5%., and the treatment volume was 1.3 mL. In addition, negative and positive controls were set. In main study, the frequency of cells with chromosome aberrations in CP treated groups was less than 5% in short-time treatment with and without metabolic activation and continuous treatment without metabolic activation. In addition, there was no statistically significant difference when compared to the negative control group. The frequency of cells with structural chromosomal aberrations in the positive control group was more than 10% compared to the negative control group, and it increased statistically significantly. In conclusion, under the conditions of this study, CP pharmacopuncture did not show the possibility of causing chromosome aberrations.
