뽕나무 버섯 균사체의 생리.생태학적 연구 (Physiological and Ecological Studies on Mycelia of Armillariella mellea)

  • Armillariella mellea의 2차균사를 potato dextrose medium, yeast extract medium, malt extract medium Hamada medium, 등에 배양해서 생리 생태적 성질을 연구했다. 그 결과를 요약하면 아래와 같다. 1. 사용했던 4종의 배치 중에서 malt extract medium에서 가장 잘 자랐고, 그 다음 yeast extract medium, potato dextrose medium 그리고 Hamada medium 순으로 생육율이 높였다. 2. $27^{\circ}C$에서 가장 잘 자랐고, $36^{\circ}C$에서는 집락의 지름이 $27^{\circ}C$와 비슷하나 균사의 밀도가 매우 낮았다. 3. pH6에서 가장 높은 생육율을 나타내었다. 4. 식물 생장 호르몬을 첨가한 경우에는 Heteroauxin의 5ppm 농도에서 대조군보다 26.2% 증가로 가장 많은 생육 증량을 보이고, 다음으로 gibberellin 100ppm, Tomatoton 10,000배액, 그리고 Adoton 5,000배액의 순서였다. 식물 생장 호르몬을 적당 농도로 사용하면 생장 증량 효과를 얻을 수 있으나, 과량 사용할 경우, 오히려 생장 억제 효과를 나타내었다. 5. Vitamin 첨가경우에 있어 200ppm 농도의 균일한 조건하(下)에서 Vit.A 첨가군에서 대조군보다 18.2%의 가장 많은 생육 증량을 보였고, $Vit{\cdot}D_2\;Vit{\cdot}E,\;Vit{\cdot}B_1\;folic\;accid\;Vit{\cdot}C$ 그리고 $Vit{\cdot}B_2$의 순으로 그 증가가 나타나 지용성 비타민 첨가 배지에서 상당한 생육율을 나타내었다.

매미눈꽃동충하초(Paecilomyces sinclairii)의 균사 생장과 자실체 형성 조건의 특성 (The Characteristics of Cultural Conditions for the Mycelial Growth and Fruiting Body Formation of Paecilomyces sinclairii)

  • 매미누꽃동충하초(Paecilomyces sinclairii)를 전라남도 백양사의 인근 숲에서 채집하여 균주를 순수 분리하고 균사의 최적 배양조건과 특성을 조사하였다. 균사생장에 적합한 pH는 $5{\sim}9$의 범위였으며 pH 8에서 가장 왕성한 균사생장을 보였고, 온도는 $25{\circ}C$가 적합하였다. 균사생장에 가장 적합한 배지는 Hamada 배지였으며 탄소 영양원은 dextrin, 질소 영양원은 glutamine인 것으로 조사되었다. 균사의 생장은 C/N비가 20:1일 때, 옥수수추출물과 우유가 혼합한 배지에서 가장 양호하였다. 자실체는 20%의 누에번데기가 첨가된 현미배지에서 500lux, $25{\circ}C$의 조건으로 배양할 때 가장 자실체 형성이 우수하였다.

  • Isolates of Ophiocordyceps heteropoda (Kobayasi) collected from Mt. Halla on Jeju-do, Korea were tested for mycelial growth on different agar media and in the presence of different carbon and nitrogen sources. Similarly, isolates were also incubated at different temperatures as well as under continuous light and dark conditions. Growth was better on Hamada agar, basal medium, and malt-yeast agar, but poor on Czapek-Dox agar. Different carbon sources such as dextrin, saccharose, starch, lactose, maltose, fructose, and dextrose resulted in better growth. Complex organic nitrogen sources such as yeast extract and peptone revealed the most effective growth. Mycelial growth was best at $25^{\circ}C$. The growth rate was faster in the dark than the light, but mycelial density was less compact in the dark.

구강암에 대해 항암효과를 나타내는 methanol 자화 방선균의 분리 및 동정 (Isolation and Identification of Methylotrophic Actinomycetes capable of Producing Anti-oral Cancer Activity)

  • An appropriate amount of samples, collected from three each paddy forest, field and riverside soil near Taegu city, was suspended in sterile water and then diluted in order to isolation of antagonistic to oral cancer. The diluted samples were inoculated on separating medium in the routing spreading method. So, seven hundred and eighteen strains were isolated on HV agar and 220 strains were on methanol medium from soil samples. So, during the screening of anti-oral cancer activity from soil, we isolated microorganisms showing powerful antagonistic activity. Among them, No. 78 strain exhibited the most strongly anti-oral cancer activity. Microbiological properties were investigated by the methods described in the Bergey's Manual of Systematic Bacteriology and experimental methods of identification of actinomycetes by Hamada et al. As a result, a methylotrophic actinomycetes strain No. 79 was estimated as Amycolatopsis sp. based on taxonomic studies.

  • Oudemansiella radicata, one of edible mushrooms belonging to Tricholomataceae of Basidiomycota, has been known to exhibit outstanding therapeutic effects on the hypertension caused by high blood pressure and inhibitory effects on the sarcoma 180 and Erhrlich carcinoma of mice. As one of preliminary experiments for producing fruiting-body of O. radicata, this study was carried out to obtain the basic information for culture conditions of mycelial growth of the fungus. The optimal temperature and pH for the mycelial growth were $25^{\circ}C$ and pH 6, respectively. The medium for favorable mycelial growth of O. radicata was shown in the Lilly medium, whereas compact mycelial density was found in Hamada medium. The carbon and nitrogen sources promoting for mycelial growth of O. radicata were xylose and alanine, respectively. The optimum C/N ratio was about 20 : 1 in case that 3% glucose was supplimented to the basal medium as a carbon source.

꽃송이 버섯의 균사생장을 위한 최적요인 (The optimal Factors for the Mycelial Growth of Sparassis crispa)

  • 꽃송이 버섯의 균사생장은 $25^{\circ}C$의 온도와 pH4의 조건에서 가장 양호하였으며, Hamada 배지에서 가장 조밀한 생장을 나타낸 반면, Hoppkins 배지에서 가장 큰 직경생장을 나타내었다. Maltose, Arabinose 및 Mannitol 같은 탄소원은 꽃송이 버섯의 양호한 생장을 촉진하였다. 또한, 질소원의 하나인 Glycine도 양호한 균사생장을 나타내었다. 배지에 탄소원을 1%로 고정시킨후 C/N비를 20:1로 하였을 때, 꽃송이 버섯 균사의 생장이 가장 양호하였다. 유기산으로서 Fumaric acid와 Lactic acid의 처리를 하였을 때, 꽃송이 버섯의 생장이 좋았다.

번데기동충하초 균사 중의 Branched-Chain Amino Acid Aminotransferase의 분리정제 및 그 특성에 관한 연구 (A Study on the Purification and Characteristics of Branched-Chain Amino Acid Aminotransferase in Cultural Mycelia of Cordyceps militaris)

  • The optimum conditions of Cordyceps militaris mycelial growth, purification and characteristics of branched-chain amino acid aminotransferase [BCAT(EC 2.6.1.42)] in this mycelium were studied. Optimum pH, temperature and medium of culture of mycelia were 5.5, $22.5^{\circ}C$ and Hamada medium (HM), respectively. BCAT in homogenate of this mycelia was precipitated by 20-40% saturated solution of ammonium sulfate and then purified by DEAE (diethylaminoethyl)-Sephadex A-50 column chromatography with linear concentration gradient and Sephadex G-200 gel filtration. A single band of purified enzyme was detected on SDS-PAGE (sodium dodecylsulfate-polyacrylamide gel electrophoresis). Optimum pH and temperature of BCAT were found to be 7.8 and $29^{\circ}C$, respectively. It showed activity toward L-leucine, L-isoleucine and L-valine as a substrate. The Km values of this enzyme for L-leucine were determined to be 5.88 mM for L-leucine.

The Optimal Culture Conditions Affecting the Mycelial Growth and Formation of Paecilomyces fumosoroseus

  • The fruiting body of Paecilomyces fumosoroseus was collected at Mt. Mani, Ganghwa Island, Korea in September, 2001. This study was carried out to obtain the basic informations for the mycelial growth and fruiting body production of P. fumosoroseus in artificial media. The optimal conditions for the mycelial growth were obtained at $25^{\circ}C$ and in the range of pH $6{\sim}9$, respectively. P. fumosoroseus showed the favorable growth on Hamada medium. The carbon and nitrogen source favorable for mycelial growth were dextrin and histidine, respectively. Optimum C/N ratio suitable for optimal growth of P. fumosoroseus was observed on the culture media adjusted to the ratio of 40:1. The mycelial growth of P. fumosoroseus was optimal on corn meal agar supplemented with 30% of silkworm pupae. The most favorable fruiting body formation of P. fumosoroseus was obtained in the medium containing unpolished rice supplemented with 20%(w/w) silk worm pupae at $25^{\circ}C$ under 100 lux.

영지속 균주의 배양적 특성 및 형태적 특성에 관한 연구 (Studies on Cultural and Morphological Characteristics of Isolate on Ganoderma species)

  • Ganoderma속균은 약효가 인정되어 많이 인공 재배되어 이용되고 있으나 본 속균에 대한 기초적인 연구가 미흡한 실정이므로 본 연구에서는 영지 속균을 채집하여 분리 동정하였으며, 또한 배양적 특성과 형태적인 특징을 조사하였다. 영지속균은 경기도와 강원도 11개 장소의 참나무, 아카시나무, 오리나무 등에서와 6재배농가에서 채집되었다. 대부분의 영지는 하나의 버섯이 그루터기에서 형성되나 오리나무에서는 다수가 형성되었다. 영지속균의 균사 생장적온은 G. applanatum, G.lucidum, G. neo-japonicum에서는 $28{\sim}30^{\circ}C$이었고, G. tsugae에서는 $26^{\circ}C$이었으나 $32^{\circ}C$에서는 천천히 생장하였다. Hamda배지를 pH 6.2로 조정하였을 때 균사생장이 가창 좋았다. 6종의 공시균주에 대한 균사의 형태적인 특정으로 G. applanatum, G. lucidum, G. neo-japonicum은 staghorn를 형성하였으나 G.oregonens, G. ualeosiacum은 포도형의 가지를 가진 staghorn균사를 형성하였다. Clamp connection은 G. applanatum, G. lucidum, G. oregonense, G. valeosiacum균의 균사에서 관찰되었으나 node형을 가진 G. neo-japonioum에서는 형성되지 않았다. 후막포자는 G. applanatum, G. neo-japonicum에서 형성되었으며, cuticular cell는 G. lucidum, G. neo-japonicum, G. oregonense, G. tsugae의 균사에서 형성되었다.

Vegetative Growth of Four Strains of Hericium erinaceus Collected from Different Habitats

  • Vegetative growth of four different strains of Hericium erinaceus was observed. The temperature suitable for optimal mycelial growth was determined to be $25^{\circ}C$, with growth observed in the extend temperature range of $20{\sim}30^{\circ}C$. The different strains of this mushroom showed distinct pH requirements for their optimum vegetative growth, with the most favorable growth observed at pH 6. Considering vegetative mycelial growth, PDA, YM, Hennerberg, Hamada, and Glucose peptone were the most favorable media, and Czapek Dox, Hoppkins, Glucose tryptone, and Lilly were the most unfavorable media for these mushroom strains. With the exception of lactose, most of the carbon sources assayed demonstrated favorable vegetative growth of H. erinaceus. For mycelial growth, the most suitable nitrogen source was alanine and the most unsuitable was histidine. Oak sawdust medium supplemented with $10{\sim}20%$ rice bran was the best for mycelial growth of the mushroom.
