Owing to the kinetic differences in ammonia oxidation among ammonia-oxidizing microorganisms (AOM), there is no standard set of kinetic values that can be used as a representative set for nitrifying wastewater treatment plant (WWTP) design. As a result, this study clarified a link between the half-saturation constants for ammonia oxidation (Ks) and the dominant ammonia-oxidizing bacterial (AOB) groups in sludge from full-scale WWTPs and laboratory-scale nitrifying reactors. Quantitative polymerase chain reaction analyses revealed that AOB affiliated with the Nitrosomonas oligotropha cluster were the dominant AOM groups in the sludge taken from the low-ammonia-level WWTPs, while AOB associate with the Nitrosomonas europaea cluster comprised the majority of AOM groups in the sludge taken from the high-ammonia-level WWTPs and nitrifying reactors. A respirometric assay demonstrated that the ammonia Ks values for the high-ammonia-level WWTPs and nitrifying reactors were higher than those of the low-ammonia-level plants. Using the Ks values of available AOM cultures as a reference, the Ks values of the analyzed sludge were mainly influenced by the dominant AOB species. These findings implied that.different sets of kinetic values may be required for WWTPs with different dominant AOM species for more accurate WWTP design and operations.

In this study, we investigated the effects of dissolved inorganic and organic nutrient on the growth of dinoflagellate Alexandrium affine (LIMS-PS-2345). The maximum uptake rates (ρmax) and half saturation constants (Ks) calculated from the uptake kinetics experiment were 77.0 pmol/cell/hr, 17.6 μM for nitrate and 15.5 pmol/cell/hr, 3.88 μM for phosphate, respectively. These results suggested that this species has high inorganic nutrient demand and a low affinity for inorganic nutrients. During the utilization of organic nutrients for A. affine, growth rates of experimental groups added by organic nitrogen (urea and glycine) and phosphorus (adenosine triphosphate and glycerol phosphate) were above 70 %, compared to the experimental groups added by inorganic nutrients. Thus, A. affine may need to utilize organic nutrients to understand the dominant strategy and advantageous position in the interspecific competition within low inorganic nutrient environments.

To supply a stable amount of standard material to detect paralytic shellfish toxin (PST), we examined possible increases in toxic content in Alexandrium catenella and A. pacificum using a light emitting diode (LED), which is one of the most eco-friendly and economical lighting method. When comparing the growth rates of organisms and wavelengths of light used, the half saturation constants (Ks) of red wavelength were higher than those of other wavelengths. In contrast, the Ks of blue wavelength were lower than those of other wavelengths. Moreover, when comparing the toxic contents and wavelengths of light used, red wavelength produced approximately 8 times more toxic content in A. catenella and approximately 3.2 times more toxic content in A. pacificum than other wavelengths. Thus, the toxic content present in the organism might be closely related to the Ks of light. The optimum light source to be used to ensure economically ef ective and productive growth in an Alexandrium culture system (photo-bioreactor) would likely consist of a two-phase culture, wherein a blue LED is used during the lag and exponential phases to increase growth rates, followed by the use of a red LED during late exponential and stationary phases to achieve increased PST yields.
The red tide-causing flagellate Chattonella anticfua can cause mass fish kills by their clogging in fish gills. Thisstudy examined the nutrient requirements of C. antiqua isolated from Korea. C. anticfua displayed maximum growthat the day five, followed by a decrease in cell density. Nitrate and nitrite were the preferred nitrogen sources, alonewith adenosine diphosphate for phosphorus compounds. In medium that contained ammonium, a significantdecrease in cell density was observed. Half-saturation constants, Ks, calculated from the maximum growth ratewere 4.94 U|M for NC>3 and 0.79 flM for P04. The growth of C. antiqua was not within the function of the N:P ratio (RU= 0.29). With an N:P ratio as low as 10, the increase in cell density was apparent, with a higher division rate. At lev-els above 50 fiM of NaNOg or 8 ;uM of NaHUPCU, the growth rates were somewhat decreased. Phosphate was thelimiting factor for C. antiqua growth since the starvation of phosphate had brought about a rapid decrease in celldensity in semi-continuous culture. Studies about the temporal modification of the efficiency of nitrate or phosphateuptake may be necessary to explain the bloom dynamics of C. antiaua.
This study was conducted to investigate the effects of light intensity and wavelength on the growth of Tetraselmis suecica and Tetraselmis tetrathele. These species were exposed to a blue light-emitting diode (LED; max=450 nm), a yellow LED (max=590 nm), a red LED (max=630 nm) and a fluorescent lamp (three wavelengths). The maximum growth rates (${\mu}_{max}$) of T. suecica and T. tetrathele under a red LED were 1.12/day and 0.95/day, respectively. Under a yellow LED, growth rates were 70% of the values for red wavelength, with low half-saturation constants (Ks). The optimum light source to ensure economically effective and productive growth in a Tetraselmis culture system (Photo-Bioreactor) would thus appear to be a three-phase culture, wherein a yellow LED is used during the lag phase and initial exponential phase to increase growth rate, followed by a red LED during the middle exponential phase to maximize growth rate, and finally a yellow LED again during the late exponential phase and stationary phase to achieve increased yield of useful bioactive substances.

This study was conducted to investigate the effects of the light quality of a Light Emitting Diode (LED) on the phosphate uptake of Prasinophytes Tetraselmis suecica and Tetraselmis tetrathele. These species were exposed to a blue LED (max = 450 nm), a yellow LED (max = 590 nm), a red LED (max = 630 nm) and a fluorescent lamp (control) at $100{\mu}mol\;m^{-2}\;s^{-1}$. The maximum uptake rates (${\rho}_{max}$) of T. suecica and T. tetrathele under the red LED were $6.35pmol\;cell^{-1}\;hr^{-1}$ and $9.85pmol\;cell^{-1}\;hr^{-1}$, respectively. The half saturation constants (Ks) of two species were $9.43{\mu}M$ and $21.2{\mu}M$, respectively. The phosphate affinity of the two species under the red LED was higher than that of other wavelengths. Thus, the optimum light source to ensure economically effective and productive growth in a Tetraselmis culture system (Photo-Bioreactor) would be red LEDs because of the high growth rate shown, regardless of relatively low nutrient conditions.

We investigated the interspecific competition between the harmful dinoflagellate Cochlodinium polykrikoides and diatom Skeletonema sp. based on the utilization and uptake of dissolved organic nutrients. C. polykrikoides and S. costatum were able to grow using dissolved organic nitrogen (DON) and dissolved organic phosphorus (DOP) as well as dissolved inorganic nitrogen (DIN) and dissolved inorganic phosphorus (DIP). This result indicates that the utilization of dissolved organic nutrients may play a role in surviving strategy in the DIN or DIP-limited environments. The half-saturation constants (Ks) of urea and glycerophosphate (glycero-P) calculated from uptake kinetics experiment of C. polykrikoides was lower than those of Skeletonema sp. This result indicates that Skeletonema sp. have higher affinity for dissolved organic nutrients, such as urea and glycero-P, than C. polykrikoides. Although Skeletonema sp. have higher affinity of dissolved organic nutrients, C. polykrikoides could effectively uptake for urea and glycero-P at sub-saturating nutrient concentrations (${\alpha}$ (${\rho}_{max}/Ks$) of C. polykrikoides was higher than Skeletonema sp.. Therefore, C. polykrikoides by utilization and effectively uptake of dissolved organic nutrients under monoculture may have an advantageous position in the interspecific competition with Skeletonema sp. in the low nutrient environments.

In the 1990s, Cochlodinium polykikoides red tide has been annually occurred in the southern coast of Korea and caused the mass damage to the fisheries with a huge amount of economic loss. The present study was done to establish the biological foundation for the elucidation of the mechanism of C. polykikoiaes red tide. The growth response of C. polykikoides to physico-chemical factors such as temperature, salinity, pH, and light intensity were examined using axenic cultures to evaluate the relative importance of these factors on the dynamics of natural populations, It was found that the highest growth conditions were $25^{\circ}C,\;40\%_{\circ}$, pH 7.5, and 7,500 lux, respectively. The tolerable salinity range of growth was relatively wide at an optimum temperature and was reduced to a much narrower range at a sub-optimum temperature. These findings indicate that C. polykikoides is an eurythermal and euryhaline organism. The organism demanded higher light intensity and oceanic pH narrow in its growth. C. polykikoides utilize inorganic nutrients, such as nitrate and ammonium as N, and phosphate as P. The nutritional requirements of C. polykikoides were $40{\mu}M$ for nitrate, $50{\mu}M$ for ammonium, and $5{\mu}M$ for phosphate. The half saturation constant (Ks) for growth was $2.10{\mu}M$ for nitrate, $1.03{\mu}M$ for ammonium, and $0.57{\mu}M$ for phosphate. These values were comparatively smaller than those of other dinoflagellates reported previously. We confirmed that the organism is characterized as an eutrophic species. However, ammonium Ks value is smaller than that of other eutrophic species, This result indicates that C. polykikoides red tide may outbreak in the waters which eutrophication is in progress rather than eutrophicated waters. C. polykikoides preferred ammonium better than nitrate as a nitrogen source when in a growth stage, Therefore, our results indicate that ammonium is more important nutrient on the growth of the organism in comparison with other inorganic nutrients and C. polykikoides red tide is related with the increased ammonium concentration in the coastal waters.

We investigated cause of non-outbreak of Cochlodinium polykrikoides blooms in the western coast of Jaran Bay during Summer, 2013, by combining chemical field data and physiological data of C. polykrikoides, which had been already published. The predominant species were mainly diatoms, and dominant species was Cerataulina pelagica, Chaetoceros spp., Navicula spp. and Nitzschia spp.. In case of dissolved inorganic nutrients in the western coast of Jaran Bay, dissolved inorganic phosphorus (DIP) was similar to that in previous outbreak period of C. polykrikoides blooms, but dissolved inorganic nitrogen (DIN) was lower. C. polykrikoides might be disadvantageous in competition with diatom species because half-saturation constants (Ks) of C. polykrikoides for inorganic nutrients was lower than those of diatoms. Also, the western coast of Jaran Bay, where DIN concentration is relatively low, was an unfavorable environment for growth of C. polykrikoides characterized by nitrogen dependence. Therefore, C. polykrikoides which have the disadvantageous position for competition of inorganic nutrient might have been suppressed by diatom blooms under environment of low nutrient in the western coast of Jaran Bay.

Serial basic tests were conducted for the determination of fundamental kinetics and for the actual application of kinetic parameter to food waste digestion with precise measurement of methane production under a thermophilic condition. The effects of food particle size, sodium ion concentration, and volatile solid (VS) loading rate on the anaerobic thermophilic food waste digestion process were investigated. Results of serial test for the determination of fundamental kinetic coefficients showed the value of k (maximum substrate utilization rate coefficient) and KS (half-saturation coefficient) as $0.24hr^{-1}$ and $700mg/{\ell}$, respectively, for non-inhibiting organic loading range. No inhibition effect was shown until $5g/{\ell}$ of sodium ion concentration was applied to a serum bottle reactor. However, the volume of methane gas was decreased gradually when the concentrations of more than $5g/{\ell}$ of sodium ion applied. All sizes of food waste particle showed the same constants (A : 0.45) but the maximum substrate utilization rate constant ($k_{HA}$) was inversely proportional to particle size. As an average particle size increased from 1.02 mm to 2.14 mm, $k_{HA}$ decreased from $0.0033hr^{-1}$ to $0.0015hr^{-1}$. The result reveals that particle size is one of the most important factors in anaerobic food waste digestion. There was no inhibition effect of sodium ion when VS loading rate was $30g/{\ell}$. And maximum injection concentration of VS loading rate was determined about $40g/{\ell}$.
