Trichophyton rubrum 및 Trichophyton raubitschekii에 의한 개 피부사상균증에 관한 연구 (Dermatophytosis by Trichophyton rubrum and T raubitschekii in dogs)

  • This study was carried out to determine the causative agent and the clinical features of dermatophytosis occurred in 3 dogs from Tae-gu, Korea between 1996 and 1997. Specimens of hair and scale, collected from skin lesions were inoculated on potato dextrose agar and mycobiotic agar supplemented with thiamine and inositol. The agar plates were incubated at $25^{\circ}C$ for 2 weeks. Growing fungi isolated were identified by the morphological and growth characterization on bromocresol purple-milk solids-glucose medium, Christensen urea broth and trichophyton media, and by hair perforation tests. Skin lesions were observed in legs, abdomen, ears, face and head with alopecia of various sizes accompanied by itching and/or inflammation. The causative agent of dermatophytosis in the 3 dogs was identified as Microsprum gypseum and Trichophyton rubrum (mixed infection, dog 1), T rubrum (dog 2) and T raubitschekii (dog 3). The dermatophytosis by T rubrum and T raubitschekii was first found on animals in Korea.

이개에 발생한 모낭암종 1예 (A Case of Trichilemmal Carcinoma in Auricle)

  • Trichilemmal carcinoma is a rare malignant neoplasm of the hair follicle from the outer root of the hair follicle sheath. This tumor can be misleading, and a false diagnosis of a squamous cell carcinoma. We report a case of trichilemmal carcinoma with a review of literature. The patient presented with an exophytic well circumscribed nodular mass on the left auricle, which was detected 6 months ago. Histopathologically, the tumor consisted of atypical clear cells which contained abundant glycogen. The tumor cells shows lobular growth pattern with necrosis, foci of trichilemmal keratinization and peripheral pallisading. Total excision and repair with full-thickness skin graft was done with minimal surgical morbidity. The patient has been free of recurrence or metastasis for 8 months.

Efficacy of Korean Red Ginseng in the Treatment of Alopecia Areata

  • Alopecia areata (AA) is an autoimmune disease that can affect any hair-bearing area. AA is known to be caused by immunological disorder but still, the pathogenesis is not fully understood. Many therapeutic modalities have been used to treat alopecia areata, with variable efficacy and safety profiles. Unfortunately, none of these agents is definitely curative or preventive alone. We studied hair growth efficacy and safety of Korean red ginseng (KRG) in AA comparing corticosteroid intra-lesional injection (ILI) alone patient group with ILI with KRG taking patient group using Folliscope 2.5 for 12 wk. Herein, we would like to report the efficacy of KRG in the treatment of AA and recommend KRG as a useful complimentary food for gaining efficacy of treatment for AA.

  • This study was conducted to evaluate the effects of Padina arborescens and 1-O-myristoyl-2-O-oleoyl-3-O-(α-D-glucopyranosyl)-glycerol(MOGG), its active component, on the prevention of hair loss. The P. arborescens extract and MOGG inhibited the activity of 5α-reductase, which converts testosterone to dihydrotestosterone(DHT), a main cause of androgenetic alopecia. When immortalized rat vibrissa dermal papilla cells were treated with MOGG, the proliferation of dermal papilla cells significantly increased. In addition, we found that the P. arborescens extract and MOGG could open the KATP channel, which may contribute to increase hair growth. Furthermore, MOGG promoted PGE2 production in HaCaT cells. The results suggest that MOGG from the P. arborescens extract has the potential to treat alopecia via 5α-reductase inhibition, the proliferation of dermal papilla, the opening of the KATP channel and/or increase of PGE2 production.

  • 최근 몇 년 동안 탈모로 인해 우울증을 경험하는 인구가 증가하고 탈모 관련 치료법도 임상의학이나 외과적인 수술법과 같이 전문화되고 있다. 탈모 방지와 발모를 촉진하기 위한 시술법과 소재들이 있으나 그 효능, 효과가 뛰어나지 못하고 일부는 부작용을 동반하는 경우가 있다. 본 총설의 마이크로니들 시술법은 약물의 흡수를 촉진시켜주는 매우 작고 섬세한 니들을 이용한 시술법이다. 이 시술을 하는 동안 마이크로니들이 두피 내부로 약물의 흡수를 도와주는 마이크로 홀을 만들어 준다. 마이크로니들 시술 약물의 주요성분으로 대두 발효물질을 분석하였으며 대두 발효 소재는 탈모증에 항산화, 항안드로겐, 항혈전의 기능을 가진다. 대두 발효물의 흡수를 증가시켜주는 마이크로니들의 우수한 약물전달시스템으로 인하여 대두 발효물이 마이크로니들과 함께 사용될 때 복잡한 탈모기전에 더욱 효과적이다. 이 시술법은 탈모증을 방지하고 관리하며 개선시켜줄 수 있는 매우 유용한 두피케어법이며 일반적인 두피관리법 보다 더 빠른 효과를 나타낼 수 있어 차별화된다. 대두 발효물을 이용한 마이크로니들 시술법은 두피케어를 위한 진일보한 기술이다.

Phloroglucinol Enhances Anagen Signaling and Alleviates H2O2-Induced Oxidative Stress in Human Dermal Papilla Cells

  • Phloroglucinol (PG) is one of the abundant isomeric benzenetriols in brown algae. Due to its polyphenolic structure, PG exhibits various biological activities. However, the impact of PG on anagen signaling and oxidative stress in human dermal papilla cells (HDPCs) is unknown. In this study, we investigated the therapeutic potential of PG for improving hair loss. A non-cytotoxic concentration of PG increased anagen-inductive genes and transcriptional activities of β-Catenin. Since several anagen-inductive genes are regulated by β-Catenin, further experiments were performed to elucidate the molecular mechanism by which PG upregulates anagen signaling. Various biochemical analyses revealed that PG upregulated β-Catenin signaling without affecting the expression of Wnt. In particular, PG elevated the phosphorylation of protein kinase B (AKT), leading to an increase in the inhibitory phosphorylation of glycogen synthase kinase 3 beta (GSK3β) at serine 9. Treatment with the selective phosphoinositide 3-kinase/AKT inhibitor, LY294002, restored the increased AKT/GSK3β/β-Catenin signaling and anagen-inductive proteins induced by PG. Moreover, conditioned medium from PG-treated HDPCs promoted the proliferation and migration of human epidermal keratinocytes via the AKT signaling pathway. Subsequently, we assessed the antioxidant activities of PG. PG ameliorated the elevated oxidative stress markers and improved the decreased anagen signaling in hydrogen peroxide (H2O2)-induced HDPCs. The senescence-associated β-galactosidase staining assay also demonstrated that the antioxidant abilities of PG effectively mitigated H2O2-induced senescence. Overall, these results indicate that PG potentially enhances anagen signaling and improves oxidative stress-induced cellular damage in HDPCs. Therefore, PG can be employed as a novel therapeutic component to ameliorate hair loss symptoms.

Allelopathic Effects of Volatile Substances from Chamaecyparis obtusa

  • The allelopathic effects of volatile substances from Chamaecyparis obtusa (S. et Z.) Endl. were examined on the germination and seedling growth of some plant species, and on the population growth of some microorganisms. The germination and seedling growth of the receptor plants were suppressed more severely by leaf and fruit essential oils than by those of other parts. Colonial growth of fungi was severely inhibited by essential oils extracted from leaves and fruits. The development of root hairs of the receptor plants was also severely inhibited by the essential oils. The cortical cells at the root tips of Lactuca sativa L. treated with essential oils showed contraction of the cytoplasm, resulting in plasma membranes becoming detached from the cell walls and the cells metamorphosing irregularly. Accumulation of lipid granules inside the contracted cytoplasm and degeneration of mitochondrial cristae were also observed.

삿갓사초 추출물의 모발 성장 기전 활성화 효과 (The Effect of Carex dispalata Extract on the Activation of Anagen Pathway)

  • Dermal papilla cells (DPCs) are present throughout the hair cycle and play an essential role in hair cycle and hair growth. In this study, we investigated the effect of Carex dispalata on the activation of anagen pathway in DPCs. C. dispalata extract increased the proliferation of DPCs and induced changes in the levels of cell cycle-related proteins. To elucidate the mechanism by which C. dispalata extract stimulates the anagen pathway related to the proliferation of DPCs, we evaluated the effect of C. dispalata extract on the activation of Akt signaling. The increase in the level of phospho-Akt by C. dispalata extract was inhibited by PI3K inhibitor (wortmannin). Wortmannin reduced the effects of C. dispalata extract on the levels of cell cycle-related proteins and proliferation of DPCs. C. dispalata extract increased the levels of Wnt/β-catenin proteins. Wnt/β-catenin inhibitor (XAV939) inhibited changes in cell cycle, cell cycle-related proteins, Wnt/β-catenin proteins, and proliferation induced by C. dispalata extract. C. dispalata extract increased the level of autophagy protein (LC3I/II), and this change was inhibited by XAV939. These results suggest that C. dispalata extract can activate PI3K/Akt, Wnt/β-catenin, and autophagy pathways in DPCs to induce cell proliferation, and thereby promote hair growth phase.

송라 추출물의 세포 수준에서 항노화 및 모유두세포 활성화 효과 (In Vitro Anti-aging and Hair Follicle Dermal Papilla Cells Activation Effects of Usnea diffracta Vain Extract)

  • 송라(Usnea diffracta Vain.)는 송라속(Usnea)에 속하는 지의류 중 하나이며, 이전 연구에서 항산화, 항균, 항염, 항종양 및 심혈관 보호 등의 약리학적 활성이 보고되어 있으나 피부 및 모발에서의 효능은 잘 알려져 있지 않다. 따라서 본 연구에서는 세포 수준에서 송라 추출물(UDE)의 항노화 및 모유두세포 증식에 대한 효과를 검증하였다. 실험 결과, 송라 추출물은 인간 섬유아세포에서 UVA에 의해 증가된 MMP-1의 발현 및 상위기전인 MAPKs (ERK, p38, JNK)와 AP-1 (c-Fos, c-Jun)의 활성을 유의적으로 감소하는 것을 확인하였다. 또한, 송라 추출물은 인간 모유두세포의 증식을 유의하게 증가시켰으며, 모발 성장인자인 VEGF 및 KGF의 mRNA 발현을 유의하게 증가시켰다. 이로 인하여, 모발 증식 및 성장인자의 발현에 관여하는 ERK/CREB의 인산화를 농도 의존적으로 증가시켰다. 송라 추출물의 주성분 확인을 위해 송라 추출물을 농축 후 Prep-LC를 이용하여 main peak로 나타난 분획을 분리 정제하였고, NMR 및 Mess 분석을 통해 diffractaic acid로 동정하였다. Diffractaic acid는 인간 섬유아세포에서 UVA에 의해 증가된 MMP-1의 발현을 유의적으로 감소시켰으며, 인간 모유두세포의 증식을 농도 의존적으로 증가시켰다. 이를 통해 송라 추출물은 항노화 및 모유두세포 활성 증가 효능을 갖는 화장품 천연소재로서의 활용 가능성을 입증하였다.

Trichophyton tonsurans의 진균학적 성상 (Mycologic Characteristics of Trichophyton tonsurans)

  • Trichophyton(T.) tonsurans has been identified for the first time in Korea in 1995 and has been increasing in number of reported in wrestlers, in judo players, and in korean - style wrestlers. Author tried mycological studies with T. tonsurans is isolated in Daegu Korea. The results are as follows: All 16 strains of T. tonsurans tested for nutritional requirement, showed strongly stimulated growth by thiamine, mild to moderate growth on casamino acid agar, poor growth on ammonium nitrates base agar. All 16 strains of T. tonsurans tested showed positive results for hair perforation and urease test. Gross findings of T. tonsurans showed that fine granular surface with light yellow hue or white color and mahogany brown color on the reverse side. Microscopic findings showed that septated hyphae, chlamydoconidia and microcondia. Microconidia were characteristic in their arrangerment and shape : round or oval shaped microconidia laterally to hyphae, some were match - head like terminal swelling.
