Electrophoretic Studies on Haemolymph Protein, Digestive Fluid Protein and Digestive Amylase Activity in the Wild silkworm, Theophila mandarina. (상잠의 형액과 소화액에 있어서 단백질과 Amylase 활성에 관한 전기영동적 연구)

  • The haemolymph protein, digestive fluid proteins and digestive fluid amylase activity of wild silkworm, Theophila mandarina those of the were studied by polyacrylamide gel electrophoresis. In addition, they was also compared with silkworm. 1. 6 main protein bands in female and 7 main protein bands in male were detected in the larval haemolymph of T. mandarina where as 8 and 7 main protein bands in female and male of B. mori were observed. Some differences in the haemolymph protein ands of T. mandarina and B. mori were observed. 2. 15 protein bands and 12 protein bands were found in the larval digestive fluid of T. mandarina and B. mori respectively. Some differences in the mobility of digestive fluid proteins of T. mandarina and B. mori were noticed. 3. Larval digestive fluid amylases were anionic and moved near the tracking dye in both T. mandarina and B. mori. Mobility of the digestive fluid amylases relative to bromophenol blue were 0.019 and 0.020 in T. mandarina and B. mori respectively.

  • Tolerance limits of the adult crab were determined by exposing them to different temperatures (5~2$0^{\circ}C$) and salinities(12~32$\textperthousand$). The tolerance limit for high temperature was determined at 14$^{\circ}C$ and 16$^{\circ}C$ for male and female snow crab, respectively and that for salinity was 21 $\textperthousand$ at 5 $^{\circ}C$. Osmolality concentration, Na$^{+}$ and $K^{+}$ ions in the haemolymph of especially, the females was positively related to the salinity changes. Increased water temperature resulted in a marked decreased in ionic and osmolality concentrations near the limit of tolerance in male, however, such a clear pattern was not apparent for the females. The concentration of glucose in the haemolymph was positively related to water temperature but negatively to salinity.ity.

  • The experiment was carried out to investigate the effects of juvenile hormone analogue on changes of protein and amino acids in haemolymph of silkworm larva. Juvenile hormone analogue was topically administered to larvae at dose of 1$\mu\textrm{g}$ 10$\mu\textrm{g}$ per gm of body weight at 60hr. of the 5th instar. The results obtained were as follows ; 1. Larval duration of the fifth instar was extended about 1 day by JHA-1 and 4 days by JHA-10 as compared with the control. 2. Cocoon weight and cocoon layer weight by topical application of JHA were heavier than those of the control, but cocoon layer ratio was decreased in JHA-10 to the exclusion of JHA-1. 3. The concentration of haemolymph protein during the fifth instar was increased remarkably by the JHA application. 4. The total content of amino acids in haemolymph proteins of JHA-10 approximately doubled that of the control, with the conspicuous increase of glycine and arginine level.

  • The just molted larvae of the fifth instar were fed on with cadmium trated mulberry leaf and on the third day of the fifth instar the daily change of the total protein, activities of GOT and GPT in larvae haemoymph were sexually analyzed. The result obtained are summarized as follows : 1. The total haemolymph was decreased by cadmium treatment. A decreasing ratio of the total haemolymph protein was higher at the later development stage of the male fifth instar than the female fifth instar. 2. There seemed a slight difference in albumin content of the female blood along with the fifth larvae development between control and cadmium treatment where 7% decrease of blood albumin took place with cadmium treatment comparing to that of cotrol. Contrarily a decrease of blood globulin was made on both sexes. 3. The activities of GOT and GPT were inhibited by cadmium treatment.

  • To determine the metabolism of leucine during the cuticle formation and the sclerotization process in Pieris rapae L., $U-^3$H-leucine or $U-^4$C-tyrosine is injected into the haemolymph of newly molted pupa through dorsal cuticle of heart area. The results show that leucine as a common amino acid participates in the synthesis of cuticle protein over the first 3 hr after ecdysis. It is also shown that leucine in the haemolymph at ecdysis is freely being moved between major internal organs during the short time period post-ecdysis, providing the evidence for some involvements including haemolymph protein synthesis and storage of fat body and gut in metabolism of leucine.

  • Oral application of dimethyl sulfoxide in different concentration (1, 3 and 5 percent) to silkworm, Bomyx mori at the fourth and the fifth instar has significantly increased the commercial characters such as larval and silkgland weights, female and male cocoon weights, their shell weights and egg production. However, larval duration, cocooning and hatching percentages did not showed any significant change, as compared with that of the corresponding parameters of the carrier control. Similarly the glycogen and protein contents of the fat body and trehalose and protein contents of the haemolymph were significantly increased in 1 and 3 percent, whereas the increase of them in 5 percent concentration did not take place significantly, except haemolymph trehalose where it was found to be significant, as compared with that of carrier control.
