Effects of Polygonati Rhizoma Extracts on the Collagenase Activity and Procollagen Synthesis in Hs68 Human Fibroblasts and Tyrosinase Activity

  • Objectives : This study was designed to investigate the collagen metabolism and tyrosinase activity of Polygonati Rhizoma extracts (PR). It's effects are to tonify spleen qi and augment the spleen yin. It enrichs the yin and moisten the lung. Methods : The effect of PR on type I procollagen production and collagenase (matrix metalloproteinase-1, henceforth referred as MMP-1) activity in human normal fibroblasts Hs68 after ultraviolet B (UVB, 312 nm) irradiation was measured by ELISA method. The tyrosinase activity after treatment of PR was measured. Results : There were no cytotoxicity at concentrations of 10, 30, $100{\mu}g/ml$. The reduced type I procollagen production was recovered by PR in UVB damaged Hs68 cells at a concentration of $100{\mu}g/ml$ ($16.2{\pm}0.0$ ng/ml) from control group ($13.9{\pm}0.5$ ng/ml). However there was no statistical significance. PR reduced The increased MMP-1 activity after UVB damage at concentrations of $10{\mu}g/ml$, $30{\mu}g/ml$, and $100{\mu}g/ml$ in a dose dependent manner ($42.2{\pm}20.5%$, $44.8{\pm}8.5%$, and $22.0{\pm}5.8%$). PR $100{\mu}g/ml$ treatment showed the statistical significace (p < 0.05). PR significantly reduced the tyrosinase activity at a concentration of 10 mg/ml ($32.0{\pm}12.8%$, p < 0.05). However, the L-DOPA oxidation was not changed. Conclusion : PR showed the anti-wrinkle effects and whitening effects in vitro. Although more researches are needed to validate the efficacy, these results suggest that PR may have potential as an anti-aging ingredient in cosmetic herb markets.

Effects of Molecular Weights of Sodium Hyaluronate on the Collagen Synthesis, Anti-inflammation and Transdermal Absorption (히알루론산나트륨의 분자량 크기에 따른 Collagen 합성, 항염증 및 피부 흡수에 미치는 영향)

  • In this study, we examined the effects of various molecular weights (1, 10, 50, 100, 660, and 1500 kDa) of sodium hyaluronate (HA), which were prepared by enzyme hydrolysis, on the collagen synthesis, anti-inflammation and skin absorption. These HA did not significantly affect the viability of human dermal fibroblast Hs68 cells. Among them, 1500 kDa, 50 kDa HA most significantly increased collagen production by 59%, and 50% in the Hs 68 cells, respectively. Whereas 1500 and 660 kDa HA hardly pass through mouse transdermis membrane, lower molecular weights (1, 10, or 50 kDa) of HA showed time-dependent increase in skin permeation. HA of 50 kDa showed highest anti-inflammatory effects by reducing nitric oxide and tumor necrosis factor-${alpha}$ production in the RAW 264.7 cells, comparing to other HA (1, 10, and 100 kDa HA). Recently, there is no report about anti-wrinkle and anti-inflammatory effects and skin permeation of different molecular weights HA (1, 10, 50, 100, 660 and 1500 kDa), which were produced by enzyme hydrolysis. These results suggested that 50 kDa HA can be potent candidates for the development of effective anti-aging and anti-wrinkle cosmetic agents. The results of this study demonstrated that among those HA with different molecular weights, 50 kDa HA showed highest anti-inflammatory activity, significant capability to induce collagen synthesis and high level of skin permeation.

  • In this study, we would like to confirm the collagen recovery effect through cell experiments of the flavonoid apigenin and baicalein and propose results that can support the collagen recovery effect through comparative transcriptome analysis. As a result of the study, it was confirmed that apigenin and baicalein were effective in recovering type I collagen damaged by UV in the HS68 cell line, and that both substances resulted in changes in the expression of ECM-related genes. Not only did it show a common mechanism for regulating the ECM, but it also showed the results of changing different categories of genes, making it possible to predict that it would have various effects on cells.

Protective Effect of Saponaria Extract Against UVB-Damage in Skin Fibroblasts (UVB로 산화적 손상을 유도한 피부섬유아세포에 Saponaria 추출물의 보호효과)

  • The skin is the largest organ of the human body and protects the inside of the body. Ultraviolet rays cause various inflammatory reactions in the skin, including photoaging and oxidative damage. The purpose of this study is to investigate the protective effect of Saponaria extract by irradiating UVB on fibroblasts. In this study, the effectiveness of Saponaria showing protective activity against UVB-induced cytotoxicity, oxidative cell death, and NO and PGE2 production was evaluated. HS68 cells were irradiated with UVB(120 mJ/cm2) and treated with Saponaria extract at various concentrations of 100, 200, and 400 ㎍/mL for 24 hours. Intracellular reactive oxygen species (ROS) generated by ultraviolet B were detected using a spectrofluorometer after DCF-DA staining. Lipid peroxidation was also analyzed by measuring the level of 8-isoprostane secreted into the culture medium. As a result, treatment with Saponaria extract effectively inhibited UVB-induced cytotoxicity. Oxidative cell damage was mediated by PGE2 in UVB-induced HS68 fibroblasts, which was significantly inhibited by Saponaria extract treatment. In addition, it was evaluated that the protective effect of these extracts was mediated by the inhibition of intracellular ROS production and lipid peroxidation in a concentration-dependent manner. These results suggest that Saponaria extract can be used as an anti-aging functional material because it inhibits skin damage mediated by oxidative stress caused by UVB and exhibits a cellular protective effect.

The effect of wrinkle care and whitening by Rhus verniciflua Stokes (건칠의 항주름 효과와 미백효과 연구)

  • Objectives : Recently, the demands for the effective and safe depigmentating and anti-aging agents of the skin have increased due to the medical, pharmaceutical and cosmetic reasons. The aim of this study is to search new materials from the traditional herbal medicines which inhibit the aging process of skin in vitro.Methods : Human dermis cell (HS68) was used to test the effect of Rhus verniciflua Stokes (RVS). 80% ethanol or water extracts were screened for their inhibitory activities against elastase. Elastase inhibition effect was tested by microplate reader instrument. And MMP-1 suppression effect of RVS was tested by western blot. These cells were investigated the viability by MTS assay. And also the inhibition effect of tyrosinase by RVS was tested.Results : RVS (final concentrstion 1 ㎎/㎖ appeared over 30% of inhibition of elastase activity. So we are investigated anti wrinkle effects of Rhus verniciflua Stokes look through MMP-1 inhibition activity, also Extracts of RVS showed higher anti-tyrosinase activity than arbutin as final concentration 1 mg/ml. These results suggest that herbal medicines could be strong potential sources of inhibition of anti-aging and whitening effects for the skin.Colclusions : RVS was the best suppressor candidate of elastase activity among other control oriental drugs. It was found that RVS did not have toxicity to cells. We found proper concentration of RVS to treat in HS68 culture and investigated not only wrinkle care effect but also whitening effect of RVS.

Effects of Avocado Yoghurt, Three Beans Cookie, and Tiger Nut Galsu Drink on Fibroblasts (Hs68) and Keratinocytes (HaCaT)

  • The present study aimed to investigate the effects of skin care foods on the synthesis of pro-collagen type I C peptide and suppression of matrix metalloproteinase (MMP)-1 secretion through an in vitro study using fibroblasts (Hs68 cells) and keratinocytes (HaCaT cells). Among the three skin care foods developed in this study, three beans cookie and avocado yoghurt influenced the production of pro-collagen type I C peptide and suppressed MMP-1 secretion; however, tiger nut Galsu drink did not exhibit these effects. All skin care foods, including three beans cookie and plain yoghurt ($50{\mu}g/mL$, p<0.001) influenced the suppression of MMP-1 in addition to other commercially available breast milk production support foods examined, such as Heath Heather ($50{\mu}g/mL$, p<0.001), Happy Mama ($50{\mu}g/mL$, p<0.01), BioLys ($50{\mu}g/mL$, p<0.001), Enfamama ($25{\mu}g/mL$, p<0.0001), and Pregnagen ($25{\mu}g/mL$, p<0.001). Avocado fruit yoghurt ($25{\mu}g/mL$, p<0.05), avocado fruit jam yoghurt ($50{\mu}g/mL$, p<0.01), Enfamama ($100{\mu}g/mL$, p<0.05), and Pregnagen ($100{\mu}g/mL$, p<0.05) influenced the production of pro-collagen type I C peptide and suppressed MMP-1 secretion. This result indicates that only avocado jam yoghurt significantly influenced both the prevention of skin keratinization and acceleration of recovery of skin fibrous structure. Therefore, avocado is a favorable ingredient for nutrition-balanced dietary foods or an essential ingredient in products for revitalization of human skin.

Protective Effects of Pyrus pyrifolia NAKAI Leaf Extracts on UVB-induced Toxicity in Human Dermal Fibroblasts (자외선B 노출로 인해 손상된 피부세포에 대한 돌배나무잎 추출물의 보호효과)

  • Skin damage is mainly caused by environmental factors such as ultraviolet light, heat, and smoking. It is known that reactive oxygen species production is commonly involved in the pathogenesis of skin damage induced by these factors, causing skin aging. Pyrus pyrifolia Nakai continues to be a popular and highly consumed fruit in many countries with known beneficial effects including antitumor, antioxidative, and anti-inflammatory effects. However, there is no evidence of a therapeutic effect of Pyrus pyrifolia extract (PPE) against skin aging via inhibition of mitochondria-mediated apoptosis. In this study, we investigated PPE protective effect against photoaging induced by UVB ($50mJ/cm^2$) in HS68 human dermal fibroblasts. Lactate dehydrogenase assay showed that PPE significantly protected HS68 cells against UVB-induced damage in a dose-dependent manner. Other assays using DCF-DA demonstrated that PPE protected HS68 cells by regulating reactive oxygen species production. PPE also regulated mitochondrial dysfunction and mitochondrial membrane potential induced by UVB, and inhibited UVB-induced caspase-3 activity. These results indicate that PPE protects human dermal fibroblasts from UVB-induced damage by regulating the oxidative defense system.

A Study on infection symptom of infectious pancreatic necrosis virus(IPNV) in chinook salmon embryo cell line (연어 세포주의 전염성 췌장괴사 바이러스의 감염 증상에 관한 연구)

  • CHSE(Chinook Salmon Embryo)-214 fish cell lines was cultured in Eagle's minimal medium (MEM) supplemented with 10% fetal bovine serum and 2mM-glutamin. Optimum growth temperature of CHSE-214 cell line was $20^{\circ}C$. Infectious pancreatic necrosis virus(IPNV) was successfuly multiplied and showed the cytopathic effect in CHSE-214 cell line. Infection symptom of IPNV was observed with inverted phase contrast microscopy. At 6h-12hrs post-infection, the cells infected with IPNV were similer to normal cells. At 18-24hrs post-infection, the cells were somewhat round form and a little swollen form than normal cells. At 30hs post-infection, the cells were becoming more abnormal cells. At 48-68 post-infection, the infected cells were lysed and showed the severe cytopathic effect.

Physiological Activities of Leaf and Twig Extracts from Lindera obtusiloba Blume (생강나무 잎 및 가지 추출물의 생리활성)

  • We investigated the physiological activities of extracts from Lindera obtusiloba Blume leaf and twig (LLW: water extract from Lindera obtusiloba Blume leaf, LLE: 50% ethanol extract from Lindera obtusiloba Blume leaf, LTW: water extract from Lindera obtusiloba Blume twig, LTE: 50% ethanol extract from Lindera obtusiloba Blume twig). Total polyphenol and total flavonoid contents of LTE were 445.38 mg/g and 302.09 mg/g, respectively. The electron donating ability (95.38%) of LTE was higher than that of the LLE (93.76%), LTW (88.09%), and LLW (82.06%). The oxygen radical absorbance capacity of extracts were improved with 50% ethanol condition, rather than hot water. Superoxide radical scavenging activity and FRAP activity of the extracts were improved with an increase of treatment concentration. All the extracts($1,000{\mu}g/mL$) stimulated a production of nitric oxide (NO) in macrophage RAW264.7 cells. In particular, the NO stimulating activity of LTE was superior to that of LLE, LTW, and LLW. The antitumor activity of LTE ($500{\mu}g/mL$) in A549, HeLa and SNU719 was 55.63%, 83.87% and 68.11%, respectively. The UVB-induced MMP-1 production in HS68 cells was suppressed by the treatment of LTE (88.28%), LLE (83.96%), LTW (80.59%) and LLW (76.08%).

Anti-aging Effects of Prescription Extracts Containing Forsythia viridissima L. (연교를 함유한 처방단 추출물의 항노화 효과)

  • Forsythia fructus has been shown to have antioxidative, anti-inflammatory, antibacterial, anti-aging and whitening effects. This work was carried out to investigate the anti-aging effects of Forsythia viridissima-prescription extracts (Yeongyoseungma-tang, Gamiyeongyoseungma-tang, Hoechunyangkyeok-san) on skin. Skin anti-aging effect of Forsythia viridissima-prescription extracts (Yeongyoseungma-tang, Gamiyeongyoseungma-tang, Hoechunyangkyeok-san) was evaluated by using antioxidant assay, anti-glycation activity, inhibitory effect of tyrosinase activity, expression of type I procollagen and UVA-induced matrix metalloproteinase-1 on HS68 cells, and reduction of ${\alpha}$-MSH-induced melanin on B16F1 cells. Forsythia viridissima-prescription extracts showed anti-oxidative and anti-glycation activity. The pectinex hydrolysed extract from Yeongyoseungma-tang 75% EtOH extract and Gamiyeongyoseungma-tang 75% EtOH extract increased the type I procollagen synthesis, and decreased the UVA-induced MMP-1 expression on HS68 cells. The pectinex hydrolysed extracts of Hoechunyangkyeok-san 75% EtOH extract had the inhibitory effect of melanin synthesis on B16F1 cells. Based on these results, we suggest that Forsythia viridissima-prescription extracts may be useful as a potential source of functional anti-aging cosmetics.
