The Effects of Haedoksamul-tang on Oxidative Stress and Hyperlipidemia in LPS-induced ICR Mouse (해독사물탕(解毒四物湯)이 LPS 유도 ICR mouse의 산화스트레스 및 고지혈증에 미치는 효과)

  • Objectives: The present study was conducted to examine whether Haedoksamul-tang (HS), a traditional oriental herbal medicine, have beneficail effects on anti-inflammation and dyslipidemia in lipopolysaccharide (LPS)-induced ICR mouse. Methods: Twenty four 8-week old male ICR mouse were divided into four groups: normal untreated; LPS treatment only; HS 10 mg/kg plus LPS treatment; and HS 30 mg/kg plus LPS treatment. HS was orally administered per day for 2days. Twenty-four hours after LPS injection (10 mg/kg/day, i.p.), all the mice were sacrificed, and serological changes were evaluated. The levels of nuclear factor-${\kappa}B$ (NF-${\kappa}B$), sterol regulatory element-binding transcription protein 1 (SREBP-1) activity and cyclooxygenase-2 (COX-2), inducible nitric oxide synthase (iNOS), tumor necrosis factor a (TNF-a), monocyte chemotactic protein 1 (MCP-1), acetyl-CoA carboxylase a (ACCa) expression were analyzed in Western blot analysis. Results: HS inhibited oxidative stress in the liver of LPS-induced ICR mice. The LPS-induced ICR mice exhibited the increase of NF-${\kappa}B$ activity and COX-2, iNOS, TNF-a, MCP-1 expressions in the liver, while HS treatment significantly inhibited them. Moreover, The administration of HS significantly decreased the elevated serum triglyceride and down-regulated the levels of SREBP-1, ACCa in the liver of LPS-induced ICR mice. Conclusions: In conclusion, HS could have hepato-protective effects against the oxidative stress-related inflammation and abnormal lipid metabolism.

  • This study was performed to analyse some rick ffactors for chronic degenerative diseases of the Korean dict. The study subjects consisted of preschool children(PC), elementary school children(EC), high school students(HS), college students(CS), and adults over 30 years old living in Seoul and the surrounding areas. The subjects over 30 years old were classfied to 30-49 years, 50-69 years, and over 70 years. A dietary survey was conducted using 24-hour recall method and data were collected from 2392 subjects. The dietary risks for chronic degenerative disease such as food sources of fat consumption, high cholesterol intake, low dietary fiber intake, and low calcium intake and high protein intake were analyzed. Younger subjects had more animal fat proportion than older subjects did. HS and CS had the least fat proportion from fishes and shellfishes. Each mean cholesterol intake of PC, EC, HS, and CS was over 300mg/day, and HS load the highest level of 415mg/day. Cholesterol density of the diet of PC was the highest of 175mg/1000kcal and the level decreased when the age increased. The subjects consuming cholesterol over 100 mg/1000kca1 were about 70% of PC and HS. More than 20% of PC, EC, and HS consumed calcium under 75% of RDA with protein consumption over 125% of RDA. Dietary fiber intakes were 18g for HS, CS, 30-49 years, and 50 ∼ 69 years, 14g for EC and over 70 years, and 12g for PC. Ninety percent of PC and EC, 80% of HS and CS, and 70% of tole subjccts over 30years consumed dietary fiber under the level of 10g/1000kca1. The major food sources of dietary fiber were vegetables and grains. From these results, children and adolescents had ricks for chronic degenerative diseases on all the analyzed items. The results of this study would provide the basic information for development of dietary interventions to improve diet find health.

Gene Cloning, Expression, and Characterization of a New Carboxylesterase from Serratia sp. SES-01: Comparison with Escherichia coli BioHe Enzyme

  • The carboxylesterase-encoding gene(bioHs) of a newly isolated strain, Serratia sp. SES-01, was cloned from the genomic DNA library by detecting formation of transparent halo around the colony on LB-tributyrin agar plates. The amino acid sequence of BioHs was highly similar to the members of the BioH enzyme family involved in the biotin biosynthetic pathway; it showed the highest similarity(91%) with that of Serratia proteamaculans. To compare BioHs with other BioH enzymes, the relatively well-known bioHe gene of E. coli was cloned with PCR. After we achieved high-level expression of soluble BioHs and BioHe through the exploration of different culture conditions, the purified BioHs and BioHe enzymes were characterized in terms of specificity, activity, and stability. BioHe was generally more robust to a change in temperature and pH and an addition of organic solvents than BioHs. The two enzymes exhibited a strong preference for carboxylesterase rather than for thioesterase and were optimal at relatively low temperatures($20-40^{\circ}C$) and alkaline pHs(7.5-9.0). The results in this study strongly suggested that both the BioHs and BioHe enzymes would be potential candidates for use as a carboxylesterase in many industrial applications.

  • We have studied the blending effects of Diene NF 35 R and Alfin 1530 at various blending ratios, 100/0, 70/30, 50/50, 30/70, 0/100, and of carbon black HAF-HS and ISAF-HS at various compounding ratios of 45 PHR, 55 PHR, 65 PHR, for tyre tread rubber. As the results, it was found that; 1. For tyre tread rubber, as the blending ratio, AR 1530/Diene NF 35R, indicated 70/30, the physical properties we examined were most excellent. 2. Excellent result was obtained in the case of carbon black compounding ratio of 55 PHR. The compounding of ISAF-HS made better result than that of HAF-HS for tensile strength, but the compounding of HAF-HS made better result than that of ISAF-HS for tearing strength and best result for abrasion quantity. 3. Heat buildup obtained from compounding carbon black HAF-HS indicated low temperature than that from compounding carbon black HAP-HS. As the compounding amount of carbon black increased, and as the blending amount of AR 1530 decreased, the heat buildup increased. 4. Carbon black was more efficient to AR 1630 than io Diene NF 35 R. 5. In the physical properties, mooney viscosity and mooney scorch time, as the compounding amount of carbon black increased, the values of mooney viscosity increased, but that of mooney scorch time had a max. point at the compoundiug amount of carbon black, 55 PHR.

Impact of the Sasang Constitutional Type on the High-Sensitivity C-Reactive Protein, a Predictor of Cardiovascular Disease (사상체질(四象體質)과 심혈관질환 예측인자인 High-sensitivity C-reactive protein의 상관성에 관한 연구)

  • 1. Objectives The purpose of this study was to investigate the high-sensitivity C-reactive protein (hsCRP) related characteristics found in each Sasang Constitutional type. 2. Methods This cross-sectional epidemiological study was conducted using data from 2,842 men and women. After typing each participant into one of the four Sasang constitutional types, they were analyzed on various cardiovascular disease-related variables. 3. Results 1) The predictors of cardiovascular disease (metabolic syndrome, high risk FRS group, high risk hsCRP group) was found to show the highest prevalence in the Taeeum type. 2) In the group with metabolic syndrome, the mean hsCRP concentration in the Taeeum and Soyang types were higher than in the Soeum type. In the group without metabolic syndrome, the mean hsCRP concentration in the Taeeum type were higher than in the Soyang and Soeum types. 3) In the FRS low risk group, the mean hsCRP concentration were higher in order of Taeeum type > Soyang type > Soeum type. In the FRS high risk group, the mean hsCRP concentrations were not significantly different among the Sasang constitutional types. 4. Conclusions The results of this study suggest that the Taeeum constitutional type is a risk factor for high hsCRP and cardiovascular disease. These findings suggest that the prevention of cardiovascular disease is more important in the Taeeum type compared to other constitutional types. Moreover, preventive measures are warranted even in the Taeeum-type persons with low clinical cardiovascular risk.

  • Direct competitive enzyme linked immunosorbent assay(ELISA) with monoclonal antibodies have been studied for quantitative determination of T-2 toxin from the mold corn. The T-2HS, T-2HS-BSA, T-2HS-HRP and monoclonal antibodies against T-2 toxin produced in the studies were qualified for quantitative ELISA test of T-2 toxin. The mean recovery rate from ground com spiked T-2 toxin was 83%. The meaning range of the T-2 test was 60ng to $2{\mu}g$. According to the recovery results with the com spiked T-2, the tests proved to be suitable in the screening of the moldy feed samples for the presence of T-2 toxin and will be able to become the basis of the ELISA test for the quantitative screening kits of T-2 toxin.

Comparison of Motion Sensor Systems for Gait Phase Detection (보행주기 검출용 모션 센서 시스템의 비교)

  • Gait phase detection is important for evaluating the recovery of gait ability in patients with paralysis, and for determining the stimulation timing in FES walking. In this study, three different motion sensors(tilt sensor, gyrosensor and accelerometer) were used to detect gait events(heel strike, HS; toe off, TO) and they were compared one another to determine the most applicable sensor for gait phase detection. Motion sensors were attached on the shank and heel of subjects. Gait phases determined by the characteristics of each sensor's signal were compared with those from FVA. Gait phase detections using three different motion sensors were valid, since they all have reliabilities more than 95%, when compared with FVA. HS and TO were determined by both FVA and motion sensor signals, and the accuracy of detecting HS and TO with motion sensors were assessed by the time differences between FVA and motion sensors. Results show of that the tilt sensor and the gyrosensor could detect gait phase more accurately in normal subjects. Vertical acceleration from the accelerometer could detect HS most accurately in hemiplegic patient group A. The gyrosensor could detect HS and TO most accurately in hemiplegic patient group A and B. Valid error ranges of HS and TO were determined by 3.9 % and 13.6 % in normal subjects, respectively. The detection of TO from all sensor signals was valid in both patient group A and B. However, the vertical acceleration detected HS validly in patient group A and the gyrosensor detected HS validly in patient group B. We could determine the most applicable motion sensors to detect gait phases in hemiplegic patients. However, since hemiplegic patients have much different gait patterns one another, further experimental studies using various simple motion sensors would be required to determine gait events in pathologic gaits.

Expression of Tight Junction Molecule In The Human Serum-Induced Aggregation of Human Abdominal Adipose-Derived Stem Cells In Vitro

  • Previously we have shown that human abdominal adipose derived-stem cells (ADSCs) could aggregate during the high-density culture in the presence of human serum (HS). In the present study, we observed that human cord blood serum (CBS) and follicular fluid (HFF) also induced aggregation. Similarly, porcine serum could induce aggregation whereas bovine and sheep sera induced little aggregation. qRT-PCR analyses demonstrated that, compared to FBS-cultured ADSCs, HS-cultured cells exhibited higher level of mRNA expression of CLDN3, -6, -7, -15, and -16 genes among the tight junction proteins. ADSCs examined at the time of aggregation by culture with HS, BSA, HFF, CBS, or porcine serum showed significantly higher level of mRNA expression of JAM2 among JAM family members. In contrast, cells cultured in FBS, bovine serum or sheep serum, showed lower level of JAM2 expression. Immunocytochemical analyses demonstrated that the aggregates of HS-cultured cells (HS-Agg) showed intense staining against the anti-JAM2 antibody whereas neither non-aggregated cells (HS-Ex) nor FBS-cultured cells exhibited weak staining. Western blot results showed that HS-Agg expressed JAM2 protein more prominently than HS-Ex and FBS-cultured cells, both of latter reveled weaker intensity. These results suggest that the aggregation property of ADSCs during high-density culture would be dependent on the specific components of serum, and that JAM2 molecule could play a role in the animal sera-induced aggregation in vitro.

In Vitro Genotoxicity Assessment of a Novel Resveratrol Analogue, HS-1793

  • Resveratrol has received considerable attention as a polyphenol with various biological effects such as anti-inflammatory, anti-oxidant, anti-mutagenic, anti-carcinogenic, and cardioprotective properties. As part of the overall safety assessment of HS-1793, a novel resveratrol analogue free from the restriction of metabolic instability and the high dose requirement of resveratrol, we assessed genotoxicity in three in vitro assays: a bacterial mutation assay, a comet assay, and a chromosomal aberration assay. In the bacterial reverse mutation assay, HS-1793 did not increase revertant colony numbers in S. typhimurium strains (TA98, TA100, TA1535 and TA1537) or an E. coli strain (WP2 uvrA) regardless of metabolic activation. HS-1793 showed no evidence of genotoxic activity such as DNA damage on L5178Y $Tk^{+/-}$ mouse lymphoma cells with or without the S9 mix in the in vitro comet assay. No statistically significant differences in the incidence of chromosomal aberrations following HS-1793 treatment was observed on Chinese hamster lung cells exposed with or without the S9 mix. These results provide additional evidence that HS-1793 is non-genotoxic at the dose tested in three standard tests and further supports the generally recognized as safe determination of HS-1793 during early drug development.

Effects of cooling systems on physiological responses and intestinal microflora in early gestating sows exposed to high-temperature stress

  • This study was conducted to investigate the effect of cooling systems on reproductive performance, body temperature, blood metabolites, and the intestinal microbiome in early gestating sows exposed to high ambient temperature. In total, 39 pregnant sows (Landrace × Yorkshire; 2 parities) were randomly assigned to and maintained in the following three treatment groups (13 sows per group) over days 0 to 35 of pregnancy: (i) air cooling (AC; 26.87 ± 1.23℃), (ii) water-drip cooling (WC; 28.81 ± 0.91℃), and (iii) a lack of cooling with heat stress (HS; 30.72 ± 0.70℃). Backfat thickness was measured before and after HS. Feces were collected on day 0 and 35 d of the trial for microbiome analysis, whereas blood was taken at day 35 of pregnancy and analyzed. Reproductive performance and physiological responses were identified at day 35. Respiration rate along with rectal and skin temperatures were lower (p < 0.05) in the AC group than in the HS and WC groups. Serum blood urea nitrogen values were increased (p < 0.05) in the WC group compared with those measured in the AC and HS groups. Triiodothyronine was found at greater levels (p < 0.05) in the AC than in the HS group. Reproductive performance was not affected by the cooling systems. At the phylum level, fecal pathogenic Spirochaete and Euryarchaeota were found in higher numbers (p < 0.05) in all groups after HS. Similarly, at the genus level, the amount of Treponema was greater (p < 0.05) in all groups after HS. In conclusion, our results suggest that AC or WC can ameliorate or mitigate the adverse effects of HS on the physiological parameters of pregnant sows reared under high temperatures.
