Content and Stability of Alisol B, Alisol B acetate in Alismatis Rhizoma (택사의 Alisol B, Alisol B acetate 함량과 안정성)

  • The current definition of Arismatis Rhizoma is the tuberous root of Alisma orientale Juzepzuk in the Korean Pharmacopoeia, but there is still no marker compound. So it has difficulties in quality control. Therefore, in this study, we have established a method to analyze alisol B and alisol B acetate using HPLC as a marker compounds of Arismatis Rhizoma. As the result of the analysis, alisol B content was ranged from 0.02% to 0.50% and alisol B acetate content was ranged from 0.12% to 0.25% in 40 samples. The stability of alisol B and alisol B acetate was investigated during 24 months. As a result, alisol B acetate was significantly decreased. The marker compound of Arismatis Rhizoma is alisol B acetate and the content of 0.05% or more is suitable by applying the stability results.

Discrimination between steam processed and unprocessed Tubers of Gastrodia elata Blume by HPLC

  • In this study, to evaluate the effectiveness and safety of oral therapy using Gastrodiae Rhizoma, a new HPLC-PDA analysis method was developed for the simultaneous quantitation of the three major components: (1) gastrodin, (2) gastrodigenin, and (3) p-hydroxybenzaldehyde, in steam processed and unprocessed tubers of Gastrodia elata Blume. The clear separation of the three components was achieved on a C18 column (250 × 4.6 mm, 5 ㎛) by gradient elution using water (including 0.1 % formic acid) and acetonitrile as the mobile phase. The flow rate was 1.0 mL/min, and the UV detector wavelength was set at 270 nm. The results demonstrate satisfactory linearity, recovery, precision, accuracy, stability, and robustness. The established HPLC-PDA method was applied to quantify three major compounds in 59 samples of G. elata Blume tubers. Finally, the steam processed and unprocessed tubers of G. elata Blume were successfully distinguished by pattern recognition analysis.

Simultaneous Analysis of Cold Medicine Component by High-Performance Liquid Chromatography(HPLC) (고성능 액체크로마토그래피(HPLC)를 이용한 Cold Medicine 성분의 동시 분석)

  • In this study, for the purpose of standardized quality control of a cold medicine, we simultaneous analyzed four main chemical components of a cold medicine: acetaminophen, caffeine, methyl paraben, and propyl paraben. The sample was subjected to quantitative analysis using high performance liquid chromatography (HPLC), after pretreatment of four components. The experiment was carried out by using Isocratic elution at wavelength of 270nm. Acetonitrile and water (H2O) were used as a mobile phase at a flow rate of 1.0mL/min in a commercial C18 reversed-phase column. A volume of 10uL cold medicine were injected into the column with column oven temperature at 35℃. As a result of the experiment, the values of Resolution were 4.983, 1.596, 5.519, and 1.678 respectively-well over Rs >1.5, which indicates that the separation of four components were efficient. In addition, value of symmetry factor of the components was 1.056, 1.069, 1.032, and 1.133 respectively, to show its symmetrical stability. The calibration curve of all four components exhibits good linearity with R2 >0.9995 to 0.9999. Furthermore, the limit of detection(LOD) were between 0.0118 to 1.5973 mg/mL, while the limit of quantification (LOQ) were between 0.0353 to 4.7919 ㎍/mL with the recovery rate of 79.6% ~ 120.5%. The results of this study showed an efficient quality evaluation of a simultaneous analysis method for cold medicine components.

Observation of Molecular Relaxation Behavior of Glucose Powders with Different Structures and Particle Sizes Using Low Field Nuclear Magnetic Resonance (NMR) (NMR을 이용한 구조 및 입도 차이에 따른 분말 포도당의 molecular relaxation behavior의 관찰)

  • Molecular relaxation behaviors of crystalline glucose anhydrous, crystalline glucose monohydrate, and amorphous glucose with different particle sizes were observed by measuring spin-spin relaxation time constant $(T_2)$ at the temperature range of $-20\;to\;110^{\circ}C$ using temperature-controlled low field nuclear magnetic resonance spectroscopy. No change in $T_2$ values of crystalline glucose anhydrous was observed throughout the temperature range, whereas $T_2$ values of crystalline glucose monohydrate and amorphous glucose increased from around $45\;and\;65^{\circ}C$, respectively. These results indicate that molecular mobility of crystalline glucose anhydrous does not change even at temperature higher than $100^{\circ}C$ and that the stability of powdered glucose could be improved by increasing the particle size of materials.
