Carotenoid의 생리활성과 함량분석 (Biological Activities and Analysis of Carotenoids in Plants)

  • Carotenoids are the major pigment of pepper (Capsicum annuum) and tomato (Lycopersicon esulentum) which are very important foods in Korea. However the analysis of carotenoids is quite complicated because of their diversity and the presence of cis-trans isomeric forms of these compounds. The objective of this review is to collect the achievements on the field of the chromatographic separation of carotenoids in food and some vegetables, to describe and critically evaluate the techniques, And to compare the benefits and shortcomings of the various chromatographic methods such as adsorption and reversed-phase HPLC and thin-layer chromatography. HPLC equipped with ultra-violet or photodiode array detection is most often employed in routine use for the analysis of carotenoids. Here, the method to analyze carotenoids by HPLC separation after solvent extration and purification from pepper powder samples done in our laboratory is also mentioned.

Achiral and Chiral Determination of Benzophenanthridine Alkaloids from Methanol Extracts of Hylomecon Species by High Performance Liquid Chromatography

  • A high performance liquid chromatographic (HPLC) method was developed for the qualitative and quantitative determination of benzophenanthridine alkaloids from the methanol extracts of Hylomecon hylomeconoides and H. vernale (Papaveraceae). Achiral and chiral methods were adapted for the separation of 6-methoxydihydrosanguinarine (1), 6-acetonyldihydrosanguinarine (2) and dihydrosanguinarine (3). The achiral reversed phase HPLC method made it possible the simultaneous separation and determination of 1, 2 and 3 within 20 min on ODS column using acetonitrile-phosphate buffer (50 mM, pH 7.0) (50 : 50, v/v). The separation and determination of 1 and 2 enantiomers was available using chiral columns. The same amount of (+) and (-)-enantiomers of 1 was found from the methanol extract of specimen, indicated that 1 could be the artifact produced by the reaction of sanguinarine with methanol. H. hylomeconoides showed higher level of 1 and 3 in compared with H. vernale, especially in root samples permitting the possibility of chemical discrimination between two species.

Chiral Purity Test of Bevantolol by Capillaryelectrophoresis and High Performance Liquid Chromatography

  • Two methods for the chiral purity determination of bevantolol were developed, namely capillary electrophoresis (CE) using carboxymethyl-${\beta}$-cyclodextrin (CM-${\beta}$-CD) as a chiral selector and high-perfomance liquid chromatography (HPLC) using a chiral stationary phase. In the HPLC method, the separation of bevantolol enantiomers was performed on a Chiralpak AD-H column by isocratic elution with n-hexane-ethanol-diethylamine (10:90:0.1, v/v/v) as mobile phase. In the CE method, bevantolol enantiomers were separated on an uncoated fused silica capillary with 50 mM amonium phosphate dibasic adjusted to a pH 6.5 with phosphoric acid containing 15 mM CM-${\beta}$-CD as running buffer. Validation data such as linearity, recovery, detection limit, and precision of the two methods are presented. The detection limits of S-(-)-bevantolol were 0.1% and 0.05% for CE and HPLC method, respectively and R-(+)-bevantolol were 0.15% and 0.05% for CE and HPLC method, respectively. There was generally good agreement between the HPLC and CE results.

  • RP-HPLC에서 주요 변수인 이동상의 조성과 컬럼의 종류를 변화시켜 난백 단백질을 실험하였다. C4, C8 C18 컬럼을 비교 실험하여 C18 컬럼에서 가장 많은 피크를 보여 C18 컬럼을 선택하였다. 등용매 용리에서 ACN : water의 조성이 50 : 50에서 가장 많은 피크를 보여 이 결과를 토대로 기울기 용리를 하여 실험하였다. 기울기 용리에서 얻어진 4가지 피크를 확인하기 위하여 단일성분인 lysozyme와 ovalbumin의 체류시간을 측정하여 확인하였고 논문을 통해 2가지 피크를 예측할 수 있었다.

  • A new chiral derivatization agent with sugar moiety. 2, 3.4, 6-tetra-O-acetyl-D-galactopyranosyl isothiocyanate (GATC) was synthesized. Several $\beta$-blockers were investigated for the possible separation of the enantiomers by reversed-phase HPLC after derivatization with this new chiral derivatization agent (GATC). GATC was reacted readily with $\beta$-blockers at room temperature and the reaction mixture could directly be injected into the HPLC system. (omitted)

  • In the past two decades, separations of enantiomers (optical isomers) by high-performance liquid chromatography (HPLC) have remarkably advanced [1]. Among many commercially available chiral stationary phases (CSPs) for HPLC, polysaccharide-based CSPs are the most popular ones, which can cover the resolution of a wide range of the chiral compounds [2, 3, 4]. Here, I will explain mainly the HPLC separation of enantiomers using these CSPs. (omitted)

Kromasil HPLC 칼럼을 이용한 Ibuprofen의 분리특성 연구 (Separation Characteristics of Ibuprofen in Kromasil HPLC Column)

  • Kromasil HR100-5CHI-TBB 칼럼을 사용하여 HPLC를 통한 비스테로이드 계통의 진통 및 소염제인 racemic ibuprofen의 분리특성을 연구하였다. 이동상의 조성비 변고에 따른 분리도 (resolution), 이론단수 (number o( theoretical plates), 이론단 높이 (HETP: Height Equivalent to a Theoretical Plate), 용량인자 (capacity factor)를 계산하고, 그 결과 정성분석 기준치 이상의 분리도를 갖고 분리시간을 단축할 수 있는 hexane /t-BME의 조성이 55 / 45일 때 $1\%$의 acetic acid를 첨가했을 때의 용매를 최적의 이동상으로 결정했다. 유속에 따른 칼럼의 효율을 비교하기 위한 실험을 통해 유속의 상승은 칼럼의 효율의 감소를 야기하고, 분리도를 감소시킴을 보였다. PIM (Pulsed Input Method)을 사용하여 분석한 결과 ibuprofen 의 농도가 증가할수록 Langmuir 등온흡착식을 따르는 비선형 거동을 보임을 증명하였고, 그 결과 등온흡착식을 결정하였다. 등온흡착식은 S-form과 R-form의 경우 각각 $C_{S,S}=\frac{1.178C_{M,S}}{1+0.019C_{M,S}},\;C_{S,R}=\frac{1.866C_{M,S}}{1+0.017C_{M,S}}$로 결정하였다.

  • 식물 유래 화합물은 다양한 생리활성과 구조 다양성으로 많은 연구자에게 관심을 받고 있으나 화합물 분리는 많은 시간 소요, 과량의 유기용매 소모와 분리과정 중의 활성성분의 유실 등의 어려움을 겪게 된다. 이러한 점을 극복하기 위하여 HPLC/SPE/HPLC를 결합한 Sepbox 분리시스템을 이용하여 고추잎 추출물에서 288개 분획물을 획득하였고 이 중에서 alpha-glucosidase 저해효능을 가지는 화합물인 luteolin 7-O-glucoside를 효과적으로 확인할 수 있었다. 또한 고추잎에 풍부한 플라보노이드 다당체를 가수분해함에 따라 해당 활성이 증가함을 검증하였다. 그러므로 본 연구결과는 Sepbox 시스템이 생리활성 평가와 결합할 경우 식물 내 유용물질 탐색에 효과적임을 보여주었다.
