Fatty and organic acids, and barbaloin in various parts of Aloe species dried at different drying temperatures (건조조건(乾燥條件)에 의(依)한 알로에의 부위별(部位別), 품종별(品種別) 지방산(脂肪酸) 유기산(有機酸) 및 Barbaloin 성분(成分) 연구(硏究))

  • The fatty and organic acids, and barbaloin in various parts of aloe species dried at different temperatures were analyzed by GC and HPLC. Seven fatty acids and six organic acids were identified and quantified. In the case of fatty acids, generally, the contents of palmitic and eicosanoic acid were abundants, and compared to the total contents of seven fatty acids, Aloe arborescence variant 1 was abundant, but Aloe saponaria was poor. And six fatty acids were distributed in the aloe species with the exception of linoleic acid. The contents of malic, citric and oxalic acids in the aloe species were higher than those of other acids, and compared to the sum of contents of six organic acids, Aloe saponaria was high, but Aloe arborescence variant 1 was low. Therefore Aloe arborescence variant 1 was abundant in total fatty acids, but poor in total organic acids. The contents of fatty and organic acids in the sample dried at $65^{\circ}C$ and $80^{\circ}C$ air circulation were almost similar. The contents of fatty and organic acids in the freeze-dried samples were lower than in the other dried samples. The contents of barbaloin in Aloe arborescence and A. arborescence variant 1 were higher than those of other various samples, and barbaloin was not detected in Aloe saponaria. As the drying temperature was increased, the contents of barbaloin in the various parts of the Aloe vera decreased.

Effects of Caffeic Acid Methyl Ester from Mentha arvensis Linne var. piperascens on Function of E6 Oncoprotein of HPV 16 (박하에서 분리한 Caffeic Acid Methyl Ester가 자궁경부암 바이러스 발암단백질 E6의 기능에 미치는 영향)

  • Cervical cancer is one of the leading causes of female death. Viral oncoproteins E6 and E7 are selectively retained and expressed in carcinoma cells infected with HPV (Human pa pilloma virus) type 16 and cooperated in immotalization and transformation of primary keratinocyte. E6 and E7 oncoproteins interfere the functions of tumor suppressor proteins p53 and retinoblasoma protein (pRb), respectively. Among a lots of natural products, Mentha arvensis Linne var.piperascens have inhibitory effects on bindings between E6 oncoprotein and tumor suppressor p53, E3 ubiqutin- protein ligase (E6AP). HPV oncoprotein inhibitors from Mentha piperita L. were isolated by solvent partition and column chromatography (Silica gel, RP-18) and inhibitory compounds were finally purified by HPLC using an ELISA screening system based on binding between E6 and E6AP. The aim of this study is to identify the structure of inhibitory compounds and to investigate whether these compounds have inhibitory effects on functions of E6 oncoprotein. We investigated whether caffeic acid methyl ester (CAM) extracted from Mentha piperita L. could inhibit the function of E6 oncoprotein. CAM inhibited the in vitro binding of E6 and E6AP which are essential for the binding and degradation of the tumor suppressor p53 and also inhibited the proliferation of human cervical cancer cell lines (SiHa and CaSKi) in a dose response manner. These results suggest that CAM inhibited the function of E6 oncoprotein, suggesting that it can be used as a potential drug for the treatment of cervical cancers infected with HPV.

Application as a Cosmeceutical Ingredient of Euryale ferox Seed Extract (가시연꽃 종자 추출물의 화장품 원료로서의 특성)

  • In our search for the natural cosmetic ingredients, we found that Euryale ferox seed extract exhibited the strong antioxidative activity. Five active compounds were isolated from the ethyl acetate extract through various chromatographic methods and their structures were determined by NMR and MS spectral analysis. These compounds were identified as fucosterol (1), 3-(4-hydroxy-3-methoxybenzyl)-4-[(7'R),5'-dihydroxy-3'-methoxybenzyl]tetrahydrofuran (2), resorcinol (3), pyrogallol (4) and 4-O-methylgallic acid (5).We evaluated the antioxidative, antielastase activities and melanogenesis inhibitory effects of these compounds. The $SC_{50}$ values of compounds 2 ~ 5 for free radical scavenging activity were $17.0\;{\sim}\;100.2\;{\mu}M$ and especially compounds 4 and 5 were 6-fold more effective than ferulic acid as a positive control. And compounds 2 ~ 4 inhibited human neutrophil elastase with $IC_{50}$ values of $18.8\;{\sim}\;78.2\;{\mu}M$ and compound 3 also inhibited melanin synthesis in B16F10 melanoma cells with an $IC_{50}$ value of $492.8\;{\mu}M$. These results suggest that Euryale ferox extract having a lot of various active ingredients may be useful as a natural multi-functioning agent.

Analysis and Risk Assessment of Aflatoxin M1 in Infant Formula (분유 중 아플라톡신 M1 분석 및 위해평가)

  • To analyze aflatoxin $M_1$ ($AFM_1$), we dissolved infant formula in warm water and cleaned it by using an immunoaffinity column (IAC). The amount of $AFM_1$ was determined by high-performance liquid chromatography coupled with fluorescence detection. $AFM_1$ was detected in 281 of 439 samples. Thus, the detection rate of $AFM_1$ was 64.0%. The average concentration of $AFM_1$ in positive samples was 2.6 ng/kg (of prepared formula). The estimated daily intake (EDI) of $AFM_1$ through infant formula was 0.087-0.646 ng/kg body weight/day and the additional number of cases of liver cancer associated with exposure to $AFM_1$ would be 0.003-0.020 cancer cases/1,000,000. Because there is less than 1 cancer case/1,000,000 per year, the exposure to $AFM_1$ through infant formula in Korea is considered to be an unlikely human health concern.

Free Redical Scavenging and Cytotoxicity Activitives of Soybean Germ Saponin (대두배아 사포닌의 유리기 생성 억제 및 세포독성)

  • This study was carried out to investigate functional activities of the free radical scavenging and germ of Glycin max. Merrill fur cytotoxicity toward P338 and L1210 cells derived from mouse. Effect of crude saponin were examined to oxygen radicals and their scavenger enzymes in liver fractions of Spragae-Dawley(SD) rats. Male rats were fed basic diets of control and experiment diets of 0.5∼1.0% crude saponin. There were no significant differences in hydroxy radical($.$OH) formation of liver mitochondria and microsomes in 1.0% group, while $.$OH formations were significantly decrease in 0.5% and 1.0% saponin compared with control group. Their oxygen radical(O$_2$$\^$$.$/) scavenging activities were significantly decrease in liver cytosol of 0.5% and 1.0% saponin group compared with control group. Soybean germ saponin was isolated purified by the method of HPLC to investigate the cytotoxicity of mouse cells by using the MTT assay. SA-1 saponin fraction of soybean germ showed to inhibit toward growth cell of P338 and L1210 cells and its showed less than 50% cytotoxicity These results suggest that the saponin may play a effective role in attenuating a oxygen radical formations and increasing a scavenger enzyme activities.

Effect of Administration Method on Blood Garcinia cambogia Concentration and Antioxidative Activity (투여 방법에 따른 가르시니아 캄보지아 혈중 농도 변화와 항산화 효과 변화)

  • The objective of this study was to investigate the effects of administration methods for on Garcinia cambogia on blood Garcinia cambogia concentration and antioxidative levels. Rats were divided into three groups: G1 (normal group), G2 (one administration of Garcinia cambogia extract 2,800 mg/kg b.w.), G3 (three separate administrations every 6 h of Garcinia cambogia extract 750, 850, and 1,200 mg/kg b.w. for 18 h). Blood samples were collected every hour, and animals sacrificed 18 h after the oral administration of Garcinia cambogia extract. We examined changes in the serum concentration of Garcinia cambogia by HPLC analysis. Two hours following an oral administration of Garcinia cambogia extract (2,800 mg/kg b.w.), serum Garcinia cambogia levels reached their maximum, but gradually decreased until 10 hours when it was no longer detectable. In contrast, serum Garcinia cambogia levels under G3 administration were maintained above a certain level after 18 h. To determine whether this level of Garcinia cambogia could affect blood oxidative levels, we measured serum lipid peroxidation by TBARS levels. TBARS levels from G3 treatment were significantly lower than G1 and G2. To analyze other antioxidative activities, radical scavenging activities were measured by the DPPH and ABTS radical scavenging assays. There were no significant differences between the groups in DPPH radical scavenging activity. However, ABTS radical scavenging activity significantly increased with G3 treatment compared with G1 and G2. In conclusion, our data show that three times administration of Garcinia cambogia every 6 h may helpful for maintaining serum Garcinia cambogia levels and antioxidative effects.

Chlorogenic Acid Isomers from Sorbus commixta of Ulleung Island Origin and Their Inhibitory Effects against Advanced Glycation End Product (AGE) Formation and Radical Scavenging Activity (울릉 마가목의 클로로겐산 이성체의 최종당화산물의 생성 저해 및 라디칼 소거 활성)

  • Advanced glycation end product (AGE) formation and reactive oxygen species are potential therapeutic targets for the prevention of diabetic nephropathy and other pathogenic complications. Activity-guided isolation of an ethylacetate-soluble portion of 80% methanolic extract from fruits of Sorbus commixta of the Ulleung Island origin using AGE formation inhibition assay led to the isolation and identification of three caffeoylquinic acid derivatives of a previously known structure, 3-O-caffeoylquinic acid (neochlorogenic acid; 1), 4-O-caffeoylquinic acid (cryptochlorogenic acid; 2), and 5-O-caffeoylquinic acid (chlorogenic acid; 3). The structures of these compounds were confirmed by interpretation of nuclear magnetic resonance and mass spectrometry data. Among the isolates, the major metabolite, neochlorogenic acid (1) showed the most potent inhibitory effect against AGE formation with an $IC_{50}$ value of $167.5{\pm}3.5{\mu}M$. Furthermore, all isolated chlorogenic acid isomers were evaluated for their radical scavenging activity against peroxynitrite, and structurally related isomers 1, 2, and 3 exhibited potent inhibitory effects in this radical scavenging assay. This result suggests that the monocaffeoyl quinic acid derivatives isolated from S. commixta might be beneficial for the regulation of diabetic complications and related diseases.

Effect of Nitrogen Split Application on Growth and Quality of Okro Tea Leaves(Camellia sinensis var. sinensis) (질소 분시횟수가 옥로차엽의 생육과 품질에 미치는 영향)

  • The split application of a nitrogen fertilizer was applied at the levels to 3, 4, 5, 6 and 7 time. The results of yield and quality of Okro tea are summarized as follows: Among five treatments, the growth and yield were somewhat more excellent in the five time split treatments compared with other treatments. The chemical components such as total nitrogen, total amino acid and vitamin C were somewhat higher in the five time split treatment compared with other treatment. However, the content of components such as tannin, caffeine, chlorophyll were not different among plots of $4{\sim}7$ time split application except three time split application manuring. The content of fatty acid was producted $2,888{\sim}2,933mg/100g$. Especially, the content of oleic acid, linoleic acid and linolenic acid was the higheat in the five time split application. Sixteen kinds of free amino acid including theanine as a good taste component of Okro tea were isolated and measured by HPLC. Theanine in the five time split application was higher with value of $72{\sim}513mg/100g$ than in other treatment. In scoring test, tea appearance and interal qualities of 3 time split application become lower than any other treatment as 80.2 point, while five time application had the highest with 84.7 point. Conquently, those results meant that five time split application was optimum split application of fertilizer for high yield and good quality of Okro tea.

Analysis of Florfenicol in Meats (식육 중 항생제 florfenicol의 분석)

  • Analytical method for determination of florfenicol was developed for estimate veterinary drug residue of unestablished MRLs in meat. The method was validated in correspondence with the CODEX guideline for florfenicol residue in meat. The samples mixed with sodium sulfate were extracted with ethyl acetate. After clean-up, the residue was dissolved in mobile phase and analyzed using high-performance liquid chromatography with fluorescence detector. The calibration curve showed good linearity($r^2=0.9997$) within the concentration range of $0.05{\sim}1.0\;mg/kg$. The limit of detection(LOD) and limit of quantification(LOQ) were validated at 0.012 and 0.039 mg/kg, respectively. The recoveries in fortified meat ranged from 85.6 to 95.6%($1.1{\sim}5.3%$ RSD) at the 0.05 to 0.4 spiking levels. We monitored 150 samples of meats that were purchased in Korea(Seoul, Busan, Daegu, Daejeon and Gwangju). Among tested samples, florfenicol was detected in 1 of pig at the level of 0.040 mg/kg, and below LOQ in 1 of cattle, 2 of pig and 2 of chicken. The residues of florfenicol in the tested samples were within the MRLs.

Antioxidantive, Phospholipase $A_2$ Inhibiting, and Anticancer Effect of Polyphenol Rich Fractions from Panax ginseng C. A. Meyer (한국산 인삼의 Polyphenol 분획물의 항산화, Phospholipase $A_2$ 및 암세포증식 억제효과)

  • The polyphenol fractions of Korean ginseng were purified using Sephadex LH-20, MCI gel, Bondapak $C_{18}$ TLC, and HPLC from the 60% acetone soluble fraction. Fraction I showed 48.16%, 79.71% and 43.55% inhibition at 150 ppm against lipid oxidation in the presence of copper ion, superoxide and hydrogen peroxidation. Electron donating abilities of fraction II showed 35.17% inhibition at 200 ppm. Fraction III showed 48.49% and 25% inhibition at 150 ppm against lipid oxidation in the presence of ferrous ion and hydroxy radical ion. The phospholipase $A_2$ inhibitory effect of fraction III was 48.9% at the concentration of $60\;{\mu}g/ml$. The cytotoxic effects of fraction II was the highest (73.29% at 0.25 mg/ml) among the tested polyphenol fractions.
