The Optimization of Method for Prediction of Drug-Induced Liver Injury Using HepG2 Cells Cultured with Human Liver Microsomes (Human Liver Microsomes과 HepG2 세포를 이용한 약물유래 간독성 평가 방법의 최적화)

  • The aim of the present study was to optimize in vitro method for the prediction of drug-induced liver injury using human liver microsomes (HLM). Cytotoxicity test of cyclophosphamide and acetaminophen in HepG2 cells cultured with HLM showed that the newly established condition using 0.375 mg/ml HLM for 24 hr incubation was comparable or more sensitive than the previously established condition using 0.75 mg/ml HLM for 12 hr incubation. Although the cytotoxic effect of troglitazone was completely attenuated by 0.75 mg/ml HLM, it was augmented by 0.375 mg/ml HLM in the presence of the NADPH-generating system. The cytotoxic effect of chlormezanone, a withdrawn drug due to hepatotoxicity in human, was increased by HLM in the presence of the NADPH-generating system. In contrast, the cytotoxic effect of methapyrilene, a withdrawn drug due to hepatotoxicity in rats, was decreased by HLM in the presence of the NADPH-generating system. The present study suggests that the optimized in vitro method using HLM can be useful for the prediction of drug-induced hepatotoxicity.

Understanding and Application of Hierarchical Linear Model (위계적 선형모형의 이해와 활용)

  • A hierarchical linear model(HLM) provides advantages over existing traditional statistical methods (e.g., ordinary least squares regression, repeated measures analysis of variance, etc.) for analyzing multilevel/longitudinal data or diary methods. HLM can gauge a more precise estimation of lower-level effects within higher-level units, as well as describe each individual's growth trajectory across time with improved estimation. This article 1) provides scholars who study children and families with an overview of HLM (i.e., statistical assumptions, advantages/disadvantages, etc.), 2) provides an empirical study to illustrate the application of HLM, and 3) discusses the application of HLM to the study of children and families. In addition, this article provided useful information on available articles and websites to enhance the reader's understanding of HLM.

Effects of Substitution of Hydrolyzed Leather Meal for Fish Meal in the Broiler Diet (브로일러 사료에서의 피혁분의 어분대치 효과)

  • An experiment was carried out to evaluate the feeding value of hydrolyzed leather meal (HLM) replacing fish meal A total of 240 day-old female chicks of Maniker strain was divided into 20 groups of 12 birds ecah. Each groups was assigned to one of the five dietary treatments (0. 1. 2, 4 and 6% HLM). The feeding trial was conducted for six weeks. Results obtained are summarized as follows: 1. The HLM used in this experiment contained 74.41% crude protein. 17.0% crude ash and 2.0% chromium. Pepsin digestibility of the protein was measured to be 85%. Amino acid contents were also determined 2. The inclusion of HLM up to 4% in broiler rations did not depress the growth rate, feed consumption and feed efficiency of experimental birds. However, these performance parameters were significantly (p<0.05) depressed when fed 6% HLM. These results indicate that HLM in place of fish meal can be used up to 4% in tile broiler rations.

  • Application of residential demand response (DR) programs are currently realized up to a limited extent due to customers' difficulty in manually responding to the time-differentiated prices. As a solution, this paper proposes an automatic home load management (HLM) framework to achieve the household minimum payment as well as meet the operational constraints to provide customer's comfort. The projected HLM method controls on/off statuses of responsive appliances and the charging/discharging periods of plug-in hybrid electric vehicle (PHEV) and battery storage at home. This paper also studies the impacts of different time-varying tariffs, i.e., time of use (TOU), real time pricing (RTP), and inclining block rate (IBR), on the home load management (HLM). The study is effectuated in a smart home with electrical appliances, a PHEV, and a storage system. The simulation results are presented to demonstrate the effectiveness of the proposed HLM program. Peak of household load demand along with the customer payment costs are reported as the consequence of applying different pricings models in HLM.

  • To measure the effect of school zone on housing cost, Linear Regression Model is widely used, and school zone is known as a key determinant of housing cost in Korea. However, when the Hierarchical Linear Model (HLM) is applied with the same data, school effect on housing cost becomes statistically non-significant. It is because HLM effectively separates the effect of individual housing's attributes from the group effect. In sum, the housing cost of Kangnam, where good public schools are located, is apparently is higher than that of Kangbuk. However, the school effect on housing cost (Level 2) becomes non-significant when individual housing's attributes (Level 1) are controlled with HLM.

Reliability Improvement of Offshore Structural Steel F690 Using Surface Crack Nondamaging Technology

  • Microcracks can rapidly grow and develop in high-strength steels used in offshore structures. It is important to render these microcracks harmless to ensure the safety and reliability of offshore structures. Here, the dependence of the aspect ratio (As) of the maximum depth of harmless crack (ahlm) was evaluated under three different conditions considering the threshold stress intensity factor (Δkth) and residual stress of offshore structural steel F690. The threshold stress intensity factor and fatigue limit of fatigue crack propagation, dependent on crack dimensions, were evaluated using Ando's equation, which considers the plastic behavior of fatigue and the stress ratio. ahlm by peening was analyzed using the relationship between Δkth obtained by Ando's equation and Δkth obtained by the sum of applied stress and residual stress. The plate specimen had a width 2W = 12 mm and thickness t = 20 mm, and four value of As were considered: 1.0, 0.6, 0.3, and 0.1. The ahlm was larger as the compressive residual stress distribution increased. Additionally, an increase in the values of As and Δkth(l) led to a larger ahlm. With a safety factor (N) of 2.0, the long-term safety and reliability of structures constructed using F690 can be secured with needle peening. It is necessary to apply a more sensitive non-destructive inspection technique as a non-destructive inspection method for crack detection could not be used to observe fatigue cracks that reduced the fatigue limit of smooth specimens by 50% in the three types of residual stresses considered. The usefulness of non-destructive inspection and non-damaging techniques was reviewed based on the relationship between ahlm, aNDI (minimum crack depth detectable in non-destructive inspection), acr N (crack depth that reduces the fatigue limit to 1/N), and As.

HLM analysis of effects of Cultural capital and Social Welfare Expenditures on life satisfaction of the elderly in OECD countries

  • The object of this study is through an empirical analysis, how cultural capital at the individual level and social welfare expenditure at the national level affect the life satisfaction of the elderly. In this study method, a Hierarchical Linear Model(HLM) analysis was performed on 3,297 elderly people aged 65 and older and 9 OECD countries. As a result of analysis, first, it was confirmed that life satisfaction and social class had a significant effect. Therefore, in to increase the satisfaction of the life of the elderly, policy and practical intervention measures that can narrow the gap between social classes should be prepared. Second, the old-age pension and survivor's pension had no significant effect on life satisfaction. However, as a result of the interaction, social class has a positive effect on life satisfaction, and it was confirmed that the lower the income inequality, the more positive the life satisfaction was. In conclusion, this implies that both individuals and countries should make efforts to variously increase the life satisfaction of the elderly.

The Effect of Individual and Team Characteristics on Knowledge Creation : An Analysis by Hierarchical Linear Model (HLM) (개인과 집단의 특성이 지식창출에 미치는 영향)

  • This paper investigates the effect of stress on knowledge creation. The goal stress of resource inadequacy and job stress had negative influences on knowledge creation. However, the cohesion and mastery climate of team had positive influence on knowledge creation. Therefore this paper verifies the moderate role of the cohesion and mastery climate of team on the relationship between stress and knowledge creation. The model developed was tested using data collected from knowledge based industry with 375 members in 69 teams in 12 different firms. A Hierarchical Linear Model (HLM) was used to test the hypotheses generated from the model. Results show that job stress had a negative influence on knowledge creation as we expected but the goal stress didn't. The mastery climate of team affected knowledge creation positively and moderated the relationship between the goal stress and knowledge creation. Furthermore, the team cohesion had a positive influence on knowledge creation. The study provided some implications that practitioners should consider the stress when they design jobs for team members and suggest them the way to manage their job stress when they work.

The Effects of Cultural Capital and Social Welfare Expenditure on the Elder's Subjective Happiness

  • The purpose of this study is to introduce policy and theoretical implications by analyzing affecting factors for the elder's happiness. For this study, we analyzed data using HLM. Data include a world value survey(hereafter, WVS) as personal level analysis data and also OECD's Social Expenditure Database(hereafter, SOCX) and database from the World Bank as national level analysis data. The subjects of personal level analysis were the elder who are over 65-years od age, and they were total 3,297 people, and while the subjects of national level analysis were total 9 OECD countries. For the data analysis, hierarchial linear model(HLM) analysis was done by using HML 7.0 program. As a result of analysis, First, for the elderly's happiness, they should improve self-disposition, members of social groups, and social class. Second, the old-age pension and the survivor's pension had no meaningful effect on the happiness. but it was found that self - disposition, social class, gender, and health status showed meaningful interaction effect according to old - age pension, survivor pension, per capita GDP, income inequality. This suggests that efforts to improve the happiness of the elderly should be made at the individual level and the national level at the same time.
