FMI based Real-time CPS Distributed Simulation Framework using OMG DDS middleware (OMG DDS 미들웨어를 이용한 FMI기반 실시간 CPS 분산 시뮬레이션 프레임워크)

  • To develop highly dependable CPS, M&S(modeling and simulation) is very important. It is not easy to model any CPS whole system in a single simulation tool because each simulation tool is optimized for modeling each different part of the CPS. The FMI is the standard for M&S between different simulation tools. The DDS is a communication middleware suitable for large-scale real-time data transmission. In this paper, we proposed FMI based CPS real-time distributed simulaton framework using DDS. To evaluate the performance of the proposed framework, we performed distributed simulation using IEEE HLA/RTI and OMG DDS middleware and measured and compared the execution time of the entire simulation. From the simulation results, we can confirm that the simulation execution time using DDS is at least 1.14 times faster compared to execution time using HLA/RTI.

Isorhamnetin Attenuates Staphylococcus aureus-Induced Lung Cell Injury by Inhibiting Alpha-Hemolysin Expression

  • Staphylococcus aureus, like other gram-positive pathogens, has evolved a large repertoire of virulence factors as a powerful weapon to subvert the host immune system, among which alpha-hemolysin (Hla), a secreted pore-forming cytotoxin, plays a preeminent role. We observed a concentration-dependent reduction in Hla production by S. aureus in the presence of sub-inhibitory concentrations of isorhamnetin, a flavonoid from the fruits of Hippophae rhamnoides L., which has little antibacterial activity. We further evaluate the effect of isorhamnetin on the transcription of the Hla-encoding gene hla and RNAIII, an effector molecule in the agr system. Isorhamnetin significantly down-regulated RNAIII expression and subsequently inhibited hla transcription. In a co-culture of S. aureus and lung cells, topical isorhamnetin treatment protected against S. aureus-induced cell injury. Isorhamnetin may represent a leading compound for the development of anti-virulence drugs against S. aureus infections.

Korean Red Ginseng Significantly Slows CD4 T Cell Depletion over 10 Years in HIV-1 Infected Patients: Association with HLA

  • We have shown that long-term intake of Korean red ginseng (KRG) delays disease progression in HIV-I infected patients. In the present study to investigate whether this slow progression was associated with protective human leukocyte antigen (HLA) alleles as well as with KRG-intake, we have performed clinical analysis of 31 HIV-1 infected patients who have been living for more than 10 years without any antiretroviral therapy. Average amount of KRG-intake over $130\;{\pm}16$ months was $4,797\;{\pm}4,921\;g$ and the annual decrease in CD4 T cell (AD) was $30\;{\pm}29{\mu}L$. We observed significant correlations among amount of KRG-intake, AD(r=-0.53, P < 0.01), and plasma HIV-1 RNA copy (r=-0.35, P < 0.05), along with a significant correlation between KRG-intake and HLA score AD(r=-0.49, P < 0.01), whereas there was no significant correlation between HLA score and AD or viral load. When the 31 patients were divided into 2 groups based on the amount of KRG-intake, the $AD(14/{\mu}L)$ in the 16 patients who had taken higher amounts of KRG was significantly less than that $(49/{\mu}L)$ in the 15 patients with a little or no KRG-intake (P < 0.01). These data indicate that KRG-intake sig­nificantly slows CD4 T cell depletion in HIV-1 infected patients.

  • The present investigation demonstrates HLA-DR and PGP9.5 double positive cells accumulate thymus cortical region in normal baboon thymus and baboon lung. But, these cells disappeared in thymus and lung of bronchopulmonary dysplasia (BPD) animals. 125d GC animal model is more suitable for BPD than 140d GC animal. Anti-bombesin antibody, 2A11 treated baboon recover normal level of HLA-DR positive cells from BPD animal. In addition, thymocytes show responsiveness for bombesin. These observation suggest that blocking BLPs protects a chronic lung injury by BPD and 2A11 is possible agent for passive therapy of BPD.

  • This paper proposes a framework to construct the synthetic battlefield based aviation engagement simulation model for the distributed system. The proposed framework has the synthetic battlefield in the HLA (High Level Architecture)/RTI (Run-Time Infrastructure) based distributed system to reflect environmental effects into the aviation engagement simulation model. In an aviation engagement, the environment affects weapon systems such as detection and movement. Therefore, environmental effects are required to be reflected in the simulation. However, former researches are inadequate for complex operations of weapon systems that are requirements of the engagement simulation. Thus, the construction of the engagement simulation system of which reflects environmental effects based on environmental data is still difficult. The main objective of this paper is to propose a framework to solve the difficulty and constructs an example system based on the proposed framework.

  • Recently, as weapon systems have been more diverse and complicated, the factors of risk increase in development. Consequently, demanding reduction of acquired costs and period increase. Under the acquisition environment, more efficiently to develop weapon system, the necessity of application of defense M&S from requirement phase is on the rise. As the importance of M&S is stressed under distributed environment, so the standard of M&S(HLA, SEDRIS, etc.) and the system engineering process, namely FEDEP(Federation Development & Execution Process) have been developed. In this paper using the 5 phase expression, we constructed underwater engagement simulation(UNES) that prototype to develop naval weapon system test bed which take up integrated architecture in HLA. we developed simulators according to FEDEP for expandability and described process applying FEDEP fur UNES development.

Expression of Major Histocompatibility Complex during Neuronal Differentiation of Somatic Cell Nuclear Transfer-Human Embryonic Stem Cells

  • Human pluripotent stem cells (hPSCs) such as human embryonic stem cells (hESCs), induced pluripotent stem cells, and somatic cell nuclear transfer (SCNT)-hESCs can permanently self-renew while maintaining their capacity to differentiate into any type of somatic cells, thereby serving as an important cell source for cell therapy. However, there are persistent challenges in the application of hPSCs in clinical trials, where one of the most significant is graft rejection by the patient immune system in response to human leukocyte antigen (HLA) mismatch when transplants are obtained from an allogeneic (non-self) cell source. Homozygous SCNT-hESCs (homo-SCNT-hESCs) were used to simplify the clinical application and to reduce HLA mismatch. Here, we present a xeno-free protocol that confirms the efficient generation of neural precursor cells in hPSCs and also the differentiation of dopaminergic neurons. Additionally, there was no difference when comparing the HLA expression patterns of hESC, homo-SCNT-hESCs and hetero-SCNT-hESCs. We propose that there are no differences in the differentiation capacity and HLA expression among hPSCs that can be cultured in vitro. Thus, it is expected that homo-SCNT-hESCs will possess a wider range of applications when transplanted with neural precursor cells in the context of clinical trials.

  • A dynamic model of direct-acting OHC valve train system has been used to determine the load conditions in the system. The modified equations for calculating the friction forces between cam and HLA, and at a camshaft bearing have been defined considering the lubrication conditions. Then, to understand the frictional characteristics in the system, a parameter study has been performed. As the results of the analysis, valve spring stiffness and preload have great effects on the friction in the system, but the effects of other parameters are negligible. So, how to design the valve train system with respect to the reduction of friction is to minimize the valve spring stiffness and preload in the limit of satisfying the dynamic constraints.

  • Reactive arthritis comprises a subgroup within infection-associated arthritides in genetically susceptible hosts. Researchers and clinicians recognize two clinical forms of reactive arthritis which occurs after genitourinary tract infection and after gastrointestinal tract infection. Chlamydia infection has been implicated as the most common agent associated with post-venereal reactive arthritis. Studies have proposed Shigella infection, Salmonella infection, or Yersinia infection as the microorganisms responsible for the post-dysenteric form. The human leukocyte antigen (HLA)-B27 antigen is the best-known predisposing factor. We report a case of HLA-B27-associated reactive arthritis after Salmonella enteritis at Pusan National University hospital.

  • 사람의 항 HLA 단일군 항체 생산의 선결 조건인 가장 효과적인 시험관내 면역 조건을 확립하기 위해, 사람의 혈중 임파구를 함원으로 하여 마우스의 대식세포, 흉선세포. 이들의 조건배지, 그리고 임파구 촉진인자 등을 포함한 14가지의 다양한 배양 조건에서 세포를 배양하였으며, 마우스의 비장세포와 사람의 혈중 임파구에서 각각 항체 생산을 유도하였다. 항체의 생성 여부는 면역효소법(I섬SA)으로 조사하였다. 마우스의 비장세포는 모든 조건에서 다량의 항체가 검출되었으며, 사람의 혈중 임파구 분화에는 마우스의 조건배지보다 PWM. LPS와 같은 임파구 촉진인자가 효과적임을 알 수 있었으며. 특히 allogenic MLC(Mixed Lymphocyte Culture)에 의해 임파구 분화유도에 유용한 물질이 생성됨을 알 수 있었다.
