Antioxidant Activity and Biological Properties in Extracts of Euonymus alatus (Thnub.) Sieb. (화살나무 추출물의 항산화 활성 및 생물학적 특성)

  • Antioxidant activity and biological properties in the MeOH extracts from different plant parts of Euonymus alatus (THNUB.) were measured by DPPH free radical scavenging ability and inhibition ability against xanthine/xanthine oxidase and proliferation in HL-60 cells. DPPH free radical scavenging activities in extracts of plant parts were high such as leaf, wing, root, seed and stem, respectively. The EtOAc fractions of plant parts were purified through LH-20 column chromatography and identified by GC/MS. LH-4 fraction and LH-5 fraction of leaf, stem and root showed stronger activities than other fractions in the inhibitor activity of DPPH and xanthine/ xanthine oxidase. $IC_{50}$ values of LH-4 fraction eluted from stem extracts showed such as 2.38 and 5.32 in DPPH and xanthine/ xanthine oxidase assay. Polyphenolic compounds were identified in purified LH-20 fractions showed highest $IC_{50}$ value in DPPH and xanthine/xanthine oxidase assay The activity of POD according to sampling time was high in root harvested in May and leaf harvested in September, respectively. The activity of SOD showed only in the extracts of stem in plant parts. SOD and POD in leaf were similar in the patterns of isozyme to those of stem. The purified extracts from Euonymus alatus (THNUB.) exerted inhibition ability of proliferation in HL-60 cells.

Chemical Characteristics and Biological Activities of Herbimycin A and Dihydroherbimycin A Produced by a Soil Isolate Streptomyces sp. AO-0511 (Streptomyces sp. AO-0511이 생산하는 Herbimycin A 및 Dihydroherbimycin A의 이화학적 특성 및 생물 활성)

  • A streptomycete strain was isolated from the soil samples from Korea. The chemotaxonomy and 16S rDNA sequencing confirmed that the strain belonged to the genus Streptomyces and we named it Streptomyces sp. AO-0511. Two antibiotics, herbimycin A and dihydroherbimycin A produced by this strain were tested for their physico-chemical and biological characteristics. Both compounds were stable under acidic pH. Dihydroherbimycin A was more heat-stable and polar compared with herbimycin A. Only weak antibacterial activities were detected against Bacillus subtilus ATCC 6633 and Micrococcus luteus ATCC 9341. However, herbimycin A and dihydroherbimycin A showed strong inhibitory activities on lung cancer cells (A549 cells) and leukemia cells (HL-60). The cytotoxicity was determined using L5178Y and P388 cell lines. The results showed that herbimycin A and dihydroherbimycin A had lower toxic effects on the cells compared with the standard compounds, comptothecin and cyclosporin A. Therefore, both compounds could be good candidates for the development of new anticancer drugs.

A Comparison of Functional Fragrant Components of Cymbidium (Oriental Orchid) Species (기능성 지표물질 확인을 위한 동양란 심비디움(Cymbidium) 향기 성분 비교 분석)

  • We analyzed the functional fragrant components of three species of Cymbidium oriental orchids using gas chromatography-mass spectrometry (GC/MS). For the comparative analysis, C. goeringii 'Minchunran', 'Jugeumhwa', C. forrestii 'Chwigae', 'Songmae', 'Yongja', and C. faberi 'Choemae', 'Namyangmae', 'Hwaja' were investigated. Major fragrant components detected by GC/MS were selected on the basis of more than 3% value according to the analysis of peak area (%). We found that ${\alpha}$-bergamotene, which has a cytotoxic effect on breast cancer, cervical cancer, and glioblastoma, and nerolidol, which induces apoptosis of human hepatoma cells (HepG2), inhibits the growth of Streptococcus mutans and babesiosis, and has antibacterial properties, are common substances produced by C. goeringii L. Nerolidol and ${\beta}$-bisabolene, which is cytotoxic and suppresses the growth of malignant melanoma cells (B16-F10), HepG2, and leukemia cells (HL-60, K562), are major substances in C. forrestii R. Furthermore, ${\alpha}$-pinene, which inhibits the growth of gliobastoma cells (SF-767) and inhibits the anti-inflammatory action of hepatoma cells (BEL-7402); 1,8-cineole, which is a potential therapeutic agent for the treatment of gastric ulcers; and 1,3,7-octatriene, which functions as a pheromone, are the most common substances in C. faveri R. Thus, substances identified as major fragrant components in oriental orchid species have multiple beneficial applications in human health. This research forms the basis for further studies of the roles of major fragrant components in oriental orchids.

The Effects of Cysteamine on the Radiation-Induced Apoptosis (방사선조사에 의해 발생되는 세포고사에 대한 Cysteamine의 효과)

  • Purpose : To Investigate the pathways of radiation induced apoptosls and the effect of cysteamine (${\beta}$-mercaptoethyiamine), as a radioprotector, on it. Materials and Methods : HL-50 ceils were assigned to control, irradiated, and cysteamlne (1 mM, 10mM) pretreated groups. Irradiation was given In a single fraction of 10 Gy (6 MV x-ray) and cysteamine was administered 1 hour before irradiation. The activities of caspase-8 were measured in control and irradiated group to evaluate its relation to the radiation Induced apoptosis. To evaluate the role of cysteamine In radiation Induced apoptosis, the number of viable cells, the expression and activity of caspase-3, and the expression of poly (ADP-ribose) polymerase (PARP) were measured and compared after irradiating the HL-60 celis with cysteamine pretreatment or not. Results : The intraceliular caspase-8 activity, known to be related to the death receptor induced apoptosis, was not affected by irradiation(p>0.05). The number of viable cells began to decrease from 6 hours after irradiation (p>0.05), but the number of viable cells In 1 mM cysteamine pretreated group was not decreased after irradiation and was similar to those in the control group. In caspase-3 analyses, known as apoptosis executioner, its expression was not different but its activity was Increased by irradiation(p>0.05). However, this Increase of activity was suppressed by the pretreatment of 1 mM cysteamine. The cleavage of PARP, thought to be resulted from caspase-3 activation, occurred after irradiation which was attenuated by the pretreatment of 1 mM cysteamine. Conclusion : These results show that radiation induced apoptotic process is somewhat different from death receptor induced one and the pretreatment of 1 mM cysteamine has a tendency to decrease the radiation-induced apoptosis in HL-60 cells.

The activity of antioxidants and suppression of cancer cell proliferation in extracts of Orostachys japonicus A. Berger (바위솔 추출물의 항산화활성 및 암세포 증식억제)

  • The antioxidative activities and biological properties in the EtOAc extracts and purified extracts of Orostachys japonicus were measured by assay of DPPH, xanthine/xanthine oxidase and mammalian cells(2-12). Scavenging of DPPH radical and inhibition of xanthine/xanthine oxidase of EtOAc extracts were showed the highest activity in the arable land and in September. The S-4 fraction showed the highest activity among the silica-gel column chromatography fractions. LH-4 fraction showed higher activity than the other fractionsins in assay of DPPH and xanthine/xanthine oxidase. Fatty acids and phenolic compounds were identified by GC/MS and main compounds were 1,2,3-benzenetriol, alpha-androsta-7,14-diene in LH-4 fraction. The activities of POD and SOD in samples havested on different habitats were high such as arable land> intermountain> seashore. That of POD and SOD in crude extracts of late stage were higher than early stage. Isozyme bands of crude extracts samplinged in all habitats and all growing stages showed two bands and the signal intensity showed strongly according to passage of growing stage. The purified extracts of LH-4 fraction showed excellent inhibition effect in proliferation of HL-60 cells and markedly suppressed colony formation in mouse fibroblast cells. Dose response between partially purified extracts(400ppm) and negative control did not produced statistically significant reduction in colony formation.

The Antimutagenic and Anticancer Effect of Taxus cuspidata Extracts (주목 추출물의 발암 억제효과 및 암세포에 미치는 영향)

  • This study was Performed to determine the effects of antimutagenicity and anticancer of Taxus cuspidata produced in Korea. Extracts of Taxus cuspidata were obtained from leaves, barks and roots. All the samples tested had no effects on the mutagenicity by Ames test using Salmonella typhimurium TA98 and TA100 or rec-assay using Bacillus subtilis Hl7 and M45 strains. However the treatment of 50$\mu\textrm{g}$/plate of Taxus cuspidata extracts showed strong antimutagenicity with 98% inhibition against TA100 induced by MNNG and with 98% inhibition against TA98 and TA100 induced by 4NQO whereas 73~89% and 16~60% antimutagenic effect were shown against both strain induced by Trp-P-1 and $Benzo(\alpha)pyrene,$ respectively. The treatment of $0.5$\mu\textrm{g}$/{\mu}\ell$ root extracts had the highest cytotoxicity with 90% against liver cancer cell, Hep 3B, followed by bark extracts(87%) and leave extracts(72%), whereas $0.5$\mu\textrm{g}$/{\mu}\ell$ treatment of Taxus cuspidata extract had only 22~36% cytotoxicity on human normal liver cell WRL 68.

  • A broad spectrum of antibiotic action was studied with Centipedin purified from centipede Scolopendra subspinipes multilans L. Koch aganist gram-positive, gram-negative bacteria and fungi. The minimal inhibitory concentrations(MICs) were determined in liquid medium. The significant antibiotic activity was obtained aganist gram-negative Klebsiella pneumoniae ATCC 8308 responsible for causing infection at lung and intestine. The MIC value against Klebsiella pneumoniae ATCC 8308 was $2{\mu}g/ml$, and this Centipedin was active against Proteus vulgaris NRRL B-123. In addition, it has been shown that Centipedin blocks procaryotic RNA transcription and a little of DNA replication system in vitro. Centipedin did not exhibit any significant cytotoxicity against animal cells such as human blood leukemia (HL-60) and mouse B lymphocyte myeloma cell.

  • Essential oils from Needl Juniperrus seed and trunk (Juniperrus rigida Sieb.) and Olibanum resin (Boswellia carteii Birew) were extracted by a supercritical fluid extraction system (SFE) and immune activity of each essential oils were observed. The immune activities of each essential oil were compared. Essential oil from Olibanum resin enhanced the growth of human immune T cell up to 1.33 times, compared to control group. Each essential oils showed the potent inhibitory effect on the human cancer cell lines, and increased the secretion of cytokines, IL-6 and $TNF-{\alpha}$ from human B cell as well as the growth of human immune cells.

Comparative Evaluation of Antioxidant Activities of Ethanol Extracts and Their Solvent Fractions Obtained from Selected Miscellaneous Cereal Grains (잡곡 유래 에탄올 추출물 및 이의 유기용매 분획들의 항산화 활성 비교평가)

  • To examine the antioxidant activities of 11n selected miscellaneous cereal grains (proso millet, yellow glutinous proso millet, hwanggeumchal sorghum, glutinous sorghum, white glutinous sorghum, yellow glutinous foxtail millet, nonglutinous foxtail millet, green glutinous foxtail millet, golden foxtail millet, barnyard millet, and adlay), the free radical-scavenging activities of 80% ethanol extracts of the individual grains were investigated using 1,1-diphenyl-2-picryl-hydrazl (DPPH) and 2,2'-azino-bis (3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) methods. The ethanol extracts of hwanggeumchal sorghum, glutinous sorghum, and barnyard millet grains exhibited more potent free radical-scavenging activities as compared to the other grains. When these three ethanol extracts were sequentially fractionated with n-hexane, methylene chloride, ethyl acetate, and n-butanol, the majority of the antioxidant activities were detected in the ethyl acetate and butanol fractions in which phenolic ingredients were abundant. The ethyl acetate and butanol fractions of hwanggeumchal sorghum and the ethyl acetate fraction of glutinous sorghum showed higher antioxidant activity than that of ${\alpha}$-tocopherol. Both ferric thiocyanate (FTC) and thiobarbituric acid (TBA) methods demonstrated that these organic solvent fractions could inhibit lipid peroxidation. The ethyl acetate fractions from hwanggeumchal sorghum, glutinous sorghum, and barnyard millet grains could suppress tertiary-butyl hydroperoxide (TBHP)-induced apoptotic events, including sub-G1 peaks, ${\Delta}{\Psi}m$ loss, activation of caspase-9 and caspase-3, and cleavage of PARP and lamin B, in human HL-60 cells. These results show that the grains of hwanggeumchal sorghum (Sorghum bicolor L. Moench cv. Hwanggeumchalsusu), glutinous sorghum (Sorghum bicolor L. Moench cv. Chalsusu), and barnyard millet (Echinochloa esculenta) possess efficient antioxidant activity, which could protect cells from oxidative stress-mediated cytotoxicity.

Protective Effects of a Herbal Composition (HemoHIM) Against Apoptosis Induced by Oxidative Stress of Hydrogen Peroxide (과산화수소의 산화적 스트레스로 유도된 Apoptosis에 대한 생약복합조성물(HemoHIM)의 방호효과 평가)

  • In our previous study, a novel herb mixture (HIM-I) of Angelica gigas radix, Cnidium officinale rhizoma, and Paeonia japonica radix was developed to protect the intestinal and immune systems and promote its recovery against radiation damage. A new herbal composition (HemoHIM) with the high immune modulating activity was developed from HIM-I. HIM-I was fractionated into ethanol fraction (HIM-I-E) and polysaccharide fraction (HIM-I-P). And HemoHIM was prepared by adding HIM-I-P to HIM-I. HemoHIM showed more effective than HIM-I in immune modulation as well as radioprotection. The present study is designed to investigate the protective effects of HIM-I, HIM-I-P, and HemoHIM on hydrogen peroxide $(H_2O_2)$ induced apoptosis of human promyelocytic leukemia (HL-60) cells. It was shown that $H_2O_2$ treatment reduced the viability of cells, and increased appearance of DNA ladders, hypodiploid (subG1) cells, and phosphatidylserine translocation level. Pretreatment of HemoHIM significantly reduced the cytotoxic effect induced by $H_2O_2$, associated with reducing the translocation of phosphatidylserine, hypodiploid cells and DNA ladders. HemoHIM appeared to be more protective than HIM-I against $H_2O_2$ induced apoptosis whereas, it exhibited similar activity to HIM-I-P. These results indicated that HemoHIM might be an useful agent for protection against oxidative stress $(H_2O_2)-induced$ apoptosis as well as immune modulation, especially since it is a relatively nontoxic natural product.
