  • 2-Hydroxycinnamaldehyde is an active compound isolated from the Stem Bark of Cinnamomum cassia, a traditional oriental medicinal herb, which has been shown to inhibit tumor cell proliferation. Anticancer Effects of Leek Kimchi on Human Cancer Cells

  • The anticancer effects of leek (buchu in Korean) kimchi were evaluated in the human cancer cells: AGS gastric adenocarcinoma cells, HT-29 human colon adenocarcinoma cells and HL-60 leukemia cells. The leek kimchi (fermented for 6 days at 15$^{\circ}C$) was fractionated into 7 groups: methanol extract, hexane extract, methanol soluble extract MSE), dichloromethane (DCM) fraction (fr.), ethyl acetate fr., butanol fr. and aqueous fr. Most of the leek kimchi tractions inhibited the growth of AGS and HT-29 cancer cells in a dose dependent manner. In particular, the DCM fr. showed the highest inhibitory effect among the tractions. Treatment with the DCM fr. (0.1 mg/mL) reduced the survival rates of AGS and HT-29 cancer cells to 19% and 37% of the controls, respectively. Moreover the DCM fr. of the leek kimchi arrested G2/M phase in the cell cycle and induced apoptosis in HL-60 human promyelocytic leukemia cells. These results indicate that the leek kimchi exerted an anticancer effect on those human cancer cells, and that the DCM fr. arrested G2/M phase in the cell cycle and induced apoptosis in the leukemia cells.

Hydroxyl Radical-Mediated Commitment of HL-60 Cells to Differentiation: Modulation of Differentiation Process by Phosphodiesterase Inhibitors

  • This report shows that hydroxyl radical, generated by a Fenton reaction involving adenosine $5'-diphosphate/Fe^{2+}$ complex ($5-15\;{\mu}M$) and $H_2O_2$ ($2\;{\mu}M$), induced differentiation of HL-60 cells in a dose- and time-dependent manner. This is evidenced by the increases in 12-O-tetradecanoylphorbol 13-acetate- and fMLP-stimulated superoxide production capability. The cells exposed to hydroxyl radical for defined periods (24∼96 hr) continued to differentiate even after the hydroxyl radical generating system had been removed. The differentiated cells displayed fMLP-stimulated calcium mobilization and increased expression of myeloid-specific antigen CD11b and CD14. The extent of the differentiation was markedly reduced by desferrioxamine ($100\;{\mu}M$), dimethylthiourea (5 mM), N,N'-diphenyl-1,4-phenylenediamine ($2\;{\mu}M$), and N-acetyl-L-cysteine (5 mM). The induction of differentiation by hydroxyl radical was enhanced by 3-isobutyl-1-methylxanthine ($200\;{\mu}M$) and Ro-20-1724 ($8\;{\mu}M$), and inhibited by dipyridamole (2 ${\mu}M$). These results suggest that hydroxyl radicals may induce commitment of HL-60 cells to differentiate into more mature cells of myelomonocytic lineage through specific signal-transduction pathway that is modulated by phosphodiesterase inhibitors.

애기달맞이꽃(Oenothera laciniata Hill) 추출물의 생리활성 탐색 (Biological Activities of Oenothera laciniata Extracts (Onagraceae, Myrtales))

  • 본 실험에서는 애기달맞이꽃(Oenothera laciniata Hill)를 80% EtOH로 추출하고 추출물을 극성에 따른 용매 분획을 실시하여 분리된 분획의 총 페놀화합물 함량과 DPPH radical 소거, xanthine oxidase 저해활성, superoxide 소거활성 및 nitric oxide 소거활성 및 세포독성 등의 생리 활성을 측정하였다. 본 연구를 통해 애기달맞이꽃은 산소 유리기의 자유기를 소거할 수 있는 우수한 생리 활성물질을 다수 함유하고 있음을 확인 할 수 있었으며 분획 레벨에서의 활성이 대조구 성분 못지않게 우수하게 나타났다. 또한 총 폴리페놀 함량과 세포독성이 가장 높게 나타난 EtOAc 분획물을 농도별로 HL-60세포에 처리한 결과, 세포성장 억제는 농도 의존적으로 나타났으며, 따라서 세포성장 억제효과가 apoptosis 유도에 의한 것이지를 확인하고자, apoptosis 유도에 의하여 나타나는 현상의 하나인 DNA 단편화 현상을 비롯하여 세포 형태학적 변화 및 유동세포분석기를 통한 DNA content을 측정하였다. 그 결과, 애기달맞이꽃 추출물에 의한 세포증식 억제효과는 apoptosis유도에 의해 세포사멸이 일어남을 확인할 수 있었다.

저령의 암세포증식 억제효능 (Activity of the Extracts from Polyporus umbellatus as a Apoptosis Inducer)

  • 저령의 에틸아세테이트 추출물에서 4개의 분획을 제조한 후 백혈병 세포인 K-562, L-1210, HL-60와 U-937 세포의 세포생존 억제효능을 관찰하였다. Fr. 2는 L-1210, HL-60 세포 억제효능이 미비하였고, Fr. 3은 4종의 백혈병 세포주에 대하여 세포생존 억제효능이 있었다. 이 효능은 L-1210 세포에 대하여 가장 강하게 나타났다. 세포사멸 작용을 증명하는 DNA분절 현상을 L-1210 세포에서 관찰하였을 때, fr. 3을 30 $\mu\textrm{g}$/mL, 100 $\mu\textrm{g}$/mL로 처리한 군에서 만 DNA분절 현상이 관찰되었다. Fr. 3에 대한 DNA합성 억제능을 [$^3$H]thymidine incorporation test로 관찰하였을 때 50 $\mu\textrm{g}$/mL, 100 $\mu\textrm{g}$/mL로 처리한 군에서만 [$^3$H]thymidine incorporation이 저하되었다. 결론적으로 저령에는 백혈병세포인 L-1210과 HL-60의 세포생존 억제성분이 포함되어 있으며, 억제작용은 L-1210에 가장 뚜렷이 나타내었다. Fr. 3은 L-1210 세포에 대하여 DNA 합성능을 저해시켰고, Fr. 3가 L-1210 세포에 대하여 세포생존 억제작용을 나타내는 것은 계획된 세포사멸현상인 apoptosis에 의한 것으로 사료된다. 따라서 fr. 3에 대한 성분 연구가 요구되어진다.

Sasa quelpaertensis Leaf Extracts Induce Apoptosis in Human Leukemia HL-60 Cells

  • In east Asia, the leaves of various Sasa species have been used in folk medicine for centuries. The effects of the methanolic extract and its subsequent fractions derived from the leaves of Sasa quelpaertensis Nakai on cell proliferation in human leukemia HL-60 cells were evaluated. The ethyl acetate fraction of this extract (ESQL) significantly reduced cell viability in a dose-dependent manner ($0-250\;{\mu}g/mL$). ESQL ($IC_{50}=24.8\;{\mu}g/mL$) exhibited growth inhibition comparable to the main constituent of green tea, epigallocatechin ($IC_{50}=26.2\;{\mu}g/mL$), which was used as a positive control. ESQL treatment induced apoptosis, which was confirmed by the presence of nuclear condensation and annexin V-staining. These results demonstrate that ESQL contains chemopreventive phytochemicals that may be useful in neutraceutical applications.

  • In the present study, the protective effect of Scutellaria baicalensis against DNA damage in HL-60 cells exposed to $^{60}$ Co ${\gamma}$-rays was evaluated using micronuclei formation and alkaline single cell gel electrophoresis (SCGE, comet assay). Induction of the Intrinsic Apoptotic Pathway by 3-Deazaadenosine Is Mediated by BAX Activation in HL-60 Cells

  • 3-Deazaadenosine (DZA), a potent inhibitor of S-adenosylhomocysteine hydrolase, was previously proposed to induce intrinsic apoptosis in human leukemic cells. In the present study, we analyzed the mechanism underlying the DZA-induced intrinsic apoptotic pathway. DZA activated typical caspase-dependent apoptosis in HL-60 cells, as demonstrated by an accumulation of hypo-diploidic cells, the processing of multiple procaspases and an inhibitory effect of z-VAD-Fmk on this cell death. During DZA-induced apoptosis, cytochrome c (cyt c) was released into the cytosol. This was neither prevented by z-VAD-Fmk and nor was it associated with the dissipation of mitochondrial membrane potential (${\Delta}{\Psi}_m$). Prior to the release of cyt c, BAX was translocated from the cytosol to mitochondria and underwent oligomerization. Finally, the overexpression of BCL-XL protected HL-60 cells from apoptosis by blocking both the cyt c release and BAX oligomerization. Collectively, these findings suggest that DZA may activate intrinsic apoptosis by stimulating BAX activation and thereby the release of cyt c.

저혈청 배지에서 인간 전골수세포(HL-60)를 이용한 tPA 생산과 세포사멸기작에 관한 연구 (The Production of Tissue Type-Plasminigen Activator and Mechanism of Cell Death from Human Promyelocytes(HL-60) in Low Serum Containing Medium)

  • HL-60 was cultivated to produce tPA (tissue-type plasminogen activator) and study the mechanism of cell death. Maximum cell density and tPA production were obtained as $5.27{\times}10^6$ cells/ml and 324ng/ml, respectively under perfusion cultivation. tPA production was enhanced to 420ng/ml in adding 160nM of phorbol ester. The cells were gradually differentiated to granulocytes rather than proliferation. By Fluorescent microscope, apoptosis was prevailed except the death phase and in high agitation speed, but necrosis was prevailed in thawed cells and during the latter periods of the cultivation. It was also proved that tPA was most produced in apoptosis. To obtain higher tPA productivity, the cells must be maintained in apoptosis, not necrosis phase when the cells were dying.
