HL-60 사람 백혈병 세포에서 camptothecin이 DNA topoisomerase l과 c-myc의 발현에 미치는 영향 (Effects of camptothecin on the expression of DNA topoisomerase I and c-myc in HL-60 human leukemia cells)

  • Camptothecin (CPT) is an antitumor alkaloid that has been isolated from the Chinese tree, Camptotheca acuminata. The cytotoxicity of CPT has been correlated to its inhibition of DNA topoisomerase (Topo) I by stabilizing drug-enzyme-DNA “cleavable complex" resulting in DNA single-strand breaks and DNA-protein crosslinks. This studies were designed to elucidate whether CPT regulates Topo I mediated by CPT in DNAs containing c-myc protooncogene. We have conducted experiments on Topo I purification, pUC-MYC I cloning and Topo I assay using electrophoresis, quantitative RT-PCR and Northern blotting techniques. CPT ingibited the relaxation activity of Topo I in pUC19 DNA at various concentrations (1-1000 $\mu$M), while it enhanced the cleavage of Topo I in the pUC-MYC I by forming a cleavable complex at relatively high concentrations (100-1000 $\mu$M). In HL-60 cells treated with CPT, the expression of c-myc gene was decreased over that in the control group with no changes in the expression of Topo I mRNA. Our results suggest that Topo I is the target of CPT cytotoxicity but it does not affect Topo I extression, and the suppression of c-myc mRNA expression by CPT is due to c-myc damage resulted from formation of a cleavable complex with CPT. 인간 골수성 백혈병 세포에서 Flt-3 수용체 리간드에 의한 CD11c 발현의 증가 (Up-regulation of CD11c Expression on Human Acute Myelogenous Leukemia Cells by Flt-3 Ligand)

  • CD11c와 CD80 및 CD86과 같은 보조 수용체는 주로 수지상 세포에서 발현되는 세포 표지 인자이다. 본 연구에서는 KG-1, HL-60, NB4 및 THP-1 세포와 같은 여러 종류의 백혈병 세포를 이용하여 이들 세포에 재조합 Flt-3 리간드를 처리하였을 때 수지상 세포의 표면 인자인 CD11c의 발현에 어떠한 변화가 있는가를 조사하였다. KG-1 세포뿐만 아니라 NB4세포와 HL-60 세포에서도 Flt-3 수용체가 발현됨을 확인하였으나 THP-1 세포에서는 이들 수용체가 발현되지 않았다. KG-1 세포를 Flt-3 리간드나 granulocyte macrophage-colony stimulating factor (GM-CSF)와 tumor necrosis factor (TNF)-$\alpha$를 섞은 배양액에서 배양하였을 때 세포 증식은 억제되었으며 CD11c 발현은 현저히 증가되었다. 그러나 Flt-3 리간드를 처리한 KG-1세포에서는 GM-CSF와 TNF-$\alpha$를 처리한 세포에서와는 다르게 major histocompatibility complex (MHC)-I 및 MHC-II의 발현은 증가되지 않았다. Flt-3 리간드는 HL-60 세포와 NB4 세포의 CD11c 발현도 증가시켰으나 THP-1 세포에서는 아무런 영향이 없었다. CD11c의 발현과 비교하여 CD11b의 발현은 Flt-3 리간드에 의하여 KG-1 세포에서는 약하게 증가하였으나 NB4 세포와 HL-60 세포에서는 증가되지 않았다. KG-1 세포를 Flt-3 리간드로 처리하였을 때 extracellular signal-regulated kinase-1/2 (ERK-1/2)와 p38-mitogen-activated protein kinase (p38-MAPK)의 단백질 인산화가 증가되었으며 Flt-3 리간드에 의한 CD11c 발현의 증가는 MEK의 억제제인PD98059에 의하여 사라짐을 확인하였다. 본 연구 결과는 Flt-3 수용체 리간드의 처리에 의하여 $CD34^+$ myelomonocyte분화 단계인 KG-1 세포와 promyelocyte 분화 단계의 백혈병 세포에서 수지상 세포와 유사한 세포 형으로 분화된다는 것을 보였고 Flt-3 수용체 리간드에 의한 이들 백혈병 세포의 수지상 세포유사 세포로의 분화는 ERK-1/2의 활성화에 의하여 일어날 수 있음을 보여 준다.

In Vitro Antioxidant and Antiproliferative Activities of Novel Orange Peel Extract and It's Fractions on Leukemia HL-60 Cells

  • In the present work, novel orange peel was extracted with 100%EtOH (ethanol) and fractionated into four fractions namely F1, F2, F3, F4 which were eluted from paper chromatographs using 100%EtOH, 80%EtOH, 50%EtOH and pure water respectively. The crude extract and its four fractions were evaluated for their total polyphenol content (TPC), total flavonoid content (TFC) and radical scavenging activity using DPPH (1,1-diphenyl-2-picrylhydrazyl) assay. Their cytotoxic activity using WST assay and DNA damage by agarose gel electrophoresis were also evaluated in a human leukemia HL-60 cell line. The findings revealed that F4 had the highest TPC followed by crude extract, F2, F3 and F1. However, the crude extract had the highest TFC followed by F4, F3, F2, and F1. Depending on the values of $EC_{50}$ and trolox equivalent antioxidant capacity, F4 possessed the strongest antioxidant activity while F1 and F2 displayed weak antioxidant activity. Further, incubation HL-60 cells with extract/fractions for 24h caused an inhibition of cell viability in a concentration-dependent manner. F3 and F4 exhibited a high antiproliferative activity with a narrow range of $IC_{50}$ values ($45.9-48.9{\mu}g/ml$). Crude extract exhibited the weakest antiproliferative activity with an $IC_{50}$ value of $314.89{\mu}g/ml$. Analysis of DNA fragmentation displayed DNA degradation in the form of a smear-type pattern upon agarose gel after incubation of HL-60 cells with F3 and F4 for 6 h. Overall, F3 and F4 appear to be good sources of phytochemicals with antioxidant and potential anticancer activities.

The Cytotoxicity of Eutigosides from Eurya emarginata Against HL-60 Promyelocytic Leukemia Cells

  • Two phenolic glucosides, eutigoside Band eutigoside C were isolated from the fresh leaves of Eurya emarginata. These two phenolic glucosides exerted a significant inhibitory effect on the growth of HL-60 promyelocytic leukemia cells. Furthermore, when the HL-60 cells were treated with eutigoside C, several apoptotic characteristics such as DNA fragmentation, morphologic changes, and increase of the population of sub-G1 hypodiploid cells were observed. In order to understand the mechanism of apoptosis induction by eutigoside C, we examined the changes of Bcl-2 and Bax expression levels. The eutigoside C reduced BcI-2 protein and mRNA levels, but slightly increased Bax protein and mRNA levels in a time-dependent manner. When we examined the activation of caspase-3, an effector of apoptosis, the eutigoside C increased the expression of active form (19-kDa) of caspase-3 and the increase of their activities was demonstrated by the cleavage of poly (ADP-ribose) polymerase, a substrate of caspase-3, to 85-kDa. The results suggest that the inhibitory effect of eutigoside C from E. emarginata on the growth of HL-60 appears to arise from the induction of apoptosis via the down-regulation of BcI-2 and the activation of caspase.

세포 주기 변화에 따른 Toxoplasmu gondii의 침투 양상 (Cell cycle-dependent entry of Toxoplasma gondii into synchronized HL-60 cells)

  • Toxoplasma gondii를 HL-60 세포와 함께 in vitro 배양시 Toxopsasma 침투 정도가 각 세포에서 균일하지 않으므로 세포의 주기에 따른 일정 phase가 그 침투에 좋은 환경을 제공할 것이라는 추론으로 HL-60세포 주기를 동시화(synchronization)하여 각 stage에서 변화를 관찰하였다. 동시화는 과량의 thymidine이 DNA 합성을 억제함을 이용하여 2mM thymidine을 10시간 간격으로 각 24, 18시간 동안 처리하여 (double thymidine block method) S (synthetic) phase를 진행하는 세포를 얻었고 이후 30시 간 동안 13회의 간격을 두고 $5{\times}10^6/ml$의 Toxoplasma를 첨가하여 1시간 동안 배양하였다. 숙주세포의 동시화 정도는 (1) 3H-thymidine의 표지량 (2) mitotic index 측전 및 (3) 세포수의 증가를 통해 확인하였다. Toxoplasma의 침투 정도는 S phase중에서도 배지에서 thymidine을 제거한 후 3시간 경과시가 그 전후에 비해 6배 이상 높았으며 특히 이 시기는 DNA 합성이 최고가 되는 점과 일치하였다. 침투된 Toxoplasma 수의 변화 외에 세포의 모양도 상당한 변화가 있었고 이는 19시간 후 2번째 S phase에서도 약하나마 관찰되었다 실험 결과를 통해 특정 약 1시간 동안 일어나는 어떤 세포의 변화가 Toxoplasma 침투에 중요한 역할을 한다는 것을 알 수 있었다. 이는 원충 기생충의 숙주세포 흡착과 interiorization과정에 receptor가 관련되고 몇 receptor는 세포주기에 따라 발현이 조절되는 사실로부터 $G_1/S$ 경계부터 3시간째에 발현되면서 Togopzasma를 유인하는 receptor molecule의 존재 가능성을 시사하였다.

Echinacea angustifolia 메탄올 추출물의 암세포 증식억제 및 항산화 효과 (Antiproliferative and Antioxidative Activities of Methanol Extracts of Echinacea angustifolia)

  • Echinacea angustifolia의 부위별(꽃봉오리, 잎, 줄기 및 뿌리) 메탄을 추출물의 암세포(HepG2, 3LL, HL60, L1210)를 대상으로 한 항암 활성과 전자공여능을 검색한 결과는 다음과 같다. 1. 에키네시아 메탄을 추출물의 간암세포인 HepG2 cell 대한 MTT assay는 농도의존적으로 세포독성 효과가 증가하였으며, 인간유래 백혈암세포인 HL60 cell의 경우에 잎과 뿌리 추출물은 저농도에서부터 독성효과가 컸고, 줄기와 꽃봉오리는 저농도에서는 독성효과가 낮았으나 고농도로 갈수록 독성효과가 커짐을 알 수 있었다 폐암세포인 3LL cell과 마우스 유래 백혈암세포인 L1210 cell에 대한 경우는 세포독성효과가 없었다. 2. Hemacytometer에 의한 HepG2 cell의 암세포 성장에 미치는 효과는 배양기간이 증가함에 따라 농도의존적으로 증식억제 효과가 증가되었다. 3. HepG2 세포주의 형태학적 변화에서 대조군은 암세포가 조밀하게 중첩되어 증식되었으나 시료를 0.5 mg/mL 이상의 농도로 첨가하였을 때 세포의 결속력이 감소되어 세포주위가 흐트러지고 세포가 사멸된 것을 관찰 할 수가 있었다. 4. 전자공여능의 수준은 부위에 따라 최고의 EDA를 나타내는 농도가 달랐으며, 뿌리와 줄기부위는 저농도에서도 매우 높은 전자공여능을 나타내었다.

  • $^1H$-NMR spectroscopy was used to detect apoptosis in HL-60 cells in vitro. The relationship between cell apoptosis and NMR data was validated by the flow cytometry assay. To evaluate the NMR apoptosis results, the ratio of methylene and methyl groups caused by lipids was used. In addition, an identical analysis was applied to HepG2 cells. Detection of apoptotic cell death by NMR spectroscopy was oserved.

Binding of Galectin-1 to Neutrophils Enhanced by Activation

  • Human polymorphonuclear neutrophils undergo diaphoresis after a selectin-mediated rolling on the endothelial cells of the blood vessel wall. Extravasation is believed to be an integrin-mediated process. Galectin-1 is a small dimeric beta-galactoside-binding protein synthesized by the endotherial cells and present in the perivascular connective tissue. In this study we suggest the possible role of galectin-1 in extravasation of the activated neutrophils. MAL lectin binding study showed, that f-MetLeuPhe-activated neutrophils decrease surface sialylation and increase galectin-1 binding via exposure of new galectin-1 binding sites. Desialylated HL-60 cells also show the same decrease in MAL binding and increase in galectin-1 binding, an increase which was not observed in the presence of lactose. Galectin-1 blotting analysis detected two possible major ligands (approximately 120 and 160 kDa) of galectin-1 from the desialylated HL-60 cell lysates.

  • In a continuous effort to develop novel anticancer agents we newly synthesized and evaluated the antitumor activity of nucleoside analogues. One analogue, 4-[2-Chlor-6-(3-iodo-benzylamino)-purin-9-yl]-2,3-dihydroxy-cyclopentanecarboxylic acid methylamide (LJ-331), has been shown to exert a potent inhibition of human cancer cell growth in vitro including human lung (A549), stomach (SNU-638) and leukemia (HL-60) cancer cells. Following mechanism of action study revealed that LJ-331 induces cell cycle arrest at the G$_1$ phase in HL-60 cells and evokes apoptotic phenomena such as an increase in DNA ladder intensity and chromatin condensation by a dose-and time-dependent manner.   • We have previously shown that 2,4,3',5'-tetramethoxystilbene (TMS), a synthetic trans-stilbene analogue, is one of the most potently selective inhibitor of human cytochrome P450 1B1 in vitro and in vivo. In the present studies, the apoptotic effects of TMS were investigated in HL-60 cells. The effects of TMS on the proliferation of HL-60 cells were determined with MTT assay. TMS exhibited cytotoxicity with an $IC_50$ value of 37 nM. Cotreatment with TMS and etoposide, a well-known anticancer drug significantly enhanced the cytotoxicity. (omitted)
