Phylogenetic ANalysis of Hepatitis G Virus by Group-Specific Sequences in the 5-Untranslated Region (5'-UTR 영역의 그룹특이적 염기서열에 의한 HGV의 계통분석)

  • The nucleotide sequences of the 5'-untraslated region(5'-UTR) of Hepatitis G virus(HGV) from sera of Korean patients were determines. When compared to the previously reported isolates, the Korean isolates have higher sequence homology with the Japanese isolates indicating the geographic distribution of HGV variants. Interestingly, three discrete regions which are highly conserved among HGV isolates from the same geographical area, thus could be applied to distinguish HGV isolates from the different areas were noticed in the 5'-UTR. Based on the sequences of these group-specific regions, twenty four different HGV isolates could be classified into 5 groups. By using the group-specific regions, inconsistency in HGV typing when based on the different regions of HGV could be solved.

Genetic Analysis of the Envelope Region of Hepatitis G Vrius isolated from Korean Hepatitis Patients (한국인 간염환자에서 분리한 G형 감염바이러스(HGV)의 외피영역의 유전적 다양성 분석)

  • The genetic of a recently described virus, hepatitis G virus(HGV) was investigated. HGV envelope 1 (E1) nucleotide sequences isolated from six Korean hepatitis b virus-positive patients by using a reverse transcription-poly-merase chain reaction procedure, were analysed and compared to the seven previously reported HGV isolates. Sequence homology among the Korean isolates was 88-97% whereas among the isolates from different geographic areas was 80-92%, indicating geographical divergence of HGV. Nucleotide substitutions spread uniformly throughiut the E1 fragment. Furthermore, compared to the prototype HGV sequence, frameshift mutations were observed in most of the Korean isolating that a different translating initiation site for the polyprotein exists in the Korean type HGV.

  • The aim of this study was to develop hydroponic-cultured ginseng vinegar (HGV) containing ginsenoside Rg2 in order to its anti-obesity and anti-hyperlipidemic effects in C57BL/6J mice. HGV was prepared by two-stage fermentation. The ginsenoside Rg2 contained in acetic acid-fermented HGV increased by 4.0 times compared to that in pre-fermented HGV. To measure the anti-obesity effect of HGV, thirty two mice were randomly divided into four groups: normal diet group (ND), high-fat diet group (HFD), high-fat diet-supplemented with HGV group (HGV), and high-fat diet-supplemented with green tea extract group (GT). Body weight, fat weight, and liver weight decreased in the HGV group. The HGV group also showed lower plasma levels of low-density lipoprotein (LDL)-cholesterol and triglycerides, and higher levels of high-density lipoprotein (HDL)-cholesterol compared to the corresponding levels in the HFD group. Furthermore, there were significant decreases in plasma aspartase aminotransferase (AST) and alanine aminotransferase (ALT) levels in the HGV group compared to the corresponding levels in the HFD group. These results suggest that HGV can be used as an anti-obesity therapeutic agent or functional ingredient.

Performance of bridge structures under heavy goods vehicle impact

  • This paper presents a numerical study on the performance of reinforced concrete (RC) bridge structures subjected to heavy goods vehicle (HGV) collision. The objectives of this study are to investigate the dynamic response and failure modes of different types of bridges under impact loading as well as to give an insight into the simplified methods for modeling bridge structures. For this purpose, detailed finite-element models of HGV and bridges are established and verified against the full-scale collision experiment and a recent traffic accident. An intensive parametric study with the consideration of vehicle weight, vehicle velocity, structural type, simplified methods for modeling bridges is conducted; then the failure mode, impact force, deformation and internal force distribution of the validated bridge models are discussed. It is observed that the structural type has a significant effect on the force-transferring mechanism, failure mode and dynamic response of bridge structures, thus it should be considered in the anti-impact design of bridge structures. The impact force of HGV is mainly determined by the impact weight, impact velocity and contact interface, rather than the simplification of the superstructure. Furthermore, to reduce the modeling and computing cost, it is suggested to utilize the simplified bridge model considering the inertial effect of the superstructure to evaluate the structural impact behavior within a reasonable precision range.

The Detection and the Antigenic Analysis of the Hepatitis G Virus in Korea (한국인에서 Hepatitis G Virus (HGV) 검출 및 항원분석에 관한 연구)

  • We investigated the rate of hepatitis G virus infection among 50 patients who were not infected with the hepatitis C virus but showed symptoms of hepatitis. Viral RNA was extracted from the patients' sera and cDNA was synthesized and amplified by RT-PCR (reverse transcription-polymerase chain reaction) using random hexamer and 5 primers (470-20-1-77F, 470-20-1-211R, 470-20-1-211R-biotin, GV57-4512MF, GV57-4657MR). The amplified PCR products were confirmed by electrochemiluminescence (ECL), liquid hybridization (LH) and Southern blotting (SB). Among the 50 PCR products, by means of ECL, we found 4 samples to be positive and 5 samples to be indeterminate. The GV45-89M probe (5'-CYCGCTGRTITGGGGTGTACfGGAAGGC-3') was end-labelled with gamma-$^{32}P$ ATP and used for liquid hybridization with the PCR products. By using liquid hybridization, we detected specific bands from 4 positive sera and also from one indeterminate serum as determined by ECL. An 1.5% agarose gel electrophoresis of the 9 PCR products which were HGV positive or indeterminate as determined by ECL showed a 160bp band from 4 positive and one indeterminate serum. The 5 PCR products proved to be positive when SB was applied with the GV45-89M probe as well as when LH was applied. LH and SB were shown to have higher sensitivity and specificity than ECL. Two cases among 5 positive cases had relatively high SGOT, SGPT, ALP values when compared with other 48 cases. In summary, we confirmed hepatitis G virus infection in 5 cases among 50 Korean patients showing symptoms of viral hepatitis.

A Study of Classification of Road Tunnel for Fire Safety (안전성 향상을 위한 도로터널 등급에 관한 연구)

  • In road tunnel, in order to prevents an accident and minimize the damage of an accident in the case of fire, safety facilities and equipments are integral parts. The type and amount of safety facilities are based on tunnel type and length, traffic flow rate, etc. Therefore many countries use a tunnel classification system that categories tunnel into groups, and specifies the necessary emergency equipment for each group. In this study, for the purpose of classifying tunnel based on tunnel ist investigated the domestic and foreign standards and regulations for safety of road tunnel. As a results, we suggest the method of classification of tunnel by traffic performance, tunnel grade, the volume of traffic, fraction of HGV, rules or regulations for transports of dangerous good through tunnel.

Research and Development Trends of a Hypersonic Glide Vehicle (HGV) (극초음속 활공 비행체(HGV)의 연구개발 동향)

  • The hypersonic glide vehicle ascends to a high altitude by a rocket booster, separates it from the booster, and glides at a hypersonic speed of Mach 5 or higher at an altitude of about 30~70 km, changing its direction in the atmosphere. Since it moves on an unpredictable flight path rather than a parabolic trajectory, it is difficult to intercept with current missile defense systems. The U.S. conducted HTV-2 and AHW flight tests in the early 2010s to confirm the possibility of hypersonic gliding flights, and recently it has been developing hypersonic glide vehicle systems such as LRHW and ARRW. China has conducted several flight tests of the DF-ZF (WU-14) glide vehicle since 2014 and has been operating it with DF-17 missiles. Russia has conducted hypersonic glide vehicle research since the former Soviet Union, but it has repeatedly failed, and recently it has been successfully tested with the Avangard (Yu-71) glide vehicle mounted on the SS-19 ICBM. In this paper, the characteristics, flight test cases, and development trends of hypersonic glide vehicles developed or currently being developed in the United States, China, Russia, Japan, India, and Europe are reviewed and summarized.

The Phylogenetic Analysis of the NS-5 Region Sequence of Hepatitis G Viruses Isolated in Korea (국내에서 분리된 G형 간염바이러스 NS-5 Region 염기서열의 계통학적 분석)

  • We examined the hepatitis G virus infections among 227 Koreans who were healthy or were suspected of hepatitis and determined the phylogenetic relationship based on a part of the NS-5 region of 5 positive samples. Viral RNA was extracted from sera and cDNA was synthesized and subsequently amplified by RT-PCR (reverse transcription-polymerase chain reaction) or RT-nested PCR using random hexamer and NS-5 specific primers (470-20-1-77F, 470-20-1-211R, HGVNESTFO, HGVNESTRE). Five positives were found to belong to samples of patients showing symptoms of viral hepatitis. Primers used for PCR or nested PCR were derived from the NS-5 region. On the other hand, no amplification was detected using primers derived from the 5'-NCR (G-146F, G-401R). We performed TA cloning and sequencing of 5 amplified fragments, and their sequences were compared with those of foreign isolates of HGV. The phylogenetic analysis using MegAlign programme of DNAstar has shown that the Korean isolates are clustered on the phylogenetic tree. In summary, we confirmed the hepatitis G virus infection in 5 cases out of 12 patients showing the symptoms of viral hepatitis. The phylogenetic analysis of sequences of 5 amplified fragments showed that their relations to each other were closer than those to the foreign HGV isolates reported.

  • This paper describes the interception using fine fragments and particles to hypersonic vehicles which have a vulnerability in thermal and pressure during glide-phase flight. This interception concept is based on the fast relative velocity and the flight vulnerability of hypersonic vehicles. For the density calculation of fragmentation and particle in interception, error analysis of end-phase was performed including radar, intercept missile and target maneuvering errors. In relation to the vulnerability and error analysis, the penetration characteristics of fine fragments in high temperature were analyzed. Presented the interception in glide-phase could be applied to the concept of horizontal multi-layer defense to hypersonic vehicles.

Development and performance evaluation of SB3-level roadside barrier for highway transition zone (고속국도용 SB3등급 전이구간 방호울타리 개발 및 성능평가)

  • PURPOSES : In this research, an SB3-level roadside barrier for a highway transition zone that meets the newly established guide Installation and Management Guide for Roadside Safety Appurtenance is developed. Its performance is evaluated by a numerical simulation and real-scale vehicle impact test. METHODS : The commercial explicit dynamic software LS-DYNA is utilized for impact simulation. An FE model of a passenger vehicle developed and released by the National Crash Analysis Center (NCAC) at George Washington University and a heavy goods vehicle (HGV) model developed by the TC226/CM-E Work Group are utilized for impact simulation. The original vehicle models were modified to reflect the conditions of test vehicles. The impact positions of the passenger vehicle and truck to the transition guardrail were set as 1/2 and 3/4 of the transition region, respectively, according to the guide. RESULTS : Based on the numerical simulation results of the existing transition barrier, a new structural system with improved performance was suggested. According to the result of a numerical simulation of the suggested structural system, two sets of transition barriers were manufactured and installed for real-scale vehicle impact tests. The tests were performed at a test field for roadside safety hardware of the Korea Highway Corporation Research Institute. CONCLUSIONS : The results of both the real-vehicle impact tests and numerical simulations of the developed transition barrier satisfied the performance criteria, and the results of numerical simulation showed good correlation with the test results.
