Analysis of Performance Influencing Factor in Chemical Process Industry : A Practical Application (석유화학 산업에서의 수행영향인자 및 근본원인 분석 결과)

  • Chemical Process industry in Korea has over 30 year's of history and is likely to face potential incidents. The traditional risk analysis and control system in Chemical Process industry focuses on mechanical defects, overlooking the human performance control. Although development of automation technology and controlling technology was necessary, human decision factor is essential to preventing accidents in the Chemical Process. Almost all serious accidents take place when inappropriate humanperformance and mechanical defects of safety equipments simultaneously occurs. The AHRA(Advanced Human Reliability Analyzer) software has been developed to collect failure data and analyze human error probability (Reliability) in Chemical Process Industry in Korea. This paper describes the HRA analysis result of PIF(Performance Influencing Factor) evaluation, HEP(Human Error Probability) and root cause of accidents by applying a Chemical Process Industry related accident data. This analysis result should present a scheme that, by controlling human error factor other than putting safety management funds into the machinery in plants, can reduce cost and maximize the safety in Chemical Process Industry.

A Study on the bioactive surface modification of PEEK intervertebral fusion cage using various coating techniques (다양한 코팅 처리에 의한 PEEK 추간체유합보형재의 생체 활성 표면에 관한 연구)

  • In this study, the surface of Polyetheretherketone (PEEK) disks was modified to have a hydrophilic surface by applying a coating of Polyethylene glycol (PEG), Hyaluronic acid(HA), and Poly-Dopamine(PDA). The investigation aimed to examine whether the coated surfaces showed enhanced bioactivity for orthopedic applications compared to the pure PEEK. The microstructure, surface characteristics, and wettability of PEEK coated with PEG, HA, and PDA were analyzed using scanning electron microscopy(SEM), FT-IR spectrophotometer, Roughness Measurement System, Micro-Vickers, and Contact angle measurement. The mechanical properties were analyzed using a tensile testing machine, while the MTT assay for cell activity was analyzed using a microplate reader to measure optical density. According to the SEM and FT-IR results, the composition and crystal structure of PEG, HA and PDA coated surface were verified. Also, roughness, hardness, and contact angle were all improved in the coating group compared to the pure PEEK. We checked the HepG2 cell proliferation by using MTT assay on 7th days. In MTT assay results, HepG2 cell proliferation was increased with time, at 7 days, cell viability on discs coated with PDA was significantly higher than pure PEEK, PEG, HA coated group. PDA coated PEEK exhibited the highest surface roughness, hardness, contact angle, and cell activity. The mechanical properties were not affected by the presence of the coating.

The Effects of Cesium, Strontium and Cobalt on Cell Toxicity in the 2D and 3D Cell Culture Platforms (단층 및 입체 세포배양환경에서 세슘, 스트론튬 및 코발트가 세포 독성에 미치는 영향 분석)

  • Currently, there are 442 operating nuclear power plants in the world, and 62 more are under construction. According to this reasoning, the treatment of radioactive waste is important to prevent the environmental ecosystem including humans, animals, and plants. Especially, a leakage of radioactive waste causes not only regional problem but also serious global one. In this study, we demonstrate the effect of radioisotopes (e.g., cesium, strontium, and cobalt) on a 3D culture cell. To develop the 3D cell culture system, we used a 96-well-culture plate with biocompatible agarose hydrogel. Using this method, we can perform the 3D cell culture system with three different cell lines such as HeLa, HepG2, and COS-7. In addition, we conducted a cell viability test in the presence of radioisotopes. Interestingly, the 3D morphological cells showed 42% higher cell viability than those on the 2D against cesium. This result indicates that the 3D platform provides cells morphological and physiological characteristic similar to in vivo grown tissues. Moreover, it overcomes the limitation of conventional cell culture system that can't reflect in vivo systems. Finally, we believe that the proposed approach can be applied a new strategy for simple high-throughput screening and accurate evaluation of metal toxicity assay.

Effects of Buthus martensi Karsch on tumor promotion in two-stage carcinogenesis in mice (전갈(全蝎)이 노화(老化)에 따른 2단계(段階) 발암화(發癌化) 과정(過程)에 미치는 영향(影響))

  • To clarifiy the activating effects of Buthus martensi Karsch(BMK) on tumor promotion in two-stage carcinogenesis in mice was investigated. In vivo system, BMK was seen to gave an inhibitory activity on TPA-induced mouse ear edema. In addition, the BMK was proved to have antitumor-promoting activity in two-stage mouse skin carcinogenesis induced by DMBA and two-stage mouse lung carcinogenesis induced by 4-NQO as a initiator plus TPA and glycerol as a promoter. Moreover, BMK significantly exhibited an cytolytic effect in HepG2 cells and showed significant antitumor activity against Sarcoma-180 bearing mice by oral administration. These results suggest that BMK could be effective in adjuvant chemotherapy for human cancer.

  • 1차년도에서 이미 확립한 다양한리포좀의 제조 방법에 따라간세포를표 적하기 위해 이들 리포좀의 조성에 galactocercbroside를 함유시켜 targeted liposomes을 제조하였다. 표적리포좀의 제조 방법에 따른 봉합율 (cncapsulation)과 안정성을 조사했을 때, freezing-thawing 과정을 거친 리포좀이 encapsulation 과 stability가 가장 뛰어남을 알 수 있었다. In vitro 간세포 표적능력을 in vitro cell culture system에서 간세포 cell line (Hep G2, 2.2.15과 다른 cell line (Vero E6, J82)에 대한 표적리포좀과 control liposomes의 uptake를 FTTC 또는 CF 형광으로 측정했을 때, 표적리포좀이 간세포 cell line에 더 많이 uptake하는 것을 알았다. In vivo 실험에서는 6 - 8 주령의 mouse tail vain에 표적리포좀과 control liposomes을 주사하여 각 장기에 존재하는 리포좀의 양을 형광으로 측정했을 때, 표적리포좀 (targetcd liposomes)이 다른 장기에 비해 liver에 선택적으로 많이 분포함을 알수있었다. 또한 간암의 효과적인 치료를 위해 간세포와 암세포를 동시에 표적할 수는 double targeting liposomes을 design 하였다.

  • 전통적인 HRA(Human Reliability Analysis)방법은 특정 애플리케이션 또는 산업을 염두에 두고 있으며. 또한 이러한 방법은 종종 복잡하며, 시간이 많이 걸리고 적용하는 데 비용이 많이 들며 직접 비교하기에는 적합하지 않다. 제안된 HFHM(Human Factors Hazard Model: 인적 요인 위험 모델)은 기검증되고 시간 테스트를 거친 FTA(Fault Tree Analysis:결함 트리 분석)및 ETA(Event Tree Analysis:이벤트 트리 분석)의 확률 분석 도구 및 새로 개발된 HEP(Human Error Probability:인적 오류 확률)예측 도구와 통합되고, 인간과 관련된 PSF(Performance Shaping Factors:성능 형성 요인)를 중심으로 새로운 접근 방식으로 개발되었다. 인간-시스템은 상호작용으로 인한 재난사고 가능성을 모델링하는 위험분석 접근법 HFHM은 다음과 같은 상용 소프트웨어 도구 내에서 예시되고 자동화된다. HFHM에서 생성된 데이터는 SE 분석가 및 설계에 대한 표준화된 가이드로 사용될 수 있다. 본 연구에서는 인적 위험을 예측하는 이 새로운 접근 방식을 통해, 전체 시스템에 대한 포괄적인 재난안전 분석을 가능하게 한다.

Inhibitory Effect of Linum usitatissimum and Perilla frutescens as Sources of Omega-3 Fatty Acids on Mutagenicity and Growth of Human Cancer Cell Lines (식물성 오메가-3계 지방산 급원인 아마씨 및 들깨의 항돌연변이 및 암세포 증식 억제 효과)

  • It has been known that Linum usitatissimum and Perilla frutescens are dietary sources of possible chemopreventive compounds such as lignans and $\alpha$-linolenic acid. Here, we investigated and compared the inhibitory effects of methanol extracts from Linum usitatissimum and Perilla frutescens on mutagenicity using the Ames test, and growth of human cancer cells (AGS human gastric adenocarcinoma, HT-29 human colon cancer, Hep 3B hepatocellular carcinoma cells). In the Ames test system using Salmonella typhimurium TA100, aflatoxin $B_1$ ($AFB_1$)-induced mutagenicity was significantly inhibited by treatment with the methanol extract from either Linum usitatissimum or Perilla frutescens (p<0.05) in a dose dependent manner. As for N-methyl-N'-nitro-N-nitrosoguamidine (MNNG)-induced mutagenicity, the methanol extracts (5 mg/assay) from Linum usitatissimum and Perilla frutescens showed 63% and 78% inhibitory rates, respectively, indicating that Perilla frutescens possessed stronger antimutagenic activity than did Linum usitatissimum. Inhibitory effects of methanol extracts from Linum usitatissimum and Perilla frutescens on the growth of human cancer cells (AGS, HT-29 and Hep 3B) appeared to increase dose dependently, and the inhibition was more effective against AGS and HT-29 compared to Hep 3B cells. Our results suggested that the methanol extract from Perilla frutescens showed stronger antimutagenic activity than that from Linum usitatissimumas assayed by the Ames mutagenic test, whereas the methanol extract from Linum usitatissimum was more effective than its counterpart for growth inhibition of human cancer cells. It is concluded that intake of Linum usitatissimum and Perilla frutescens as sources of omega-3 fatty acids will be beneficial for preventing cancer.

DNA Damage and Micronuclei Induced by Di (2-ethylhexyl) phthalate in Human Breast Carcinoma MCF-7 cells (Di(2-ethylhexyl) phthalate에 의해 유도된 DNA손상과 소핵 형성)

  • Di-2-ethylhexyl phthalate (DEHP) is the most commonly used phthalate ester in polyvinyl chloride formulations including food packing and storage of human blood. DEHP is a well known as non-genotoxic carcinogen and endocrine disrupting chemical (EDC). DEHP have shown all negative results in ICH-guildeline recommended standard genotoxicity test battery. In this study, to assess the clastogenic and DNA damaging effect in human-derived tissue specific cells, DEHP was treated in human derived MCE-7 cells, HepG2 cells, LNCap cells, BeWo cells, MCE-10A cells, and female peripheral blood cells using micronucleus assay and in human breast carcinoma MCF-7 cells up to $1.28$\times$10^{-2}$ M using Comet assay. The in vitro micronucleus assay is a mutagenicity test system for the detection of chemicals which induce the formation of small membrane bound DNA fragment i.e. micronuclei in the cytoplasm of interphase cells, originated from clastogenic and/or aneugenic mechanism. The single cell gel electrophoresis assay (Comet assay) is used to detect DNA strand-breaks and alkaline labile site. In our results, DEHP increased significantly and/or dose-depentently and time-dependently micronucleus frequency at the 6 and 24 hr without metabolic activation system only in MCE-7 cells. DEHP treated with 2 hrs in MCF-7 cells using Comet assay induced DNA damage dose-depentantly.

Antiviral Effects of Natural Products on the Inhibition of Hepatitis B Virus DNA Replication in 2.2.15 Cell Culture System

  • Evaluation of plant extracts that might inhibit hepatitis B virus (HBV) replication was performed to find potent anti-HBV agents. Eighty-five species of plants from forty-three families were tested for their anti-HBV activities using HBV-producing HepG2-derived 2.2.15 cells. The anti-HBV activity of plant extracts was measured by slot blot hybridization technique and cytotoxicity was determined by crystal violet staining procedure. All plants were extracted with methanol and the extracts were partitioned into n-hexane, ethyl acetate and aqueous layer. The ethyl acetate fractions of Rhus verniciflua $(stem:\;EC_{50},\;8.2{\mu}g/ml;\;CC_{50},\;9.4{\mu}g/ml)$, Gastrodia elata $(root:\;EC_{50},\;17.7{\mu}g/ml;\;CC_{50},\;>20{\mu}g/ml)$, Raphanus sativus $(seeds:\;EC_{50},\;17.3{\mu}g/ml;\;CC_{50},\;>20{\mu}g/ml)$, and Angelica gigas $(root:\;EC_{50},\;8.3{\mu}g/ml;\;CC_{50},\;15.6{\mu}g/ml)$ revealed the anti-HBV activity in 2.2.15 cell culture system and these fractions are under the process of further sequential fractionation by column chromatography to find the active principles against HBV.

Allozyme Diversity and Population Genetic Structure in Korean Endemic Plant Species : II. Hosta yingeri (Liliaceae)

  • Levels of genetic diversity, population genetic structure, and gene flow in Hosta yingeri, a herbaceous perennial endemic to Taehuksan, Sohuksan, and Hong Islands, were investigated. Starch gel electrophoresis was conducted on leaves for 101 plants collected from three populations. Although the distribution of thespecies is restricted in the islands, it maintains high levels of genetic variatin; 64% of polymorphic loci in at least one population (Ps), the mean number of alleles per locus (Ap) of 1.92, and the mean effective number of alleles per locus (Aep) of 1.52. Overall, mean genetic diversity (Hep=0.250) was substantially higher than mean estimate for species with very similarlife history traits (0.102). Large populaton size, the persistence of multiple generations within populations, high fecundity, predominantly outcrossing breeding system, large size of pollinator visitation areas may be explanatory factors contributing the higher level of genetic diversity maintained within populations. Analysis of fixation indices showed an overall slight excess of heterozygotes (mean FIS=-0.066) relative to Hardy-Weinberg expectations, which may in part be due to the near self-incompatible breeding system in the species. Significant differences in allele frequencies among populaitns were found for 14 out of 16 polymorphic loci (P<0.05). Slightly more than 80% of the total variation in the species was common to all populations (GST=0.198). As expected, indirect estimate of the number of migrants per generation (Nm=0.45, calculated from mean GST) and nine private alleles found in the three populations indicate that gene movement among three isolated island populations was low.
