The Activation of PPAR-α and Wnt/β-catenin by Luffa cylindrica Supercritical Carbon Dioxide Extract

  • Luffa cylindrica (LC) is a very fast-growing climber and its fruit have been considered as agricultural wastes. We conducted to check the comparative qualities of ethanol solvent extraction (LCE) and supercritical carbon dioxide extraction (LCS) of L. cylindrica fruit and seed. LCS had higher antioxidant and polyphenol contents than LCE. LCS were significantly increased peroxisome proliferator-activated receptor (PPAR)-a and involucrin expression as epidermal differentiation marker in 3D skin equivalent model. LCS also showed antimicrobial activity against Staphylococcus aureus, a causative bacteria in atopic dermatitis. In addition, LCS inhibited the adipocyte differentiation of 3T3-L1 cells. When treated with the extract at a concentration of 100 ㎍/mL, the Wnt/β-catenin pathway reporter luciferase activity of HEK 293-TOP cells was increased approximately by 2-folds compared to that of the untreated control group. These results indicate that L. cylindrica supercritical carbon dioxide extract may serve as a cosmeceutical for improving skin barrier function and the treatment of obesity.

Non-canonical targets play an important role in microRNA stability control mechanisms

  • MicroRNAs (miRNAs) regulate gene expression by guiding the Argonaute (Ago)-containing RNA-induced silencing complex (RISC) to specific target mRNA molecules. It is well established that miRNAs are stabilized by Ago proteins, but the molecular features that trigger miRNA destabilization from Ago proteins remain largely unknown. To explore the molecular mechanisms of how targets affect the stability of miRNAs in human Ago (hAgo) proteins, we employed an in vitro system that consisted of a minimal hAgo2-RISC in HEK293T cell lysates. Surprisingly, we found that miRNAs are drastically destabilized by binding to seedless, non-canonical targets. We showed that miRNAs are destabilized at their 3' ends during this process, which is largely attributed to the conformational flexibility of the L1-PAZ domain. Based on these results, we propose that non-canonical targets may play an important regulatory role in controlling the stability of miRNAs, instead of being regulated by miRNAs.

  • 전계효과 트랜지스터를 이용한 바이오센서는 하나의 칩 위에 많은 센서 소자를 집적할 수 있으므로, 같은 종류의 센서를 다수 배열함으로써 다차원화할 수 있고, 다른 종류의 센서를 여러개 배열함으로써 다기능화할 수 있다. 또한 지능회로와 함께 집적하여 지능화하거나, 관련회로 및 장치들을 함께 집적함으로써 시스템화할 수 있기 때문에 최첨단 센서로 각광을 받고 있다. 그러나, 전계효과 트랜지스터를 이용한 바이오센서는 게이트 영역에 생체 분자를 고정시키는 것이 어렵고, 고정되더라도 생체 분자의 양이 미량이어서 재현성이 떨어지며, 생체 분자가 발생시키는 시그널이 적어 전류 세기 변화에 대한 검출감도가 저하되는 문제점이 있다. 본 연구에서는 반도체 리소그래피 공정을 이용하여 생체 분자를 물리 화학적 처리 없이 게이트 영역에 집중적으로 고정시킬 수 있는 기술에 대해 연구하였다. 산화막이 증착된 기판 위에 포토레지스트를 도포한 뒤 리소그래피공정을 이용하여 패터닝 하였으며 기판 위에 human embryonic kidney(HEK)-293 세포를 배양하였다. 연구결과, 친수성인 포토레지스트보다 소수성인 산화막 영역에 다수의 세포가 선택적으로 집중 배양됨을 확인하였다. 따라서 본 연구결과를 바이오센서에 적용할 경우 센서의 검출감도를 향상시킬 수 있을 것으로 기대된다.

Extracellular Zinc Modulates Cloned T-type Calcium Channels

  • In the present study, we investigated effects of extracellular zinc (Zn$\^$2+/) on T-type Ca$\^$2+/ channel isoforms (${\alpha}$lG, ${\alpha}$lH, and ${\alpha}$lI) stably expressed in HEK 293 cells. Ca$\^$2+/ currents were measured using 10 mM Ca$\^$2+/ as a charge carrier under whole cell-ruptured patch configuration. Zn$\^$2+/ blocked the ${\alpha}$lH currents with a 100- and 200-fold higher potency (IC$\sub$50/ = 2.5 ${\mu}$M) when compared with those for blockade of the ${\alpha}$1G and ${\alpha}$1I currents, respectively.(omitted)

  • Cysteine and histidine-capped water-dispersible ZnS:Mn nanocrystals (ZnS:Mn-Cys and ZnS:Mn-His) were synthesized and their effects on E. coli and human cells were investigated. Particle sizes of these nanocrystals were found from HR-TEM images to be 3.5 nm and 4.0 nm, respectively. Their solution photoluminescence spectra showed identical broad emission peaks at 580 nm. ZnS:Mn-His significantly suppressed the growth of E. coli at $100{\mu}g/mL$ and 1 mg/mL concentrations, something not observed with ZnS:Mn-Cys. Consistent with this, greater inhibition of cell proliferation and viability were observed in HEK293 and IMR90 cells in ZnS:Mn-His at $100{\mu}g/mL$ and 1 mg/mL concentrations.

  • Recently, decades of robust researches on degenerative brain disorder have been highlighted on the interactive connection of gut and brain. In terms of inflammatory cytokine production, others have shown that Nucleotide-Binding Oligomerization Domain Containing 2 (NOD2) is involved with Leucine-Rich Repeat Kinase 2 (LRRK2). HEK293T cells were transiently co-transfected with Myc-tagged LRRK2 and Flag-tagged NOD2 and then followed by co-immunoprecipitation assay. In this study, we provide the novel finding of physical protein-protein interaction between NOD2 and LRRK2. G2019S variant has shown stronger interactions against NOD2 than those of wild type LRRK2. In an axis of NOD2-LRRK2 communication, it is believed to pave a new way in the understanding of the bidirectional molecular mechanism of brain disorder, including Parkinson's disease into gut inflammatory disease, including Crohn's disease.

  • 본 연구에서는 고분해능 및 고감도화된 시스템 개발을 위하여 AOTF를 이용하여 하이퍼스펙트럼 영상기법을 활용한 전임상 영상장비에 대한 연구를 수행하였다. 제작된 고감도 하이퍼스펙트럼 분자영상 시스템의 생물학적 적용을 위하여, AOTF의 파장 또는 진동수를 변화시키면서 GFP가 발현된 HEK 293 세포의 이미지를 촬영하였다. 또한, 제작된 실험 대상물 이미지화 시스템을 이용해서 실험용 쥐의 이미지를 촬영하였다. 실험용 쥐를 크세논 아크램프 사용 전 후 이미지를 촬영한 결과 크세논 아크 램프 사용 후에는 청색의 선명한 영상을 얻을 수 있었다.

  • The dopamine D2 receptor (D$_2$R) is target for antipsychotic drugs and associated with several neuropsychiatric disorders. The internalization (sequestration) of G protin-coupled receptor is caused by agonist-induced receptor phosphorylation mediated by GRK, followed by the interaction with ${\beta}$-arrestin. In this study, we examined the agonist-dependent sequestration/internalization of dopamine D$_2$R, which were transiently expressed in HEK 293 cells with of without GRK co-expression. Co-expression of GRK2 or GRK3 markedly enhanced the sequestration of D$_2$R. (omitted)

Elucidation of their differential functional characteristics is important for understanding their roles in brain.
