Decreased Expression of TRPV4 Channels in HEI-OC1 Cells Induced by High Glucose Is Associated with Hearing Impairment

  • Purpose: Previous reports have shown that hyperglycemia-induced inhibition of transient receptor potential vanilloid sub type 4 (TRPV4), a transient receptor potential ion channel, affects the severity of hearing impairment (HI). In this study, we explored the role of TRPV4 in HI using HEI-OC1 cells exposed to high glucose (HG). Materials and Methods: HEI-OC1 cells were cultured in a HG environment (25 mM D-glucose) for 48 hours, and qRT-PCR and Western blotting were used to analyze the expression of TRPV4 at the mRNA and protein level. TRPV4 agonist (GSK1016790A) or antagonist (HC-067047) in cultured HEI-OC1 cells was used to obtain abnormal TRPV4 expression. Functional TRPV4 activity was assessed in cultured HEI-OC1 cells using the MTT assay and a cell death detection ELISA. Results: TRPV4 agonists exerted protective effects against HG-induced HI, as evidenced by increased MTT levels and inhibition of apoptosis in HEI-OC1 cells. TRPV4 overexpression significantly increased protein levels of phosphorylated p38 mitogen-activated protein kinase (p-p38 MAPK), while TRPV4 antagonists had the opposite effect. Our results indicated that TRPV4 is a hyperglycemia-related factor that can inhibit cell proliferation and promote cell apoptosis by activating the MAPK signaling pathway in HEI-OC1 cells. Conclusion: Our results show that the overexpression of TRPV4 can attenuate cell death in HEI-OC1 cells exposed to HG.

  • In this research we have developed a reliable, effective and feasible HEI(High-Energy Ion Implantation) process 3D-simulation tool, and then by using it we can predict and analyze the effect of HEI process on characteristics of the standard CMOS device. high-energy ion implantation above 200 keV is inevitable process to form retrograde well and buried layer to prevent leakage current, to conduct field implant for field isolation, and to perform after-gate implantation. The feasible analysis tool is a product of the HEI process modeling verified by comparison of the SIMS experiments with the simulation results. Especially, in this paper, we present the predicting capability of HEI-induced impurity and damage profiles compared with the published SIMS data in order to acquire the reliability of our results ranging from few keV to several MeV for phosphorus and boron implantation.

Evaluation of dietary quality of adolescents using Healthy Eating Index

  • The aim of this study was to use the Healthy Eating Index-2005 (HEI-2005) to assess diet quality and determine the relationship between the HEI-2005 and the energy and nutrient intakes of adolescents. A cross-sectional study was conducted on 1,104 healthy adolescents, mean age of $15.8{\pm}1.24$ years. Dietary intake was measured with the 24-hour dietary recall method, and dietary quality was assessed by means of the HEI-2005. Diet quality scores ranged from 23.7 to 77.5. The mean score was found to be $51.5{\pm}9.07$ according to the HEI-2005. There were no differences according to gender, 42.8% had a poor diet and 57.2% had a diet that needs improvement. No subjects had a "good diet". Lower mean subgroup scores were found for whole grains, total vegetables, total fruits, dairy products, and meat and beans. Fruits and vegetables scores were significantly high in girls, whereas sodium, oil, and meat and beans scores were significantly high in boys. Total HEI-2005 scores were increasingly associated with parental education level when age and gender were adjusted. There was a negative correlation between HEI-2005 scores and age, total energy intake, and fat intake. Positive correlation was only observed in the HEI-2005 scores for protein and dietary fibre intakes. Consequently, the overall diet quality and nutritional habits of Turkish adolescents need modification and improvement. In the family, measures should be initiated by the government, including advertisements and campaigns.

Protective Effect of Ursolic Acid from Corni fructus on the Hydrogen Peroxide-induced Damage of HEI-OC1 Auditory Cells (산수유로부터 추출한 ursolic acid가 과산화수소로 손상된 HEI-OC1 청각세포보호에 미치는 영향)

  • The fruits of Cornus officinalis have been used in traditional Oriental medicine for treatment of inner ear diseases, such as tinnitus and hearing loss. In the present study, we showed that the ursolic acid obtained from Corni fructus protected HEI-OC1 auditory cells from hydrogen peroxide cytotoxicity in a dose-dependent fashion. In addition, to investigate the protection mechanism of ursolic acid on hydrogen peroxide cytotoxicity toward HEI-OC1, we measured the effects of ursolic acid on lipid peroxidation and activities of superoxide dismutase (SOD), catalase (CAT), and glutathione peroxidase (GPX) in hydrogen peroxide treated cells. Ursolic acid (0.05 - 2 ${\mu}g/ml$) had protective effect against the hydrogen peroxide-induced HEI-OC1 cell damage and reduced lipid peroxidation in a dose-dependent manner. Pre-treatment with ursolic acid significantly attenuated the decrease in activities of CAT and GPX, but SOD activity was not affected by the ursolic acid or hydrogen peroxide. These results indicate that ursolic acid protects hydrogen peroxide-induced HEI-OC1 cell damage through inhibition of lipid peroxidation and induce the antioxidant enzymes CAT and GPX.

Anti-oxidant Effects of the Water Extracts from the Inonotus Obliquus against Cisplatin- Induced Damage in HEI-OC1 Cells (차가버섯 물 추출물의 cisplatin에 의해 유도된 HEI-OC1세포 손상에 대한 항산화효과)

  • The medicinal mushroom Inonotus obliquus is a traditional and widely used multi-functional fungus. In this study, we have investigated whether Inonotus obliquus (Chaga mushroom) extracts exerts anti-oxidant effects on cisplatin-induced cytotoxicity in auditory cell line, HEI-OC1 cells. First of all, Chaga extracts has no harmful effects on viability of HEI-OC1 cells in the dose range of $65{\sim}125{\mu}g/m{\ell}$. Moreover, it shows cyto-protective effects on the cells treated with cisplatin-induced cytotoxicity in HEI-OC1 cells and the damage of hair cells arrays of the rat primary organ of Corti explants in the presence of cisplatin. Pretreatment with Chaga extracts inhibited the cell death, reactive oxygen species generation (ROS), lipid peroxidation induced by cisplatin. These effects were associated with the induction of antioxidant enzyme by Chaga extracts. We further investigated the effects of Chaga extracts on expression of antioxidant enzymes such as Cu, Zn superoxide dismutase (SOD 1) and Mn SOD (SOD 2) by RT-PCR. In addition, Chaga extracts shows SOD activity and SOD protein expression in cisplatin treated group induced similar to control group. Taken together, these results indicate that Chaga extracts can prevent cisplatin-induced cytotoxicity by radical-scavenging activity (SOD activity) in HEI-OC1 cells. It might be an effective as antioxidant and further studies on the chemo-preventive mechanisms of Inonotus obliquus are needed.

Breeding of Black Tomato 'Hei-G' Suitable for Protected Cultivation (시설재배용 흑색 토마토 '헤이-G' 육성)

  • 'Hei-G' is a new tomato cultivar originating from the cross of two inbred lines, the maternal plant TKUI-3 separated from 'Kame' in 2007, and the paternal plant AL-1 developed by pedigree breeding. The new cultivar was registered as 'Hei-G' named 'JTB026 (Jeonnam-2)' based on characteristics and yield trials carried out in the spring of 3 years, from 2010 to 2012. 'Hei-G' is an indeterminate type with strong plant vigor. The fruit is round and the ripening fruit is blackish red. The average fruit weight of 'Hei-G' is about 43.4 g, and its marketable yield is $4,944kg{\cdot}10a^{-1}$. The lycopene content of 'Hei-G' ($11.8mg{\cdot}100g^{-1}$) is higher than that of other tomatoes (e.g., cv. Rapido, 3.1 mg). In addition, 'Hei-G' is suitable for environment-friendly protected cultivation because of its resistance to powdery mildew.

  • 'Hei' is a new cultivar of red with black flesh; originated from the cross of two inbred lines, mother plant, TKUI separated from 'Kame' in 2007 and father plant, TLB separated from tomato collected in Europe by pedigree breeding method. 'Hei' was tested for specific character and productivity of lines of tomatoes in spring for 2 years, 2008 to 2009. This variety had resistance to powdery mildew. 'Hei' is an indeterminate type. The fruit shape is round and the skin color is blackish red. The average fruit weight of 'Hei' is about 127.9 g and marketable yield is $5,715kg{\cdot}10a^{-1}$. Lycopene contents of 'Hei' are about $18.5mg{\cdot}100g^{-1}$, high compared with other tomato ($3.1mg{\cdot}100g^{-1}$). 'Hei' is suitable for environment-friendly protected cultivation because of resistance to powdery mildew.

A Study on the Channel Length and the Channel Punchthrough of Self-Aligned DMOS Transistor (자기정렬 DMOS 트랜지스터의 채널 길이와 채널 Punchthrough에 관한 고찰)

  • A general closed form expression for the channel length of the self-aligned double-diffused MOS transistor is obtained from the 2-dimensional Gaussian doping profile. The proposed model in this paper is composed of the doping concentration of the substrate, the final surface doping concentration and the vertical junction depth of the each double-diffused region. The calculated channel length is in good agreement with the experimental results. Also, the optimum channel structure for the prevention of the channel puncthrough is obtained by the averaged doping concentration in the channel region. A correspondence between the results of device simulation of channel punchthrough and the estimations of simplified model is confirmed.

Associations Between XRCC1 Arg399Gln, Arg194Trp, and Arg280His Polymorphisms and Risk of Differentiated Thyroid Carcinoma: A Meta-analysis

  • Background: Associations between Arg399Gln, Arg194Trp and Arg280His polymorphisms of the XRCC1 gene and risk of differentiated thyroid carcinoma (DTC) have been widely studied but the findings are contradictory. Methods: We performed a meta-analysis in the present study using STATA 11.0 software to clarify any associations. Electronic literature databases and reference lists of relevant articles revealed a total of 10, 6 and 6 published studies for the Arg399Gln, Arg194Trp and Arg280His polymorphisms, respectively. Results: No significant associations were observed between Arg399Gln and DTC risk in all genetic models within the overall and subgroup meta-analyses, while the Trp/Trp vs Arg/Arg and recessive model of the Arg194Trp polymorphism was associated with DTC susceptibility, and the dominant model of Arg280His polymorphism contributed to DTC susceptibility in Caucasians. Conclusions: Our meta-analysis suggests that XRCC1 Arg194Trp may be a risk factor for DTC development.
