Phenotypic characterization of pre-harvest sprouting resistance mutants generated by the CRISPR/Cas9-geminiviral replicon system in rice

  • Pre-harvest sprouting is a critical phenomenon involving germination of seeds in the mother plant before harvest under relative humid conditions and reduced dormancy. In this paper, we generated HDR mutant lines with one region SNP (C/T) and an insertion of 6 bp (GGT/GGTGGCGGC) in OsERF1 genes for pre-harvest sprouting (PHS) resistance using CRISPR/Cas9 and a geminiviral replicon system. The incidence of HDR was 2.6% in transformed calli. T1 seeds were harvested from 12 HDR-induced calli and named ERF1-hdr line. Molecular stability, key agronomic properties, physiological properties, and biochemical properties of target genes in the ERF1-hdr line were investigated for three years. The ERF1-hdr line showed significantly enhanced seed dormancy and pre-harvest sprouting resistance. qRT-PCR analysis suggested that enhanced ABA signaling resulted in a stronger phenotype of PHS resistance. These results indicate that efficient HDR can be achieved through SNP/InDel replacement using a single and modular configuration applicable to different rice targets and other crops. This work demonstrates the potential to replace all genes with elite alleles within one generation and greatly expands our ability to improve agriculturally important traits.

  • 본 논문에서는 110Mbps의 전송속도를 가지는 eHDR-WPAN( enhanced High Data Rate-Wireless Personal Area Networks)시스템의 성능향상 방안으로 인터리버를 적용하였다. 실내무선채널에서 발생하는 페이딩 현상은 연집화 오류 성향을 증가시킨다. 그러므로 인터리빙 기법을 이용하여 오류의 연집화 성향을 산발화 시킴으로써 시스템의 오류성능을 개선 할 수 있다. 본 논문에서는 컨볼루셔널, 블록, 랜덤 인터리버를 적용하였으며, 심벌 간 거리를 9와 27로 설정하였다. 블록 인터리버의 경우, 10$^{-4}$에서 약 0.6dB 정도 E$_b$/N$_o$의 성능향상을 보였다. 그러므로 eHDR-WPAN 시스템에서는 심벌 간 거리가 9인 블록인터리버가 최적의 인터리버임을 확인하였다.

국부 영상 활동도에 적응적인 칼만 필터를 이용한 HDR 세부 영상 레이어의 잡음 제거 (Noise Reduction of HDR Detail Layer Using a Kalman Filter Adapted to Local Image Activity)

  • In High Dynamic Range (HDR) image processing, tone mapping is the process to compress an input image into a Low Dynamic Range (LDR) image. In most cases, the reason that detail preservation is prior to take over tone mapping is that the dynamic range is significantly different between input and output images. In the case of iCAM06, details are separated by using a bilateral filter, however, it causes noise amplification at the dim surround region. Thus, we suggest that the detail signal, which is separated from the bilateral filter, is combined with the base signal after an adaptive Kalman filter is applied according to the local standard deviation. We confirmed that the proposed method enhances the HDR images quality by checking the noise reduction in a dim surround region.

Dynamic loading tests and analytical modeling for high-damping rubber bearings

  • High-damping rubber bearings (HDRB) are commonly used as seismic isolation devices to protect civil engineering structures from earthquakes. However, the nonlinear hysteresis characteristics of the HDRB, such as their dependence on material properties and hardening phenomena, make predicting their behavior during earthquakes difficult. This study proposes a hysteretic model that can accurately predicts the behavior of shear deformation considering the nonlinearity when designing the seismic isolation structures using HDR bearings. To model the hysteretic characteristics of the HDR, dynamic loading tests were performed by applying sinusoidal and random waves on scaled-down specimens. The test results show that the nonlinear characteristics of the HDR strongly correlate with the shear strain experienced in the past. Furthermore, when shear deformation occurred above a certain level, the hardening phenomenon, wherein the stiffness increased rapidly, was confirmed. Based on the experimental results, the dynamic characteristics of the HDR, equivalent stiffness, equivalent damping ratio, and strain energy were quantitatively evaluated and analyzed. In this study, an improved bilinear HDR model that can reproduce the dependence on shear deformation and hardening phenomena was developed. Additionally, by proposing an objective parameter-setting procedure based on the experimental results, the model was devised such that similar parameters could be set by anyone. Further, an actual dynamic analysis could be performed by modeling with minimal parameters. The proposed model corresponded with the experimental results and successfully reproduced the mechanical characteristics evaluated from experimental results within an error margin of 10%.

히스토그램 변형을 이용한 HDR 영상 렌더링을 위한 인지기반 톤 맵핑 기법 (Perception-Based Tone Mapping Technique for Rendering HDR Image Using Histogram Modification)

  • 본 논문에서는 HDR (high dynamic range) 영상 렌더링을 위한 히스토그램 변형 기법을 이용한 인지기반 톤 맵핑(tone mapping) 기법을 제안한다. HDR 톤 맵핑 알고리듬은 넓은 동적 영역을 갖는 영상을 LDR (low dynamic range) 영상 장치에 표현하기 위해 사용된다. 인간의 시각특성을 이용한 톤 맵핑 알고리듬은 상당히 효과적이지만 고정된 형태의 맵핑 함수를 사용하기 때문에 모든 영상에서 동일한 성능을 얻지 못한다. 제안하는 방법은 인지기반 톤 맵핑 함수와 히스토그램 변형기법을 적용하여 HDR 영상의 동적 영역을 압축한다. 인지기반 톤 맵핑 함수를 이용해서 원 영상의 히스토그램을 제한하고 보상과정을 통해서 적응적인 톤 맵핑 함수를 얻을 수 있다. 이는 화소값에 따라 서로 다른 제약조건들을 사용함으로써 contrast를 향상시킬 뿐만 아니라 원 영상의 디테일도 보존한다. 또한, 히스토그램 변형기법을 사용함으로써 과도한 히스토그램 평활화로 인한 왜곡을 개선할 수 있고, 더욱이 제어파라미터를 이용하여 영상의 contrast를 조정할 수 있다. 시뮬레이션 및 실험을 통해 주관적 및 객관적 성능을 비교하고 기존 방법에 비해 제안한 방법이 우수함을 입증한다.

Damping of a taut cable with two attached high damping rubber dampers

  • Due to their low intrinsic damping, stay cables in cable-stayed bridges have often exhibited unanticipated and excessive vibrations which result in increasing maintenance frequency and disruption to normal operations of the entire bridges. Mitigation of undesired cable vibration can be achieved by attaching an external damping device near the anchorage. High Damping Rubber (HDR) dampers have many advantages such as compact size, better aesthetics, easy maintenance, temperature stability, and cost benefits; therefore, they have been widely used to increase cable damping. Although a single damper has been shown to reduce cable vibrations, it is not the most effective method due to geometric constraints. This paper proposes the use of two HDR dampers to improve effectiveness and robustness in suppressing cable vibration. Oscillation parameters of the cable-dampers system were investigated in detail by modeling the stay cable as a taut string and each HDR damper as complex-valued impedance and by using an analytical formulation of the complex eigenvalue problem. The problem of two HDR dampers arbitrarily located along a cable is solved and the solution is discussed. Asymptotic formulas to calculate the damping ratios of the cable with two HDR dampers installed near the anchorage(s) are proposed and compared with the exact solutions. Further, a design example is presented in order to justify the methodology. The results of this study show that when the two HDR dampers are installed close to each other on the same end of the cable, some interaction between the dampers leads to reduced damping ratio. When the dampers are on the opposite ends of the cable, they are effective in increasing damping ratio and can provide better vibration reduction to multiple modes.

효과적인 역 톤 매핑을 위한 필터링 기법 (Image Filtering Method for an Effective Inverse Tone-mapping)

  • 본 논문에서는 가이디드 영상 필터 (guided image filter: GIF)를 이용하여 컨볼루션 신경망 (convolutional neural network; CNN)을 이용한 역 톤 매핑 (inverse tone-mapping) 기법의 결과를 향상시킬 수 있는 필터링 기법을 제안한다. 저동적범위 (low dynamic range; LDR) 영상을 고동적범위 (high dynamic range; HDR) 디스플레이에서 표현할 수 있도록 변환하는 역 톤 매핑 기법은 지속적으로 제안되어왔다. 최근 들어 컨볼루션 신경망을 이용하여 단일 LDR 영상을 HDR 영상으로 변환하는 알고리듬이 많이 연구되었다. 그 중엔 제한된 동적범위 (dynamic range)로 인해 화소가 포화되어 기존 화소 정보가 손실되는데 이를 학습된 컨볼루션 신경망을 이용해서 복원하는 알고리듬이 존재한다. 해당 알고리듬은 비포화 영역의 잡음까지는 억제하지 못하며 포화 영역의 디테일까지는 복원하지 못한다. 제안한 알고리듬은 입력 영상에 가중된 가이디드 영상 필터 (weighted guided image filter; WGIF)를 사용해서 비포화 영역의 잡음을 억제하고 포화 영역의 디테일을 복원시킨 다음 컨볼루션 신경망에 인가하여 최종 결과 영상의 품질을 개선하였다. 제안하는 알고리듬은 HDR 정량적 화질평가 지표를 측정하였을 때 기존의 알고리듬에 비해 높은 화질평가 지수를 나타내었다.

Molecular cloning and characterization of 1-hydroxy-2-methyl-2-(E)-butenyl 4-diphosphate reductase (CaHDR) from Camptotheca acuminata and its functional identification in Escherichia coli

  • Camptothecin is an anti-cancer monoterpene indole alkaloid. The gene encoding 1-hydroxy-2-methyl-2-(E)-butenyl 4-diphosphate reductase (designated as CaHDR), the last catalytic enzyme of the MEP pathway for terpenoid biosynthesis, was isolated from camptothecin-producing Camptotheca acuminata. The full-length cDNA of CaHDR was 1686 bp encoding 459 amino acids. Comparison of the cDNA and genomic DNA of CaHDR revealed that there was no intron in genomic CaHDR. Southern blot analysis indicated that CaHDR belonged to a low-copy gene family. RT-PCR analysis revealed that CaHDR expressed constitutively in all tested plant organs with the highest expression level in flowers, and the expression of CaHDR could be induced by 100 ${\mu}M$ methyl-jasmonate (MeJA), but not by 100 mg/L salicylic acid (SA) in the callus of C. acuminata. The complementation of CaHDR in Escherichia coli ispH mutant MG1655 demonstrated its function.

Evolution of CRISPR towards accurate and efficient mammal genome engineering

  • The evolution of genome editing technology based on CRISPR (clustered regularly interspaced short palindromic repeats) system has led to a paradigm shift in biological research. CRISPR/Cas9-guide RNA complexes enable rapid and efficient genome editing in mammalian cells. This system induces double-stranded DNA breaks (DSBs) at target sites and most DNA breakages induce mutations as small insertions or deletions (indels) by non-homologous end joining (NHEJ) repair pathway. However, for more precise correction as knock-in or replacement of DNA base pairs, using the homology-directed repair (HDR) pathway is essential. Until now, many trials have greatly enhanced knock-in or substitution efficiency by increasing HDR efficiency, or newly developed methods such as Base Editors (BEs). However, accuracy remains unsatisfactory. In this review, we summarize studies to overcome the limitations of HDR using the CRISPR system and discuss future direction.

log 촬영과 HDR을 이용한 실사 360 영상 제작흐름 연구 (A Study for Virtual Reality 360 Video Production Workflow with HDR Using log Shooting)

  • 최근 VR 콘텐츠 제작은 크게 3가지 방법이 사용된다. CG를 이용한 방법, 게임엔진을 이용한 방법 그리고 실사촬영 방법이다. 이중 가장 보편적인 제작방법은 실사촬영이다. 하지만 지금까지 실사촬영은 대부분 액션캠(actioncam)을 이용한 촬영으로 영화, 드라마 같은 전문적인 영상 제작 방식과는 차이가 있다. 본 논문에서는 영상제작 전문가 방식과 액션캠 촬영 방식의 차이를 지적하고 대안을 제시하고 있다. 제안된 방법은 log 촬영을 통해 HDR이 가능한 촬영과 이를 활용한 편집 방법이다. 실험 촬영, 편집 결과 제안한 방법이 기존 액션캠 촬영 방법보다 더 많은 색정보를 확보할 수 있고 이를 통해 액션캠이 구현하기 어려운 고화질 영상을 구현하였다.
