해변 염생식물군집에 대한 생태학적 연구(III) 인천 간척지의 토지환경, 종의 다의성 및 염류순환에 대하여 (Ecological Studies on the Halophyte Communities at Western and Southern Coast in Korea (III) On the Soil Properties, Species Diversity and Mineral Cyclings in Reclaimed Soil in Incheon)

  • Physicochemical properties of soil, mineral cyclings, production of plants, and relationship between sodium(Na) content and progresses of plant communities were studied in a coastal salt marsh in Incheon. Contents of Na, available phosphorus(A-P) and value of electric conductivity of soil decreased in order of Salicornia herbacea, Limonium tetragonum, Phragmites communis and Zoysia sinica communities, but contents of organic matter, total nitrogen(T-N) and calcium(Ca) of soil were vice versa. Specise diversity index decreased with increase of Na content of soil with correlation coefficient of -0.82. The aboveground biomass of plant communities were 2,981 g.dw/$m^2$ in P. communis, 1,471 g.dw/$m^2$ in Z. sinica, 189g.dw/$m^2$ in S. herbacea and 71 g.dw/$m^2$ L. tetragonum, respectively. Seasonal changes of contents of inorganic nutrients per unit land area coincided with those of biomass of plant communities, however, the maximum contents of K occured earlier than the maximum biomass. Amounts of inorganic nutrients absorbed by plant were directly proportion to its biomass and it was true to reverse in restored amounts of them to soil. In turnover times of nutrients of the communities, it took the shortest time for P but the longest for Ca and P. communis community took the shortest but L. tetragonum the longest. For example, in P. communis turover time of P took one year and that of Na 1,440 years. Lack of P element, therefore, was expected in this study area.

In vitro Multiplication of Haloxylon recurvum (Moq.) - a Plant for Saline Soil Reclamation

  • Haloxylon recurvum (Locally known as Khar) is drought and salt tolerant plant of Thar Desert. This plant is a major biomass producer and has economic and ecological importance for the region. There is need for study on biology, propagation and genetic improvement for utilization of this plant for reclamation of saline soils. We report here on in vitro propagation of Haloxylon recurvum (Moq.) using nodal explant. Secretion of phenolic compound from explants was a major constraint for establishment of culture. This was checked by thorough washing and quick transfer of explant on fresh culture medium. Juvenile nodal explant with leaves was found suitable for culture establishment. Benzy-ladenine($4.0\;{\mu}M$) incorporated in Murashige and Skoog (MS) medium with additives (50 mg/L ascorbic acid and 25 mg/L each of adenine sulphate, arginine and citric acid) induced multiple shoots from nodal explant. Addition of $1.0\;{\mu}M$ naphthalene acetic acid (NAA) in combination with $4.0\;{\mu}M$ BAP improved the growth of axillary shoots. Further shoot amplification was achieved by repeated subculture of mother explants on fresh medium. Forty percent of the micropropagated shoots rooted on half-strength MS medium with $4.0\;{\mu}M$ indolebutyric acid (IBA) and 100 mg/L activated charcoal, at $28{\pm}2^{\circ}C$ and $60\%$ RH. Sixty percent of these plantlets were hardened in green house.

  • To get basic data for the utilization of S. herbacea L. as a raw material in food and Chinese herbs, chemical compositions of its leaves, stem and root were investigated. Leaves had the highest level of moisture and the lowest bevel of total sugar. The crude protein and crude lipid contents of the stem were similar to those of the root. Crude ash and salt contents (dry basis) in leaves were considerably higher than those of the stem and root. Total amino acid contents of leaves, stem and root were 1,270 mg/100 g, 1,525 mg/100 g, and 1,569 mg/100 g, respectively. Although the amino acid compositions of loaves, stein, and root were different, their major amino acids were glutamic acid, leuicine, isoleucine and aspartic acid. The rich minerals in leaves, stem and root were Na, K and Ca.

Anti-Inflammatory Activity of the Solvent-Partitioned Fractions from Spergularia marina in LPS-Stimulated RAW 264.7 Cells

  • As a part of ongoing research to elucidate and characterize antioxidant and anti-inflammatory nutraceuticals, solvent-partitioned fractions from Spergularia marina were tested for their ability to scavenge radicals and suppress inflammation. The results of the 2',7'-dichlorofluorescein diacetate assay indicate that solvent-partitioned fractions from S. marina scavenged intracellular radicals in $H_2O_2$-stimulated mouse macrophages. The tested fractions decreased the generation of nitric oxide (NO) and the expression of inflammation mediators, namely, inducible nitric oxide synthase (iNOS) and interleukin (IL)-6, by lipopolysaccharide (LPS)-induced mouse macrophages, indicating that S. marina decreases inflammation. Among all tested fractions [i.e., $H_2O$, n-buthanol (n-BuOH), 85% aqueous methanol (aq. MeOH), and n-hexane], the 85% aq. MeOH fraction showed the strongest antioxidant and anti-inflammatory response. The 85% aq. MeOH fraction scavenged 80% of the free radicals produced by $H_2O_2$-induced control cells. In addition, NO production was 98% lower in 85% aq. MeOH fraction-treated cells compared to LPS-induced control cells. The mRNA expression of iNOS and IL-6 was also suppressed in 85% aq. MeOH fraction-treated cells. The results of the current study suggest that the phenolic compound components of S. marina are responsible for its antioxidant and anti-inflammatory effects.

Comparison of Biological Activities of Korean Halophytes

  • Halophytes are expected to possess abundant secondary metabolites and various biological activities because of habitat in extreme environments. In this study, we collected 14 halophytes (Asparagus oligoclonos, Calystegia soldanella, Carex pumila, Chenopodium glaucum, Elymus mollis, Glehnia littoralis, Limonium tetragonum, Messerschmidia sibirica, Rosa rugosa, Salsola komarovii, Spergularia marina, Suaeda glauca, Suaeda maritima, and Vitex rotundifolia) native to Korea and compared their total polyphenol contents, antioxidant and anti-inflammatory activities. The total polyphenol contents of R. rugosa (27.28%) and L. tetragonum (13.17%) were significantly higher than those of the other 12 halophytes and L. tetragonum, R. rugosa, and M. sibirica showed significantly greater antioxidant activities than the other 11 halophytes, as determined by DPPH (2,2-diphenyl-1-picrylhydrazyl). A. oligoclonos, E. mollis, and C. pumila showed significantly greater anti-inflammatory activities than the other 11, as determined by NO (Nitric oxide) and $PGE_2$ (Prostaglandin $E_2$) levels. In contrast, these three extracts had normal and low total polyphenol contents among the 14 halophytes. Consequently, the total polyphenol content in the 14 studied halophytes appeared to be related to antioxidant, but not anti-inflammatory activity levels.

A report of 20 unrecorded bacterial species in Korea, isolated from soils of coastal areas in 2022

  • To obtain unrecorded bacterial species in Korea, various soils of coastal areas were collected from the Republic of Korea in 2022. After plating the samples on marine agar and incubating aerobically and anaerobically, approximately 1,700 bacterial strains were isolated and identified using 16S rRNA gene sequences. A total of 20 strains showed ≥98.7% 16S rRNA gene sequence similarity with validly published bacterial species but not reported in Korea, indicating they are unrecorded bacterial species in Korea. The unrecorded bacterial strains belonged to four phyla, six classes, 15 orders, 16 families, and 19 genera which were assigned to Blastomonas and Sphingomonas of the class Alphaproteobacteria; Pseudidiomarina, Kushneria, Salinicola, and Salinisphaera of the class Gammaproteobacteria; Evansella, Virgibacillus, and Paenibacillus of the class Bacilli; Cyclobacterium of the class Cytophagia; Pedobacter of the class Sphingobacteriia; and Demequina, Ornithinimicrobium, Blastococcus, Jatrophihabitans, Kineococcus, Glaciihabitans, Aeromicrobium and Streptomyces of the class Actinomycetes. The details of the 20 unreported species, including Gram reaction, morphology, biochemical characteristics, and phylogenetic position are also provided in the description of the strains.

Trimethylsilyl 유도체화 방법을 적용한 함초 추출물의 GC-MS 분석 (GC-MS analysis of Salicornia herbacea L. extracts with trimethylsilyl derivatization)

  • Salicornia herbacea, a noted halophyte, has been widely researched for its diverse physiological activities. The continuous exploration of its compounds is essential. This study employed gas chromatography (GC) coupled with mass spectrometry (MS) for qualitative analysis. This involved pretreatment including trimethylsilyl (TMS) derivatization of the S. herbacea extract, facilitating its GC analysis. Five compounds, including various fatty acids and β-sitosterol, were identified by direct analysis of the methanol extract of S. herbacea without pretreatment by GC-MS. Conversely, the analysis of the TMS-derivatized extract revealed 28 distinct peaks. Quantitative analysis further indicated that the predominant compounds in the S. herbacea extract were sugars and sugar derivatives, notably glucose, fructose, and glucitol. The collective concentration of these sugars and its derivatives amounted to 116.45 mg/g, representing 11.6% of the dry weight of the raw S. herbacea. Following sugars and their derivatives, fatty acids constituted the next most abundant group of compounds. However, the presence of amino acids and organic acids was relatively low.

A report of 20 unrecorded bacterial species isolated from island and coastal ecosystems in 2023

  • Various samples from island and coastal ecosystems in South Korea were investigated to discover unrecorded bacterial species. Soils from these areas, along with seawater samples, were plated on marine agar and R2A agar (containing 3% sea salt). From these samples, approximately 1,070 bacterial strains were isolated as single colonies and identified using 16S rRNA gene sequences. A total of 20 strains, which exhibited at least 98.7% similarity in their 16S rRNA gene sequences to those of validly published bacterial species not yet reported in Korea, were identified as unrecorded bacterial species. These strains belonged to three phyla, six classes, 10 orders, 14 families, and 16 genera. These were assigned as follows: Thioclava, Breoghania, Acidovorax, Erythrobacter, Paracoccus, Jiella, Aurantimonas, and Qipengyuania within the class Alphaproteobacteria; Pseudomonas, Cobetia, and Rheinheimera within the class Gammaproteobacteria; Aequorivita, Leeuwenhoekiella, and Polaribacter within the class Flavobacteriia; Algoriphagus within the class Cytophagia; and Microbacterium within the class Actinobacteria. The unreported species underwent further taxonomic characterization, which included assessments of Gram reaction, colony and cell morphology, biochemical and phylogenetic characteristics.

가사도 폐염전의 식생 현황에 관한 생태학적 연구 (Ecological Studies on the Vegetation of Abandoned Salt field in Gasado)

  • 본 조사는 2002년 7월부터 9월까지 진도군 가사도에서 도서연안 폐염전의 식생 현황을 조사하기 위하여 Braun-Blanquet and Elenberg의 방법에 따라 21개의 방형구(1${\times}$1m, 1${\times}$2m)를 설치하여 생태학적 조사하였다. 조사된 폐염전의 관속식물은 17과 48종이며, 이중에서 염생식물은 20종(42%), 중성식물은 28종 (58%)이 나타났다. 또한 폐염전에서 서식지의 교란은 염생식물의 교란으로 나타났으며, 이 지역에서 귀화식물은 오리새, 달맞이꽃, 망초, 코스모스 등의 23종이 분포하고, 이중에서 중성식물이 20종(71%) 분포하고 있었다. 식생군락은 염생식물 초지군락은 갯질경군락, 갯개미자리군락, 칠면초군락, 습지 초지군락은 갈대군락, 천일사초군락, 가는잎모새달군락, 중성 초지군락은 실망초군락과 갯당근군락 등으로 구분되었다. 각 군락들의 폐염전에서 식생의 구조, 분포, 변화에 대해서 설명하고, 식생표와 현존식생도를 작성하였다.

Inhibition of MMP-2 and MMP-9 Activities by Limonium tetragonum Extract

  • Matrix metalloproteinases (MMPs) are crucial extracellular matrices degrading enzymes that take important roles in metastasis of cancer progression as well as other significant conditions such as oxidative stress and hepatic fibrosis. Natural products are on the rise for their potential to provide remarkable health benefits. In this context, halophytes have been of interest in the nutraceutical field with reported instances of isolation of bioactive compounds. In this study, Limonium tetragonum, an edible halophyte, was studied for its ability to inhibit MMP-2 and -9 using HT1080 fibrosarcoma cells. Results showed that L. tetragonum extract was able to inhibit the enzymatic activity and mRNA expression of MMP-2 and -9 according to gelatin zymography and RT-PCR assays, respectively, but it was not able to significantly change the MMP pathway related factors such as tissue inhibitors of metalloproteinases. Also, Mitogen-activated protein kinases pathway-related protein levels and their phosphorylation were assayed. While the phosphorylated p38 levels were decreased, extracellular signal-regulated kinase and c-Jun N-terminal kinase were not affected by L. tetragonum treatment. In conclusion, it was suggested that L. tetragonum contains substances acting as MMP inhibitors on enzymatic activity rather than intracellular pathway intervention, which could be useful for further utilization of L. tetragonum as a source for anti-MMP agents.
