This experiment was conducted to obtain the have higher contents of pharmaceutical constituents as well as higher yield from colchicine induced diploid and tetraploid extracts of Platycodon grandiflorum. In order to determine the biological activity, this study was focused to evaluate the cytotoxicity, antimicrobial on the bronthus disease bacteria, antioxidant enzyme activity of diploid and tetraploid extracts in P. grandiflorum. The activities of antioxidant enzyme according to different solvent extracts were measured as superoxide dismutase (SOD), catalase (CAT), peroxidase (POD), and ascorbate peroxidase (APX). The cytotoxicity of methanol extracts of P. grandiflorum showed significant differences between tetraploid and diploid. That is, the cytotoxic effect against human cancer cell was higher in tetraploid than in diploid. At all extracts concentration, tetraploid samples showed high toxicity and the $IC_{50}$ (concentration causing 50% cell death) value showed the highest on HCT-116 cell ($105.91{\mu}g/mL$), and exhibited significant activity against the Hep 3B cell ($140.67{\mu}g/mL$), SNU-1066 cell ($154.01{\mu}g/mL$), Hela cell ($158.37{\mu}g/mL$), SNU-601 cell ($182.67{\mu}g/mL$), Calu-6 cell ($190.42{\mu}g/mL$), MCF-7 cell ($510.19{\mu}g/mL$). Antimicrobial activities of diploid P. grandiflorum were relatively low compared to tetraploid P. grandiflorum on most of the bacterial strains. In tetraploid P. grandiflorum, K. pneumoniae showed the clear zone formation (18~19 mm) of growth inhibition, followed by the clear zone formation of 13~15 mm on C. diphtheria and S. pyogenes. The antimicrobial activities in diploid P. grandiflorum were the highest on K. pneumonia (14~15 mm), and showed the clear zone formation of 11~12 mm on C. diphtheria and 12~13 mm on S. pyogenes. The antimicrobial activity is thought to look different depending on the bacterial strains and the polyploidy of P. grandiflorum. The root extract of P. grandiflorum had the highest (97.2%) SOD enzyme activity in ethyl acetate partition layer of tetraploid while water partition layer of diploid showed the lowest (48.6%) SOD enzyme activity. The activity of CAT showed higher values in the root of tetraploid than in the diploid of P. grandiflorum in all partition layers except butyl alcohol. The activities of APX and POD showed higher values in the root of tetraploid than in the diploid of P. grandiflorum in all fraction solvents except water layer. These results indicate that the tetraploid P. grandiflorum can be used as a source for developing cytotoxic agent and antimicrobials which can act against bronchus diseases bacterial strains.

This study was carried out to identify the potential protected areas recognized by experts to conform to the international definition of protected areas for effective implementation of CBD Aichi Target-11. A policy forum of 13 experts on the protected area was formed and conducted four forums, two surveys, and examination in the context of international perspective to identify four potential protected areas including the water source protection area, riparian buffer zone, fishery resources protected area, and urban natural park area. Excluding the existing protected areas, the total size of the potential protected areas was about $5,643.9km^2$ composed of the terrestrial areas of $3,117.9km^2$ and the marine areas of $2,526km^2$. As such, we can expect the expansion of coverage of national protected areas by up to 3.11% of the terrestrial land and up to 0.67% of the terrestrial marine area. This study is meaningful in that it provides the fundamental information to achieve the national target of protected areas in response to CBD Aichi Target-11. Further research on improving the protected areas qualitatively and quantitatively and identifying and linking with other effective regional OECM are required to enhance the national protected area system.

Preharvest sprouting seriously reduces milling and baking quality of hard winter wheat (Triticum aestivum L.) grain. To determine the effect of nitrogen fertilizer application on decreasing of preharvest sprouting, several levels of N-fertilization were conducted in two winter wheat cv. Keumkang and Jokyung, grown in Iksan. Nitrogen fertilization is used to increase grain yield and protein content. Grain yield increased at 108kg/ha (50% increased nitrogen to the standard) application and decreased as more nitrogen was applied. There was a linear increase in grain protein contents with increasing level of nitrogen application. Germination rate, germination index and ABA sensitivity were gradually reduced by increasing of nitrogen application level. Preharvest sprouting showed a significantly correlation to germination rate but could not be correlated to protein content and falling number. A significant positive correlation was detected between preharvest sprouting and different additional nitrogen fertilizer levels.
This study was carried out to investigate the effects of Ca-gluconate (Ca-glu) on fruit firmness and softening enzyme activities of hydroponically grown tomato (Solanium esculentum Mill.). The obtained results revealed that the rate of weight loss was markedly increased from at storage to 5 days after storage (DAS) in control, and was constantly increased until 7 DAS as 4.1% in Ca-glu treatment. Fruit firmness was more rapidly decreased in Ca-glu induced fruit compared to control. Results showed that fruit firmness in control and Ca-glu treated fruit were 0.67 and $0.95kg{\cdot}{\varphi}12mm^{-1}$, respectively. In our investigation, no difference was revealed in Hunter's 'a' value between control and Ca-glu treated fruit. Total carotenoids content of control fruit were rapidly increased while the Ca-glu treated fruit were gently increased. Lycopene content was higher ($63.3{\mu}g{\cdot}g^{-1}\;FW$) in control than Ca-glu treatment ($56.8{\mu}g{\cdot}g^{-1}\;FW$). The activity of Polygalacturonase (PG) was rapidly increased with increasing storage period as from 0.4 to 1.2 units whereas the PG activity of Ca-glu treatment was gently increased from 1 to 7 DAS, and rapidly increased from 7 to 11 DAS. However, the pectinesterase (PE) activity was rapidly increased in control fruit, when the storage period was increased, but interestingly, the Ca-glu treated fruit was slowly increased from 1 to 7 DAS, and rapidly increased 7to 11 DAS. ${\beta}$-galactosidase activity of Ca-glu induced fruit was rapidly increased from 1 to 7 DAS as from 1.6 to 3.0 units, and gently increased from 7 to 11 DAS. ${\beta}$-galactosidase activity of control were higher than Ca-glu treatment.

The roots of Platycodon grandiflorum species either dried or fresh, are used as an ingredient in salads and traditional cuisine in Korea. To interpret the root proteins, a systematical and targeting analysis were carried out from diploid and tetraploid roots. Two dimensional gels stained with CBB, a total of 39 differential expressed proteins were identified from the diploid root under in vivo condition using image analysis by Progenesis Same Spot software. Out of total differential expressed spots, 39 differential expressed protein spots (${\geq}\;1.5$-fold) were analyzed using LTQ-FTICR mass spectrometry. Except two proteins, the rest of the identified proteins were confirmed as down-regulated such as Isocitrate dehydrogenase, Proteasome subunit alpha type-2-B. However, the most of the identified proteins from the explants were mainly associated with the oxidoreductase activity, nucleic acid binding, transferase activity and catalytic activity. The exclusive protein profile may provide insight clues for better understanding the characteristics of proteins and metabolic activity in various explants of the economically important medicinal plant Platycodon grandiflorum.
102 ESTs (Expressed Sequence Tags) were obtained by sequencing clones from a library of rainbow trout kidney cDNAs. Of the sequences generated, 55.8% of the ESTs were represented by 37 known genes. The 45 clones of unknown gene products potentially represent 40 novel genes. The genes involved in structural function (14.5%) and transcription/translation (11.6%) account for the major gene expression activities in the kidney Microarray experiment was conducted to compare gene expression of the unique ESTs in young and adult rainbow trout kidneys. While mitochondrion, cytochrome b, rho G, spastin protein, and three unknown genes were down-regulated in the mature fish kidney, calponin 1, calcium binding protein, histone deacetylase 1, and an unknown gene were up-regulated in the mature fish kidney. This research demonstrates the feasibility and power of functional genomics in rainbow trout.
This study investigates the cultivation processes of major nuts and fruits such as Ginkgo biloba L., Vitis coignetiae, Actinidia arguta Planch, Rubus coreanus Miq., and Rubus crataegifolius and analyzes standard income in order to provide foundational data necessary to make reasonable policies related with the criteria of compensating the loss of forest products or diagnosis of forestry management. According to the research results, the types of cultivation by items can be largely divided into open field culture and mountainous culture. Regarding the average annual income per unit area (3.3) by items, the open field culture of Ginkgo biloba L. was 1,060 Won, mountainous culture of Ginkgo biloba L. was 618 Won, open field culture of Vitis coignetiae was 5,891 Won, mountainous culture of Actinidia arguta Planch was 8,113 Won, open field culture of Rubus coreanus Miq. was 14,701 Won, and open field culture of Rubus crataegifolius was 17,482 Won or so; thus, the average annual income of Rubus coreanus Miq. and Rubus crataegifolius was very higher than that of the other items. In particular, because the production of Rubus crataegifolius starts two years after the planting and they rely on family labor mainly for the cultivation, the expenses for employment and labor were analyzed to be low. In addition, the analysis shows that at most of the nuts and fruits cultivation, initial investment costs for facilities or seedlings occupy a great part as about 30% averagely out of the management cost, so it is thought that the government should provide proper support to vitalize the cultivation.
This study was conducted to investigate somatic embryogenesis capacity using callus derived from bud meristems in sweetpotato. Shoot apical meristem explants $(height:150{\mu}m;base:\;350{\mu}m)$were cultured on MS medium supplemented with 1 mg/L 2/4-D. Embryogenic callus were observed in five cultivars when their shoot apices were cultured on MS medium supplements with 1 mg/L 2,4-D. After 6 weeks of culture, greater than 80% of the survived explants produced embryogenic calli and the calli gave rise to somatic embryos at frequencies of 72% (Yulmi), 60% (Shinhwangmi), 78% (Geonmi), 70% (KoKei 14), 40% (Sinjami). The regenerated plants developed into whole plantlets after they were transferred onto the fresh hormon-free MS medium of 74% (Yulmi), 82% (Shinhwangmi), 86% (Geonmi), 74% (Kokei 14), 41% (Sinjami) respectively.

Overlay network eliminates the need to change the application-layer tree when the underlying network changes and enables the overlay network to survive in environments where nonmember nodes do not support multicast functionality. An overlay protocol monitors group dynamics, while underlying unicast protocols track network dynamics, resulting in more stable protocol operation and low control overhead even in a highly dynamic environment. But, if overlay multicast protocols does not know the location information of node, this makes it very difficult to build an efficient multicasting tree. So, we propose a Hierarchical Overlay Multicast Architecture (HOMA) with the location information. Because proposed architecture makes static region-based dynamic group by multicast members, it is 2-tired overlay multicasts of application layer that higher layer forms overlay multicast network between members that represent group, and support multicast between multicast members belonging to region at lower layer. This use GPS, take advantage of geographical region, and realizes a region-sensitive higher layer overlay multicast tree which is impervious to the movements of nodes. The simulation results show that our approach solves the efficiency problem effectively.

A dynamo set for a high-power induction motor drive is expensive and needs a long time to manufacture. Therefore, the development of a simulator that functions as the induction motor and load equipment is required. A load simulator of an inverter for a high-power three-phase induction motor consists of a reactor and three-phase PWM inverter. Therefore, it cannot simulate the dynamic characteristics of an induction motor and functions only as a load. In this paper, a real-time simulator is proposed to simulate a model of an induction motor and the load characteristics based on an LCL filter and three-phase PWM rectifier for a three-phase induction motor. The currents of a PWM inverter that simulate the stator currents of the motor are controlled by the inductor currents and capacitor voltages of the LCL filter. The capacitor voltages of the LCL filter simulate the induced voltages in the stator windings by the rotating rotor fluxes of the motor, and the capacitor voltages are controlled by the inductor currents and a PWM rectifier. The rotor currents, the stator and rotor flux linkages, the electromagnetic torque, the slip frequency, and the rotor speed are derived from the inverter currents and the motor parameters. The electrical and mechanical model characteristics and the operation of vector control were verified by MATLAB/Simulink simulation.
