A Study on 'Tang', Traditional Soup Originated in Kyoung-Nam Province (경남지방의 향토요리에 관한 연구 -탕류를 중심으로-)

  • Currentry, the originality of traditional soups in Kyoung-Nam province is vanishing by the inflex of processed and instant food, food industrial development. Especially, such factors as the influence of alien cooked, the develoment of transportation, the movement of population have made it hard to preserve the traditionnal food preparations. The purpose of this thesis was to seek out the various kinds of Kyoung-Nam province soups preparation. To do this, about 60 kinds of soups recipe were reviewed and charactered into several categories. The findings of the study are as follows. 1) Soup is made by fish or shellfish, freshwater fish. Meat and fowls is rarely used in Kyoung-Nam province (coastal region). 2) Soups of inland area is made by most vegetables and these soup were always harmonized with soybean powder or perilla powder. 3) Various soup season with the mag-jang, mul-jang sauce, hab-ja-jang sauce. 4) These soups were in general to make much red pepper and much salt.

A Case Report of Verruca Vulgaris Patient, Treated only with Korean Medicine (한약 단독 치료를 이용한 심상성 사마귀 1례 보고)

  • Objectives : The purpose of this study is to report a case of verruca vulgaris patient, treated only with Korean medicine. Methods : A 6-years-old female patient was treated only with Korean medicine due to verruca vulgaris on left hand. Photographs and size measurements of lesions were used to evaluate the changes in symptoms. Results : After 3 months of treatment, the patient's lesions showed almost complete clearance and there were no adverse effects. Conclusions : According to the results, Korean medicine with Bojungikgi-tang Hab Saengmaek-san Gamibang could be an effective and safe treatment for verruca vulgaris in a pediatric patient.

  • 국내 이상기후로 인해 여름철 하천과 호소에서 빈번하게 발생되는 조류의 과대성장이 매년 문제가 되고 있다. 하천 조류는 일차생산자로서 매우 중요한 역할을 하지만, 하천조류 중 유해남조류가 생장하면서 발생하는 악취 유발 물질과 독성물질의 배출로 문제를 야기하고 있다. 국내에서는 조류경보제와 수질예보제를 시행하여 국민의 안전을 도모하기 위해 최선을 다하고 있으며, 발령 기준은 유해남조류세포수에 따라 발령이 되기 때문에 유해남조류 측정은 매우 중요하다. 현재 조류의 분석방법은 현장에서 조류샘플을 채취하고 실험실에서 현미경을 통해 조류샘플을 검경하고 녹조류, 남조류, 규조류의 세포수 또는 우점종을 산정한다. 조류검경은 개인의 역량에 따라 오차가 생길수 있고 시간이 많이 소요된다. 최근 많은 연구자들이 이런 문제를 해결하기 위해 인공위성, 광학영상, 초분광영상 등을 통해 녹조류와 남조류 대체 인자인 Chlorophyll, Phycocyanin을 통해 조류농도를 추정하고 있으나, 조류세포수 분석에 대한 연구는 미비한 실정이다. 본 연구에서는 매년 조류 발생으로 문제가 되는 물금취수장 인근 하천에서 발생한 조류를 채취하고 조류검경을 통해 얻은 남조류세포수와, LISST-HAB를 통해 얻은 Phycocyanin농도, 초분광영상을 활용한 조류스펙트럼 데이터를 통해 남조류세포수 추정하고 남조류세포수와 비교분석을 진행하였다. 본 연구를 통해 조류측정 원격탐사 연구의 기초자료로 제공하고자 한다.

Increasing Sulforaphane Formation in Broccoli Sprouts by Radish Sprouts Additions

  • Cruciferous plants such as broccoli and radish contain glucosinolate, which is a bioactive precursor that is most often used in Korean foods and is unique as a food ingredient. In addition, it contains various other phytochemicals and is promising as a health-oriented food material. In particular, Sulforaphane is a hydrolyzate of the glucosinolate, which has a more beneficial effect on the human body. Glucosinolate may be hydrolyzed by enzymes called myrosinase, which is voluntarily possessed by cruciferous plants. However, the ESP(Epithiospecifier protein) in broccoli sprouts could acts competitively with myrosinase, and convert to the less bioactive sulforaphane nitrile form. Therefore, we improved the yielding of sulforaphane in broccoli sprouts with a new method. We induce inactivation of the ESP protein by heat treatment. At this time, a myrosinase was introduced from the radish sprout because myrosinase in broccoli sprouts is also denatured by heat treatment. According to the results, we have confirmed by GC / MS that formation of sulforaphane increases more than 7 fold using set heating and mixing conditions.

Rapid and exact molecular identification of the PSP (paralytic shellfish poisoning) producing dinoflagellate genus Alexandrium

  • The marine dinoflagellate genus Alexandrium comprise PSP producing A. acatenella, A. angustitabuzatum, A. catenella, A. fundyense, A. minutum, A. ostenfezdii, A. tamiyavanichii and A. tamarense. In monitoring toxic Alexandrium, rapid and exact species identification is one of the significant prerequisite work, however we have suffered confusion of species definition in Alexandrium. To surmount this problem, we chose DNA probing, which has long been used as an alternative for conventional identification methods, primarily relying on morphological approaches using microscope in microbial field. Oligonucleotide DNA probes targeting rRNA or rDNA have been commonly used in diverse studies to detect and enumerate cells concerned as a culture-indetendent powerful tool. Despite of the massive literature on the HAB species containing Alexandrium, application of DNA probing for species identification and detection has been limited to a few documents. DNA probes of toxic A. tamarense, A. catenella and A. tamiyavanichii, and non-toxic A. affine, A. fraterculus, A. insuetum and A. pseudogonyaulax were designed from LSU rDNA D1-D2, and applied to whole cell-FISH. Each DNA probes reacted only the targeted Alexandrium cells with very high species-specificity within Alexandrium. The probes could detect each targeted cells obtained from the natural sea water samples without cross-reactivity. Labeling intensity varied in the growth stage, this showed that the contents of probe-targeted cellular rRNA decreased with reduced growth rate. Double probe TAMID2S1 achieved approximately two times higher fluorescent intensity than that with single probe TAMID2. This double probe did not cross-react with any kinds of microorganisms in the natural sea waters. Therefore we can say that in whole-cell FISH procedure this double DNA probe successfully labeled targeted A. tamiyavanichii without cross-reaction with congeners and diverse natural bio-communities.

Characteristics of Algicide Produced by Micrococcus luteus SY-13 Inhibiting Cochlodinium polykrikoides and the Effects on Marine Organisms (적조생물 Cochlodinium polykrikoides를 저해하는 Micrococcus luteus SY-13이 생산하는 살조물질의 특성과 해양생물에 미치는 영향)

  • Algicidal bacterium was isolated from sea water during the declining period of Cochlodinium polykrikoides blooms and this bacterium had a significant algicidal activity against C. polykrikoides. In this study, algicidal bacterium was identified on the basis of biochemical and chemotaxonomic characteristics, and analysis of 16S rDNA sequences. The algicidal bacterium showed 98.6% homology with Micrococcus luteus ATCC $381^T$. Therefore, this bacterium was designated Micrococcus luteus SY-13. The optimal culture conditions of the algicidal bacterium was $25^{\circ}C$, initial pH 8.0, and 3.0% NaCl concentration. M. luteus SY-13 is assumed to produce secondary metabolites which have algicidal activity. When 10% culture filtrate of this strain was applied to C. polykrikoides ($1.0\;{\times}\;10^4\;cells/ml$) cultures, over 98% of C, polykrikoides cells were destroyed within 6 hours. The culture filtrate of M. luteus SY-13 exhibited similar algicidal activity after heat-treatment at $121^{\circ}C$ for 15 min. While algicidal activity remained in filtrates with pH adjusted to 8.0, loss of algicidal activity occurred when the pHs of filtrates were adjusted to over 9.0 or heat-treated at $121{\times}180^{\circ}C$ for 1 hour. M. luteus SY-13 showed significant algicidal activities against C. polykrikoides (98.9%) and a wide algicidal range against various harmful algal bloom (HAB) species. However, there was no algicidal effect on diatom and marine livefood organisms except Isocrysis galbana. These results suggest that M. luteus SY-13 could be a candidate for use in the control of HABs.

Methods for sampling and analysis of marine microalgae in ship ballast tanks: a case study from Tampa Bay, Florida, USA

  • Ballasting and deballasting of shipping vessels in foreign ports have been reported worldwide as a vector of introduction of non-native aquatic plants and animals. Recently, attention has turned to ballast water as a factor in the global increase of harmful algal blooms (HABs). Many species of microalgae, including harmful dinoflagellate species, can remain viable for months in dormant benthic stages (cysts) in ballast sediments. Over a period of four years, we surveyed ballast water and sediment of ships docked in two ports of Tampa Bay, Florida, USA. Sampling conditions encountered while sampling ballast water and sediments were vastly different between vessels. Since no single sample collection protocol could be applied, existing methods for sampling ballast were modified and new methods created to reduce time and labor necessary for the collection of high-quality, qualitative samples. Five methods were refined or developed, including one that allowed for a directed intake of water and sediments. From 63 samples, 1,633 dinoflagellate cysts and cyst-like cells were recovered. A native, cyst-forming, harmful dinoflagellate, Alexandrium balechii (Steidinger) F. J. R. Taylor, was collected, isolated, and cultured from the same vessel six months apart, indicating that ships exchanging ballast water in Tampa Bay have the potential to transport HAB species to other ports with similar ecologies, exposing them to non-native, potentially toxic blooms.

Lessons from the Sea : Genome Sequence of an Algicidal Marine Bacterium Hahella chehuensis (적조 살상 해양 미생물 Hahella chejuensis의 유전체 구조)

  • Harmful algal blooms (HABs or red tides), caused by uncontrolled proliferation of marine phytoplankton, impose a severe environmental problem and occasionally threaten even public health. We sequenced the genome of an EPS-producing marine bacterium Hahella chejuensis that produces a red pigment with the lytic activity against red-tide dinoflagellates at parts per billion level. H. chejuensis is the first sequenced species among algicidal bacteria as well as in the order Oceanospirillales. Sequence analysis indicated a distant relationship to the Pseudomonas group. Its 7.2-megabase genome encodes basic metabolic functions and a large number of proteins involved in regulation or transport. One of the prominent features of the H. chejuensis genome is a multitude of genes of functional equivalence or of possible foreign origin. A significant proportion (${\sim}23%$) of the genome appears to be of foreign origin, i.e. genomic islands, which encode genes for biosynthesis of exopolysaccharides, toxins, polyketides or non-ribosomal peptides, iron utilization, motility, type III protein secretion and pigment production. Molecular structure of the algicidal pigment was determined to be prodigiosin by LC-ESI-MS/MS and NMR analyses. The genomics-based research on H. chejuensis opens a new possibility for controlling algal blooms by exploiting biotic interactions in the natural environment and provides a model in marine bioprospecting through genome research.

Assessment of the Organic and Nitrogen Fractions in the Sewage of the Different Sewer Network Types by Respirometric Method (미생물 호흡률 측정에 의한 관거시스템 유형별 하수의 기질 분율 평가)

  • Respirometric analysis of domestic sewage by measuring oxygen uptake rate(OUR) was carried out for the experimental assessment of the organic and biomass fractions. The data of the organic and biomass fractions in sewage is essential for the activated sludge model to optimize the biological treatment plant. As a result of this study, the fractions of readily biodegradable substrate($S_S$), slowly biodegradable substrate($X_S$), inert soluble substrate($S_I$), inert particular substrate($X_I$) and heterotrophic biomass($X_{HAB}$) were about 26.6%, 41.5%, 8.5%, 14.7% and 8.7% on the basis of chemical oxygen demand($COD_{Cr}$), respectively. And the fractions of nitrogen were also studied. The fractions of soluble nitrate nitrogen($S_{NO}$), soluble ammonia nitrogen($S_{NH}$), soluble nonbiodegradable organic nitrogen($S_{NI}$), soluble biodegradable organic nitrogen($S_{ND}$) and slowly biodegradable organic nitrogen($X_{ND}$) were about 3.7%, 64.9%, 4.7%, 9.4% and 17.4%, respectively.
