Assessment of hemagglutination activity of porcine deltacoronavirus

  • Porcine deltacoronavirus (PDCoV) is an emerging swine enteric coronavirus that causes diarrhea in piglets. However, the biological characteristics of PDCoV are unclear. In this study, the hemagglutination (HA) abilities of two PDCoV strains (CH-01 and HNZK-04) were investigated. Our results showed that PDCoV has the ability to agglutinate rabbit erythrocytes after virion pretreatment with trypsin or neuraminidase. Additionally, the HA assay results showed a significant positive correlation with the infectious viral titer. Our results suggest that assessing the HA activity of PDCoV may be a useful diagnostic method for investigating and surveilling PDCoV infections.

Effects of Squalene on the Immune Responses in Mice(I): Humoral Immune Responses of Squalene

  • Effects of squalene on humoral immune system in mice were investigated. Squalene exhibited significant increases in the circulating leukocyte counts and relative spleen and thymus weights of the mice. However, the relative liver weight was slightly decreased. Hemagglutination titers (HA) were signficantly enhanced by squalene while Arthus reaction was not affected. Splenic plaque forming cells (PFC) were also greatly increased by squalene, especially at doses of 50 and 100 mg/kg of it.

The Analysis of the Value of the Thyroid Autoantibody Measured by Radioimmunoassay (방사면역측정법에 의한 갑상선 자가항체 측정의 기본적 및 임상적 검토)

  • To evaluate the values of the thyroid autoantibody measured by radioimmunoassay (RIA) and compare it with hemagglutination method (HA) in the normal and the thyroid disease, data were obtained from total 618 persons; 236 healthy persons, 217 patients with Graves' disease (including 113 patients with undertreated Graves' disease), 100 Hashimoto's disease, 31 thyroid nodule, and 34 simple goiter. RSR kit made in England was used and could be detected to at least 3 U/ml. The positive rates of normal group were antimicrosomal antibody (AMA) 31.8%, antithyroglobulin antibody (ATA) 44.5% by RIA and there was no considerable change in sex and age distribution. In Graves' disease, the positive rates of AMA and ATA were 90.4, 76.9% by RIA, 85, 39% by HA. In Hashimoto's disease, 94,91 % by RIA, and 87,48% by HA, respectively. The autoantibody titer by RIA in thyroid autoimmune disease as well as in normal group was more senisitive than that by HA, especially in ATA. There were linear relationships between the titer of RIA and that of HA in AMA of Graves' disease and AMA and ATA of Hashimoto's disease. There was no relationship among thyroid autoantibody, free $T_4$ index, TBII, and TSH. The titers of AMA and ATA were found to decrease in patients with Graves' disease during the course of antithyroid drug therapy. Of the 236 normal subjects, thirty-seven (15.7%) had concentrations of above 7.5 U/ml in AMA, forty. four (18.6%) above 9 U/ml in ATA. These values were considered as the upper limit for the normal range. In Graves' disease, 82.7, 53.8% were above 7.5, 9 U/ml, respectively; In Hashimoto's disease, 82, 79% were positive. We conclude that RIA was more sensitve than HA in measuring the thyoird autoantibody, but we will study further more for determining the normal range and its interpretation.

Studies on serological tests for pullorum disease (추백리의 혈청학적 진단법에 관한 연구)

  • In order to establish a sensitive and specific diagnostic method for detection of antibody to Salmonella pullorum, a enzyme-linked immunosorbent assay(ELISA) was designed and standardized. The diagnostic efficacy of the established ELISA was compared with that of the serum plate agglutination test and immunodiffusion test for pullorum disease. 1. The chicken hyperimmune sera to Salmonella pullorum, S gallinarum, S typhimurium and S typhi were shown the cross reaction to S pullorum antigen by serum plate agglutination test. 2. When compared the cross reaction titer of microplate agglutination test for chickens hyperimmune sera, it was found that the titer were 64 in S pullorum, 32 in S gallinarum, 4 in S typhimurium and 8 in S typhi, respectively. 3. When compared the specificity of various antigen(HA, EA, PA and SA) by the immunodiffusion test, the most suitable antigen was phenol-treated bactrium. 4. The optimal concentration of S pullorum antigen for ELISA was 1 : 160 dilution of bacterium. 5. The efficacy of the ELISA for detection of S pullorum antibody was compared with serum Plate agglutination test and immunodiffusion test in chickens infected with S pullorum. The antibody was first detected at 6 days after infection using three tests examined. The antibody was alldetected at 9 days by ELISA, at 12 days by serumplate agglutination test, at 15 days by immunodiffusion test.

Anti-inflammatory and Analgesic Activities of Water Extract of Root Bark of Ulmus parvifolia (참느릅나무 근피수침엑스의 소염.진통작용)

  • Ulmus parvifolia has been used as a traditional folk medicine to treat the carbuncle in deep skin. In this study, the effect of water extract of root bark of Ulmus parvifolia (WUP) on the carbuncle, pain, inflammation and hypersensitivity was evaluated in animal models. The administration of WUP significantly decreased the size of Staphylococcus aureus ($10^8$ cells/mouse)-induced carbuncle, and also exhibited analgesic activity in the HAc-induced writhing syndrome at doses of 50-500 mg/kg. It also showed significant anti-inflammatory activity in the carageenin- and complete Freund's adjuvant-induced inflammation. In the histamine-induced anaphylaxis, it decreased the percent of mortality by protecting mice treated with Bordetella pertussis. In the immune responses in the mice sensitized and challenged with sheep red blood cells, the Arthus reaction determined by swelling of foot pad at 4 h after challenge, HA titer, HY titer and PFC which can be used to evaluate the humoral immune response were significantly suppressed by oral administration of WUP at doses of 100 and 200mg/kg. The cellular immune responses in the same mice such as delayed type hypersensitivity determined by swelling of foot pad at 24 h after challenge and RFC were also significantly suppressed in the same manner.

Effects of Evening Primrose Oil on the Immune Responses in Mice (월견초종자유가 생쥐의 면역반응에 미치는 영향)

  • The purpose of this experiment was to investigate both the immunomodulatory effect of evening primrose(EP) oil and the effects of EP oil on immunoregulation by cyclophosphamide in mice. EP oil at doses of 0.1, 0.2 and 0.4 ml/kg were orally administered to ICR male mice once daily for 28 consecutive days. Cyclophosphamide was injected intraperitoneally to ICR mice with a single dose of 5 mg/kg at 2 days before secondary immunization. Mice were sensitized and challenged with sheep red blood cells(S-RBC). Immnune responses were evaluated by humoral and cellular immune responses and non-specific immune response. The results of this study were summarized as follows; (1) The humoral immune responses such as hemagglutination titer(HA), hemolysin titer(HY), Arthus reaction and plaque forming cell(PFC) were significantly enhanced in the low dose EP oil administered groups(0.1 and 0.2 ml/kg). However, in the high dose EP oil administered group(0.4 ml/kg) the responses were significantly lowered. (2) In the case of cellular immune responses, delayed type hypersensitivity reaction(DTH) was significantly decreased in EP oil whereas rosette forming cell(RFC) was remarkably enhanced. (3) Activities of natural killer cells and phagocyte were generally enhanced in EP oil. In addition, serum albumin and globulin were also increased.

  • To find compounds of immunomodulating and anti-allergic function, effects of protein-bound polysaccharides extracted from Phellinus igniarius (PI), Fomitella fraxinea (FF) and Agrocybe cylindracea (AC) on hemagglutinin titer (HA), hemolysin titer (HY), plaque forming cell (PFC), rosette forming cell (RFC) and phagocytosis were investigated in BALB/C mice. The oral administration of the protein-bound polysaccharides of PI, FF and AC for 10 days resulted in the enhanced phagocytic activity of peritoneal exudate cells (PEC), spleen cells (SC) and blood lymphocyte cells (BLC). Moreover, PI showed the activating effect on the phagocytosis of PEC and AC in SC. In the experiment of PFC and RFC, the results of the experimental group which was given each samples as compared to the control group, showed the enhanced level of activity such as PI 130%, FF 90% and AC 70%. Generally, HY and HA showed from ten to hundred times of level in each sample groups, as compared to the control group.

  • This study was performed to investigate the effect of olive oil diet on the immune response in ICR male mice. Experimental diets of 4 groups were fed ad libitum to the ICR male mice for 27 days. The results of this study were summarized as followings: 1. 10% Olive oil diet group as compared with the control diet group significantly decreased liver weight rate but significantly increased hemagglutination titer (HA), Arthus reaction, delayed type reaction (DTH), rosette forming cell (RFC), and phagocyte activity. 2. 20% Olive oil hypersensitivity diet group as compared with the control diet group significantly increased body weight gain, liver weight rate, and HA but significantly decreased Arthus reaction, DTH, RFC, phagocyte activity, and peripheral circulating white blood cell (WBC). 3. 30% Olive oil diet group as compared with the control diet group significantly increased liver weight rate but significantly decreased body weight gain, Arthus reaction, plaque forming cell (PFC), DTH, RFC, phagocyte activity, and WBC. The results showed that the increase of olive oil doses significantly decreased humoral and cellular immune responses, phagocyte activity, and WBC.

Virological studies on the puppies experimentally infected with canine parvovirus isolated in Korea (환견으로 부터 분리한 canine parvovirus의 실험적 감염 자견에 대한 바이러스학적 연구)

  • To investigate the pathogenicity and virological properties of TJ-89-2 strain of canine parvovirus(CPV -2) that was isolated from the diseased puppies in Korea, seven puppies-were inoculated intraorally with the isolate at the HA titer of 8,192. All of the puppies showed the signs of anorexia, vomiting, diarrhea and died at the 5th to 10th days post inoculation (pi). All of the fecal samples from the puppies revealed significantly high HA titers afterward the 5th days pi. Body temperature and the number of total leucocytes were slightly increased at the early stage of infection, but extremly decreased at the stage of collapse. HI titers of the sera began to increase at the 3rd to 4th days pi, reaching 512 to 1,024 at the 4th to 5th days pi.

  • A lectin has been purified from the bark of the legume Maackia fauriei. This lectin, MFA, was found to agglutinate human ABO erythrocytes at a titer of 256. The results from electrophoretic analyses, gel-filtration chromatography, and enzyme linked lectinsorbent assay indicate that MFA is an acidic glycoprotein, and exists as a tetramer of 30 kDa subunits that are linked by noncovalent bonds. The activity of MFA is critically dependent upon CaC1$_2$. MFA demonstrated high homogeneity with the lectins from M. amurensis, which is the only legume source of lectins that bind to sialoglycoconjugate, in its N-terminal amino acid sequence and amino acid composition, (omitted)
