DNA barcode and phylogenetic study of the tribe Desmodieae (Fabaceae) in Korea (한국산 도둑놈의갈고리족(콩과)의 DNA 바코드 및 계통학적 연구)

  • Species identification for the Korean tribe Desmodieae was conducted using the DNA barcoding genes rbcL, matK (from chloroplast DNA) and ITS (from nuclear ribosomal DNA). A total of 25 taxa (n = 75) in five genera were sequenced, and neighbor-joining trees were constructed using different combinations of DNA barcodes. When comparing these phylogenetic trees, a tree with all loci combined (rbcL + matK + ITS) showed the highest rate of identification success (72%). On this tree, two subtribes and five genera within the tribe were supported as monophyletic. In the Desmodiinae clade, Desmodium and Hylodesmum were more closely related to each other than to Ohwia. In the Hylodesmum clade, H. oldhamii was found to be a sister to H. podocarpum complex, and all taxa within the complex were identified successfully. Subsp. fallax, regarded as a variety of subsp. oxyphyllum, is closely clustered with subsp. podocarpum. Although var. mandshuricum has been regarded as a synonym of var. oxyphyllum, this taxon is supported as a distinct variety. For the Lespedezinae clade, all species of Kummerowia were monophyletic, while nine of 16 Lespedeza taxa were identified successfully. In particular, the resolution of Macrolespedeza (28.5%) was lower than that of Junceae (77.8%). Among the Lespedeza taxa, L. cuneata was distinguishable from L. lichiyuniae, despite morphological similarities. It has been suggested that both L. maritima and L. inschanica are hybrids. The former is thought to be an independent species. While it is difficult to determine whether the latter originated via hybridization, this study showed that it is closely related to L. juncea.

Evaluation of Korean Native Ducks on Production Efficiency Factor, Carcass Yield, Partial Meat Ratio and Meat Quality with Weeks (토종오리 대형종의 주령별 생산지수, 도체 수율, 부분육 비율 및 육질 평가)

  • This work was carried to evaluate production efficiency factor, carcass yield and meat quality with weeks of Large-type Korean native ducks. Korean native ducks (n = 90) from National Institute of Animal Science (RDA, Korea) were used in this work. Ninety ducks were divided into 6 groups (15 birds/group) and were fed with meat-type duck diets for 8 wk old. When ducks grew at specific wk (6, 7 and 8 weeks), 2 ducks per group were slaughtered at 6, 7 and 8 wk old. Production efficiency factor, carcass yield, partial meat and meat quality were researched in this work. There was no significant difference on livability with weeks, but body weight at 7 and 8 wk old was higher than that at 6 wk old (P<0.05). Feed conversion ratio at 6, 7 and 8 wk old were 2.25, 2.69 and 3.21, respectively, so there was significant difference with weeks (P<0.05). Production efficiency factor at 6, 7 and 8 wk old were 256.6, 199.8 and 153.0, respectively, so there was significant difference with weeks (P<0.05). Carcass yield at 8 wk old was higher than that at 6 and 7 wk old as 73.5% (P<0.05). Lightness at 6, 7 and 8 wk were 41.8, 39.0 and 38.1, respectively, and that at 6 wk old was the higher than other weeks (P<0.05). There was no significant difference on redness at 6, 7, and 8 wk old (P>0.05) and yellowness at 8 wk old was higher compared to other weeks (P<0.05). Cooking loss was the highest at 6 wk old as 31.6%, but water holding capacity was the highest at 8 wk old (P<0.05). There was no significant difference on shear force among weeks. pH at 6 wk old was the lower than that of other weeks as 5.84. Moisture content significantly decreased with weeks (P<0.05) and fat content at 8 wk was the highest as 1.88% (P<0.05). Protein content significantly increased with weeks until 20.9% at the age of 8 wk (P<0.05). Ash content at 7 and 8 wk old was the higher than that at 6 wk old (P<0.05). There was no significant difference on juiciness, tenderness, and flavor with weeks. Finally, these results may provide that shipping time at 7 wk old preferred to that at 6 and 7 wk old, but further research was needed because of deficiency of data.

Effects of Coastal Mud-Flat Bacteria Origin Pretense Supplementation on Laying Performance, Nutrient Digestibility and Total Protein Concentration of Serum in Laying Hens (갯벌 미생물 유래 단백질 분해 효소제의 급여가 산란계의 산란 생산성, 영양소 소화율 및 혈청내 총 단백질 함량에 미치는 영향)

  • This study was conducted to investigate the effects of coastal mud-flat bacteria origin protease supplementation on laying performance, nutrient digestibility and total protein concentration of serum in laying hens. A total of 252 laying hens were randomly allocated into three treatments with seven replications for eight weeks. Dietary treatments included 1) CON (basal diet), 2) PRO1 (basal diet + 0.05% protease) and 3) PRO2 (basal diet + 0.1% protease). During the entire experimental period, hen-day egg production was not affected by treatments (P>0.05). Difference of yolk height was increased in PRO1 treatment compared with CON treatment (P<0.05). Difference of egg weight was increased in PRO2 treatment compared with CON and PRO1 treatments (P<0.05). Shell quality, yolk color unit, haugh unit and egg yolk index were not affected by treatments (P>0.05). DM digestibility was improved in CON and PRO2 treatments compared with PRO1 treatment (P<0.05). N digestibility was improved in PRO2 treatment compared with CON treatment (P<0.05). Total protein concentration in serum were not affected by treatments (P>0.05). In conclusion, mud flat bacteria origin protease was effective for improving egg weight, yolk height and nutrient digestibility in laying hens.

Yields of oyster mushroom(Pleurotus ostreatus) on addition rate of Juglans mandshurica sawdust (가래나무(Juglans mandshurica) 톱밥의 첨가량이 느타리버섯의 수량에 미치는 영향)

  • This study was carried out to investigated optimum mixing ratio of Korean natural Juglans mandshurica using as functional plants. Total nitrogen and carbon source of J. mandshurica was 0.21% and 46.0%, respectively and C/N ratio was 219. Total nitrogen source and pH of substrate mixed with J. mandshurica was 2.3~2.9 and 5.0, respectively. The contents of CaO, MgO and $Na_2O$ at J. mandshurica media were higher at harvest media than inoculation media. Mycelial growth was the fastest at J. mandshurica 20%, and slower by increase of J. mandshurica substrate. Yields of fruiting body show the highest to 163 g/850 mL of medium which are addition 10% of J. mandshurica sawdust, and diameter and thick of pileus were the highest, too. The L value of pileus and stipes were increased by increase of J. mandshurica substrate, but there was no significant difference in the a-value and the b-value. The contents of $P_2O_5$ and $K_2O$ of fruiting body were increased at J. mandshurica substrate, but there was no significant difference in contents of CaO, MgO and $Na_2O$. The contents of Cu of fruiting body was decreased by increase of J. mandshurica substrate, but Fe, Mn and Zn were increased.

Legal Issue in Case of Death or Injury of an International Crew While on Board (국제항공운송 승무원이 항공기내에서 사상(死傷)을 당한 경우 법률관계 - 국내외 판례의 분석을 중심으로 -)

  • Air passengers may be compensated for damages based on the above agreement when the passenger suffers an accident to the extent that they are recognized as an accident under Article 17 of the Montreal Convention in 1999. If a flight or cabin crew and passengers both undergo an accident, passengers are subjected to compensation under the Montreal Convention however flight cabin crews will be compensated by the Labor Law, which is the governing law in the labor contract with the airline. The flight or cabin crew boarding the aircraft work is on a work contract, not a passenger transport contract. Therefore, if the flight or cabin crew on the aircraft is injured due to an accident, and the air carrier is liable for default due to a labor contract, the Labor Law, workers or survivors claim damages due to illegal acts against the employer. In which case, civil law will apply. In this regard, if a Chinese cabin crew working for a Chinese airline dies due to an accident in the Republic of Korea, whether the family of the deceased claims damages against the Chinese airline or not has international court jurisdiction in the Republic of Korea, which is the place of tort. We examined whether it is the law of the Republic of Korea or whether it's the Chinese law, the law applicable to the work contract, is applied. Also, Seoul District Court 1995.5.18. The sentence 94A 14144 was found that if the injured crew during the flight work was not satisfied with the insurance compensation under the Labor Standards Act and the Industrial Accident Compensation Insurance Act, he could claime to damage under the civil law against an air carrier or third parties responsible for the accident. This law case shows that you can claim a civil damage as a cause. In case of death due to an existing illness while on the way to work, the Korea Workers'Compensation and Welfare Service did not recognize the death of the deceased as an occupational accident, and the trial was canceled by the parents of the deceased for the survivor's benefit and funeral expenses. (Seoul Administrative Court 2017.8. 31. Although the sentence was judged as an occupational disaster in 2016, the 2016 8816 Decision), it was defeated in the appeals court (Seoul High Court 2018.7.19.Sentence 2017 No. 74186) and I criticized the judgment of the appeal by analyzing the deceased's disease and related the cause of it to workload. Sometimes, a flight or cabin crew is on board not for the flight duty such as transferring to another flight or returning to the home base or lay-over place after their scheduled flight, this is called "Deadheading". If the crew who is not considered the same as a passenger, but is not on duty, is injured in an accident, does the crew claim compensation for damages under the labor contract or whether the Montreal Convention is applied to the passenger. In conjunction with the discussion, there was a similar case, In re Mexico City Aircrash of October 31, 1979, 708 F.2d 400 (9th Cir. 1983), Demanes v. United Airlines, 348 F.Supp. 13 (C.D.Cal. 1972), Sulewski v. Federal Express Corp., 749 F.Supp. 506 (S.D.N.Y. 1990) and reviewed by the European Court of Justice (CJEU) at Wucher Helicopter GmbH and Euro-Aviation Versicherungs AG v. After examining several acts in several countries it's undeniably crucial to clearly understand the definition of "passenger" as stated in the Fridolin Santer case.

Physicochemical Properties of Olive Flounder Paralichthys olivaceus, Red Seabream Pagrus major and Jacopever Sebastes schlegeli (넙치(Paralichthys olivaceus), 참돔(Pagrus major) 및 조피볼락(Sebastes schlegeli)의 이화학적 성분 특성)

  • Olive flounder (Paralichthys olivaceus) including red seabream (Pagrus major) and jacopever (Sebastes schlegeli) were known to be suitable characteristics for preparing a processed food because of white meat fish containing low fat in muscle. In this study, the physicochemical properties of olive flounder (46-50 cm of length, 950-1,050 g of weight), red seabream (30-33 cm of length, 1,250-1,350 g of weight) and jacopever (20-23 cm of length, 550-650 g of weight) were studied to obtain a basic data for the development of a new processed food. Head, scale, bone and viscera of each samples were removed, and then grinding with homogenizer before experiment. Moisture content of red seabream is lower than that of olive flounder and jacopever, therefore, crude protein and crude lipid are higher content. Red seabream showed higher redness, yellowess and color difference value than those of olive flounder and jacopever except lightness. The major total amino acid in olive flounder, seabream and jacopever were glutamic acid, lysine and aspartic acid in order. Especially, the highest content of free amino acid was lysine for olive flounder and jacopever and alanine for red seabream. The highest content of mineral in olive flounder, seabream and jacopever was potassium (K) and then phosphorous (P) and sodium (Na) in order. The highest amount of fatty acid in olive flounder, red seabream and jacopever was polyenoic acid in which especially contained 6.8, 7.1 and 6.7% for EPA, and 15.2, 15.2 and 17.1% for DHA, respectively. From the result of sensory evaluation, there was no significant difference in color and odor, but not in taste and texture.

Changes of characteristics of livestock feces compost pile during composting period and land application effect of compost (축분 퇴비화과정 중 특성변화와 축분퇴비 이용효과)

  • Composting of livestock feces is economic and safe process to decrease the possibility of direct leakage of organic pollutants to ecosystem from commercial and environmental point of view. This study was conducted with three different experiments related to composting of livestock feces. The purpose of experiment 1 was to investigate changes of characteristic of compost pile during composting period by low temperature in cold season. To compare composting effect of experimental compost pile and control pile exposed in cold air, experimental compost piles were warmed up by hot air until their temperatures were reached at $35^{\circ}C$. Sawdust, Ricehull and Ricestraw were mixed with livestock feces as bulking agent. The highest temperatures of compost pile during composting period were in sawdust, rice hull, rice straw, and control were $75^{\circ}C$, $76^{\circ}C$, $68^{\circ}C$, $45^{\circ}C$ respectively. Moisture content, pH, C/N and volume of compost were decreased during composting period. Experiment 2 was carried out to study utilization effect of compost by plant. A corn was cultivated for 3 years on fertilized land with compost and chemical fertilizer. The amount of harvest and nutrition value of corn were analyzed. In first year of trial, the amount of harvest of corn on land treated with compost was lower by 20% than that of land treated with chemical fertilizer. In second year, there was no difference in yield of com between compost and chemical fertilizer. In third year, the yield of com on land fertilized with compost was much more than that of land fertilized with chemical fertilizer. The purpose of experiment 3 was to estimate the decrease of malodorous gas originating from livestock feces by bio-filter. Four types of bio-filters filled with saw dust, night soil, fermented compost and leaf mold were manufactured and tested. Each bio-filter achieved 87-95% $NH_3$ removal efficiency. This performance was maintained for 10 days. The highest $NH_3$ removal efficiency was achieved by leaf mold on the first day of operation period. It reduced the concentration of $NH_3$ by about 95%. Night soil and fermented compost showed nearly equal performance of 93 to 94% for 10 days from the beginning of operation. The concentration of hydrogen sulfide and methyl mercaptan originating for compost were equal to or less than $3mg/{\ell}$ and $2mg/{\ell}$, respectively. After passing throughout the bio-filter, hydrogen sulfide and methyl mercaptan were not detected.

Studies on the Laboratory Test of Lime Requirement (실험실(實驗室)에서의 석회소요량(石灰所要量) 측정방법(測定方法)에 관(關)한 연구(硏究))

  • A simplest and best method to find out lime requirement for Korean soils is described. Twenty six top soil samples were collected from various localities and chemical characteristics were dertermined. Three different buffer curve methods were employed as the standard to find out the most reliable test for lime requirement. In addition, the lime requirement test by the Shoemaker's and Woodruff's as well as the O.R.D. kit method were compared with the most reliable buffer curve method. The results may be summarized as follows. 1. The regression equation of the most reliable lime requirement by buffer curve method, y, on percent base saturation of various soils, X, was y=9.69-0.106 X and the negative correlation coefficient r=-0.84 was statistically significant at 5% level. 2. Lime requirement test by the best, buffer curve, method was more well correlated with the sum of exchangeable hydrogen and exchangeable aluminum contents of the soil samples than the exchangeable hydrogen content only. 3. The ordinary buffer curve method for lime requirement is more reasonable than the lime requirement test by the buffer curve prepared after addition of 1 normal potassium chloride or calcium chloride solutions. 4. The lime requirement tests by the most reasonable buffer curve method, X, and by the Woodruff's, the Shoemaker's and O.R.D. kit were compared. The best method for laboratory lime requirement test was the Shoemaker's method and the O.R.D. kit method also was good enough for the field test and these two method are recommended as the unified method of lime requirement test in each soil testing laboratories throughout Korea.

Microbiological Contamination Levels in the Processing of Korea Rice Cakes (떡류의 제조공정별 미생물학적 오염도 평가)

  • This study was conducted to evaluate microbial contamination levels of Korea traditional rice cakes such as Sirutteok, Garaetteok and Gyeongdan in the manufacturing process and environment. The microbial contamination levels such as total aerobic bacteria, fungi, coliforms, Escherichia coli, Staphylococcus aureus, Bacillus cereus and Clostridium perfringens of rice cake products were analyzed. The contamination levels of total aerobic bacteria, coliforms, fungi and B. cereus in raw materials were in the range of 2.4~4.5, ND~1.9, 1.2~2.1 and 1.0~2.1 log CFU/g, respectively. The microbial contamination levels of total aerobic bacteria, coliforms, fungi and B. cereus in manufacturing process of rice cakes were increased in the soaking and grinding steps and were decreased in steaming step. E. coli, S. aureus and C. perfringens were not detected in any manufacturing process and environment. The microbial contamination levels of raw materials and final products of rice cake were suitable for microbial safety standard in Korea. However, the manufacturing environment such as equipments and employee's sanitation were in trouble for microbial safety. The results of this study suggest that safety educatio n for personal hygiene and safetymanagement in processing environment are continuously required to assure safety in working environment and employee's individual hygiene.

Growth Performance of Pangasiid Catfish, Silver carp and Catla in Polyculture (복합양식에서 pangasiid catfish, silver carp과 catla의 성장률)

  • The growth performance of pangasiid catfish (Pangasius hypophthalmus), silver carp (Hypophthalmichthys molitrix) and catla (Catla catla) was evaluated in polyculture at varied combinations and stocking ratios in earthen pond for a period of 16 weeks. Combinations of pangasiid catfish, silver carp and catla with the total density of 30,000 fish per hectare were evaluated. A commercial pelleted feed containing 28% crude protein and 6% lipid was fed only to pangasiid catfish at the rate of 8% of body weight during the first six weeks, 6% during the second six weeks and 4% thereafter twice daily. The growth and feed utilization efficiency of pangasiid catfish were found similar in all treatments. The growth of silver carp was markedly decreased upon the increase of its own stocking density. The highest growth of silver carp achieved in polyculture of pangasiid catfish, silver carp and catla at 2:1:1 ratio where density of silver carp was lowest. The growth of catla was found to be affected by the presence of silver carp. The growth rate of catla markedly declined with the increase of silver carp density. The highest growth of catla obtained in polyculture of pangasiid catfish only with catla stocked at 1:1 ratio. The highest yield and economic return achieved in polyculture of pangasiid catfish, silver carp and catla at the ratio of 2:1:1 due to higher growth of silver carp and higher market value of catla. The water quality conditions in all different treatment ponds were within optimum ranges throughout the culture period and the values did not show any significant variation (p>0.05) among the treatments.
