This study was carried out to identify the effect of sucrose, calcium, and boron on the increase of pollen germination frequency in peach $(Prunus\;persica\;_{SIEB})$ under various temperature levels. The highest pollen germination was obtained at $20^{\circ}C$. Pollen germination was completed in 3.5 hrs. after planting of pollen on the basal medium which was composed of 1% agar and 10% sucrose, and conditioned pH 5.5. The optimum concentrations of sucrose and $H_3BO_3$ at $25^{\circ}C$ were 10% and $100\;mg\;{\cdot}\;L^{-1}$, respectively. However, $CaCl_2$ inhibited pollen germination in peach. Eight peach cultivars showed significantly different germinabilities at $10^{\circ}C$ on the basal medium. 'Nagasawa-Hakuho' showed the highest germinability of 26.1%, and 'Baekmijosaeng' showed the lowest germinability of 0.1%. $H_3BO_3$ strongly promoted pollen germination frequency when added to the germination medium at $10^{\circ}C$. Optimum concentration was $100\;mg\;{\cdot}\;L^{-1}$.
In an attempt to develop an unique enzyme (inulinase) for fructan utilization. bacterial strains were isolated [yom soil. Stram 96-11 secreting inulinase o[ high activity was tentatively identificated as Arthrobacter protophmmiae/ranwsus. The optimum culture conditions o[the slnin for the production of the inulinase were as follow: inorganic saIl basal medium contained sources fl % (w/v) inulin, 1 % (w/v) tryptone, and 1 % (w/v) $NH_4Cl$]. $35^{\circ}C$, initial pH 7.5. aeration 1 vvm and agitation 200 rpm.
Six protein-degrading bacteria were isolated from digestive organ of Harmonia axyridis. These isolates were categorized as Staphylococcus sciuri subsp. sciuri (3 strains), Bacillus subtilis (1 strain), and Bacillus thuringiensis (2 strains) by 16S rRNA gene sequence analysis. The Staphylococcus sp. CB2-3 was selected as a protease-producing bacterium which showed the highest protease activity of 58.5 U/ml at the pH 5.0 medium. The optimal pH and temperature of protease activity were pH 5.0 and $40^{\circ}C$, respectively. This acid protease had a relatively high stability of 80% between $30-50^{\circ}C$ at broad temperature range. The opimal medium compositions of carbon, nitrogen and mineral source for cell growth and protease activity were investigated. When sorbitol (0.5%) was used as carbon source, enzyme activity was increased about 2 times than that of the basal medium. When skim milk (0.5%) was used as nitrogen source, activity was increased about 2.5 times than that of the control. Cell growth and enzyme activity were increased by mineral source such as KCl, $K_2HPO_4$, $FeSO_4$, but was completely inhibited by divalent ions such as $Co^{2+}$, $Zn^{2+}$, $Mn^{2+}$, $Cu^{2+}$.
The production of single cell protein using the amylolytic fusant obtained from cell fusion between Hansenula anomala and Saccharomyces cerevisiae was studied. The fusant12 strain was selected for single cell protein production from starchy materials among five fusants. Optimum nitrogen source and its concentration for the growth of fusant12 were ammonium sulfate and 0.1%, respectively. Optimum concentration of soluble starch and optimum pH of the basal medium were lord and pH 5.6, respectively. Autolysis of fusant12 was effectively carried out by addition of 5% (v/v) ethyl acetate to the cell suspension and liquidization for 30 min before incubation for 24 hr at 3$0^{\circ}C$. Coculture of fusant12 and non-amylolytic yeast, Torulopsis candida YA-l5, resulted in the increase of the mass as compared to the monoculture of fusant12. The cell mass on tapioca medium was increased about 2.5 times as on soluble starch medium. The content of crude protein and nucleic acid of the dry cell were 39% and 5.8%, respectively.

To obtain functional materials from a submerged culture of Hericium erinaceum, a suitable basal medium for flask culture was screened and the optimal culture conditions in a jar fermenter were investigated with the addition of ginseng extracts (GE) to the basal liquid medium. Of all tested basal liquid media, the mushroom complete medium (MCM) supplemented with 0.5% of GE produced the highest mycelial dry weight (MDW) of 5.91 g/L in the flask, which reached a plateau at $25^{\circ}C$, pH 5.5 after 10 days. The submerged culture conditions for the mass production of mycelia in a 50 L jar fermenter were also optimal at $25^{\circ}C$, pH 5.5, 120 rpm agitation speed and 0.4 vvm aeration rate. Under these conditions, the maximum MDW was produced, which reached a value of 4.28 g/L within 5 days. When we investigated the effects of the amount of GE in the MCM on the production of MDW in the jar fermenter, the addition of 5% GE (HE-GE-5) under the optimal culture conditions produced the maximum MDW (4.93 g/L). In addition, the crude polysaccharide of HE-GE-5 contained mainly neutral sugars (63.2%) with considerable amounts of uronic acid (19.3%) and a small amount of proteins (8.8%) and it had potent immunostimulation properties.

It was investigated under several cultural conditions to produce biomass directly from starch by an strictly aerobic and amylolytic yeast, Sporobolomyces holsaticus FRI Y-5. Its optimal temperature and initial pH of medium for growth were $23^{\circ}C$ and 6.9, respectively. Activation energy, Ea, for growth was calculated to be 17.33 Kcal/mole from the Arrhenius relationship. When each of 13 nitrogen sources was added to the basal medium, $(NH_2)_2CO$ had the best effect, on which concentration of cell after 3 day incubation was 10.6 g/l and cell yield was 0.451. The yeast growth was affected by $MgSO_4,\;K_2SO_4\;and\;ZnSO_4$ as a mineral source and was best on the medium containing all of them. The addition of yeast extract (5g/l) could enhance the production of biomass and cell yield to 77% and 32%, respectively.
For the selection of powerful antagonistic bacterium for biological control of soil borne Eminia carotovora subsp. carotovora causing rot of vegetable, excellent strains (S4, S14, 565) were selected from 1,196 strains of bacteria which were isolated from rhizosphere in vegetable root rot-suppresive soil. Strains were identified to be Pseudomonas species with Api 20NE kit. Antagonistic substance was produced in 523 synthetic broth medium at pH 7~8 and $30^{\circ}C$ during 3 days culture. The substance was stable in the pH range of 6 to 9. When the basal medium was supplemented with mannitol and sorbitol as carbon source and calcium chloride as metal salt, the production of the inhibitory substance was increased. The inhibitory acitivity was increased by the addition of fertilizer in soil. The isolated strains were resistant to the agricultural chemical such as benomyl and fosethyl-Al-folpet, and the antibiotics such as penicillin and lincomycin. We had found that Pseudomonas sp. S14 strain had a single plasmid. After treated with acridin orange for curing, we confirmed the existence of antagonistic gene in the chromosomal DNA.
The culture conditions for Bacillus sp. DP-152 in the flask were investigated for the production of polysaccharide locculant, DP-152. The optimum pH and temperature for the locculant production were 8.0 and $30^{\circ}C$, respectively. The avorable substrates for flocculant production were soluble tarch and ammonium nitrate. The medium composition was optimized as follows: 30 g soluble starch, 0.75 g $NH_4NO_3,\; 2.0g\; K_2\;HPO_4,\; 0.1\; g KH_2PO_4,\; 0.2g\; MgSO_4.\; 7H_2O,\; and\; 0.2g\; MnSO_4~5H_2O$ in 11 of distilled water. Under this optimized condition, flocculating activity has been improved 4-fold compared with that of the basal medium. In the culture flask, the highest flocculating activity was obtained after 70 h of cultivation and the amount of bioflocculant DP-152 yielded was 12.4 g/$\ell$. The solution of bioflocculant DP-152 showed non-Newtonian characteristics. Bioflocculant DP-152 exhibited apparently higher viscosity at all concentrations compared to that of zooglan (from Zoogloea ramigera), and it was stable over a wide range of temperatures and pHs.
The hairy root culture of Raphanus sativus cv. Chungpihongsim was established by transformation with Agrobacterium rhizogenes $A_4$. The transformed roots grew well in adjusted Murashige and Skoog medium to 1/2 basal salts, pH 5.2, 3% sucrose. Agropine and mannopine, opine synthesized in the transformed tissue were detected in the extract of hairy roots. When 2, 4-D and kinetin were added in culture medium of hairy roots, the synthesis of anthocyanin was induced with disorganization of hairy root. Especially, addition of $0.45\;\mu\textrm{M}$ 2, 4-D and $2.3\;\mu\textrm{M}$ kinetin showed the maximum synthesis of anthocyanin. Pattern of anthocyanin synthesized in transformed roots was somewhat different from that of ordinary roots. However, aglycone part of all anthocyanin was identified as pelargonidin. The content of total anthocyanin in this sample was tentatively calculated 0.49 mg/g fresh weight.weight.
We investigated the capability of the phosphate-solubilizing fungus, Penicillium sp. GL-101, to solubilize in vitro some insoluble rock phosphate via possible mechanisms: acidification of the medium, production of chelating metabolites, redox activity, and so on. GL-101 was able to solubilize rock phosphate (mostly calcium phosphate) in a liquid potato dextrose broth(PDB) medium, as determined by spectrophotometric analyses. Acidification was the major mechanism of solubilization since the pH of cultures fell below 4.0 and in cultures containing 1.0%(w/v) loess the pH dropped from 7.0 to 3.2. More than 10 mg/mL concentrations of citric acids were detected by high-performance liquid chromatography(HPLC) in the culture supernatants. Also this fungus showed the phosphatase activity (over 1.3 unit) to contribute partially releasing phosphate from rock phosphate, when supplemented with 1.0% loess in culture broth. The chelating activity of GL-101 in culture supernatants was not present because 2-ketogluconic acid, a chelating agent for the phosphate, was produced only a basal level. Therefore, the solubilization mechanism of rock phosphate by Penicillium sp. GL-101 involves both acidification due to citric acid production and phosphatase activity.
