The effects of basal media, carbon, nitrogen, phosphate and some major macro elements on growth and shikonin production in Lithospermum erythrorhizon hairy root culture were studied. Among examined media, growth of hairy root cultured in B5 liquid medium was rapid, whereas shikonin production was high in MS liquid medium. Under B5 basal medium, sucrose concentration for optimal growth and shikonin production was 9% and 4% respectively. The growth and shikonin production on pH changes in B5 medium resulted little effect in pH 5.8 to pH 8.8 ranges, whereas growth was decreased dramatically in both above 8.8 and under 5.8. Nitrogen source and concentration effected on the growth and shikonin production. The highest growth rate was in B5 medium (50 mM $KNO_3$ and 1 mM $NaH_2PO_4)$, whereas the highest shikonin production was in the condition supplemented with 5 mM $KNO_3$ and 10 mM $NaH_2PO_4$.

Dendrobium candidum Wallich ex Lindley is a traditional Chinese medicine plant and has been widely used for medicinal and ornamental purposes. In this study, several different factors affecting micro propagation of protocorm-like bodies (PLBs) such as basal media, plant growth regulators, and carbon sources. The proliferation PLB derived from seeds was the best in $H_3P_4$ basal medium containing $0.1mg{\cdot}L^{-1}$ NAA and $0.1mg{\cdot}L^{-1}$ Kinetin. PLB growth was the best when $10g{\cdot}L^{-1}$ sucrose was added to the carbon atoms in the medium. The rate of shoot formation from the propagated PLB was the highest in 1/4 MS or $H_1P_2$ medium containing $10g{\cdot}L^{-1}$ sucrose, and the shoot length was longer than the others.
Effects of the special media on the mycelium growth in Agaricus campestris has been studied. The results are summarized as follows; 1. The mycelium growth of Agaricus campestris were considerably Stimulated on the Carrot (Dancus carota L.) basal medium which was added 4ml. of carrot extract, Cucumber(Cucumis sativus L.) basal medium added 3ml of cucumber extract, and Radish (Rhaphanus sativus L.) basal medium added 3ml. of radish extract during the culture for 144 hours. 2. The mycelium of Agaricus campestris on the media which was added the several kinds of vegetable extracts shows a lot of growth for 144 hours. The orders are as follows; Carrot basal medium(4ml/100ml)>Cucumber basal medium (3ml/100ml)>Radish basal medium (3ml/100ml)>Lettuce basal medium (2ml/100ml)>Cabbage basal medium (2ml/100ml). However, the Lettuce (Lactuca scariota L.) basal medium and the Cabbage (Brassica chinensis L.) basal medium among these five media are no significant differences.
The optimum concentrations of pH, sucrose and vitamins for the growth and tropane alkaloid production of hairy root clone DTLA9 (best growth line) were investigated. The optimum pH in growth and tropane alkaliod production of DTLA9 clone in SH (Schenk and Hildebrandt, 1972) basal medium without growth regulator were pH 6.3 and 6.5, respectively. Also, the optimum sucrose concentration in growth and tropane alkaliod production in the same medium were 3.0 and 2.8%, respectively. The optimum concentrations of ascorbic acid, D-pantothenate, nicotinic acid, pyridoxine, riboflavin, and thiamine on the growth of DTLA9 clone in SH basal medium without vitamins were 0.1 mM, 0.003 mM, 0.07 mM, 0.002 mM, 0.025 mM, and 0.01 mM, respectively. In particular, supplement of 0.1 mM ascorbic acid to SH basal medium without vitamins stimulated the tropane alkaloid production.
In this study, Pullulan Production from whey medium by Aureohsidium pullulans was investigated. When the Pullulan production from whey medium was carried outs final cell concentration was similar to sucrose basal medium but Pullulan concentration was less than 5g/$\ell$. For pullulan production from whey medium, adaptation culture technique was tried on lactose and galactose base medium When the adaptation culture technique was not applied, the maximum concentration of pullulan was 3.4g/$\ell$ for lactose, 2.5g/$\ell$ for galactose. Adapted strains produced 10~12g/$\ell$ from lactose, 10~11g/$\ell$ from galactose. The pullulan production from lactose basal medium was 13.5g/$\ell$ for lactose adapted strains and 18.6g/$\ell$ for galactose adapted strains. When the adapted strains were inoculated on whey medium, maximum pullulan production was obtained at initial pH 3.0.
1) The comparative studies of the quantitative measurement of growth characteristics and utilization of substrates by Euglena gracilis var. bacilla 10616 in the light and in darkness have been carried out. Eodogenous respiration, effect of respiratory inhibitors and responses to the added substrates for the exogenous respiration are also investigated. 2) All cultures are grown in the open air under the continuous illumination of fluorescent light of 3500 lux at room termperature, the growth rate of the culture in the basal medium added 0.5% lactate is found to be the highest. The growth rate decreases successively for the cultures of 0.5% sucinate, 0.5% Na-acetate, 0.5% malate, and control. There is no growth in the basal meidum added 0.5% butyrate and 0.5% hydroquinone. The similar results are obtained for the mentioned cultures in the darkness. However, the growth rate in basal medium added 0.5% glucose and 0.5% sucrose does seem to increase in the darkness unlike the illumination. 3) The endogenous rate of respiration for the organism cultured photosynthetically is about 12.94ul 02/mg/hr, in basal medium and the respiratory quotient is about 0.84. The rate is decreased by starvations to 6.5ul 02/mg/hr, about to a half, but the respiratory quotient does net change. 4) The oxygen consomption during initial 2 hours in suspending solution ranging from pH 4.5 to pH 9.3 is highest at pH 4.5 in which the algae had grown, at pH 5.5 and at pH 6.9. 5) Endogenous respiration of the cells is strongly inhibited by 0.1M of potassium cyanide, malomic acid, sodium fluoride and iodo-acetic acid. It is also strongly inhibited by 0.01M of potassium cyanide. 6) The respiratory response to added substrates for the exogenous respiration in the organism is coincided with the rate in the basal medium added the substrate in light and in darkness, whether the cells are fed or starved. 7) According to the results of this study, there seems to be the flexibility of the interconversion between photosynthesis and chemosynthesis, heterotropic mode of metabolism, in Euglena gracilis var. bacillaris, and that this organism utilizes the lactate most. It also may be suggested that the enayme systems linked in the each steps of Embden-Myerhof-Parnas path way and TCA cycle seem to exist in this organism.
Penicillium sp I which produces a powerful hydrolysing enzyme was isolated from putrefid and dry Jerusalem artichoke medium. The strain was used to study on the optimum culture conditions for enzyme production. The results obtained are as follows: 1. Penicillium sp I was a vigorous strain to produce inulase. 2. The optimum culture conditions of the strain was examined in the Jerusalem artichoke extract medium and the synthetic medium. 3. Inulase productivity in the Jerusalem artichoke extract medium was higher than that of the synthetic medium. 4. The optimum culture period of the Jerusalem artichoke extract medium was four days, whereas that of the synthetic medium was five days. 5. The optimum temperature, pH and concentration in the Jerusalem artichoke extract medium were $30^{\circ}C$, 5.0 and 4.0% (W/V), respectively. Meanwhile, the optimum temperature, pH and concentration in the synthetic medium were $30{\sim}33^{\circ}C$, $5.0{\sim}6.0$, and $1.0{\sim}1.5%$ (W/V), respectively. 6. Corn steep liquor, peptone, $(NH_4)_2HPO_4,\;NH_4H_2PO_4,\;(NH_4)_2SO_4$, etc. were favorable as nitrogen sources. Of these, especially, Corn steep liquor and peptone as organic nitrogen sources caused an increase in inulase production in the synthetic medium. 7. All sugars except for inulin have no effect upon the inulase production. 8. KCl, $MgSO_4\;and\;FeSO_4$ were favourable mineral sources for inulase production.
The aims of this study were, firstly, to analyze the biochemical compositions of serum and follicular fluid (FF) from prepubertal gilts after PMSG (1,000 IU) treatment. The concentrations of total proteins, lipids, cholesterol, glucose and sex hormones (progesterone, $P_4$; estradiol-$17{\beta}$, $E_2$; testosterone, T) were measured. Secondary, the effects of porcine FF (pFF) addition (40% and 100%) in IVM media and different culture conditions [Exp. 1: mBMOC-2+20% porcine serum (PS), fresh IVM medium, filtered IVMconditioned medium, or rabbit oviducts; Exp. 2: mBMOC-2+20%PS or stepwise medium replacement procedures (SMRP) cocultured with or without cumulus cells] on the in vitro development (IVD) of porcine oocytes were also examined. Results showed that no significant differences were found in total protein levels between serum and pFF from different sizes (large, >7 mm; medium, ~5-7 mm; small, <3-5 mm) of follicles (75-85 and 49-90 mg/dl; p>0.05). Total lipid concentrations remained constant in serum (395-472 mg/dl), and reduced significantly in the pFF from large follicles (287 mg/dl) at 132 h after PMSG treatment when compared to those at other time points (441-480 mg/dl). Basal cholesterol levels in serum and pFF at 12 h were similar (153-161 mg/dl), but increased at 36 h (186-197 mg/dl). Basal P4 and E2 levels in serum (0.1 ng/ml and 5.5 pg/ml) were low, but increased from 0.34 ng/ml and 12.13 pg/ml at 24 h to 0.81 ng/ml and 61.70 pg/ml at 98 h, respectively, after PMSG treatment (p<0.05). P4 levels increased linearly in pFF from large follicles during 12 through 132 h (138-1,288 ng/ml). A similar increase was also observed in $E_2$ levels (22-730 pg/ml) before 60 h post PMSG treatment, and then dropped afterwards (730-121 pg/ml). The development of the oocytes fertilized in 40% pFF-medium was greater than that in 100% pFF-medium group without gonaodtropin addition (31% vs 10%, p<0.05). However, both were lower than those in mBMOC-2+20%PS and in rabbit oviducts (p<0.05). When cocultured with cumulus cell monolayers, a greater cleavage rate was observed in the group cultured in filtered IVM-conditioned medium than the SMRP group (36% vs 18%, p<0.05). A similar phenomenon was also observed in the culture without cumulus cell monolayers (33% vs 19%, p<0.05). It is concluded that neither the fresh IVM nor filtered IVM-conditioned medium has positive effect on the IVD of oocytes. Coculture with cumulus cell monolayers and the SMRP were not beneficial to the development of IVF pig oocytes.
A potent lytic strain was selected by an extensive screening test of microorganisms isolated from soils and sewages on the medium containing baker's yeast as a carbon source. This strain (M-10) was identified to a strain of Humicola sp. by the Genera of Fungi (Clements, 1964). The strain was cultured on the basal medium composed of 2% of baker's yeast, 0.3% of $K_2HPO_4$, 0.01% of $MgSO_4{\cdot}7H_2O$, 0.1% of yeast extract in a shaking incubator. Cultural conditions for lytic enzyme production has been studied, and the results obtained were as follows: 1. The Optimal conditions for lytic enzyme production were: initial pH 5.5 to 6.0, temperature $33^{\circ}C$ in shaking culture. 2. Among the various carbon sources, baker's yeast (4%) was the best for lytic enzyme production, increasing the level of activity eight, times higher than when grown on glucose (1%). 3. The most effective concentration of $K_2HPO_4\;and\;MgSO_4{\cdot}7H_2O$ in the basal medium for lytic enzyme production was 0.1% and 0.01% respectively. 4. When the strain was cultured under the optimal conditions, the production of lytic enzyme was maximized in 72 hours.
- I aimed to isolate trans-acting factors involved in the basal expression level of eukaryotic genes. One of the yeast histidine biosynthetic gene, HIS5 was taken as a model for this study. HIS5 gene has a substantial basal level in amino acid rich medium and is derepressed if starved for any single amino acid. The derepression is mediated by cis-acting DNA sequences 5'-TGACTC-3' found in 5' non-transcribed region of the gene and trans-acting factors including GCN4 as positive factor and its negative factor GCDI 7, and GCNZ as a negative factor of GCD17. I first investigated the role of these trans-acting factors in HIS5 basal expression level by using HIS5-pH05 fusion in which expression of pH05 gene encoding inorganic phosphate-repressible acid phosphatase (APase) is regulated by HIS5 promoter. Strain with gcn2 or gcn4 mutation showed 3 to 4 fold lower APase activity than wild type. The level of APase activity was similar in gcn2 and gcn4 mutants. Trans-acting factors involved in basal level were identified by isolating 14 mutants showing increased expression of HISSPH05 fusion from gcn4 background. All the mutants carry a single nuclear recessive mutation and fall into four complementation groups, designated as bell (basal expression level), be12, be23 and be14.
