Water and Electrolyte Metabolism of Korean Buddhist Nuns (한국여승(韓國女僧)의 수분(水分) 및 전해질대사(電解質代謝))

  • The relationship between water and electrolyte metabolism, and dietary intake were studied in 45 healthy Buddhist nuns who were vegetarians aged 20-34, and 28 nursing students aged 20-22 who stayed at the dormitory of Kyungpook Medical School in the Fall, 1981. The Buddhist nuns obtained significantly higher carbohydrate and total caloric intakes but significantly lower protein and lipid intakes than the female students. The Buddhist nuns excreted significantly higher urine output($1,697{\pm}68\;ml/day$, p<0. 05) and lower osmolality ($616{\pm}18\;mOsm/kg\;H_2O$, p<0.05) than the students ($1,505{\pm}67\;ml/day$ and $688{\pm}36\;mOsm/kg\;H_2O$). However, both groups excreted similar amounts of urinary $Na^+$, $K^+$ and total osmolar contents. Free water clearance of the Buddhist nuns was higher(p<0.05) than that of the students but the osmolar clearance was about the same in the two groups. Daily urine output showed good correlation with Na output (r=0.76) and osmolar clearance but not with free water clearance. Both groups showed similar values of plasma concentration of creatinine, daily excretion of creatinine and clearance. Urinary excretion of urea for Buddhist nuns was 6.4 g/day, and was significantly higher than that of the students (5.1g/day).

Changes of Glycosidase Activity and Fertilizing Ability in Vitro by Incubation of Frozen-Thawed Spermatozoa in the Pig (돼지 동결정액의 배양에 따른 체외수정능력과 Glycosidase Activity의 변화)

  • This study has evaluated effect of the spermatozoa incubation on the glycosidase activity and fertilizing ability in vitro in the pig. To identify sperm glycosidases specific for sugar residues found in the zona pellucida of pig oocytes, the spermatozoa were treated experimentally and assayed for activities of $\alpha$-L-fucosidase, $\alpha$-D-mannosidase, $\beta$-D-galactosidase and N-acetyl-$\beta$-D-glucosaminidase ($\beta$-GlcNAc'ase). The glycosidases activity were higher in spermatozoa incubated for 2h than without incubation. The $\beta$-GlcNAc'ase activity was at least two-fold higher than other glycosidase regardless of spermatozoa incubation. In the same glycosidases, the activity had a tendency to increase as time of spermatozoa incubation was prolonged, but there were no differences in spermatozoa incubated during the various periods (4~24h). The percentages of spermatozoa that reached acrosome reaction were affected by glycosidases in the medium (P<0.05, for mannosidase), and were higher in spermatozoa with that than without incubation. On the other hand, the spermatozoa motility were decreased with incubation periods, but no effects by different glycosidases on the change of sperm motility during the various periods of incubation. In other experiment, the binding and penetration of pig spermatozoa were tested with oocytes matured in vitro in the presence of various glycosidase. The penetration rates were decreased with incubation of spermatozoa when oocytes were inseminated in medium with different glycosidases. These rates were higher in spermatozoa non-incubated than with incubation for 2h (P<0.05 for GlcNAc'ase; P<0.01 for control group). The sperm-zona binding rate in control group were higherthan in medium with glycosidases. In addition, the highest binding rate were obtained in medium with GlcNAc'ase. In all glycosidases, the sperm-zona binding rate in spermatozoa without incubation were higher than incubation for 2h. The significant differences were obtained in spermatozoa treated with $\alpha$-D-mannosidase (P<0.05). These results suggest that $\beta$-GlcNAc'ase is present mainly in the plasma membrane of pig spermatozoa. It was also shown that the glycosidase activity were increased in all glycosidases in spite of low sperm-zona binding rate and penetration rates by spermatozoa incubation.

  • The pool size of plasma glucose, turnover rate and other concerned items for glucose metabolism in normal laying hen were investigated by a single-injection method using $U-C^{14}-glucose$. The 11.6 nCi of pure dose was injected to a hen normally fed through the wing vein. The glucose concentration in plasma sample taken at 5 minutes after injections was 214mgper 100ml. From the plottings of logarithmic standard specific activities of plasma taken from 5 to 120 minutes against the time after injection and from the regresion analysis, metabolic states were determined. The pool size was 1.07g, turnover rate was 0.024 per minute, turnover time was 41 minutes, utilization rate was 26mg/min. (0.83 g/hr/kg B.W. 3/4) and glucose space(extracellular fluid volume) was 25.3 per cent of body weight. The values obtained from. 10-50 minutes samples were similar to those described above, which we from 5-120 minutes samples.

Effects of Spent Composts of Se-Enriched Mushrooms on Carcass Characteristics, Plasma GSH-Px Activity, and Se Deposition in Finishing Hanwoo Steers (셀레늄급원으로 셀레늄강화버섯 폐배지의 급여가 거세한우의 도체특성, 혈중 GSH-Px활성 및 조직내 셀레늄축적에 미치는 영향)

  • This study was conducted to determine effects of spent composts of Se-enriched mushrooms (Se-SMC) as the dietary selenium source on carcass characteristics, plasma glutathione peroxidase(GSH-Px) activity and Se deposition in finishing Hanwoo steers. In combination with both Se-SMC and normal SMC, experimental treatment diets were formulated to contain 0.1, 0.3, 0.6 and 0.9 ppm of Se on a dry matter basis. A total of 20 finishing Hanwoo steers (average BW = 613 kg, average age = 20 to 24 mo) were allotted to treatments in four groups of five steers per pen for 12 wk preceding slaughter. While the experiment is employed, blood samples were taken to analyze Se concentration and GSH-Px activity, and muscle and liver samples were collected for analyses of Se contents in their tissues after slaughter. DMl and BW gain were not affected by dietary Se level and any toxic symptoms in treatments with a higher level of Se were not observed. No differences were noted for carcass characteristics. Se concentration in whole blood and plasma GSH-Px activity were linearly increased with the increasing level of dietary Se (P < 0.01). Se content in the hind leg for Se-SMC supplemented groups significantly increased (P < 0.05) upon dietary Se level, with 0.27, 0.37, 0.40 and 0.46 !1g1g dry, respectively. However, Se content in the loin was not affected by dietary Se levels. Se content in the liver was significantly increased(P < 0.05) as dietary Se increased, with 0.79, 1.40, 2.39 and 3.10 !1g1g dry, respectively. These results suggested that Se in the Se-SMC was highly bioavailable, and Se-SMC might be used not only as an inexpensive way of providing Se for ruminants but also as another way of producing Se-fortified beef.

The Production Mechanism of TNF-${\alpha}$ and IL-6 by Group IIA Phospholipase $A_2$ (IIA형 Phospholipase $A_2$에 의한 TNF-${\alpha}$와 IL-6 생성 기전)

  • Background: Secretory phospholipase $A_2$ ($sPLA_2$) are a group of extracellular enzymes that release fatty acids at the sn-2 position of phospholipids. Group IIA $sPLA_2$ ($sPLA_2$-IIA) has been detected in the inflammatory fluids, and its plasma level increases in the inflammatory disease. This study examined the effect of $sPLA_2$-IIA on mouse macropahges in order to investigate the potential mechanism of $sPLA_2$-induced inflammation. Materials and Methods: Wild type $PLA_2$ and mutant H48Q $PLA_2$ were purified from HEK293 cells transfected with the corresponding plasmids, and the $PLA_2$ activities were measured using 1-palmitoyl-2-[1-$^{14}C$]linoleoyl-3-phosphatidylethanolamine as substrates. The TNF-${\alpha}$ and IL-6 released in the supernatants were determined by ELISA. In addition, the TNF-${\alpha}$ and IL-6 mRNA were analyzed by RT-PCR. Results: $sPLA_2$-IIA stimulated the production of TNF-${\alpha}$ and IL-6 in a dose- and time-dependent manner. In addition, the effect of $sPLA_2$-IIA on cytokine production from the macrophage was found to be associated with the accumulation of their specific mRNA. The mRNA levels of TNF-${\alpha}$ and IL-6 peaked at 2 and 6 hours in a time-dependent manner, respectively. Conclusion: In conclusion, the production of proinflammatory cytokine might be mediated by the binding of $sPLA_2$-IIA to the receptors.

Formation Behavior and Properties of PEO Films on AZ91 Mg Alloy in 0.1 M NaOH + 0.05 M NaF Solution Containing Various Na2SiO3 Concentrations (AZ91 마그네슘 합금의 플라즈마 전해산화 피막 형성 및 물성에 미치는 0.1 M NaOH + 0.05 M NaF 용액 중 Na2SiO3 농도의 영향)

  • Effects of Na2SiO3 concentration added into 0.1 M NaOH + 0.05 M NaF solution on the formation behavior and properties of PEO films on AZ91 Mg alloy were investigated under 1200 Hz of alternating current (AC) by voltage-time curves, in-situ observation of arc generation behavior and measurements of film thickness, surface roughness and micro vickers hardness. In the absence of Na2SiO3 in the 0.1 M NaOH + 0.05 M NaF solution, about 4 ㎛ thick PEO film was formed within 1 min and then PEO film did not grow but white spots were formed by local burning. Addition of Na2SiO3 up to 0.2 M caused more increased formation voltage and growth of PEO film with uniform generation of arcs. Addition of Na2SiO3 from 0.2 M to 0.4 M showed nearly the same voltage-time behavior and uniform arc generation. Addition of Na2SiO3 more than 0.5 M resulted in a decrease of formation voltage and non-uniform arc generation due to local burning. PEO film growth rate increased with increasing added Na2SiO3 concentration but maximum PEO film thickness was limited by local burning if added Na2SiO3 concentration is higher than 0.5 M. Surface roughness of PEO film increased with increasing added Na2SiO3 concentration and appeared to be proportional to the PEO film thickness. PEO film hardness increased with increasing added Na2SiO3 concentration and reached a steady-state value of about 930 HV at more than 0.5 M of added Na2SiO3 concentration.

A Novel Polyclonal Antiserum against Toxoplasma gondii Sodium Hydrogen Exchanger 1

  • The sodium hydrogen exchanger 1 (NHE1), which functions in maintaining the ratio of $Na^+$ and $H^+$ ions, is widely distributed in cell plasma membranes. It plays a prominent role in pH balancing, cell proliferation, differentiation, adhesion, and migration. However, its exact subcellular location and biological functions in Toxoplasma gondii are largely unclear. In this study, we cloned the C-terminal sequence of T. gondii NHE1 (TgNHE1) incorporating the C-terminal peptide of NHE1 (C-NHE1) into the pGEX4T-1 expression plasmid. The peptide sequence was predicted to have good antigenicity based on the information obtained from an immune epitope database. After induction of heterologous gene expression with isopropyl-b-D-thiogalactoside, the recombinant C-NHE1 protein successfully expressed in a soluble form was purified by glutathione sepharose beads as an immunogen for production of a rabbit polyclonal antiserum. The specificity of this antiserum was confirmed by western blotting and immunofluorescence. The antiserum could reduce T. gondii invasion into host cells, indicated by the decreased TgNHE1 expression in T. gondii parasites that were pre-incubated with antiserum in the process of cell entry. Furthermore, the antiserum reduced the virulence of T. gondii parasites to host cells in vitro, possibly by blocking the release of $Ca^{2+}$. In this regard, this antiserum has potential to be a valuable tool for further studies of TgNHE1.

A Study on the Heavy Metal Contamination of paddy Soil in the Vicinity of the Seosung Pb-Zn Mine (서성 연-아연광산 주변 농경지 토양의 중금속 오염 연구)

  • Fifty seven soil samples were collected from the paddy soil filled with tailings in the vicinity of the Seosung Pb-Zn mine. Those samples were analyzed for As, Cd, Co, Cr, Cu, Pb, and Zn in order to investigate heavy metal pollution levels in the paddy soil. Analyses of the soil samples were carried out using Inductively Coupled Plasma Atomic Emission Spectrometry(ICP-AES) . Paddy soils show pH range from 6.55 to 8.26. X-ray diffraction analyses of the paddy soil indicate that the soils consist predominantly ankerite, siderite, quartz, mica, and clay minerals with minor amounts of amphibole and chlorite. The mineral composition of the waste rocks consists of massive galena, sphalerite, and minor amounts of pyrite, arsenopyrite, chalcopyrite, calcite, siderite, Pb-sulfosalt, and marcasite. The paddy soils were significantly contaminated by heavy metals(average concentrations, As: 334.4 ppm, Cd: 37.6 ppm, Co: 15.7 ppm, Cu: 214.1 ppm, Pb: 4,612 ppm, and Zn: 4,468 ppm).

Effects of Season and Split-sex Feeding on Performance, Stress and Carcass Quality of Finishing Pigs (계절과 성분리 사육이 비육돈의 생산성, 스트레스 및 도체품질에 미치는 영향)

  • This study was designed to determine the effect of season and split-sex feeding on performance, stress and carcass quality of finishing pigs. Two hundred and seventy cross-bred finishing pigs $(82.1{\pm}0.82kg)$ were randomly divided into six groups in 2 seasons (summer and winter) ${\times}$ 3 split-sex feeding (female only, castrated male and mixed) factorial arrangement of treatments. In winter, feed intake and weight gain of finishing pigs were better than summer (p<0.05). Split-sex feeding was affected the feed conversion, feed conversion of finishing pigs were superior in winter and mixed groups. Plasma concentrations of cortisol as a stress indicators was greater in the mixed groups vs female only and castrated male groups. There were no interactions between the season and split-sex feeding in their effects on these blood variables. The season was affected the carcass characteristics and PSE incidence of the finishing pigs, PSE incidence was low in winter and mixed groups. Meat quality did not show any significant difference due to the difference of season and split-sex feeding of finishing pigs. There was no significant difference in the pH, drip loss and meat color of pork loin between the season and split-sex feeding of finishing pigs. In conclusion, the results of this study suggest that we need more precise on-farm management practice in summer and application of split-sex feeding skills to reduce the stress and improve welfare status of finishing pigs.

Eliminatory Effect of Mixture including Hot Water Extract of Dendropanax morbifera Lev. on Alcohol-induced Blood Alcohol Concentration and Hangover in Rat (흰쥐에서 황칠나무 열수 추출물을 포함한 혼합물의 혈중 알코올 농도와 숙취 해소 효과)

  • This study was performed to investigate the ameliorating effect of a hangover beverage mixture (SBJ) that contains Dendropanax morbifera Lev. and several medicinal plant extracts, on hepatoprotection and alcohol-metabolizing enzymes in alcohol-induced hangover in both in vitro and in vivo models. In human hepatoma cell line, HepG2, 300 mM of ethanol-induced hepatotoxicity was significantly improved by pretreatment of SBJ by dose-dependent manner. In the in vivo study, administration of alcohol to rats raised to the concentration of blood alcohol and lactate dehydrogenase (LDH). Blood alcohol and LDH levels in SBJ-treated rats significantly decreased at 0.5 h and 8 h after acute ethanol administration (40%, 4.6 g/kg body weight) as compared to alcohol-treated rats. Hepatic alcohol dehydrogenase (ADH) and aldehyde dehydrogenase (ALDH) activity were significantly higher in SBJ-treated rats than in alcohol-treated rats. SBJ supplementation reduced formation of malondialdehyde (MDA), and inhibited reductions of hepatic superoxide dismutase (SOD), hepatic glutathione (GSH), glutathione-S-transferase (GST), glutathione reductase (GR) and glutathione peroxidase (GPx) levels, compared with rats administered alcohol. Plasma catalase (CAT), aspartate aminotransferase (AST) and alanine aminotransferase (ALT) levels showed unaltered resulted in all experimental groups compared with the control group. These results suggest that SBJ exhibit hepatoprotective properties by enhancing ADH, ALDH activity and stimulating the antioxidant defense system in alcohol-induced hangover.
