Killer yeast, Hansenular capsulata S-13 were treated with heat, ethylmethane sulfonate and N-methyl-n'-nitro-n-nitrosoguanidine and a mutant(S13-E1), showing 2-fold higher killer toxin activity than that of parent strain to killer sensitive strain, Saccharomyces cerevisiae ATCC 38026 was obtained. Hansenular capsulata S13-E1 showed strong killer toxin activity to Saccharmyces mellis and Saccharomyces sal년 and four strains of gas-producing yeasts from traditional Doenjang and Kochujang. The culture condition for killer toxin production by Hansenular capsulata S13-E1 was optimized to be 1.0% potato extract, each 0.5% of peptone and glucose, and 0.025% MgSO4 with initial pH 4.5 at 3$0^{\circ}C$ and 36 hr of batch cultivation.
The chromosomal aberration of the trichothecene mycotoxins such as T-2 toxin (T-2), HT-2 toxin (HT-2), nivalenol (NIV) and deoxynivalenol (DON) which are one of the most important food borne contaminants produced by Fusarium species fungi, was investigated in the chinese hamster lung cells. These trichothecene mycotoxins showed high cytotoxicity in order of T-2, HT-2, NIV, and DON to the chinese hamster lung cells. Nevertheless high cytotoxicity of these trichothecene mycotoxins, no clastogenicity of T-2 and HT-2 in the range of 0.01-0.0025 ng/ml, of NIV in that of 0.3-0.075ng/ml, and of DON in that of 1.0-0.25 ng/ml was observed in both with and without metabolic activation system.
The production of interleukin-1(IL-1)and nitric oxide(NO) by cultured fibroblast cells of human nasal turbinate was revealed by biological assay respectively. The cells were incubated for various periods of time in the presence of staphyloccocal toxic shock syndrome toxin-1(TSST-1) and house dust mite(Dermatophagoides farinae, HDM), and the culture supernatants were harvested. There was a little difference in the activities of IL-1beta and the amount of NO produced by the cells when stimulated with 0.002-0.1$\mu$g/ml of TSSTO-1 and 0.02-1$\mu$g/ml of HDM. The shapes of the time course curves for the production of IL-1beta and NO by the cells were different. Groups stimulated with TSST-1 or HDM produced more IL-beta in 2 h than no exposure group(Control). A certain mixed group(TSST-1, 10ng+mite, 100 ng) continued to produce IL-1beta highly throughout the entire incubation period. The cells stimulated with TSST-1 or HDM produced more NO in 2 h and 6 h than that produced in the end of incubation(48 h). Also, the mixed groups were generally similar. There results suggest that induction of IL-1beta by a certain mixed condition(TSST-1+mite) in fibroblast cell in vivo may play a role in inflammation.
Foreign proteins, $endo-{\beta}-1,4-glucanase$ of Bacillus subtilis, preS1+S2 region of hepatitis B virus large surface antigen, human ${\beta}_2-adrenergic$ receptor ($h{\beta}_{2}AR$), and bovine growth hormone (bGH) were expressed in Saccharomyces cerevisiae and secreted into the medium. These proteins were expressed using the alcohol dehydrogenase I (ADH1) promoter of Saccharomyces cerevisiae and secreted by signal sequence of the 97 K killer toxin gene of doublestranded linear DNA plasmid (pGKL1) of S. cerevisiae. All these proteins underwent severe modifications; in particular, N-glycosylation in the case of $endo-{\beta}-1,4-glucanase$, $h{\beta}_2AR$, and preS1+S2. Seventy four percent of the expressed $endo-{\beta}-1,4-glucanase$ was secreted into the culture medium. Highly modified proteins were detected in the culture medium and in the cell. Expressed $h{\beta}_2AR$, which has seven transmembrane domains, remained in the cell. The degrees of secretion and modification and the states of proteins in the culture medium and in the cell were quite different. These results indicated that the nature of the protein has a critical role in its secretion and modifications.

Seventy-four Shiga toxin-producing Escherichia coli (STEC) isolates belonging to the serotype O91:H21 were isolated from 1,643 asymptomatic human carriers in a STEC outbreak at Gwangju in Korea. Although the isolates did not cause any symptoms, all of them produced Shiga toxins 1 (Stx1) and 2 (Stx2). In order to determine why these strains cause no symptoms, we explored the differences in virulence potential between the asymptomatic STEC O91:H21 isolates and symptomatic STEC O91:H21 strains (ATCC 51435 and ATCC 51434). The asymptomatic STEC O91:H21 isolates showed strongly reduced cytopathic effects compared with the symptomatic strains when intact bacterial cells were used as an inoculant. Moreover, we found a reduced adherence phenotype when testing asymptomatic strains on HeLa cells. Real-time quantitative PCR results suggest that transcriptional repression of the genes encoding type-1 fimbriae occurs in the asymptomatic isolates but not in the symptomatic strains.
The determination of fungal flora in some kinds of cereals have been carried out in other to obtain an appropriate information of the population of fungi and toxin productivity The results were summarized as follow; 1. The predominant genera were Aspergillus, Penicillium, Mucor, Rhizopus, Alternaria, Fusarium. 2. Six of Aspergillus flavus were aflatoxin-producing strains. 3. Sample barleys were found to contain the highest content of aflatoxin. 4. In electron microscopic studies of liver cells from mouse which had been injected with crude toxin, the liver cells showed the cytoplasmic change.

Escherichia coli O157:H7 cause hemorrhagic colitis and the extraintestinal complication of hemolytic-uremic syndrome, with their higher incidence occurring in children. Lipopolysaccharide (LPS) of E. coli O157:H7 is very important to make IgG anti-LPS with bactericidal activity. To identify the characteristic of E. coli OI57:H7, we isolated 60 MDa plasmid and amplified stx genes of shiga-like toxin (Stx) 1, 2 of E. coli O157:H7 by polymerase chain reaction (PCR) method. Using the simple purification method which contained phenol extract, ethanol precipitation and gel filtration steps, the LPS of E. coli O157:H7 was isolated and purified. Finally, we confirmed the purity of LPS through SDS-PAGE and silver nitrate staining.

The study was to determine the effects of diverse concentrations of T-2 toxin in broiler diet. Three hundred 1-day-old chicks with initial body weight of 46 ± 0.52 g were chosen and randomly assigned into five dietary treatments with 5 replicate cages and 12 broilers per cage for 42 d feeding trial. Dietary treatments were prepared with basal diets containing 0 (T1), 50 (T2), 100 (T3), 150 (T4), 200 (T5) ppb T2-toxin. Significant results were observed in the decreased intake of feed, feed conversion ratio (FCR), body weight gain (BWG), level of serum protein, cholesterol and hemoglobulin of broilers in increased concentration of the T-2 toxin in diet (150 and 200 ppb) groups than control. Also, observed that the uric acid, serum glutamic pyruvic transferase (SGPT), serum glutamic oxaloacetic transferase (SGOT) and Heterophil/Lymphocyte (H/L) ratio value were significantly higher (p < 0.05) in groups T4 and T5 than control. However, the BWG, feed intake and FCR, as well blood biochemical profiles of serum protein, cholesterol, hemoglobulin, uric acid, SGPT, SGOT and H/L ratio in groups T2 and T3 were statistically similar to control diet of broilers. It was concluded that the results showed that no adverse effects on growth performance and blood biochemical parameters in broilers feed with T-2 toxin (50 and 100 ppb) during the entire trial.
Toxin produced by Colletotrichum falcatum Went, the incitant of red rot of sugarcane was isolated, purified and assayed to determine host specificity and identify its chemical nature. The toxin was found to be not host specific as it inhibited germination of various seeds(gram, greengram, blackgram, pea, cowpea, rice and sugarcane) as well as different seedlings viz. tomato, coriander, pea and rice. The toxin consists of two distinct fraction-one fraction having $R_f$, value at 0.36 producing identical red rot lesion when inoculated at leaf midrib of sugarcane, and the other having $R_f$, value at 0.72 not showing any red rot lesion. Chromatogram of high performance liquid chromatography(HPLC) of the red rot lesion causing fraction showed a sharp peak at 1.62 min of retention time(RT), and spectral analysis indicated the presence of following chemical $CH_3$ - groups-C-H, C=O, C-N, $-CH_3,\;-CH_2$ -CH and molecular mass of the compound was 203. - ($M^+,\;C_{11}H_{11}N_2O_2$).
A new method developed for simultaneous purification of enterotoxin A and C from Staphylococcus aureus strain L 350/1 consisted of chromatography on carboxymethyl (CM)-cellulose using a buffer of variable pH, gel filtration on Ultro gel, and fast protein liquid chromatography(FPLC) using a buffer of variable pH. The enterotoxin A and C were purified by three steps: batchwise adsorption from culture supernatant on Amberlite CG-50; chromatography on CM-cellulose using a buffer of constant pH and molarity; and gel filtration on Sephadex G-75. The purified enterotoxin appeared homogeneous by gel diffusion and polyacrylamide gel electrophoresis. Upon treatment with CM-cellulose using a elution of variable pH, enterotoxin A and C were so close that they were not separated completely. After elution from gels, the enterotoxins appeared as a single peak at the same position. Gel filtration gave a reaction of complete identity to enterotoxin A and C in Ouchterlony immunodiffusion. In FPLC using a CM-cellulose, enterotoxin A and C were simultaneously separated at pH 8.6 and 6.8. When each fraction was performed to gel immunodiffusion, at peak of enterotoxin A and C were not detected each other. In a method of elution by pH-gradient was to be more efficient as a simultaneous separation method in terms of speed, yields and simplicity. The purified toxin A and C were identical to type A and C reference enterotoxin on both disc electrophoresis and Ouchterlony gel diffusion.
