Taxonomic review of the umbelliferous taxa Heracleum moellendorffii complex in Korea based on molecular phylogenies of nuclear ribosomal ITS sequences

  • Taxonomy of umbelliferous taxa Heracleum moellendorffii complex has been unclear in their species delimitation in the far-eastern Asian regions. In both Korea and China Heracleum moellendorffii was adopted for their description while H. sphondylium was chosen to describe Japanese Heracleum. From Genbank accessions, taxa collected from Kamtchatka and Promorskiy, Russia were defined as H. maximum, endemic taxon to North America. In this study, we reviewed the taxonomy of Heracleum moellendorffii complex in Korea and neighboring countries on the basis of molecular phylogenies derived from sequences of nuclear ribosomal DNA internal transcribed spacer (ITS) regions. From three Korean accessions of Heracleum investigated in this study, two types of ITS sequences were obtained; two accessions were related to Chinese H. moellendorffii var. moellendorffii and North American H. maximum without forming a clade while the other one was identical to accession from H. maximum from Primorskiy, Russia. In the other hand, Japanese H. moellendorffii (=H. sphondylium ssp. sphondylium var. nipponicum in the flora of Japan) was closely related to H. maximum accessions from Korea and Russia, not nested within the clade comprising several subspecies of H. sphondylium. In order to delimit species boundaries among putatively closely related Heracleum species in fareastern Asian countries, more samples and much more rapidly evolved DNA regions must be investigated with interpretation of morphological and anatomical features.

P-V 곡선법을 활용한 어수리의 수분특성 분석 (Water Relations Parameters of Heracleum moellendorffii Hance Obtained from Pressure-Volume Curves)

  • This study was carried out to establish a proper cultivation site and to diagnose the drought tolerance of Heracleum moellendorffii leaves by using pressure-volume curves. As a result of analysing data measured, the leaf of H. moellendorffii showed the osmotic pressure at full turgor (Ψosat) was -1.0MPa, and that at incipient plasmolysis (Ψotlp) -1.2MPa. Then, the value of maximum bulk modulus of elasticity Emax was 28MPa, showing the sightly strong drought tolerance of H. moellendorffii. Furthermore, the values of relative water contents RWCtlp and RWC* were above 88%, showing that the function of osmoregulation is somewhat better. Thus, responses to water relations such as Ψosat, Ψotlp, Emax, RWCtlp and RWC* of H. moellendorffii showed it's slightly high drought tolerance property.

Open Channel Block of hKv1.5 by Psoralen from Heracleum moellendorffii Hance

  • A furocoumarin derivative, psoralen (7H-furo[3,2-g][1]benzopyran-7-one), was isolated from the n-hexane fraction of Heracleum moellendorffii Hance. We examined the effects of psor-alen on a human Kv1.5 potassium channel (hKv1.5) cloned from human heart and stably expressed in Uk- cells. We found that psoralen inhibited the hKv1.5 current in a concentration-, use- and voltage-dependent manner with an IC$_{50}$ value of 180 $\pm$ 21 nM at +60 mV. Psoralen accelerated the inactivation kinetics of the hKv1.5 channel, and it slowed the deactivation kinetics of the hKv1.5 current resulting in a tail crossover phenomenon. These results indicate that psoralen acts on the hKv1.5 channel as an open channel blocker. Furthermore, psoralen prolonged the action potential duration of rat atrial muscles in a dose-dependent manner. Taken together, the present results strongly suggest that psoralen may be an ideal antiarrhythmic drug for atrial fibrillation.

대기-엽 수증기압차(VPD)에 의한 어수리와 눈개승마의 광합성 반응 비교 (Comparison of Photosynthetic Responses in Heracleum moellendorffii and Aruncus dioicus var. Kamtschaticus in Relation to Atmosphere-Leaf Vapor Pressure Deficit)

  • 산채의 일종으로 현재 임업농가에서 활발히 재배되고 있는 눈개승마와 어수리의 수증기압차(vapor pressure deficit; VPD)에 따른 순광합성 속도 등 생리반응을 조사하여 효율적인 생산과 재배관리를 위한 기초자료를 제공하고자 하였다. 건조한 High-VPD 환경에서 눈개승마가 어수리에 비해 증산량을 줄이면서 수분이용효율을 높이는 기작을 통해 높은 순광합성 속도를 유지하는 것을 알 수 있었으며, 어수리에 비해 눈개승마는 엽의 수분 퍼텐셜의 감소가 비교적 크지 않고, 엽의 수분 불포화도 역시 큰 변화가 없는 것으로 보아 건조한 대기 환경에 따른 수분스트레스의 적응력이 비교적 양호한 종임을 알 수 있었다. 어수리는 VPD가 3.0kPa 이상으로 높게 나타나는 건조한 대기상태가 되면 순광합성 속도의 감소가 뚜렷히 나타나고 수분 퍼텐셜의 급속한 저하 및 엽의 수분 불포화도가 10배 이상 높게 나타나 상대적으로 건조한 대기 환경에서 생육하기 어려운 종임을 알 수 있었다. 따라서 재배지의 환경조건이 VPD 3.0kPa 이상의 건조한 환경이 자주 형성되는 곳은 어수리의 생육에 불리할 수 있으며, 눈개승마의 경우 어수리 보다 다소 건조한 대기환경에 더 높은 적응력을 지닌 것을 알 수 있었다.

범용성 DNA 바코드(matK, rbcL) 분석을 통한 독활(獨活) 유전자 감별용 Marker Nucleotide 발굴 (Identification of Marker Nucleotides for the Molecular Authentication of Araliae Continentalis Radix Based on the Analysis of Universal DNA Barcode, matK and rbcL, Sequences)

  • Objectives : Araliae Continentalis Radix and Angelicae Pubescentis Radix have been used as the same medicinal name Korean and Chinese traditional medicines, respectively. The authentic Araliae Continentalis Radix is described only the root of Aralia continentalis in the Korean Pharmarcopoeia. However, the dried root of Angelica biserrata, Levisticum officinale, or Heracleum moellendorffii also has been distributed adulterants of Araliae Continentalis Radix. To develop a reliable method for identifying Araliae Continentalis Radix from adulterants, we carried out the analyses of universal DNA barcode sequences.Methods : Four plants species were collected from different habitate and nucleotide sequences of matK and rbcL were analyzed. The species-specific sequences and phylogenetic relationship were estimated using entire sequences of two DNA barcodes, respectively.Results : In comparative analysis of matK sequences, we were identified 104 positions of marker nucleotide for Ar. continentalis, 3 for An. biserrata, 4 for L. officinale and 8 for H. moellendorffii enough to distinguish individual species, respectively. Furthermore, we obtained marker nucleotides in rbcL at 42 positions for Ar. continentalis, 5 for An. biserrata and 2 for H. moellendorffii, but not for L. officinale. The phylogenetic tree of matK and rbcL were showed that all samples were clustered into four groups constituting homogeneous clades within the species.Conclusions : We confirmed that species-specific marker nucleotides of matK sequence provides distinct genetic information enough to identify four species. Therefore, we suggest that matK gene is useful DNA barcode for discriminating authentic Araliae Continentalis Radix from inauthentic adulterants.

Multiplex PCR을 이용한 독활 류 식물로부터 Aralia continentalis 감별 (Discrimination of Aralia continentalis from other Herbs Identified as 'Angelicae Pubescentis Radix' by Multiplex Polymerase Chain Reaction (PCR))

  • 'Angelicae Pubescentis Radix' (APR) is an important oriental medical preparation. In Korea, Aralia continentalis has been recognized as the source plant of APR. Aralia cordata, which is difficult to distinguish from A. continentalis, and Heracleum moellendorffii, which is frequently used in lieu of A. continentalis, are traded in Korean herbal markets. In contrast, in China, Angelica pubescens is recognized as the source plant of APR. In this study, we devised a method not only to discriminate A. contientalis from A. cordata, but also to discriminate both A. contientalis and A. cordata from H. moellendorffii and A. pubescens. Based on the discrepancy in the sequences of specific regions of ITS, we designed a Cont F/ Cont R primer set to amplify a 173 bp PCR band that appears only in A. continentalis. Additionally, we designed an Ara F/ Ara R primer set to amplify a 278 bp PCR band that appears in both A. continentalis and A. cordata. Using these primer sets and the ST R primer to confirm the PCR amplification results, we developed a simple multiplex PCR method for differentiating A. continentalis from A. cordata and to concurrently differentiate both A. continentalis and A. cordata from other APR herbs.

어수리분말 첨가 국수의 품질특성 및 저장성 (Quality Characteristics and Shelf Life of Noodles Prepared with Heracleum moellendorffii(Hogweed) Powder)

  • 어수리의 실용적인 활용방법을 모색하기 위하여 어수리 분말을 첨가하여 생면을 제조하고. 이들 면의 품질특성과 저장성을 비교 검토하였다. 어수리 분말의 첨가량이 증가 할수록 국수의 중량, 부피 및 수분흡수율이 감소하였다. 어수리 분말의 첨가량이 증가할수록 국수 국물의 탁도와 조리중 고형분 손실량이 대조구에 비해 낮았다. 어수리 분말 첨가에 의해 생면의 명도(L)와 적색도(a)는 감소하였으며, 황색도(b)는 증가하였다. 어수리 분말 첨가에 의해 국수의 기호성은 증가하였으며, 0.6%첨가 국수가 기호성 즉 맛, 조직감, 종합적기호도에서 가장 우수하였다. 총 폴리페놀 함량은 어수리 분말의 첨가량이 증가할수록 증가하였으며, 전자공여능은 대조구 22%, 어수리 분말 0.6%와 0.9%첨가구는 각각 40.22%와 49.42%이었다. 저장 중 미생물의 변화는 저장 6일째까지 대조구와 첨가구(0.3 - 0.6%)간에 뚜렷한 차이를 나타내지 않았으며, 저장 12일째 대조구, 0.3% 그리고 0.6%첨가구는 $10^6$ CFU/mL이었으나, 어수리 분말 0.9%첨가 국수는 $10^3$ CFU/mL이었다. 저장 중 pH의 변화는 대조구와 0.3%, 0.6%첨가구는 유사하였으나 0.9% 첨가구는 전 저장기간 동안 pH의 변화가 지연되었다.
