Characteristics of New Microsporidia S80 Isolated from Silkworm, Bombyx mori L. in Korea (가잠(家蠶)으로부터 분리(分離)된 새로운 Microsporidia S80의 특성(特性))

  • The new microsporidia S80 isolated from, Bombyx mori L. in Korea showed ovoid in the morphology of the spores and the size were measured $2.9{\pm}0.28{\mu}$ in length and $1.7{\pm}0.29{\mu}$ width. No other microsporidian spore like this has not been so far isolated from Silkworm. The length of the polar filament extruded in hydrogen peroxide ($H_2O_2$) at $30^{\circ}C$ was $26{\mu}$ of a round cytoplasm on the top. The spores were partly stained with Giemsa, Safranin-O and Gram as the same staining properties as Nosema bombycis, Microsporidia K 79 and other microsporidian spores. The fine structures were observed under scanning eleceron microscope through ultrathin sectioning. The spore wall was composed of three layers ; the thin exospore of an electron dense rippled layer, the thick electron lucent endospore which was thinning considerably at the polar filament insertion point, and the inner limiting membrane. Polar cap present at the sporeapex, with a long polar filament of 12-13 coils, subtending angle of $60^{\circ}$ to spore axis, which is tubular made up of a multilayered and are a benes core, light ring structure enclosing the dance core, the dark ring structure enclosing the inner light ring structure and the other than and light ring structure bounded from cytoplasm. Lamellate polaroplast occupied the anterior part of the spore, and the two neclei with dense nucleoplasm bounded by a double nuclear envelope were cited in the slight downer middle portion of spore. From the characteristics of the shape, size and fine structures, it is certain to reason the Microsporidia S80 belong to the phylum Microspora, class Microspora, order Microsporida, order Microsporida. The shape of two nuclei cited seems to be genus Nosema, but in the classification for the suborder it should be defined wheather pansporoblasts be formed or not and for the genis especial attempts have been made to define the characters which distinguish the disporous genera in the life cycle. Survey through the infection of the bad cocoons during 1980 to 1982 in South Korea the areas contaminated with new microsporidia were revealed 5 provinces of Kyung-Gi, Kang-Won, Chung-Nam and Chun-Nam. Pathological effects inoculated per os at second instar larvae of silkworm, the LD 50 was $7.1{\times}10^7/ml$ as lower pathogenecity than that of Nosema bombycis Naegeli of $1.2{\times}10_7/ml$. While on the other hand the inoculation of the microsporidia at fourth instar larvae lowerd the whole cocoon weight and cocoon shell weight and significant at 1% level. The microsporidia S80 defined it can not be transmitted transovarially from the result of predictive and collective examination of 21 egg batches from the infected female moth.

Serotonin Synthesis and Metabolism in Dissociated Cultures of Fetal Rat Brainstem (흰쥐 태아 뇌간의 일차 세포배양에서 Serotonin의 합성 및 대사에 대한 연구)

  • We established an in vitro system of central serotonergic neurons by culturing dissociated rat embryonic (El4) brainstem cells to 14 days in vitro and monitored the serotonergic neuronal growth by measuring 5-hydroxytryptamine (5-HT) and 5-hydroxyindoleacetic acid (5-HIAA) contents in the cells with hish performance liquid chromatography with electrochemical detection (HPLC-EC). We studied also tile effects of various drugs on the contents of 5-HT and 5-HIAA, confirming in vivo reports. The 5-HT content (13 ng/mg protein) and 5-HT turnover rate (17 pmol/mg protein/h) at 14 days in vitro were in good agreement with those reported in the adult rat brain. The 5-HT content was more easily depleted with p-chlorophenylalanine, a tryptophan hydroxylase inhibitor than with NSD 1015 (3-hydroxybenzylhydrazine), an aromatic L-amino acid decarboxylase (AADC) inhibitor. Incubation of the cultures with tryptophan or 5-hydroxytryptophan increased the rate of serotonin formation implying that neither tryptophan hydroxylase nor AADC is saturated with its amino acid substrate in this in vitro system . The 5-HT content was depleted by reserpine. The 5-HT and 5-HIAA contents were increased and decreased, respectively, by monoamine oxidase inhibitors. All the above results indicate that the biochemical properties of the central serotonergic neurons in this culture system reflect reliably those of central serotonergic neurons in vivo. We suggest that measuring 5-HT and 5-HIAA contents in the primary cultured dissociated brainstem-cells with HPLC-EC is useful in the study of pharmacology as well as toxicolgy of the central serotonergic neurons.

Nutrients and Decomposition Rate Accumulated on Soil Layers in Quercus mongolica Forest of Mt. Songnisan National Park (속리산 신갈나무림의 토양층별 영양염류 함량과 분해율)

  • The content of nutrients such as organic carbon, nitrogen, phosphorus and potassium accumulated on soil layers in Quercus mongolica forest of Mt. Songnisan National Park located at central part of Korea was measured, and then the decomposition constants and decay times of the nutrients were also calculated by the negative coefficience model(O1son,1963). The quantities of organic carton of L-layer, F-layer, H-layer and $A_1$-layer of the forest stand were 231.25 g $m^{-2}$, 291.50 g $m^{-2}$,166.91 g$m^{-2}$ and 174.51 g $m^{-2}$, respectively. The content of organic carbon and nitrogen contained in L-layer and F-layer showed large quantity than those of other layers. The large amount of phosphorus and potassium was observed at the B-layer and $A_1$-layer. On the other hand, the decomposition constants(k) of soil organic matter were as follows : organic carbon (k = 0.3657), nitrogen (k = 0.3319), phosphorus (k = 0.2050), and potassium (k = 0.0934) and the decay times needed to 99% decomposition of nutrients in soil organic matter were as follows: that is, organic carton, nitrogen, phosphorus and potassium was 13.94 years, 15.18 years, 24.79 years, and 55.11 years, respectively. By the application of Turbo Pascal Program on the inflowed and outflowed nutrients to the forest stand,87.67% (714.84 g $m^{-2}$) of organic carbon inflowed was decomposed and 81.62% (1,594.62 g $m^{-2}$) of organic carbon accumulated was decomposed. And 84.98% of nitrogen inflowed was decomposed and 70.26% of nitrogen accumulated was also decayed.50.00% of phosphorus input and 40.31% of potassium input were decomposed, and 38.40% of phosphoyus and 33.03% of potassium accumulated were also decayed, respectively. Therefore, it is suggested that Quercus mongolica forest surveyed in the present study is maintaining in steady state because input and output amounts of nutrients is shown a similar pattern.

Thermal-Denaturation of File Fish Myofibrillar Protein and Protective Effect of Sucrose, Sorbitol and Amino Acids (말쥐치 근원섬유단백질의 열안정성과 및 가지 첨가제의 영향)

  • Thermal-denaturation of myofibrillar protein of dorsal skeletal muscle from file fish was investigated by measuring denaturation constant($K_D$) and thermodynamic parameters at various temperatures. The protective effects of sucrose, sorbitol and amino acids when added individually or combined were also discussed. The denaturation rate as reflected in inactivation of myofibrillar protein Ca-ATPase was followed the first order reaction. The $K_D$ values at $25^{\circ}C,\;30^{\circ}C,\;and\;35^{\circ}C$ were $19.52{\times}10^{-5},\;112.25{\times}10^{-5},\;and\;247.20{\times}10^{-5}$, respectively. The activation energy of the reaction at $30^{\circ}C$ was 43 kcal/mole. The protective effects of sucrose, sorbitol, glycine, alanine and Na-glutamate were increased with the concentration but the effects of sorbitol and Na-glutamate decreased beyond 1.0 mole. Basic amino acids such as arginine and lysine did not revealed any protective effect on the thermal denaturation. In case of mixed addition, the effects of Na-glutamate to glycine, sorbitol to glycine, and sorbitol to sucrose or sorbitol to Na-glutamate were enhanced 1.2 to 7.0 times as much as that of control (ratio of mixing; 1:1, range of concentration; 0.5 to 1.25 mole). Under the frozen condition at $-20^{\circ}C$, two mixtures such as Na-glutamate to glycine and sorbitol to sucrose apparently revealed the protective effects.

Quality Comparison of Emulsion- Type Sausages Made from Rhus verniciflua Stokes Fed Pork and Extract (옻 급여 돈육과 추출물로 제조한 유화형 소시지의 품질 비교)

  • This study was designed to evaluate the quality comparison of emulsion-type sausages made from different Rhus verniciflua Stokes (RVS) sources. The pigs were fed a supplemented concentrate diet with a RVS supplement of $4\%$ feed for 5 weeks before slaughter. The RVS extract was prepared from 100g of RVS sawdust and 1L of distilled water for 48 hours. Emulsion-type sausages were made using lean meat or dietary RVS han1 lean $(51.07\%)$, ice water or RVS extract $(19.63\%)$, back fat $(26.60\%)$ and other additives $(2.70\%)$. The treated sausages were divided into non-dietary meat with water (T1, Control), dietary RVS meat with water (T2), non-dietary meat with RVS extract (T3), and dietary meat with RVS extract (T4). The crude fat was significantly lower (p<0.05) in dietary RVS meat-added sausages (T2, T4) than in control sausage (T1). The lightness ($L^{\ast}$) and redness ($a^{\ast}$) were significantly lower (p<0.05) in RVS extract-added sausages (T3, T4) during refrigerated storage. The $a^{\ast}$ value was higher (p<0.05) in T2 than in the other treatments. The VBN and TBARS values were significantly lower in RVS extract-added sausages (T3, T4) during refrigerated storage. The hardness, adhesiveness, gumminess, and chewiness were significantly lower (p<0.05) in T2 than in the other treatments. This results showed that feeding of RVS in diet and/or RVS extract had a significant impact on the quality of emulsion-type sausage. The RVS extract-added emulsion-type sausages (T3, T4) showed dark and reddish color although they were more effective in delaying the protein deterioration and lipid oxidation. Consequently, the sausage prepared from pigs fed $4\%$ RVS with water (T2) was more effective in increasing the $a^{\ast}$ value, textural properties, and delaying the protein deterioration, lipid oxidation than that without RVS in diet.

Physicochemical Characteristics of Sweetpotato (Ipomoea batatas (L.) Lam) Starch Depending on Cultivation Periods (재배기간에 따른 고구마(Ipomoea batatas (L.) Lam) 전분의 이화학적 특성)

  • This study was performed to understand the effect of cultivation periods on the physicochemical characteristics of the starch of five sweetpotato cultivars, cultivated in Muan, Korea. Starch, protein, and ash contents increased with increased cultivation period, whereas amylose content decreased. Rapid viscosity analysis showed that the pasting temperature, peak viscosity, breakdown, setback and final viscosity increased with increased cultivation period. However, trough and final viscosity decreased. Although the onset temperature and peak temperature values increased, the conclusion temperature did not show any consistent patterns by differential scanning calorimetry. X-ray diffraction showed that the starch samples had C-type crystallinity irrespective of the cultivation period and cultivar. The starch granules were dominantly round and oval, or polygonal irrespective the cultivation period. The bigger the particle size was, the longer the cultivation period was.

Chondrogenesis of Mesenchymal Stem Cells Derived from Human Umbilical Cord Blood (사람 제대혈 유래 간엽줄기세포로부터 연골세포 분화)

  • In the current study, the mesenchymal stem cells (MSCs) isolated and propagated from the human umbilical cord blood (UCB) were tested for their capabilities of differentiation into chondrocytes in vitro. The mesenchymal progenitor cells (MPCs) collected from UCB were cultured in a low glucose DMEM medium with 10% FBS, L-glutamine and antibiotics. The human MSC colonies were positively stained by PAS reaction. When the immunophenotypes of surface antigens on the MSCs were analyzed by fluorescence-activated cell sorter (FACS) analysis, these cells expressed positively MSC-related antigens of CD 29, CD44, CD 90 and CD105, whereas they did not express antigens of CD14, CD31, CD34, CD45, CD133 and HLA-DR. Following induction these MSCs into chondrocytes in the chondrogenic differentiation medium for 3 weeks or more, the cells were stained positively with safranin O. We clearly confirmed that human MSCs were successfully differentiated into chondrocytes by RT-PCR and immunofluorescent stain of type-II collagen protein. These data also indicate that the isolation, proliferation and differentiation of the hUCB-derived MSCs in vitro can be used for elucidating the mechanisms involved in chondrogenesis. Moreover this differentiation technique can be applied to developing cell-based tissue regeneration or repair damaged tissues.

Effects of Rice Hull Addition and Bin Wall Characteristics on Pig Slurry Composting Properties (왕겨 이용 방법과 옹벽이 돈분 퇴비화에 미치는 효과)

  • This work was carried out to investigate the effects of rice hull continuously utilized and/or replenished on the composting properties and to obtain the fundamental data between an unsupported wall and a soil supported wall during the period of composting with pig slurry in winter season. There were no the temperature holding effects in soil supported wall. New compost facility design for the temperature holding effects from a soil supported wall was required. The results were as follows; 1. Composting 1㎥ of pig slurry caused to save on 0.31㎥ of bulking agent in the unsupported wall in comparison with a soil supported wall in the rice hull single addition, and 0.45㎥ in the rice hull gradual addition. 2. The pile in the rice hull single addition had a high temperature in 4 days of composting indicating $71^{\circ}C$ and had a tendency in repeating periodically between $40^{\circ}C$ and $65^{\circ}C$ till 43 days of composting. And also the temperature of the pile was maintained between $48^{\circ}C$ and $28^{\circ}C$ after 50 days of composting. The pile of a rice hull gradual addition had the lower point of the temperature high increasingly according to adding up rice hull during the 35 days of composting. 3. The pH recorded in the rice hull single addition was higher(8.35∼10.02) compared to the rice hull gradual addition(8.6∼9.8). The pile of a rice hull single addition had a tendency in abruptly decreasing pH of the unsupported wall during the period of between 0.363$\textrm m^3$ and 0.537$\textrm m^3$ as a unit of pig slurry per rice hull. EC depending upon the way in adding rice hull was changed between 1.10 mS/$\textrm {cm}^3$ and 1.87 mS/$\textrm {cm}^3$. 4. The organic matter in an unsupported wall of the hull single addition was maintained the level of 55% during the period between 0.119㎥ and 0.363㎥ as a unit of pig slurry per rice hull while in the soil supported wall between 48 and 70. Water soluble C:N ratio was maintained between 1 and 2 in the rice hull single addition, while between 1 and 3 in the rice hull gradual addition. 5. Fertilizer constituents were detected higher level in the unsupported wall than in the soil supported wall in all treatments. This was dependant upon the input of pig slurry.

Evaluation of Air Quality in the Compost Pilot Plant with Livestock Manure by Operation Types (축분 퇴비화시스템 운용방식에 따른 실내 대기오염 평가)

  • Air quality in the livestock waste compost pilot plant at the Colligate Livestock Station was assessed to quantity the emissions of aerial contaminants and evaluate the degree of correlation between them for different operation strategies; with the ventilation types and agitation of compost pile, in this study. The parameters analyzed to reflect the level of air quality in the livestock waste compost pilot plant were the gaseous contaminants; ammonia, hydrogen sulfide, and odor concentration, the particulate contaminants; inhalable dust and respirable dust, and the biological contaminants; total airborne bacteria and fungi. The mean concentrations of ammonia, hydrogen sulfide, and odor concentration in the compost pilot plant without agitation were 2.45ppm, 19.96ppb, and 15.8 when it was naturally ventilated, and 7.61ppm, 31.36ppb, and 30.2 when mechanically ventilated. Those with agitation were 5.50ppm, 14.69ppb, and 46.4 when naturally ventilated, and 30.12ppm, 39.91ppb, and 205.5 when mechanically ventilated. The mean concentrations of inhalable and respirable dust in the compost pilot plant without agitation were 368.6${\mu}g$/$m^3$ and 96.0${\mu}g$/$m^3$ with natural ventilation, and 283.9${\mu}g$/$m^3$ and 119.5${\mu}g$/$m^3$ with mechanical ventilation. They were also observed with agitation to 208.7${\mu}g$/$m^3$ and 139.8${\mu}g$/$m^3$ with natural ventilation, and 209.2${\mu}g$/$m^3$ and 131.7${\mu}g$/$m^3$ with mechanical ventilation. Averaged concentrations of total airborne bacteria and fungi in the compost pilot plant without agitation were observed to 28,673cfu/$m^3$ and 22,507cfu/$m^3$ with natural ventilation, and 7,462cfu/$m^3$ and 3,228cfu/$m^3$ with mechanical ventilation. They were also observed with agitation to 19,592cfu/$m^3$ and 26,376cfu/$m^3$ with the natural ventilation, and 18,645cfu/$m^3$ and 24,581cfu/$m^3$ with the mechanical ventilation. It showed that the emission rates of gaseous pollutants, such as ammonia, hydrogen sulfide, and odor concentration, in the compost pilot plant operated with the mechanical ventilation and with the agitation of compost pile were higher than those with the natural ventilation and without the agitation. While the concentrations of inhalable dust and total airborne bacteria in the compost pilot plant with the natural ventilation and with the agitation, the concentrations of respirable dust and total airborne fungi in the compost pilot plant with the mechanical ventilation and agitation were higher than those with the natural ventilation and without the agitation of compost pile. It was statistically proved that indoor temperature and relative humidity affected the release of particulates and biological pollutants, and ammonia and hydrogen sulfide were believed primary malodorous compounds emitted from the compost pilot plant.

Studies of Eri-Silk Cultring in Korea (한국피마잠사개발에 대한 연구)

  • Eri-silkworm is known as a tropical insect where as poly-voltine type in that area. It eats caster oil plant leaves which are cultivated as an every year cultivatable seed oil use in this country, even though it grows for many years in tropical countries. That is why, farmers have freedom for its cultivation in any year if they want. Therefore, eri-silkworm rearing service is flexible for its diet procurment as wish of farmer. The eri-cocoon price or economical fluctuation may be reactable for the rearing work not like as mulberry cocoon. Fortunately, it also eats cynthia tree leaves. Standing from such a easy condition, the authors have studied about this problem since 1963 to develope a culturing method of eri-silkworm rearing in this country and the authors brought out the matters to be produced as an industry scale. Here, the authors summarized their works of the results covering with thirty three work tables. The obtained results are as follows.
