Bioanalytical method validation for determination of arsenic speciation in dog plasma using HPLC-ICP/MS (Dog 혈장 중 HPLC-ICP/MS를 이용한 비소 화학종 분석법 검증)

  • The approach presented in this article refers to the bioanalytical method validation for the detection and quantitative determination of arsenic species including arsenite (As(III)), arsenate (As(V)), dimethylarsinic acid (DMA) and monomethylarsonic acid (MMA) in dog plasma by high-performance liquid chromatography inductively coupled plasma mass spectrometry (HPLC-ICP/MS). The arsenic species were separated using an agilent As speciation column by a mobile phase of 2 mM sodium phosphate monobasic, 0.2 mM ethylenediaminetetraacetic acid disodium salt dehydrate, 10 mM sodium acetate, 3 mM sodium nitrate and 1 % ethyl alcohol at pH 11 (adjusted with 1M NaOH). The method validation experiment was obtained selectivity, linearity, accuracy, precision, matrix effect, recovery, system suitability, dilution integrity and various stabilities. All calibration curves showed good linearity (R2>0.999) within test ranges. The lower limit of quantitation (LLOQ) was 5 ng/mL for As(III), As(V) and DMA, and 20 ng/mL for MMA. The system suitability and dilution values were within 6.5 % and 7.7 %. Subsequently, the developed and validated HPLC-ICP/MS method was also successfully applied to determine the arsenic speciation in dog plasma samples, and the recoveries for the spiked samples were in the range of 91.5–102.2 %. Therefore, this method could be applied to the evaluation of arsenic exposure, health effect assessment and other bio-monitoring studies in biological samples.

  • Heterogeneous cation exchange membrane(HCEM) was prepared with LLDPE(Linear Low Density Poly-ethylene) as binder, powdered cation exchange resins($diameter{\leq}149{\mu}m$) as ion-exchange material and glycerol as additive for electrodialysis and electrodeionization system. The weight ratio of (binder/ion exchange)/glycerol was (60%/40%)/5%, (55%/45%)/5%, (50%/50%)/5% and (40%/60%)/5%. The characterization of prepared HCEM was evaluated on mechanical, electrochemical, morphology and ion permeable properties. It was compared with commercial membrane. Electrochemical properties of HCEM of (50%/50% )/5% were very similar to value of IONPURE(commercial membrane), in which ion exchange capacity, ion transfer number and membrane resistance were to be 1.733meq/g, 0.96 and $16.08{\Omega}/cm^2$, respectively. Ion permeability of the membrane was better than that of IONPURE membrane. Compared with IONPURE membrane, the HCEM had a higher tensile strength and lower elongation and modulus, in which HCEM had tensile strength of $62.33kg/cm^2$, elongation of 87.42% and modulus of $658.53kg/cm^2$. The HCEM of (50%/50% )15% was optimum combination.

Effect of Type I Collagen on Hydroxyapatite and Tricalcium Phosphate Mixtures in Rat Calvarial Bony Defects

  • To repair bone defects in the oral and maxillofacial field, bone grafts including autografts, allografts, and artificial bone are used in clinical dentistry despite several disadvantages. The purpose of this study was to evaluate new bone formation and healing in rat calvarial bone defects using hydroxyapatite (HA, $Ca_{10}[PO_4]_6[OH]_2,\;Bongros^{(R)}$, Bio@ Co., KOREA) and tricalcium phosphate (${\beta}-TCP,\;Ca_3[PO_4]_2$, Sigma-Aldrich Co., USA) mixed at various ratios. Additionally, this study evaluated the effects of type I collagen (Rat tail, BD Biosciences Co., Sweden) as a basement membrane organic matrix. A total of twenty, 8-week-old, male Sprague-Dawley rats, weighing 250-300g, were divided equally into a control group (n=2) and nine experimental groups (n=2, each). Bilateral, standardized transosseous circular calvarial defects, 5.0 mm in diameter, were created. In each experimental group, the defect was filled with HA and TCP at a ratio of 100:0, 80:20, 70:30, 60:40, 50:50, 40:60, 30:70, 20:80, and 0:100 with or without type I collagen. Rats were sacrificed 4 and 8 weeks post-operation for radiographic (standardized plain film, Kodak Co., USA), histomorphologic (H&E [Hematoxylin and Eosin], MT [Masson Trichrome]), immunohistochemical staining (for BMP-2, -4, VEGF, and vWF), and elementary analysis (Atomic absorption spectrophotometer, Perkin Elmer AAnalyst $100^{(R)}$). As the HA proportion increased, denser radiopacity was seen in most groups at 4 and 8 weeks. In general radiopacity in type I collagen groups was greater than the non-collagen groups, especially in the 100% HA group at 8 weeks. No new bone formation was seen in calvarial defects in any group at 4 weeks. Bridging bone formation from the defect margin was marked at 8 weeks in most type I collagen groups. Although immunohistochemical findings with BMP-2, -4, and VEGF were not significantly different, marked vWF immunoreactivity was present. vWF staining was especially strong in endothelial cells in newly formed bone margins in the 100:0, 80:20, and 70:30 ratio type I collagen groups at 8 weeks. The calcium compositions from the elementary analysis were not statistically significant. Many types of artificial bone have been used as bone graft materials, but most of them can only be applied as an inorganic material. This study confirmed improved bony regeneration by adding organic type I collagen to inorganic HA and TCP mixtures. Therefore, these new artificial bone graft materials, which are under strict storage and distribution systems, will be suggested to be available to clinical dentistry demands.

Effects of Bujasasim-tang Ethanol Extract on Oxidative Stress, Inflammation and Osteoarthritic Rat Model (부자사심탕(附子瀉心湯)이 산화적 손상, 염증 및 골관절염 병태모델에 미치는 영향)

  • Objectives This study was performed to investigate the effects of Bujasasim-tang ethanol extract (BST) on oxidative stress, inflammation and osteoarthritic rat model. Methods To ensure safety of BST, heavy metal levels were measured and cytotoxicity test was done. In vitro, To evaluate antioxidative effects of BST, total phenolic contents, 1,1-diphenyl-2-picryl-hydrazyl (DPPH), 2,2'-azino-bis-(3-ethylbenzothiazoline-6-sulfonic acid) (ABTS) scavenging activity, reactive oxygen species (ROS) levels were measured. Also, to evaluate anti-inflammatory effects of BST treated group, total nitric oxide (NO) and pro-inflammatory cytokines (IL-$1{\beta}$, IL-6, TNF-${\alpha}$) levels were measured in lipopolysaccharide (LPS)-stimulated RAW 264.7 cells. In vivo, We injected MIA $50{\mu}l$ (60 mg/ml) into knee joints of rats to induce osteoarthritis. Rats were divided into total 3 groups (normal, control, BST treated group, each n=7). Normal group was not treated at all without inducing osteoarthritis and taken normal diet. Control group was induced osteoarthritis by MIA and taken with 2 ml of distilled water once a day for 4 weeks. BST treated group was induced osteoarthritis by MIA and taken BST 2 ml (200 mg/kg/mouse) once a day for 4 weeks. We evaluated dynamic weight bearing with the Incapacitance Test Meter. At the end of experiment, the rats were sacrificed to observe the functions of liver and kidney, changes of WBC, neutrophil, lymphocyte, monocyte levels in blood, to evaluate the levels of pro-inflammatory cytokines, tissue inhibitor of metallopreteinases-1 (TIMP-1), matrix metalloproteinase-9 (MMP-9), prostaglandin $E_2$ ($PGE_2$), leukotriene $B_4$ ($LTB_4$) within serum. We observed change of articular structures by Hematoxylin & Eosin (H&E), safranin-O staining method and measured amount of cartilage by micro CT-arthrography. Statistical analysis was done by unpaired student's t-test with significance level at p<0.05 in SPSS 11.0 for windows. Results 1. Safety of the BST was identified. 2. AST, ALT, BUN, creatinine levels of BST treated group were within normal limit. In vitro, 1. DPPH and ABTS free radical scavenging activities of BST showed dose-dependent increase. 2. ROS production were significantly decreased. 3. Total nitric oxide (NO) and IL-$1{\beta}$ production were decreased. 4. IL-6 and TNF-${\alpha}$ production were significantly decreased. In vivo, 1. Weight bearing ability was significantly increased. 2. WBC, neutrophil, lymphocyte, monocyte levels in blood were decreased. 3. IL-$1{\beta}$ and TNF-${\alpha}$ levels in serum were significantly decreased. and the IL-6 level was decreased. 4. TIMP-1, MMP-9, $LTB_4$, $PGE_2$ levels in serum were significantly decreased. 5. Cartilage volume of BST treated group was significantly increased. Also changes of cartilage, synovial membrane, fibrous tissue were suppressed. Conclusions The results obtained in this study Bujasasim-tang have effects of antioxidative, anti-inflammatory, relieve pain and protection of cartilage. Therefore we expect that Bujasasim-tang is effective treatment for osteoarthritis.

Characteristics of Water Quality In the Shihwa Lake and Outer Sea (시화호 및 주변해역의 수질 특성)

  • The operation of tidal power facility may induce severe changes of water quality in Shihwa Lake. Current water quality data are quite important to water quality management policy of Shihwa Lake. Thus, the water quality data of Shihwa Lake and its adjacent sea in 2010 were presented to characterize the temporal and spatial changes of water parameters such as pH, SS, DO, COD, dissolved nutrients, chlorophyll-a, TN and TP. Highest levels of water quality parameters were observed near the Shihwa and Banweol industrial complexes and the levels of water quality parameters were on a decreasing trend to those near the water gate. It suggests that the horizontal distributions of water quality levels are mainly controlled by the supply of fresh water from streams and the inflow of outer seawater by operation of water gate. Although the higher concentrations of TN and TP were observed in the location being affected by Sorae port, the levels of water quality parameters in outer sea of Shihwa Lake were lower than those in Lake. In summer season, hypoxic condition was well developed in bottom water by strong stratification and active decomposition of organic matter. Thus, the vertical distributions of dissolved nutrient, TN and TP concentrations showed the concentrations to be higher in bottom seawater than those in surface seawater whereas the vertical distributions of chlorophyll-a, COD and POC concentrations showed the concentrations to be higher in surface seawater than those in bottom water. Results of Pearson's correlation matrix for surface seawater demonstrated that salinity showed negatively good correlation with not only dissolved nutrients except for ammonium but chlorophyll-a, COD and POC This result indicates that the supply of dissolved nutrients through several streams might significantly affect phytoplankton bloom and increase of COD concentration in surface seawater.

Formation of Au Particles in Cu2-xICu2IIO3-δ (x ≈ 0.20; δ ≈ 0.10) Oxide Matrix by Sol-Gel Growth

  • Formation of Au particles in nonstoichiometric $Cu_{2-x}{^I}Cu{_2}^{II}O_{3-{\delta}}$ ($x{\approx}0.20$; ${\delta}{\approx}0.10$) oxide from aniline + hydrochloric acid mixtures and chloroauric acid in the ratios 30 : 1; 60 : 1; 90 : 1 (S1-S3) by volume and 0.01 mol of copper acetate, $Cu(OCOCH_3)_2.H_2O$, in each case is performed by sol-gel growth. Powder x-ray diffraction (XRD) results show Au particles are dispersed in tetragonal nonstoichiometric dicopper (I) dicopper (II) oxides, $Cu_{2-x}{^I}Cu{_2}^{II}O_{3-{\delta}}$ ($x{\approx}0.20$; ${\delta}{\approx}0.10$). Average crystallite sizes of Au particles determined using Scherrer equation are found to be in the approximate ranges ${\sim}85-140{\AA}$, ${\sim}85-150{\AA}$ and ${\sim}80-150{\AA}$ in S1-S3, respectively which indicate the formation of Au nano-micro size particles in $Cu_{2-x}{^I}Cu{_2}^{II}O_{3-{\delta}}$ ($x{\approx}0.20$; ${\delta}{\approx}0.10$) oxides. Hysteresis behaviour at 300 K having low loop areas and magnetic susceptibility values ${\sim}5.835{\times}10^{-6}-9.889{\times}10^{-6}emu/gG$ in S1-S3 show weakly ferromagnetic nature of the samples. Broad and isotropic electron paramagnetic resonance (EPR) lineshapes of S1-S4 at 300, 77 and 8 K having $g_{iso}$-values ${\sim}2.053{\pm}0.008-2.304{\pm}0.008$ show rapid spin-lattice relaxation process in magnetic $Cu^{2+}$ ($3d^9$) sites as well as delocalized electrons in Au ($6s^1$) nano-micro size particles in the $Cu_{2-x}{^I}Cu{_2}^{II}O_{3-{\delta}}$ ($x{\approx}0.20$; ${\delta}{\approx}0.10$) oxides. Broad and weak UV-Vis diffuse reflectance optical absorption band ~725 nm is assigned to $^2B_{1g}{\rightarrow}^2A_{1g}$ transitions, and the weak band ~470 nm is due to $^2B_{1g}{\rightarrow}^2E_g$ transitions from the ground state $^2B_{1g}$(${\mid}d_{x^2-y^2}$>) of $Cu^{2+}$ ($3d^9$) ions in octahedral coordination having tetragonal distortion.

  • The use of synovial fluid-derived mesenchymal stem cells (SFMSCs) obtained from patients with degenerative arthropathy may serve as an alternative therapeutic strategy in osteoarthritis (OA) and rheumatoid arthritis (RA). For treatment of OA and RA patients, autologous transplantation of differentiated MSCs has several beneficial effects for cartilage regeneration including immunomodulatory activity. In this study, we induced chondrogenic differentiation of SFMSCs by inhibiting protein kinase A (PKA) with a small molecule and microRNA (miRNA). Chondrogenic differentiation was confirmed by PCR and immunocytochemistry using probes specific for aggrecan, the major cartilaginous proteoglycan gene. Absorbance of alcian blue stain to detect chondrogenic differentiation was increased in H-89 and/or miRNA-23b-transfected cells. Furthermore, expression of matrix metalloproteinase (MMP)-9 and MMP-2 was decreased in treated1 cells. Therefore, differentiation of SFMSCs into chondrocytes through inhibition of PKA signaling may be a therapeutic option for OA or RA patients.

Mechanical Properties of Wood Flour Polypropylene Composites: Effect of Cycled Temperature Change (Wood Flour 폴리프로필렌 복합재료의 기계적 특성: 반복적 온도 변화의 영향)

  • The effect of cycled temperature change on the mechanical properties of wood flour(50 wt.% and 70 wt.%) polypropylene WPC(Wood Plastic Composites) was investigated in this study. Flexural modulus and flexural strength of the WPC showed a decrease due to the degradation of interfacial adhesion between polymer matrix and wood flour by the freeze-thaw test regardless of the cycled number. At the higher loading level of wood flour, the reduction of the flexural modulus was remarkable. After the cycled heat-freeze test, it was found that the flexural modulus and flexural strength of the WPC were lower at the high temperature ($60^{\circ}C$) and higher at the low temperature ($-20^{\circ}C$). At the low temperature ($-20^{\circ}C$) which is below glass transition temperature of polypropylene ($-10^{\circ}C$), WPC is in a glassy state which brings about the high stiffness and strength. At the high temperature ($60^{\circ}C$), the flexural modulus and flexural strength of the WPC with 50 wt.% wood flour were lower because of the increase of polymer ductility.

Quorum-Sensing Mechanisms in Bacterial Communities and Their Potential Applications (세균의 의사 소통(Quorum-Sensing) 기구와 그 잠재적 응용성)

  • Although microorganisms are, in fact, the most diverse and abundant type of organism on Earth, the ecological functions of microbial populations remains poorly understood. A variety of bacteria including marine Vibrios encounter numerous ecological challenges, such as UV light, predation, competition, and seasonal variations in seawater including pH, salinity, nutrient levels, temperature and so forth. In order to survive and proliferate under variable conditions, they have to develop elaborate means of communication to meet the challenges to which they are exposed. In bacteria, a range of biological functions have recently been found to be regulated by a population density-dependent cell-cell signaling mechanism known as quorum-sensing (QS). In other words, bacterial cells sense population density by monitoring the presence of self-produced extracellular autoinducers (AI). N-acylhomoserine lactone (AHL)-dependent quorum-sensing was first discovered in two luminescent marine bacteria, Vibrio fischeri and Vibrio harveyi. The LuxI/R system of V. fischeriis the paradigm of Gram-negative quorum-sensing systems. At high population density, the accumulated signalstrigger the expression of target genes and thereby initiate a new set of biological activities. Several QS systems have been identified so far. Among them, an AHL-dependent QS system has been found to control biofilm formation in several bacterial species, including Pseudomonas aeruginosa, Aeromonas hydrophila, Burkholderia cepacia, and Serratia liquefaciens. Bacterial biofilm is a structured community of bacterial cells enclosed in a self-produced polymeric matrix that adheres to an inert or living surface. Extracellular signal molecules have been implicated in biofilm formation. Agrobacterium tumefaciens strain NT1(traR, tra::lacZ749) and Chromobacterium violaceum strain CV026 are used as biosensors to detect AHL signals. Quorum sensing in lactic acid bacteria involves peptides that are directly sensed by membrane-located histidine kinases, after which the signal is transmitted to an intracellular regulator. In the nisin autoregulation process in Lactococcus lactis, the NisK protein acts as the sensor for nisin, and NisR protein as the response regulator activatingthe transcription of target genes. For control over growth and survival in bacterial communities, various strategies need to be developed by which receptors of the signal molecules are interfered with or the synthesis and release of the molecules is controlled. However, much is still unknown about the metabolic processes involved in such signal transduction and whether or not various foods and food ingredients may affect communication between spoilage or pathogenic bacteria. In five to ten years, we will be able to discover new signal molecules, some of which may have applications in food preservation to inhibit the growth of pathogens on foods.

Effect of Immersion in Water and Thermal Cycling on the Mechanical Properties of Light-cured Composite Resins (광중합형 수복용 복합레진의 기계적 성질에 미치는 수중침적과 Thermal Cycling의 영향)

  • This study was performed to investigate the effec% of immersion in water and thermal cycling on the mechanical peoperties of light cured restorative composite resins. Five commerically available light-cured composite resins(Photo Clearfil A : CA, Lite-Fil A . LF, Clearril Photo Posterior CP, Prisms AP.H.. PA, 2100 : ZH) were unto The specimens of 12 m in diameter and 0.7 m in thickness were made, and an immersion in $37^{\circ}C$ water for 7 days and a thermal cycling of 1000 cycles at 15 second dwell time each in $5^{\circ}C$ and $55^{\circ}C$ baths were performed. Biaxial flexure test was conducted using the ball-on-three-ball method at the crosshead speed of 0.5mm/min. In order to investigate the deterioration of composite resins during the thermal cycling test, Weibull analysis for the biaxial flexure strengths was done. Fracture surfaces and the surfaces before and after the thermal cycling test were examined by SEM. The highest Weibull modulus value of 10.09 after thermal cycling tests which means the lowest strength variation, was observed in the CP group, and the lowest value of 4.47 was obsered in the LF Group. Biaxial flexure strengths and Knoop hardness numbers significantly decreased due to the thermal cycling ($\textit{p}$< 0.01), however, they recovered when specimens were drie4 The highest biaxial flexure strength of 125.65MPa was observed in the ZH group after the thermal cycling test, and the lowest value of 64.86MPa was observed in the CA group. Biaxial flexure strengths of ZH and CP groups were higher than those of PA, CF, and CA groups after thermal cycling test($\textit{p}$< 0.05). Knoop hardness numbers of CP group after the thermal cycling test was the highest(95.47 $\pm$ 7.35kg/$mm^2$) among the samples, while that of CA group was the lowest(30.73 $\pm$ 2.58kg/$mm^2$). Knoop hardness numbers showed the significant differences between the CP group and others after the thermal cycling test(($\textit{p}$< 0.05). Fracture surfaces showed that the composite resin failure developed along the matrix resin and the filler/resin interface region, and the cracks propagated in the conical shape from the maximum tensile stress zone.
