Plant cell immobilization can protect plant cells from shear forces and increase the stability of gene. An additional advantage of immobilization is the easiness for performing continuous culture with cell recycling. Therefore plant cell immobilization can overcome the limitations of plant cell applications. In addition, target protein should be selected from pharmaceutical proteins to get rid of low expression level problem. The enhanced production of human granulocyte-macrophage colony-stimulating factor (hGM-CSF) was investigated in immobilized Nicotiana tabacum suspension cell cultures. When the cells were immobilized in polyurethane foam, specific production of hGM-CSF was higher than that in alginate bead immobilization. Optimum continuous culture condition was the addition of 60 g/L sucrose in growth media with exchanging media every 6 day. Under the same condition, specific hGM-CSF production was 7 times higher in a 500-mL spinner flask than that in 100-mL Erlenmeyer flasks. Therefore, development of an effective immobilization process would be possible when the advantage of easy cell recycling was used. Consequently, enhanced production of target proteins could be possible in immobilized continuous cultures when the advantages of immobilization were applied.
Continuous concrete beams are commonly used as structural members in the reinforced concrete constructions. The use of fiber reinforced polymer (FRP) bars provide attractive solutions for these structures particularly for gaining corrosion resistance. This paper presents experimental results of eight two-span continuous concrete beams; two of them reinforced with pure glass fiber reinforced polymer (GFRP) bars and six of them reinforced with combinations of GFRP and steel bars. The continuous beams were tested under monotonically applied loading condition. The experimental load-deflection behavior and failure mode of the continuous beams were examined. In addition, the continuous beams were analyzed with a numerical method to predict the load-deflection curves and to compare them with the experimental results. Results show that there is a good agreement between the experimental and the theoretical load-deflection curves of continuous beams reinforced with pure GFRP bars and combinations of GFRP and steel bars.
본 논문에서는 암모니아의 전해 분해를 위한 분리막 반응기의 음극방 및 양극방에서 물의 전해에 따른 암모니아 용액의 pH 변화가 고찰되었으며, 단위 전해 셀이 적층된 다단 전해 반응기에서의 암모니아의 연속식 분해 특성이 평가되었다. 분리막을 가지는 반응기에서 암모니아 용액의 전해 반응 시, 양극에서는 pH가 8 이하에서부터 수소 이온이 생성되는 물 분해 반응이 일어나며, 음극에서는 pH가 11 이상에서부터 수산기 이온이 생성되는 물 분해 반응이 일어나 암모니아 용액의 pH를 변화시켜 암모니아 전해 분해에 영향을 크게 미쳤으며, 음이온 교환막을 사용하는 경우가 양이온 교환막을 사용하는 경우보다 양극방에서 암모니아 분해에 유리한 알카리 분위기를 보다 효과적으로 유지할 수 있었다. 분리막 전해 반응기의 특성을 이용하여 자체 pH 조정 기능을 가지는 연속식 암모니아 전해 반응기가 새롭게 고안하였고, 여기서는 pH-조정조 탱크 용액을 두고 이의 용액 일부를 음극방으로 순환시킴으로써, 양극방으로 주입되는 pH-조정조의 용액의 pH를 높여 암모니아 분해율을 높일 수 있었다. 또한, 그러한 반응기를 이용한 salt-free 연속식 암모니아 전해 분해 공정이 제시되었으며, 이러한 공정에서는 염소 이온을 가지는 암모니아 용액의 80%까지 연속적으로 암모니아를 환경에 무해한 질소로 분해 시킬 수 있었다.
In order to investigate the effects of continuous and pH cyclic remineralization, artificial enamel carious lesions were produced on sections of about 2mm thickness of bovine incisor. 0.1M lactic acid solution adjusted to a pH of 4.3 and saturity of $2.98{\times}10^{-8}$ was used as demineralizing solution. The lesions were remineralized with the solution of pH 7.0 containing 1.5mM $CaCl_2$, 0.9 mM $KH_2PO_4$ and 20mM Hepes. In continuous remineralization procedure, the specimens were immersed in remineralizing solution continuously for 72 hours. In pH cyclic remineralization procedure, the sections were immersed in remineralizing solution for 6 hours and in dermineralizing solution for 20 minutes alternately during 72 hours. The effect of remineralization was determined by polarizing microscopy and electron microprobe analysis. The obtained results were as follows. 1. In case of continuous remineralization, the lesion depth was reduced partially or unchanged. 2. In pH cyclic condition, the thichness of surface layer and the lesion depth were increased simultaneously. 3. The mineral content of lesion was recovered to the level of sound enamel by remineraiization.
Both lactic acid productivity and cell growth were linearly correlated with yeast extract supplementation in batch fermentation. During conventional continuous operation, although fresh feed was introduced into the bioreactor with a significantly low dilution rate (0.04 $h^{-1}$), the amount of yeast extract employed was not enough to maintain the growth of microorganism. However, when the fresh feed contained 100 g/l glucose and 2 g/l yeast extract during cell-recycle continuous operation at a dilution rate of 0.04 $h^{-1}$, more than 90 g/l lactic acid was continuously produced, with the average productivity of 3.72 $g/l{\cdot}h$. In this experiment, 82 g of yeast extract (77% of reduction yield) could be reduced for the production of 1 kg of lactic acid compared with batch fermentation of a similar volumetric productivity.
Hog manure slurry amended with sawdust was composted in pilot-scale reactor vessels using continuous aeration nuder different C/N ratios and pH conditions during composting high rate (decomposition) process. For each material two replicated piles were built and monitored over a period of three weeks. The compost piles had an initial volume of 0.18 ㎥. In this study we evaluated the temperature in compost O2 and CO2 evolution, aeration rate, NH3 concentration etc. and investigated the stability of compost during composting high rate process. The maximum NH3 concentration for the lower C/N(18∼19) and pH value of 6 reached 574∼1,063 ppm by the 16th through 11th days and the NH3 concentration during continuous aerated composting high rate process, it was more important to manage NH3 gas so that compost odor is reduced.

The production of chitosan from the mycelia of Absidia coerulea was studied to improve cell growth and chitosan productivity. Culture conditions were optimized in batch cultivation (pH 4.5, agitator speed of 250 rpm, and aeration rate of 2 vvm) and the maximum chitosan concentration achieved was 2.3g/L under optimized conditions. Continuous culture was carried out successfully by the formation of new growth spots under optimized conditions, with a chitosan productivity of 0.052g/L(sup)-1 h(sup)-1, which is the highest value to date, and was obtained at a dulution rate of 0.05h(sup)-1. Cell chitosan concentrations reached about 14% in the steady state, which is similar to that achieved in batch culture. This study shows that for the continuous culture of Absidia coerulea it is vital to control the medium composition.
Lactobacillus brevis BH-21의 균체를 고정화하여 column형 reactor를 이용하여 $\gamma$-aminobutyric acid의 연속적 생산을 검토하였다. Lactobacillus brevis BH-21에 의한 $\gamma$-aminobutyric acid 생산조건은 고정화 담체로는 3.0% sodium alginate가 좋았고 직경 2.2 mm의 bead를 사용하여 기질의 주입속도는 10 mL/h이 최적 조건이었다. Lactobacillus brevis BH-21의 고정화 균체와 유리 균체를 사용하여 $\gamma$-aminobutyric acid의 생산을 최적 조건하에서 1, 2, 3, 및 4차 발효 동안 48시간마다 신선한 기질을 주입하면서 비교ㆍ조사한 결과 고정화 균체를 사용 시에 생산량이 더 많았다. Column형 reactor에 의한 연속 발효 시 발효 25일까지 생산이 우수하였고 발효 30일까지는 $\gamma$-aminobutyric acid를 생산할 수 있었다.
