Butyrylcholinesterase Inhibitory Guaianolides from Amberboa ramosa

  • Phytochemical investigation of the whole plant of Amberboa ramosa led to the isolation of six sesquiterpene lactones which could be identified as $8{\alpha}$-hydroxy-$11{\beta}$-methyl-$1{\alpha}H,\;5{\alpha}H,\;6{\beta}H,\;7{\alpha}H,\;11{\alpha}H-guai-10(14)$, 4(15)-dien-6, 12-olide(2), $3{\beta},\;8{\alpha}-dihydroxy-11{\alpha}-methyl-1{\alpha}H,\;5{\alpha}H,\;6{\beta}H,\;7{\alpha}H,\;11{\beta}H-guai-10(14)$, 4(15)-dien-6, 12-olide (2), $3{\beta},\;4{\alpha},\;8{\alpha}-trihydroxy-4{\beta}(hydroxymethyl)-1{\alpha}H,\;5{\alpha}H,\;6{\beta}H,\;7{\alpha}H-guai-10(14)$, 11(13)-dien-6, 12-olide (3), $3{\beta},\;4{\alpha},\;8{\alpha}-trihydroxy-4{\beta}-(chloromethyl)-1{\alpha}H,\;5{\alpha}H,\;6{\beta}H,\;7{\alpha}H-guai-10(14)$, 11(13)-dien-6, 12-olide(4), $3{\beta},\;4{\alpha},\;dihydroxy-4{\beta}-(hydroxymethyl)-1{\alpha}H,\;5{\alpha}H,\;6{\beta}H,\;7{\alpha}H-guai-10(14)$, 11(13)-dien-6, 12-olide(5), $3{\beta},\;4{\alpha}-dihydroxy-4{\beta}-(chloromethyl)-8{\alpha}-(4-hydroxymethacrylate)-1{\alpha}H,\;5{\alpha}H,\;6{\beta}H,\;7{\alpha}H-guai-10(14)$, 11(13)-dien-6, 12-olide (6) by spectroscopic methods. All of them showed inhibitory potential against butyrylcholinesterase.

PEBAX-ZIF 복합막에 의한 Propylene/Propane의 기체투과 특성 (Gas Permeation Characteristics of Propylene/Propane in PEBAX-ZIF Composite Membranes)

  • PEBAX[poly(ether-block-amide)에 ZIF-8(zeolitic imidazolate framework)의 함량을 0, 1, 3, 7, 10, 20 wt%으로 하여 PEBAX-ZIF 복합막을 제조하였다. 기체투과 실험은 압력 $6kgf/cm^2$ 하에서 25, 35, $40^{\circ}C$로 온도를 달리하여 진행되었고, PEBAX-ZIF 복합막의 ZIF-8 함량 변화에 따른 $C_3H_6$$C_3H_8$의 기체투과도와 선택도($C_3H_6/C_3H_8$)를 조사하였다. $C_3H_6$$C_3H_8$의 투과기체에 대해 ZIF-8 함량 0~7 wt% 범위에서는 함량이 증가할수록 기체 투과도가 증가하다가 7~20 wt% 범위에서 함량이 증가하면 감소하는 경향을 보였다. 선택도($C_3H_6/C_3H_8$)는 PEBAX-ZIF 7 wt% 복합막에서 3.6~3.8의 값을 가지며, 가장 낮은 활성화에너지 값을 나타냈다.

In vitro and in vivo inhibition of Helicobacter pylori by Lactobacilllus paracasei HP7

  • The efficacy of standard therapeutic strategies for Helicobacter pylori (H. pylori) infection is decreasing over time due to the emergence of drug-resistant strains. As an alternative, the present study investigated the capacity of Lactobacilllus paracasei (L. paracasei) HP7, isolated from kimchi, to inhibit H. pylori growth. The effects of L. paracasei HP7 on H. pylori adhesion and H. pylori-induced inflammation were examined in AGS human gastric adenocarcinoma epithelial cells and a mouse model of H. pylori SS1 infection. L. paracasei HP7 reduced H. pylori adhesion to AGS cells and suppressed the inflammatory response in infected cells by downregulating interleukin-8. H. pylori colonization in the stomach of C57BL/6 mice was demonstrated by rapid urease test, and results showed significant decrease in mice post-treated with L. paracasei HP7. Additionally, L. paracasei HP7 decreased gastric inflammation and epithelial lesions in the stomach of H. pylori-infected mice. These results demonstrate that L. paracasei HP7 treatment can inhibit H. pylori growth and is thus a promising treatment for patients with gastric symptoms such as gastritis that are caused by H. pylori infection.

농도, 열처리 및 pH가 Caseinate의 거품성에 미치는 영향 (Effects of Protein Concentration, Heat Treatment and pH on the Foaming Properties of Caseinate)

  • Sodium caseinate의 농도, 가열온도 및 pH가 거품성에 미치는 영향을 조사하기 위하여 농도를 1.0, 3.0, 5.0, 7.0, 및 10.0%로, 열처리온도를 $55^{\circ}C$$65^{\circ}C$로, 그리고 pH를 6.0, 7.0 및 8.0으로 각각 조절하여 표면장력, 겉보기점도, 혼탁도, 거품형성능 및 거품안정성을 측정하였다. 표면장력은 농도, 열처리온도 및 pH에 따라 전체적으로 $0.0455{\sim}0.0496\;Nm^{-1}$로써 큰 변화가 없었고 겉보기점도는 농도 및 가열온도에 따라 pH 6.0, 7.0 및 8.0에서 각각 $0.0019{\sim}0.1050$, $0.0021{\sim}0.0440$$0.0022{\sim}0.0490Pa{\cdot}s$이었으며 혼탁도는 pH 6.0, 7.0 및 8.0에서 각각 $70.3{\sim}97.4$, $44.4{\sim}94.8$$36.6{\sim}93.9$이었다. 전 pH 영역을 통하여 가열온도에 따른 거품형성능의 증감은 있었으나 어떤 규칙적인 변화는 없었고 pH 6.0에서가 7.0 및 8.0 영역에서보다 거품형성능이 양호한 경향이었으며 거품형성능을 위한 적당한 단백질 농도는 $3.0{\sim}7.0%$이었다. 거품안정성은 전체적으로 10.0% 농도구에서, pH는 6.0 영역에서 가장 양호하였다. pH 6.0 및 7.0에서는 가열처리구가 대조구보다 거품안정성이 떨어지는 경향이었으나 pH 8.0의 10.0% 농도구의 경우는 모두 가열처리구가 대조구보다 안정성이 증가하였다(p<0.05). 표면장력, 겉보기점도 및 혼탁도의 증감에 따른 거품성의 규칙적인 변화는 나타나지 않았다.

한국산 톱다리개미허리노린재 집합페로몬 성분의 조성과 배합비율별 유인력 (Composition of the Aggregation Pheromone Components of Korean Bean Bug and Attractiveness of Different Blends)

  • 톱다리개미허리노린재(Riptortus clavatus Thunberg) (Heteroptera: Alydidae) 수컷 성충은 (E)-2-hexenyl (Z)-3-hexenoate (E2HZ3H), (E)-2-hexenyl (E)-2-hexenoate (E2HE2H) 및 tetradecyl isobutyrate(TI)의 세 성분으로 조성되는 집합페로몬을 분비하고, 그 페로몬에 동종의 암수 성충과 약충을 유인되며, 난기생봉인 Ooencyrtus nezarae (Hymenoptera: Encyrtidae)는 이 페로몬을 기주탐색에 이용한다. 본 연구에서는 한국의 진주와 익산, 일본의 Tsuhba와 Kumamoto지역에서 채집한 톱다리개미허리노린재의 집합페로몬 성분의 조성비를 조사하고, 각 성분의 배합비율별 유인력을 진주와 Kumamoto에서 조사하였다. 진주와 익산계통의 집합페로몬 조성 비율(E2HZ3H:E2HE2H:TI)은 각각 1:1.4:0.2, 1:0.8:0.2였으며, Tsuhba와 Kumamoto계통은 각각 1:2.8:0.2, 1:1.5:0.1이었다. 야외 유인력 검정에서, 1:1:1 (E2HZ3H:E2HE2H:TI=16.7:16.7:16.7mg/고무격막) 또는 1:1:0.5 (E2HZ3H:E2HE2H:TI=20:20:10mg/고무격막)로 배합한 페로몬이 1:5:1 (E2HZ3H:E2HE2H:TI = 7.1:35.7:7.1mg/고무격막)로 배합한 것보다 유인력이 높았다.

산성 pH가 박테리오로돕신의 분광학적 성질에 미치는 효과 (The Effect of Acidic pH on the Spectral Properties of Bacteriorhodopsin)

  • Halobacterium halobium으로부터 분리 정제된 퍼플멤브레인을 7.5% 폴리아크릴아미드겔에 혼합시켰다. 이 겔을 사용하여 pH 변화에 따른 흡수 스펙트라와 원편광 이색성스펙트라를 얻었다. pH 7.0에서 보여준 이들 스펙트라의 성질들은 수용액내에 부유하고 있는 퍼플멤브레인으로부터 얻은 것들과 동일하였다. pH 2.7에서는 최대흡광도를 605nm에서 나타내었으며 pH 0.8에서는 565nm에서 보여주었다. 광에 노출되지 않았던 겔의 경우는 광에 노출되었던 겔과는 달리 등흡광점을 보여주었다. pH 2.7과 pH 0.8에서 측정한 원편광 이색성스펙트라는 pH 7.0에서 보여준 bilobed형을 유지하였으며 UV영역에서 분자편광도나 스펙트라의 모양도 pH에 따라 큰 영향을 받지 않았다. pH 2.7에서 생성된 bR605acid 가 퍼플멤브레인의 정상 광화학 순환기 중간 생성물인 $O^{640}$와 유사한 성질을 가진종이 아닐까 추측된다.

Kinetic Study on Aminolysis of Y-Substituted-Phenyl Picolinates: Effect of H-Bonding Interaction on Reactivity and Transition-State Structure

  • A kinetic study is reported on nucleophilic substitution reactions of Y-substituted-phenyl picolinates (7a-7h) with a series of cyclic secondary amines in 80 mol % $H_2O$/20 mol % DMSO at $25.0{\pm}0.1^{\circ}C$. Comparison of the kinetic results with those reported previously for the corresponding reactions of Y-substituted-phenyl benzoates (1a-1f) reveals that 7a-7h are significantly more reactive than 1a-1f. The Br${\o}$nsted-type plot for the aminolysis of 4-nitrophenyl picolinate (7a) is linear with ${\beta}_{nuc}=0.78$, which is typical for reactions proceeding through a stepwise mechanism with expulsion of the leaving group being the rate-determining step. The Br${\o}$nsted-type plots for the piperidinolysis of 7a-7h and 1a-1f are also linear with ${\beta}_{lg}=-1.04$ and -1.39, respectively, indicating that the more reactive 7a-7h are less selective than the less reactive 1a-1f to the leaving-group basicity. One might suggest that the enhanced reactivity of 7a-7h is due to the inductive effect exerted by the electronegative N atom in the picolinyl moiety, while the decreased selectivity of the more reactive substrates is in accord with the reactivity-selectivity principle. However, the nature of intermediate (e.g., a stabilized cyclic intermediate through the intramolecular H-bonding interaction for the reactions of 7a-7h, which is structurally not possible for the reactions of 1a-1f) is also responsible for the enhanced reactivity with a decreased selectivity.

  • 1M $NaCIO_4$용액에서 다중몰리브덴산염 음이온 평형에 대한 온도의 영향을 20~50$^{\circ}$C온도 범위 내에서 고찰해 보았다. 이 온도 범위에서 생성된 다중몰리브덴산염 음이온은 평형 온도에 관계없이 헵타몰리브덴산염($Mo_7O_{24}^{-6}$) 이온과 헵타몰리브덴산염의 양성자화된 꼴($H_LMo_7O_{24}^{(6-L)-}$) 이었다. 실렌의 방법을 사용하여 계산한 헵타몰리브덴산염 형성에 대한 평형상수는 지적한 평형 온도에서 각각 다음과 같다. $8H^{+}+7MoO_4^{2-}=Mo_7O_{24}^{6-}+4H_2O$, $k_{7.8}=2.77{\times}10^{53}:20^{\circ}C= 9.29{\times}10^{51}:40^{\circ}C$,$k_{7.8}= 4.22{\times}10^{52}:30^{\circ}C = 9.29{\times}10^{51}:50^{\circ}C$ 위 반응에 대하여 반트호프 방정식을 이용하여 계산한 엔탈피 변화는 31.50kcal/mole이었다. 프로톤화헵타몰리브덴산염 이온 형성에 대한 평형상수 계산법을 유도하여 프로톤화헵타몰리브덴산염 형성에 대한 평형상수를 계산하였으며 그 결과는 다음과 같다.$ LH + Mo_7O_{24}^{-6} = H_LMo_7O_{24}^{(6-L)-} : L = 1\;or\;2$, $k_1 = 2.31{\times}10^4=2.53{\times}10^4=2.76{\times}10^4= 3.10{\times}10^4$, $k_2 = 6.19{\times}10^7\;20^{\circ}C = 7.80{\times}10^7\;30^{\circ}C = 1.22{\times}10^8\;40^{\circ}C = 2.03{\times}10^8\;50^{\circ}C$ 다음 단계 반응에 대하여 반트호프 방정식에 의하여 계산한 엔탈피 변화는 다음과 같다. $H^{+}+Mo_7O_{24}^{6-}= HMo_7O_{24}^{5-}\;{\Delta}H^{\circ}=1.900 kcal/mole$, $2H^{+}+Mo_7O_{24}^{6-}=H_2Mo_7O_{24}^{4-}\;{\Delta}H^{\circ}=7.500kcal/mole$

The hyaluronan synthesis inhibitor 7-hydroxy-4-methylcoumarin inhibits LPS-induced inflammatory response in RAW 264.7 macrophage cells

  • 7-Hydroxy-4-methylcoumarin (7H-4MC) inhibits hyaluronan production in multiple cell lines and tissue types both in vitro and in vivo. It is a commercially available drug approved for human use, called hymecromone, in European and Asian countries to prevent biliary spasms. Nevertheless, as the pharmacological efficacy of 7H-4MC has not yet been reported in macrophages, this study investigated its anti-inflammatory effects and mechanism of action using lipopolysaccharide (LPS)-induced RAW 264.7 macrophages. LPS-induced RAW 264.7 cells were treated with various concentrations of 7H-4MC (62.5, 125, 250, and 500 μM). The application of 7H-4MC significantly reduced nitric oxide and prostaglandin E2 production without cytotoxic effects. Additionally, 7H-4MC strongly decreased the expression of inducible nitric oxide synthase and cyclooxygenase. Furthermore, 7H-4MC reduced the production of proinflammatory cytokines, such as tumor necrosis factor-α, interleukin (IL)-1β, and IL-6. Finally, 7H-4MC exerted its potent anti-inflammatory actions via the upregulation of IκB-α production, which led to the inhibition of nuclear factor-κB (NF-κB) activity. These results, obtained in macrophage cell lines, suggest that 7H-4MC prevents inflammatory diseases via the NF-κB signaling pathway and that its use could be beneficial for human health. Ultimately, this is the first report describing the anti-inflammatory activity of 7H-4MC in a macrophage cell line.

발효유제품내에서 Escherichia coli O157:H7과 Salmonella ser. typhimurium의 생존 (Survival of Escherichia coli O157:H7 and Salmonella ser. typhimurium in Fermented Milk Products)

  • Escherichia coli O157:H7 and Salmonella ser. typhimurium are pathogens involved in food poisoning in numerous countries. This study aimed to obtain knowleges on the survival of E. coli O157:H7 KSC 109 and S. ser. typhimurium ATCC 14028 in fermentedmilk products which were on sale in Suwon Yakult supplier. To the final concentration of 103~104 cfu/$m\ell$ of E. coli O157:H7 KSC 109 or S. wer. typhimurium ATCC 14028 in the fermented milks, Metchnikoff, Ace, Yakult, Mastoni and Super 100 were inoculated with these pathogens and then were stored at 4$^{\circ}C$ and viable cells of these pathogens were periodically counted. The results showed that the survival of two pathogens differed in the different types of fermented milks tested. Number of suriviving E. coli O157:H7 KSC 109 and S. ser. typimurium ATCC 14028 cells (initial inoculum, 103~104 cfu/$m\ell$) were decreased to 101, 102 cfu/$m\ell$ in Ace after 100 hours, and were decreased gradually to 101 cfu/$m\ell$ in Yakult after 250 hours. In the other fermented milks, viable cells of E. coli O157:H7 KSC 109 was not drastically decreased but those of S. ser. typhimurium ATCC 14028 was decreased gradually to 102 (Mastoni), and to 101 cfu/$m\ell$ (Super 100) after 250 hours. It appeared that S. ser. typhimurium ATCC 14028 was more susceptible than E. coli O157:H7 KSC 109 at low pH. Vibale cells of E. coli O157:H7 KSC 109 was not drastically decreased in most of fermented milks tested except Ace and Yakult, but in general, S. ser. typhimurium ATCC 14028 was drastically decreased in most of the fermented milks. The major ingibition factor against these pathogens in the fermented milks during storage at 4$^{\circ}C$ appeared to be the acidity and the metabolites produced by the starters bacteria used in fermented milk products.
