A Strategy to Increase Microbial Hydrogen Production, Facilitating Intracellular Energy Reserves

  • Overexpression of the genes encoding phosphoeneolpyruvate carboxykinase (pckA) and NAD-dependent malic enzyme (maeA) facilitates higher intracellular ATP and NAD(P)H concentrations, respectively, under aerobic conditions in Escherichia coli. To verify a hypothesis that higher intracellular energy reserves might contribute to H2 fermentation, wild-type E. coli strains overexpressing pckA and maeA were cultured under anaerobic conditions in a glucose minimal medium. Overexpression of pckA and maeA enabled E. coli to produce 3-times and 4-times greater H2 (193 and 284 nmol, respectively) than the wild type (66 nmol H2). The pckA and maeA genes were further overexpressed in a hydrogenase-3-enhanced E. coli strain. The hydrogenase-3-enhanced strain (W3110+fhlA) produced 322 nmol H2, whereas the ATP-enhanced strain (W3110+fhlA+pckA) produced 50% increased H2 (443 nmol). Total H2 in the NAD(P)H-enhanced strain (W3110+fhlA+maeA) was similar to that in the control strain at 319 nmol H2. Possible explanations for the contribution of the increased cellular energy reserves to the enhanced hydrogen fermentation observed are discussed based on the viewpoint of metabolic engineering strategy.

  • This paper deals with a robust ${\mathfrak{H}}_{\infty}$ controller design problem for waypoints tracking of large displacement unmanned underwater vehicles (LDUUVs) in Takagi-Sugeno fuzzy form. The LDUUV model uses a rudder to control its horizontal motion. We determine the order of waypoints based on their priorities and consider only surge force. A fuzzy controller in state-feedback form is taken and its design condition of is represented in terms of linear matrix inequalities. A numerical simulation is included to show the effectiveness of the theoretical development.

  • This paper deals with $H_{\infty}$ fuzzy control problem of discrete-time nonlinear Markovian jump systems with time delay. The Takgi and Sugeno fuzzy model is employed to represent a delayed nonlinear system that possesses Markovian jump parameters. A stochastic mode dependent Lyapunov function is employed to analyze the stability and $H_{\infty}$ disturbance attenuation performance of the Markovian jump fuzzy system with time delay. Stochastic Lyapunov function is dependent on the operation modes of the system. A sufficient condition for the existence of fuzzy $H_{\infty}$ controller are given in terms of matrix inequalities. Also numerical example is presented to illustrate the efficient of the proposed design methods.

Effects of pH Change by CO2 Induction and Salinity on the Hatching Rate of Artemia franciscana

  • To understand the effects of lower pH levels due to elevated $CO_2$ and salinity, we designed and constructed a pH-control system that included automatic $CO_2$ infusion and measured the hatching rate of a crustacean model species, Artemia franciscana. The pH-control system was cost-effective and capable of performing animal tests in which pH fluctuated around $8.0{\pm}0.1$, with the temperature around $27{\pm}0.5^{\circ}C$. Hatching rate was observed under four different pH levels (7.0, 7.3, 7.6, and untreated control) combined with three salinity ranges (15, 25, and 35 ppt). The results demonstrated that lower pH levels led to decreased hatching rates regardless of salinity, and the minimum hatching rate was detected at pH 7.0 compared to the control (pH $8.0{\pm}0.1$), supporting the idea that OA has adverse effects on hatching rates and increases the risk of juveniles being introduced in the ecosystem. In contrast, salinity changes exhibited no synergistic effects with pH and had independent effects.

  • In this paper, the genetic algorithm is used to design a mixed H₂/ H/sub ∞/ controller Two kinds of controller forms, Youla's form and the general form, are considered to design a mixed H₂/ H/sub ∞/ controller. Efficient searching methods are sought to minimize the given H₂cost function under the H∞ constraint. It is verified by an example that the developed algorithm can provide stable results in the region where unstable results are shown by the conventional gradient method.

Effects of Intraperitoneally Administered Lipoic Acid, Vitamin E, and Linalool on the Level of Total Lipid and Fatty Acids in Guinea Pig Brain with Oxidative Stress Induced by H2O2

  • The aim of our study was to investigate the protective effects of intraperitoneally-administrated vitamin E, dl-alpha lipoic acid, and linalool on the level of total lipid and fatty acid in guinea pig brains with oxidative stress that was induced by $H_2O_2$. The total brain lipid content in the $H_2O_2$ group decreased when compared to the $H_2O_2$ + vitamin E (p<0.05), $H_2O_2$ + linalool (p<0.05), ALA (p<0.05), control (p<0.01), linalool (p<0.01), and vitamin E (p<0.01) groups. While the proportion of total saturated fatty acid (${\Sigma}SFA$) in the $H_2O_2$ group significantly increased (p<0.005) when compared to the vitamin E group, it only slightly increased (p<0.01) when compared to the control and $H_2O_2$ + vitamin E groups. The ratio of the total unsaturated fatty acid (${\Sigma}USFA$) in the $H_2O_2$ groups was lower (p<0.05) than the control, vitamin E, and $H_2O_2$ + vitamin E groups. The level of the total polyunsaturated fatty acid (${\Sigma}PUEA$) in the $H_2O_2$ group decreased in when compared to the control, vitamin E, and $H_2O_2$ + vitamin E groups. While the proportion of the total w3 (omega 3), w6 (omega 6), and PUFA were found to be lowest in the $H_2O_2$ group, they were slightly increased (p<0.05) in the lipoic acid group when compared to the control and $H_2O_2$ + lipoic acid groups. However, the level of ${\Sigma}SFA$ in the $H_2O_2$ group was highest; the level of ${\Sigma}USFA$ in same group was lowest. As the proportion of ${\Sigma}USFA$ and ${\Sigma}PUFA$ were found to be highest in the linalool group, they were decreased in the $H_2O_2$ group when compared to the control group. Our results show that linalool has antioxidant properties, much the same as vitamin E and lipoic acid, to prevent lipid peroxidation. Additionally, vitamin E, lipoic acid, and linalool could lead to therapeutic approaches for limiting damage from oxidation reaction in unsaturated fatty acids, as well as for complementing existing therapy for the treatment of complications of oxidative damage.

  • This paper presents a design technique of a Takagi-Sugeno (T-S) fuzzy-model-based $H_{\infty}$ controller for autonomous underwater vehicles (AUVs). The design procedure aims to render the stabilizing controller which satisfies performance of the diving control for AUVs in the presence of the disturbance. A nonlinear AUV is modeled by the T-S fuzzy system through the sector nonlinearity. By using Lyapunov function, the sufficient conditions are derived to guarantee the performance of robust depth control in the format of linear matrix inequality (LMI). To succeed for diving control of AUV, we add the constraints on the diving and pitch angles in the LMI conditions. Through the simulation, we confirm the effectiveness of the proposed methodology.

Hydrogen Sulfide Alleviates Seed Germination Inhibition in Oilseed Rape (Brassica napus L.) Under Salt Stress

  • The germination process is critical for plant growth and development and it is largely affected by environmental stress, especially salinity. Recently, hydrogen sulfide (H2S) is well known to act as a signaling molecule in a defense mechanism against stress conditions but poorly understood regulating seed germination. In this study, the effects of NaHS (the H2S donor) pretreatment on various biochemical (hydrogen peroxide (H2O2) content and amylase and protease activity) and physiological properties (germination rate) during seed germination of oilseed rape (Brassica napus L. cv. Mosa) were examined under salt stress. The seed germination and seedling growth of oilseed rape were inhibited by NaCl treatment but it was alleviated by NaHS pretreatment. The NaCl treatment increased H2O2 content leading to oxidative stress, but NaHS pre-treatments maintained much lower levels of H2O2 in germinating seeds under salt stress. Amylase activity, a starch degradation enzyme, significantly increased over 2-fold in control, NaHS pretreatment, and NaHS pretreatment under NaCl during seed germination compared to NaCl treatment. Protease activity was highly induced in NaHS-pretreated seeds compared to NaCl treatment, accompanied by a decrease in protein content. These results indicate that NaHS pretreatment could improve seed germination under salt stress conditions by decreasing H2O2 accumulation and activating the degradation of protein and starch to support seedling growth.

Significance of varying concentrations of T-2 toxin on growth performance, serum biochemical and hematological parameters in broiler chickens

  • The study was to determine the effects of diverse concentrations of T-2 toxin in broiler diet. Three hundred 1-day-old chicks with initial body weight of 46 ± 0.52 g were chosen and randomly assigned into five dietary treatments with 5 replicate cages and 12 broilers per cage for 42 d feeding trial. Dietary treatments were prepared with basal diets containing 0 (T1), 50 (T2), 100 (T3), 150 (T4), 200 (T5) ppb T2-toxin. Significant results were observed in the decreased intake of feed, feed conversion ratio (FCR), body weight gain (BWG), level of serum protein, cholesterol and hemoglobulin of broilers in increased concentration of the T-2 toxin in diet (150 and 200 ppb) groups than control. Also, observed that the uric acid, serum glutamic pyruvic transferase (SGPT), serum glutamic oxaloacetic transferase (SGOT) and Heterophil/Lymphocyte (H/L) ratio value were significantly higher (p < 0.05) in groups T4 and T5 than control. However, the BWG, feed intake and FCR, as well blood biochemical profiles of serum protein, cholesterol, hemoglobulin, uric acid, SGPT, SGOT and H/L ratio in groups T2 and T3 were statistically similar to control diet of broilers. It was concluded that the results showed that no adverse effects on growth performance and blood biochemical parameters in broilers feed with T-2 toxin (50 and 100 ppb) during the entire trial.

A study of struvite control using CO2 in sewage treatment process (하수처리공정에서 이산화탄소를 이용한 스트러바이트 제어에 관한 연구)

  • Sludge transporting pipes in wastewater treatment plant are easy to be clogged with struvite when the digested sludge and dehydrated filtrate are transported through the pipes, which lowers the efficiency of sludge treatment system in a WWTP. pH is one of the most important factors in struvite formation, and carbon dioxide separated from biogas can be used to control pH and struvite formation. By controlling pH, the amount of dehydrating agent can be reduced by about 10%, which saves the budget for facility maintenance. As $CO_2$ is reused and dehydrating chemicals are saved, the approach can contribute to global warming gas reduction.
