Deep Learning Based Digital Staining Method in Fourier Ptychographic Microscopy Image (Fourier Ptychographic Microscopy 영상에서의 딥러닝 기반 디지털 염색 방법 연구)

  • In this study, H&E staining is necessary to distinguish cells. However, dyeing directly requires a lot of money and time. The purpose is to convert the phase image of unstained cells to the amplitude image of stained cells. Image data taken with FPM was created with Phase image and Amplitude image using Matlab's parameters. Through normalization, a visually identifiable image was obtained. Through normalization, a visually distinguishable image was obtained. Using the GAN algorithm, a Fake Amplitude image similar to the Real Amplitude image was created based on the Phase image, and cells were distinguished by objectification using MASK R-CNN with the Fake Amplitude image As a result of the study, D loss max is 3.3e-1, min is 6.8e-2, G loss max is 6.9e-2, min is 2.9e-2, A loss max is 5.8e-1, min is 1.2e-1, Mask R-CNN max is 1.9e0, and min is 3.2e-1.

EXPERIMENTAL STUDY ON THE EFFECT OF X-IRRADIATION IN THE RAT PERIODONTAL LIGAMENT (X선조사가 발육치근막에 미치는 영향에 관한 실험적 연구)

  • The author studied on the effects of X-ray irradiation to the development of periodontal ligament in gestation rats. They were irradiated in their abdomen with 100, 200 and 300 rads respectively in one shot irradiation with deep radiation therapy equipment (MAXIMAR 250-Ⅲ), In 7th, 14th, 21th and 28th day after delivery, those new born rats were respectively sacrificed with ether anesthesia and removed of their mandibles. After removal, those mandibles were fixed in 10% neutral buffer formalin, decalcified with 5% trichloroacetic acid for 5 days and embedded with paraffine. Staining was performed with H-E, Van Gieson, Mallory azan, Bielshowsky-Gomori silver stain and Halmi's oxytalan fiber stain. The results were as follows: 1. Before tooth eruption, all the fiber: components in dental sac were almost always oriented near the outer enamel epithelial \layer. But in irradiated new born rats, those collagen fiber orientation was more irregular than those of control groups, and this phenomenon was more severe in proportion to the amount of irradiation in the gestation period. 2. Before tooth eruption, the connective tissue fibers in periodontal ligament were stained with lighter in the irradiated groups than those of control groups. Oxytalan fibers of irradiated groups were thin and splitting pattern of their fiber morphology to compare with those of control groups. 3. After tooth eruption, the periodontal ligament fibers of irradiated groups were oriented functionally and their morphology was thick, fine and heavy staining. Oxytalan fibers were revealed with oblique parallel arrangement in the periodontal ligament of irradiated groups.

  • It is known that radiation therapy is a kind of treatment choices of the maxillofacial tumors. This study is designed to investigate the effects of irradiation on rat's tongue tissues as functional tissues which relate to taste, mastication, and pronunciation. 88 rats (Sprague Dawley branch, male) were divided into control group of 4 and experimental group of 84. Experimental group was singly exposed to Co-60 irradiation with 8, 13, 18 Gy in the head and neck region. Animals were sacrificed on 1 hour, 3 hours, 6 hours, 1day, 3 days, 7 days, and 28 days after the irradiation. The specimens were observed by histopathological examination employing H-E stain and Van-Gieson stain. The results were follows; 1. The tongue tissue were severely swollen on the 1 hour after irradiation, but gradually decreased in course of time. 2. The basal cells of epithelium of tongue proliferated at initial stage of irradiation, but gradually decreased. The Keratin layer were gradually increased. 3. The muscles within the tongue were severely degenerated at initial stage of irradiation, but gradually recovered almost normally. 4. The tissue changes after irradiation were gradually increased by the degree of irradiation.

Potential of Epicoccum purpurascens Strain 5615 AUMC as a Biocontrol Agent of Pythium irregulare Root Rot in Three Leguminous Plants

  • Epicoccum purpurascens stain 5615 AUMC was investigated for its biocontrol activity against root rot disease caused by Pythium irregulare. E. purpurascens greenhouse pathogenicity tests using three leguminous plants indicated that the fungus was nonpathogenic under the test conditions. The germination rate of the three species of legume seeds treated with a E. purpurascens homogenate increased significantly compared with the seeds infested with P. irregulare. No root rot symptoms were observed on seeds treated with E. purpurascens, and seedlings appeared more vigorous when compared with the non-treated control. A significant increase in seedling growth parameters (seedling length and fresh and dry weights) was observed in seedlings treated with E. purpurascens compared to pathogen-treated seedlings. Pre-treating the seeds with the bioagent fungus was more efficient for protecting seeds against the root rot disease caused by P. irregulare than waiting for disease dispersal before intervention. To determine whether E. purpurascens produced known anti-fungal compounds, an acetone extract of the fungus was analyzed by gas chromatography mass spectrometry. The extract revealed a high percentage of the cinnamic acid derivative (trimethylsiloxy) cinnamic acid methyl ester. The E. purpurascens isolate grew more rapidly than the P. irregulare pathogen in a dual culture on potato dextrose agar nutrient medium, although the two fungi grew similarly when cultured separately. This result may indicate antagonism via antibiosis or competition.

  • It is known that radiation therapy is a kind of treatment choices of the maxillofacial tumors. This study is designed to investigate the irradiation effects on rat's periodontal tissues as functional tissues which relate to tooth-support, hard tissue formation and destruction. 20 rats (Sprague-Dowley branch, male) were devided into control group of 4 and experimental groupe of 16. Experimental group was singly exposed to Co-60 irradiation with 10 Gy in the head and neck region. Animals were sacrificed on 2 days, 1 week, 2 weeks and 3 weeks after the irradiation. The specimens were observed by histopathological examination employing H-E stain, van-Gieson stain and PA-ACH fluorescent stain. The results were as follows: 1. Cementoblasts and osteoblasts were gradually lost and rearranged along the external surfaces of the cementum and alveolar bone, but osteoclasts were almost not affected. 2. The cell numbers of the periodontal ligament were decreased due to the cellular atrophy and degeneration, but recovered almost normally on the 3rd week after irradiation. 3. The collagen fibers within the periodontal ligament were irregularly oriented, became finer and decreased in number. 4. The vessels of the periodontal ligament were decreased at the initial stage but increased again on the 2nd week after irradiation, and the hemorrhagic appearances, occurred within the tissues, due to the arterial destruction, were lasted until 3 weeks after irradiation. 5. The glycogen within the periodontal ligament was gradually increased and stored in the matrices of the cemental side on the 1st week after irradiation, but recovered almost normally on the 3rd week after irradiation.

  • $TGF-{\beta}$ is one of growth factors that may be involved in the formation of bone and cartilage. Multiple studies demonstrate that $TGF-{\beta}$ is involved in regulating cell proliferation, differentiation and matrix synthesis, events observed in frature healing. The apperance of $TGF-{\beta}$ in the fracture during healing was evaluated by immunohistologic localization of $TGF-{\beta}$ using antibody. Twenty Sprauge-Dawley strain white male rats, each weiging about 150grams were used and divided two groups. The one group, the $2{\times}2mm$ bony defect was formed in the right femur. The other group, $4{\times}2mm$ bony defect was formed in right femur. Both group were sacrificed at 3day, 1, 2, 3, 4 week and femur were harvested, paraffin sections were stained with H & E, MT stain, immunihistochemical staining with $TGF-{\beta}$ antibody and observed under light microscope. The result were as follows: 1. New bone formation and cartilagenous tissue was seen at 3day. And in the $2{\times}2mm$ bony defect group, $TGF-{\beta}$ stained the cell surounding new bone. 2. The osteoclast and trabecular was seen at 1week. $TGF-{\beta}$ stained the osteoblast and in the $2{\times}2mm$ bony defect group was stained more than $4{\times}2mm$ bony defect group. 3. The lamella bone and trabecule was seen from 3, 4week, and $TGF-{\beta}$ stained almost negative. From the above finding, we could concluded that $TGF-{\beta}$ stained the osteoblast at early stage and 1week, the peak stain was seen from 1week, and then decreased, almost negative stain was seen at 3, 4week.

Leukocyte-Endothelial Cell Adhesion Induced by Ischemia and Reperfusion Observed with in vivo Videomicroscopy (허혈-재관류에 의해 유도된 백혈구-혈관내피세포 유착에 대한 Videomicroscopy 영상소견)

  • Purpose : Recent evidence suggests a possible role for leukocytes in brain injury following ischemia and reperfusion. This study examined the temporal profile of ischemic tissue damage and leukocyte response after transient middle cerebral artery occlusion(MCAO) with reperfusion in the mouse. Methods : Focal cerebral ischemia was made by temporary occluding of the stem of the proximal MCA. Two groups of the mouse were investigated : (1) sham operation(n=10), and (2)those having the arterial occlusion released after 90 minute(n=20). By 4 hours(n=10) and 24 hours(n=10) after the onset of ischemia-reperfusion, fluorescein videoimages were under-taken in the pial venules of the mouse using a closed cranial window technique. Rhodamine 6G was administered as a $80-100{\mu}l/min$ i.v. loading dose and a $30-40{\mu}l/min$ i.v. maintenance dose in saline to selectively label circulating leukocytes. Neuropathologic evaluation for brain injury was accomplished using the histochemical stain 2,3,5-triphen-yltetrazolium chloride(TTC) and hematoxylin and eosin(H & E) stain. Results : The mean number of adherent leukocytes to cerebral venules in the 90 minutes MCAO and 24 hours reperfusion group were $306{\pm}24$ compared with $72{\pm}8$ in the sham operation group. In the TTC staining method, the cortical infarct affecting 34.8% of hemispheric volume were created in all of animals (n=10) undergoing 90 minute MCAO with 24 hours reperfusion, but the infarcted area were not found in the other(sham operation and 90 minute MCAO with 4 hours reperfusion)groups. In the H & E stain, the brain tissue following 90 minute MCAO with 4 hours reperfusion revealed only a pyknosis of the nuclei with shrunken cytoplasm, but infiltrated leukocytes were not observed. After 24 hours of reperfusion, a many leukocytes were infiltrated within parenchyma and blood vessles. Conclusions : These findings demonstrate the feasiblity of continous in vivo monitoring of leukocyte adherence in cerebral venules and suggest that reperfusion induced leukocyte adherence to venular endothelium may contribute to tissue injury following focal cerebral ischemia.

Histological Comparison Study with Primo Node and Immature Liver Tissue on Liver Surface in Rat (흰쥐의 간 장기 표면에서 관찰되는 프리모 노드와 미성숙 간조직과의 조직학적 비교연구)

  • 목적 : 프리모 모드와 미성숙 간조직에 대한 조직학적 특성을 비교하고자 하였다. 방법 : 흰쥐에서 간절제한 후, 간장기 표면에서 관찰되는 프리모 모드와 그와 유사한 미성숙 간조직을 H&E, Oil red O, Masson trichrome and van Gieson 염색방법을 통해서 비교 연구하였다. 결과 : 상기 실험 연구를 진행한 결과, 다음과 같은 결론을 얻었다. 1. 조직의 일반적인 특징을 관찰할 수 있는 H&E 염색결과, 프리모 노드에서는 많은 수의 조그마한 동(sinus)이 관찰되었고 적혈구나 영양을 공급하는 혈관의 분포는 관찰되지 않았다. 이와 반대로 미성숙 간조직에서는 동이 관찰되지 않았으며 혈관의 분포와 적혈구가 관찰되었다. 2. 양 조직의 지방성분의 유무를 관찰하기 위한 Oil red O 염색결과, 프리모 노드에서는 지방성분이 관찰되지 않았으나 미성숙 간조직에서는 관찰되었다. 3. 양 조직의 콜라겐성분의 유무를 관찰하기 위한 Masson trichrome and van Gieson 염색결과, 프리모 노드에서는 약간의 콜라겐 성분이 관찰되었으나 elastic 성분은 관찰되지 않았으며, 미성숙 간조직에서는 콜라겐 성분과 elastic 성분이 관찰되지 않았다. 결론 : 상기 결과는 프리모 노드와 간조직과는 조직학적 성질이 다른 것으로 사료되며, 특히 콜라겐 성분의 적은 결과는 프리모 노드가 불규칙한 형태를 이루고 있는 이유에 대한 실마리를 제공하였다. 이러한 결과는 프리모 노드의 특성에 대한 기본 자료로 활용될 수 있을 것이다.

Allergy Prevention Effect of Oral Administration of Quercetin on Picryl Chloride-induced Contact Dermatitis in BALB/c Mice (BALB/c mice에서 quercetin의 경구투여가 picryl chloride로 유발된 접촉성 피부 알레르기의 예방에 미치는 영향)

  • The number of patients with immune- mediated dermatitis such as contact dermatitis is increasing year by year. Allergic contact dermatitis is a complex phenomenon that involves resident epidermal cells, fibroblasts, and endothelial cells, as well as invading leukocytes that interact with each other under the control of a network of cytokines and lipid mediators. It is a cell-mediated immune reaction, which occurs after susceptible individuals are exposed to sensitizing chemicals, and characteristic eczematous reaction is seen at the point of contact with an allergen. In this study, we investigated the allergy prevention effects of quercetin on mast cell-mediated allergic inflammation in BALB/c mice. BALB/c mice were sensitized with 40 ${\mu}l$ of 1.5% picryl choloride (PCL) to the left and right ear each. Total serum IgE levels and histamine levels were measured by the sandwich ELISA method using mouse IgE, histamine measuring kit. For histopathological examination, paraffin sections were stained with hematoxylin and eosin(HE) or toluidine blue(TB). Ear swelling responses were much weaker in the high-dose group (100 mg/kg) than the control group (0 mg/kg). The number of mast cells showed a significant decrease in the high-dose group (100 mg/kg) compared to the control group (0 mg/kg). Degranulation of mast cells was also confirmed by Toluidine Blue (TB) staining method. Both total serum IgE and histamine levels were significantly decreased in the high-dose group (100 mg/kg) compared to other groups. These findings suggest a certain relationship between the elevation of IgE, histamine levels and the degranulation of mast cells. These results show that the pharmacological actions of quercetin indicate its potential activity for prevention of allergic inflammatory diseases through the down-regulation of mast cell activation.

A Study on Natural Dye Having the Effects on the Atopic Dermatitis (Part II) - Pine Needles Extract - (아토피성 피부염병변에 치료효과가 있는 천연염료 연구(II) - 솔잎 추출물 -)

  • The purpose of this study was to investigate usefulness of the pine needles extract on Atopic Dermatitis(AD)-like skin lesions. To investigate the effect of pine needles extract in vivo, atopic dermatitis (NC/Nga) mice using DNCB (2.4-Dinitrochlorbenzene) was used. NC/Nga mice were challenged with DNCB during 2 weeks to develope AD-like skin lesion. After that, pine needles extract was applied to AD-like skin lesion on the backs of the NC/Nga mice during 3 weeks. The efficacy of pine needles extract in the NC/Nga mice was evaluated by measurement of the skin lesion severity(NC mouse score), the serum IgE level, epidermal thickness changes, and mast cell number. Blood was collected from the retro-orbital area and the level of IgE in the blood was measured. The epidermal thickness and mast cell number were observed by microscopic method after H&E stain. The serum IgE levels were decreased after treatment with pine needles extract. The epidermal thickness and mast cell number were decreased after treatment with pine needles extract. To conclude, the topical application of pine needles extract suppressed the progression of AD-like skin lesion.
