석이중 오르시놀 정량 및 급성독성 (Quantitative Analysis of Orcinol and Acute Toxicity of Gyrophora esculenta)

  • In previous study, Gyrophora esculenta showed significant inhibitory effect on $\alpha$-glucosidases in vitro and blood glucose elevation in vivo. In the isolating process of active substance, orcinol was separated from Gyrophora esculenta. Orcinol is known to be toxic, therefore, in this study, it was analysed by the TLC densitometry method for quantitative determination from Gyrophora esculenta. The average amount of orcinol of Gyrophora esculenta was 0.2%. For the purpose of removing orcinol, the water extract of Gyrophora esculenta was sequentially fractionated by organic solvents, and the acute toxicity of each fraction was assessed in mice. Among them, the LD50 of butanol fraction was 1.19 g/kg(p.o.) and the weight increase of mice in that group was somewhat retarded.

석이에서 분리한 GE974의 ${\alpha}-Glucosidase$ 저해효과 (Inhibitory Effect of GE974 isolated from Gyrophora esculenta on ${\alpha}-Glucosidase$)

  • This study was conducted to search for the ${\alpha}-glucosidase$ inhibitor from the natural products. In the previous study, the water extract of Gyrophora esculenta exhibited a potent inhibitory effect on ${\alpha}-glucosidase$ activities. Then, by bioassay-guided fractionation followed by chromatographic separation of the water extract of Gyrophora esculenta, ${\alpha}-glucosidase$ inhibitor was isolated as GE974. GE974 showed significant inhibitory activities on some kinds of ${\alpha}-glucosidase$ in vitro. Its inhibitory mechanism seemed to be competitive for disaccharides. Also, it markedly inhibited ${\alpha}-glucosidases$ of intestine separated from both nondiabetics and diabetics.

석이버섯 용매 추출물의 항산화 및 아질산염 소거작용 (Antioxidative and Nitrite-scavenging Effects of Solvent Extracts from Gyrophora esculenta)

  • This study was conducted to investigate electron donating ability, antioxdative activity and nitrite-scavenging effect of solvent extracts from Gyrophora esculenta. The phenolic compounds of diethylether, butanol, petroleum ether and hexane extracts were 194.20$\mu\textrm{g}$/ml, 98.80$\mu\textrm{g}$/ml, 1.60$\mu\textrm{g}$/ml and 0.20$\mu\textrm{g}$/ml, respectively. Electron donating abilities of diethyleter and butanol extracts from Gyrophora esculaenta were 80.79% and 72.81%, respectively. Absorption of DPPH by diethyleter and butanol extracts sharply increaed at initial stage. Antioxidative activities of fiethylether and butanol extracts by peroxide values are rather high as compared to those of control, but low s compared to those of BHA. Nitrite-scavenging abilities of diethylether, butanol and petroleum ether from Gyrophora esculenta were 96.07%, 77.39% and 15.07%, respectively.

목이 및 석이 메틸 알콜 추출물의 항돌연변이원성 (Antimutagenic Effects of Methyl Alcohol Extracts from Auricularia auricula and Gyrophora esculenta)

  • 목이(Auricularia auricula)와 석이(Gyrophora esculenta)로부터 각각 얻어진 메틸 알콜 추출물에 대하여 E. coli PQ37/pKM101 균주를 이용한 SOS chromotest 방법으로 시료자체의 변이원성 유무 및 4가지 양성변이원 물질에 대한 항돌연변이성을 검토하였다. 시료자체의 돌연변이원성 유무에 있어서는 SOS induction factor (IF) 값의 증감이 없었으므로 돌연변이원성이 없는 것으로 인정되었다. 4가지 양성 돌연변이원 물질에 대한 항돌연변이성 실험결과, 4-nitroquinoline-1-oxide (4NQO)에서는 목이, 석이 각각 52%, 59%의 억제효과를 나타내었고, N-methyl-N‘-nitro-N-nitro-soguanidine (MNNG)에 대해서는 49%, 58%, mitomycin C (MMC)에 대해서는 53%, 64% 그리고 3-amino-1,4-dimethyl-5H-pyrido-(4,3-b) indol (Trp-P-l)에 대해서는 각각 61%, 64%의 억제활성을 나타내었다. 이 결과는 SOS chromotest에서는 양성변이원 4종 모두에 있어서 돌연변이 억제활성이 인정되었고, 특히 석이가 목이보다 대체로 강한 항돌연변이활성을 나타내었다.

목이 및 석이 메탄올 추출물의 유전독성 억제효과 (Antigenotoxic Effects of Methyl Alcohol Extracts from Auricularia mesenterica and Gyrophora esculenta)

  • This study was designed to demonstrate the antigenotoxic potential of methyl alcohol extracts from Auricularia mesenterica and Gyrophora esculenta against the frequency of micronucleate polychromatic erythrocyte(MNPCE) produced by benzo($\alpha$) pyrene in vivo. We used the mouse bone marrow test system to measure the effect of single and multiple treatments of each sample. Genotoxicity of benzo ($\alpha$) pyrene(150mg/kg, i.p.) as positive control was the highest at 36 hours. However, each sample per dose was not genotoxic, showing MNPCE values in the range of the control level. Treatments of methyl alcohol extracts both of Auricularia mesenterica and Gyrophora esculenta showed significant decreased frequencies of NMPCE induced by benzo($\alpha$) pyrene within 12 hours by single treatment(100mg/kg, oral). And also, the MNPCE level produced benzo($\alpha$) pyrene was decreased by the treatment of benzo($\alpha$) pyrene(5 to 200mg/kg, oral) of each sample, but significantly different redults were obtained with 100mg/kg. In the multiple treatment, the highest antigentoxic effects were demonstrated with 20mg/kg in the each sample, a range which induced inhibition indices of 54.2 and 56.3%, respectively.

담자균 추출물의 항균작용 및 항산화작용 (Antibacterial and Antioxidative Activities of Various Extracts from Basidiomycetes)

  • 여러 가지 담자균 자실체의 물과 methanol 추출물을 이용하여 5가지 식중독균에 대해 항균실험을 실시한 결과, 석이버섯, 상황버섯, 영지버섯 추출물에서 항균활성이 있었으며, 석이버섯 methanol 추출물이 가장 높은 항균력을 나타내었다. 높은 항균력이 확인된 석이버섯, 상황버섯의 80% methanol추출물을 diethylether, chloroform, ethyl acetate, n-butanol, 물의 순으로 분획물을 얻어서 항균 및 항산화 작용을 조사하였다. 시료 중 석이버섯의 diethylether, ethyl acetate, n-butanol 분획물에서 높은 항균효과가 있었으며는 것으로 나타났으며, 상황버섯의 경우 ethyl acetate, n-butanol 분획물에서 항균효과를 나타내었다. 400ppm의 석이버섯 분획물의 전자공여 능은 ethyl acetate와 chloroform 분획물에서 70% 이상, butanol과 diethylether 분획물에서 60% 이상의 전자공여 능을 나타내었다. 아질산염 소거능은 석이버섯 ethyl acetate 분획물에서 약 40%의 소거능을 나타내었다.

  • GE974, isolated from Gyrophora esculenta, showed significant inhibitory effect on several ${\alpha}-glucosidases$ in vitro in previous study. In the present study, GE974 showed significant inhibitory effect on blood glucose elevation in mice loaded with maltose, sucrose, starch and lactose. Also, it exhibited similar effect in alloxan and streptozotocin induced diabetic mice, and genetic diabetic mice(db/db mice) loaded with maltose in dose dependent manner. These results suggest that GE974 may possess hypoglycemic effect that inhibit competitively ${\alpha}-glucosidase$ activities.

  • Sprague-Dawley계(系) 숫놈 흰쥐에게 석이버섯 분말을 사료에 첨가 (0.5%, 1.0%, 3.0%, 5.0%, (群)) 하여 6주간 사육한후 혈장 및 간장중에 콜레스테롤의 변화(變化)를 측정(測定)한 결과 다음과 같다. 1) 석이버섯 분말 첨가군에 있어 기본식이군이나 대조군에 비하여 체중증가나 사료섭취량에 특별한 차이나 영향을 나타내지 않았다. 2) 그러나 혈장 및 간장지질량, 혈장 및 간장콜레스테롤치에 있어서는 현저하게 저하했음을 보였다. 3) 이러한 저하(低下)는 첨가량 증가에 따라 저하율이 뚜렸했으나 간지질량 측정에서 3.0% 첨가군이 가장 효과를 나타냈다.

Inhibitory Effects of Crude Drugs on $\alpha$-Glucosidase

  • The inhibitory activity of several crude drugs on $\alpha$-glucosidases, which are the key enzymes for carbohydrate digestion and the prevention of diabetic complications, was investigated. Several crude drugs including Terminaliae Fructus, Mori Cortex Radicis, Caesalpiniae Lignum and Cyrophora esculenta potently inhibited maltase and sucrase isolated from rat intestine, while Arecae Semen and Corni Fructus remarkably inhibited $\alpha$-amylase. Caesalpiniae Lignum and Gyrophora esculenta exhibited significant reductions of blood glucose elevation in mice loaded with maltose and sucrose.

  • The effect of the addition of the white powder separated from Sogi (Gyrophora esculenta) by the extraction methods with ethanol and acetone was checked by the feeding experiment in rats fed with a high cholesterol diet. From this experiment, it was confirmed that this material, which was assumed to be the mixture of gyrophoric acid and the decomposition products, could lower both liver and plasma cholesterol levels.
