Expression Analysis of Anthocyanin Biosynthetic Genes of Tassel and Silks in Gwangpyeongok and Dacheongok (광평옥과 다청옥의 수이삭과 수염에서 안토시아닌 생합성 유전자 발현 분석)

  • Anthocyanins are known to be involved in various functions such as antioxidant and antibacterial activities in plants. Although studies on anthocyanins in corn have been conducted recently, basic research related to anthocyanin biosynthesis is insufficient. In this study, we examined the molecular biological and physicochemical properties related to anthocyanin biosynthesis in the tassel and silks of Gwangpyeongok and Dacheongok cultivars. Anthocyanins were not synthesized in either the tassel or silks in Gwangpyeongok, whereas were synthesized in both in Dacheongok. The total anthocyanin content was approximately 30 times higher in the tassel and silks of Dacheongok than in those of Gwangpyeongok. In addition, C-3-G was measured only in the tassel of Dacheongok, and C-3-G, Pg-3-G, and M-3-G were 45.2 times, 27.3 times, and 37.6 times higher, respectively, in the silks of Dacheongok than of Gwangpyeongok. Expression of F3'H, DFR, and GST genes decreased in the tassel, and that of F3'H and DFR genes decreased in the silks of Gwangpyeongok. It was further confirmed that transcription factor P1 and R1 regulate the expression of anthocyanin biosynthetic genes in the tassel and silks, respectively, in Gwangpyeongok. Linoleic acid (C18:2) decreased by 6.6% and 10.9%, and linolenic acid (C18:3) increased by 8.5% and 8.5%, in the tassel and silks, respectively, of Gwangpyeongok compared to those of Dacheongok. Palmitic acid (C16:0) increased by 4.1% and oleic acid (C18:1) decreased by 2.1% in the silks of Gwangpyeongok compared to that in Dacheongok. In addition, the total fatty acid content in the tassel and silks increased by 10.3% and 30.4%, respectively, in Gwangpyeongok compared to that in Dacheongok. However, no significant results were observed in the analysis of phytosterol components. These results may be utilized as useful resources for the development of functional corn containing a large amount of anthocyanins.

Physiological and Proteome Responses of Korean F1 maize (Zea mays L.) Hybrids to Water-deficit Stress during Tassel Initiation (옥수수 영양생장기 한발 스트레스에 의한 광합성의 생리적 반응 및 프로테옴 변화 분석)

  • Severe droughts in spring have occurred frequently in Korea in recent years, exerting a critical impact on corn yield. Therefore, it is necessary to find physiological and/or molecular indicators of the response to drought stress in maize plants. In this study, we investigated the effects of water-deficit stress on two Korean elite F1 maize hybrids, Ilmichal and Gwangpyeongok, by withholding water for 10 days at tassel initiation. The water deficit drastically reduced the relative leaf water content, leaf number, leaf area, and stem length, leading to dry matter reduction. Moreover, it reduced the SPAD values and stomatal conductance of leaves in drought-stressed plants of both hybrids. Importantly, the number of leaves and SPAD value were non-destructive and easy to investigate in response to water-deficit stress, suggesting that they may be useful indicators for screening drought-tolerant genetic resources. We detected more than 100 spots that were differentially accumulated under drought stress. Of these spots, a total of 21 protein spots (≥1.5-fold) from drought-exposed maize leaves were successfully analyzed by MALDI-TOF-TOF mass spectrometry. Functional annotation using Gene Ontology analysis revealed that most of the identified proteins were involved in carbohydrate metabolism, stress response fatty acid catabolism, photosynthesis, energy metabolism, and transport. The protein expression levels were increased in both Ilmichal and Gwangpyeongok, except for triosephosphate isomerase, fructose-bisphosphate aldolase, and an uncharacterized protein. The lactoylglutathione lyase delta (3,5)-delta (2,4)-dienoyl-CoA isomerase was overexpressed in Gwangpyeongok only. The results obtained from this study suggest that the drought-specific genes may be useful as molecular markers for screening drought-tolerant maize genotypes.

Evaluation of Growth Characteristics and Forage Yield of Domestically Bred Silage Corn Varieties

  • This experiment was conducted to evaluate the growth characteristics and productivity of silage corn varieties developed in Korea. Corn cultivation was carried out using the experimental field in the Pyeongchang campus of Seoul National University (550 m above sea level). There have 10 domestic cultivars (Gwangpyeongok, Dacheongok, Yanganok, Jangdaok, Cheongdaok, Daanok, Sinhwangok, Sinhwangok II, Pyeonggangok, and Hwangdaok) with one imported cultivar (P1543) which tested as a control, and randomized block design with three replications. Among the 100-grains weight of the seeds, Dacheongok was the heaviest, and the germination rate for each variety was 74.6% on average, while that of Daanok and Sinhwangok were over 90%. Sinhwangok was the fastest in tasseling and silking date. The number of days required to be silking date was as slow as 85 days in Dacheongok, Cheongdaok and Pyeonggangok, and as fast as 80 days or less in Sinhwangok, Sinhwangok II and Hwangdaok. The plant height of P1543 was the highest as 344cm, and Hwangdaok and Daanok were short. In terms of the ratio of ears, Daanok had the highest rate of 60.18%, and Jangdaok and Dacheongok had the lowest. There was no significant difference in dry matter content in stover, but P1543 was generally higher in ear and total dry matter content. The dry matter yield was highest in P1543, and the yield of TDN was significantly higher in P1543 and Yanganok. There was a significant difference in the crude protein content of ears and the dry digestibility of stems (p<0.05), while there was no significant difference in the content of each part or element. Combining the above results, Yanganok was the highest in terms of yield, and Dacheongok, Sinhwangok and Pyeonggangok were also recommended for domestically grown corn varieties in the mountainous regions of Gangwon-do.

Selection of Forage Corn Varieties Adapted to High Latitude (The South of Mt. Suyang) (고위도 기후대 재배 적합 국산 사료용 옥수수 품종 선발)

  • Since maize (Zea mays L.) originated in central and south America, it requires warm climate conditions throughout its growing season. Growth halts when night-time temperatures drop below 10℃, and the plant may die if temperature reach -1.7℃. Thus, temperature should be maintained between 10 and 30℃ from seeding to maturity. The germination temperature for maize should be at least 8-11℃, whit an optimal range 32-34℃. Since temperature significantly affects the germination rate and period, it plays a crucial role in maize growth. In this study, we evaluated the quantity and feed value of 11 major varieties to determine those best suited for maize cultivation as feed in higher latitude, specifically in Democratic People's of Republic of Korea, below 38 degrees north. A cultivation test was also conducted in Suwon in Republic of Korea, to assess adaptability in areas south of Mt. Suyang. Among the varieties tested, Shinhwangok2 reached silking the fastest, in 65 days, while Gwangpyeongok took the longest at 75 days. The stem length of all varieties exceeded 230 cm. Gwangpyeongok had the tallest stems, while Daanok and Shinhwangok2ho displayed the highest ear ratios. Dacheongok presented the highest values in both dry matter and TDN quantity, with 31,420 kg/ha and 21,66 kg/ha respectively. Pyeonggangok had the highest crude protein content at 8.0%. TDN (%) ranged from 57-68%, with Hwangdaok reaching up to 68%. Based on these findings, Dacheongok and Pyeonggangok appear to be the most suitable varieties for cultivation in terms of both quantity and feed value.

Seven Days of Consecutive Shade during the Kernel Filling Stages Caused Irreparable Yield Reduction in Corn (Zea mays L.)

  • In monsoon climates, persistent shade is a troublesome weather condition with an impact on the growth and yield of corn (Zea mays L.). We imposed 7, 14, 21, and 28 days of consecutive shade (CS) on Gwangpyeongok and P3394 corn hybrids at the beginning of the kernel filling stages. Shade had little impact on leaf area and dry matter accumulation in the stem and leaves. However, dry matter accumulation in the ear was severely reduced by approximately 28% and 53% after 14 and 28 days of CS, respectively. For the components of grain yield, 7 and 14 days of shade did irreparable damage to the number of filled kernels, the kernel number per ear row, and the percent of filled kernels, but did little damage or reversible damage after removal of the shade to the 100-grain weight and the row number per ear. Shade significantly reduced the relative growth rate (RGR) due to a decrease in the net assimilation rate (NAR). These results suggest that source activity limitation by shade during the kernel filling stages leads to the inhibition of sink activity and size. The yield of biomass, ear, and grain logistically declined as the length of CS increased. Probit analysis revealed that the number of days of CS needed to cause 25% and 50% reductions in grain yield were 3.7 and 23.1, respectively. These results suggest that the plant yield loss induced by shade at the beginning of the kernel filling stages is mainly achieved within the first 7 days of consecutive shade.

Potato-maize double cropping using paddy field in southern plain of Korea

  • In order to reduce rice cultivation area in paddy fields and to increase domestic self-sufficiency of imported upland grain, it is necessary to increase double cropping area of upland crops in paddy field in accordance with decrease of rice consumption in Korea. The double cropping of spring potato-summer grain maize can be combined because of enough growing season in the southern plain of Korea. Spring potato, which is profitable, can be planted in the late February and harvested in the late May as the main crop. Subsequent grain maize can be planted in early June and harvested in November (maturity in the early October). Spring potato (variety Soomi) yielded $2,544kg\;10a^{-1}$ (tuber) when planted in late February, 2016. When maize was planted in June as the second cropping crop, though growth of plant decreased much, grain yield decreased slightly compared to normal planting in April or May. There was enough time to dry maize ear in the field after maturity before harvesting, which saved labor and time for grain drying, since there is no autumn planting at the double cropping of spring potato-summer grain maize. When grain maize (variety Gwangpyeongok) was planted in the early June (June $10^{th}$), average grain yield of above $860kg\;10a^{-1}$ over 2 years of 2015 and 2016 was obtained, and the annual total yield (potato tuber + maize grain) of 3,400 kg $10a^{-1}$ was obtained. The result indicates that the double cropping of spring potato-summer maize using paddy fields in southern plain of Korea, could contribute to the self-sufficiency of upland crops through the maximum production.

An Open Top Chamber for Forage Maize to Study the Effect of Elevated Temperature by Global Warming

  • The increase in temperature due to climate warming is predicted to affect crop yields in the future. Until now, various types of OTC (open top chamber) that simulate the future climate condition have been developed and used to study the effect of temperature increase due to global warming on maize growth. However, in most OTCs, high equipment and maintenance costs were required to artificially increase the temperature. This study was carried to develop a cost-effective and simple OTC suitable for climate warming experiments for forage maize. Three octagonal OTCs with a height of 3.5 m × a diameter of 4.08 m and a partially covered top were constructed. The lower part of OTC covered film was opened at a height of 26 cm (OTC-26), 12 cm (OTC-12) from the ground surface, or not opened (0 cm, OTC-0). Mean air temperatures during the daytime on a sunny day in OTC-0, OTC-12 and OTC-26 increased to 3.23℃, 1.33℃, and 0.89℃, respectively, compared to the ambient control plot. For a pilot test, forage maize, 'Gwangpyeongok' was grown at OTCs and ambient control plots. As a result, in the late maize vegetative growth phase (July 30), the plant height was increased more than 45% higher than the ambient control plot in all OTC plots, and the stem diameter also increased in all OTC plots. These results indicate that it is possible to set the temperature inside the OTC by adjusting the opening height of the lower end of the OTC, and it can be applied to study the response of forage maize to elevated temperature. An OTC, with its advantages of energy free, low maintenance cost, and simple temperature setting, will be helpful in studying maize growth responsiveness to climate warming in the future.

Effects of formic acid and lactic acid bacteria inoculant on main summer crop silages in Korea

  • To improve the fermentation quality of silage and reduce the nutrients loss of raw materials during the ensiling process, silage additives are widely used. The effect of additives on silage is also affected by the species of crop. Therefore, this study was designed to explore the effects of formic acid (FA) and lactic acid bacterial inoculant on the quality of main summer crop silage. The experiment was consisted on split-plot design with three replications. The experiment used the main summer forage crops of proso millet ("Geumsilchal"), silage corn ("Gwangpyeongok"), and a sorghum-sudangrass hybrid ("Turbo-gold"). Treatments included silage with Lactic acid bacterial Inoculant (Lactobacillus plantarum [LP], 1.0 × 106 CFU/g fresh matter), with FA (98%, 5 mL/kg), and a control (C, without additive). All silages were stored for 60 days after preparation. All additives significantly increased the crude protein content and in vitro dry matter digestibility (IVDMD) of the silages and also reduced the content of ammonia nitrogen (NH3-N) and pH. Corn had the highest content of IVDMD, total digestible nutrients and relative feed value among silages. Compared with the control, irrespective of whether FA or LP was added, the water soluble carbohydrate (WSC) of three crops was largely preserved and the WSC content in the proso millet treated with FA was the highest. The treatment of LP significantly increased the lactic acid content of the all silage, while the use of FA significantly increased the content of acetic acid (p < 0.05). The highest count of lactic acid bacteria (LAB) was detected in the LP treatment of corn. In all FA treatment groups, the total microorganism and mold numbers were significantly lower than those of the control and LP groups (p < 0.05). In conclusion, both additives improved the fermentation quality and nutritional composition of the main summer forage crops. The application of FA effectively inhibited the fermentation of the three crops, whereas LAB promoted fermentation. So, both FA and LP can improve the quality of various species of silage.

Application Effects of Bacterial Inoculants Producing Chitinase on Corn Silage

  • This study was aimed to isolate bacterial inoculants producing chitinase and evaluate their application effects on corn silage. Four corn silages were collected from four beef cattle farms to serve as the sources of bacterial inoculants. All isolates were tested against Fusarium graminearum head blight fungus MHGNU F132 to confirm their antifungal effects. The enzyme activities (carboxylesterase and chitinase) were also measured to isolate the bacterial inoculant. Based on the activities of anti-head blight fungus, carboxylesterase, and chitinase, L. buchneri L11-1 and L. paracasei L9-3 were subjected to silage production. Corn forage (cv. Gwangpyeongok) was ensiled into a 10 L mini silo (5 kg) in quadruplication for 90 days. A 2 × 2 factorial design consists of F. graminearum contamination at 1.0104 cfu/g (UCT (no contamination) vs. CT (contamination)) and inoculant application at 2.1 × 105 cfu/g (CON (no inoculant) vs. INO (inoculant)) used in this study. After 90 days of ensiling, the contents of CP, NDF, and ADF increased (p<0.05) by F. graminearum contamination, while IVDMD, acetate, and aerobic stability decreased (p<0.05). Meanwhile, aerobic stability decreased (p<0.05) by inoculant application. There were interaction effects (p<0.05) on IVNDFD, NH3-N, LAB, and yeast, which were highest in UCT-INO, UCT-CON, CT-INO, and CT-CON & INO, respectively. In conclusion, this study found that mold contamination could negatively impact silage quality, but isolated inoculants had limited effects on IVNDFD and yeast.

Comparison of Growth Characteristics and Productivity of Summer Forage Crops in Sihwa Reclaimed Land (시화 간척지에서 하계 사료작물의 초종 및 품종에 따른 생육특성 및 생산성 비교)

  • This study was conducted to investigate the effects of species and varieties of summer forage crops on growth characteristics and productivity in Sihwa reclaimed land. The summer forage crops used in the trial were silage corn, sorghum×sudangrass hybrid(SSH), and proso millet. For each forage species, Gwangpyeongok(GPO), P15453, P1952 and P2088 were used for silage corn, and 877F, Green star, Honey chew, and Turbo gold cultivars were used for SSH. For proso millet, Ibaekchal, Geumsilchal and Manhongchal developed by the National Institute of Crop Science were used. Silage corn and SSH were sown on May 21, 2019 and proso millet on June 4, and harvested on September 2. There was no significant difference in plant and ear height of silage corn among varieties. P1543 was the highest and P2088 was the lowest in yield of silage corn, but there was no significant difference among treatments. Among the SSH, the plant height of 877F was the highest and Turbo gold variety had the smallest (p<0.05). As for the dry matter(DM) yields, 877F had the highest at 3,862 kg/ha and Green star had the lowest at 2,669 kg/ha (p<0.05). The fresh matter yield of proso millet was 15,778 kg/ha, which was higher than that of corn or SSH, The average dry matter yield was 4,780 kg/ha, and Ibaekchal variety had the highest DM yield compared to other varieties (p<0.05). P2088 had the highest TDN content and GPO was the lowest (p<0.05). As for the SSH, the TDN content of Green star and Honey chew varieties was significantly higher, and the RFV value was the lowest in Turbo gold. The average crude protein content of proso millet was 7.03%, and the highest TDN and RFV values were 64.36% and 106 in Geumsilchal. In the experiment of the germination rate of summer forage crops according to salt concentration, silage corn showed a germination rate of 83.1% even at 0.4% salinity. In particular, P2088 and P1921 varieties had more than 80% germination rate even at 0.6% salt concentration. As for the SSH, the germination rate of 877F was 93.3% even at 0.8% salinity, and 88.3% with Honey chew, indicating higher resistance to salt concentration compared to other varieties. Proso millet showed a high germination rate of 84.0 to 88.7% even at a salt concentration of 0.6%. Considering the above results, proso millet was recommended as the most suitable forage crop species in the Sihwa reclaimed land with high salt concentration, and the Ibaekchal variety is recommended as a suitable forage crop due to its high yield.
