Effect of Lycopene on the Insulin-like Growth Factor-I Receptor Signaling Pathway in Human Colon Cancer HT-29 Cells (인간의 대장암 HT-29 세포주에서 라이코펜이 Insulin-like Growth Factor-I Receptor Signaling Pathway에 미치는 영향)

  • Epidemiological data suggest that lycopene has anticancer activities in humans. Insulin-like growth factor-I receptor (IGF-IR) is a transmembrane tyrosine kinase that mediates the biological actions of IGFs and may play an active role in cancer progression. Because our previous in vitro studies have indicated lycopene inhibits HT-29 cell growth, the aim of this study was to determine whether lycopene induces apoptotic cell death and the inhibitory effect of lycopene on HT-29 cell growth is related to changes in IGF-IR levels and the receptor's intracellular signalling pathways. HT-29 cells were incubated for 4 days in serum-free medium in the presence of 0, 25, 50, or 100 $\mu$M lycopene, and the DNA fragmentation assay was performed. Cells treated with lycopene produced a distinct oligonucleosomal ladder with different sizes of DNA fragments, a typical characteristic of cells undergoing apoptosis. HT-29 cells were cultured for 4 days in serum-free medium in the presence of 0~100 $\mu$M lycopene and IGF-I (10nM) was added for 0~60 minutes immediately prior to lysate preparations. Western blot analysis of total lysates revealed that lycopene decreased the levels of IRS-1, Akt, phosphatidylinositol 3-kinase (PI3K), and IGF-IR $\beta$-subunit, and increased the levels of the IGF-IR precursor dose dependently. Lycopene also decreased IGF-I-induced phosphorylation of IGF-IR$\beta$, IRS-1 and Akt, which were, at least in part, due to decreased expression of these proteins. These results suggest that lycopene induces apoptosis of HT-29 cells by inhibiting IGF-IR signaling thereby interfering with an IGF-II-driven autocrine growth loop, which is known to exist in this cell line.

  • An experiment was conducted to assess the effects of dietary 3,5,3'-triiodo-L-thyronine ($T_3$) at 0, 10, 20, 50, 100 ppm on growth and survival rate in juvenile black seabream held at $27.9{\pm}1.1^{\circ}C$ for 50 days. Fish were fed the $T_{3}$ experimental diet for 4() days by hand to satiation in $2{\~}4$ times per day. After 50 days period, food intake rate (${\%}$), feed efficiency (${\%}$), survival rate (${\%}$), growth of weight and length (specific growth rates), and condition factor were measured. Food intake rate was inversely related to the dietary $T_{3}$ level. But feed efficiency was not changed by $T_{3}$ level. $T_{3}$ slightly improved survival rate of larvae. Survival rate of larvae from 100 ppm was significantly higher than that of fish from control. Dietary $T_{3}$ influenced growth in length and weight. Growth of black seabream fed a diet containing 10 ppm of $T_{3}$ was significantly higher than that of fish fed control. However 100 ppm of $T_{3}$ induced the inhibition for length and weight growth. The condition factor was inversely related to the dietary $T_{3}$ content.

Wnt5a attenuates the pathogenic effects of the Wnt/β-catenin pathway in human retinal pigment epithelial cells via down-regulating β-catenin and Snail

  • Activation of the Wnt/β-catenin pathway plays a pathogenic role in age-related macular degeneration (AMD) and is thus a potential target for the development of therapeutics for this disease. Here, we demonstrated that Wnt5a antagonized β-catenin response transcription (CRT) induced with Wnt3a by promoting β-catenin phosphorylation at Ser33/Ser37/Thr41 and its subsequent degradation in human retinal pigment epithelial (RPE) cells. Wnt5a decreased the levels of vascular endothelial growth factor (VEGF), tumor necrosis factor-α(TNF-α), and nuclear factor-κB (NF-κB), which was up-regulated by Wnt3a. Furthermore, Wnt5a increased E-cadherin expression and decreased cell migration by down-regulating Snail expression, thereby abrogating the Wnt3a-induced epithelial-mesenchymal transition (EMT) in human RPE cells. Our findings suggest that Wnt5a suppresses the pathogenic effects of canonical Wnt signaling in human RPE cells by promoting β-catenin phosphorylation and degradation.   • This paper discussed motivation, some theories of motivation and motivation of library personnel. The most important aspect of any organization is its workforce. Motivation arouses, energizes, influences and sustains behaviour and performance of employees. Motivating factors include extrinsic factors which pertain to the conditions under which a job is performed and intrinsic motivators which contribute to job satisfaction and subsequently increase productivity. Two levels of motivation were identified in the library: the need-factor motivation and the profession-related motivation. Need-factor motivation is well-pronounced in the lower cadre of library staff such as library assistants, clerical staff, and porters who set their priority on economic needs. The profession-related motivation occurs among professionally trained librarians whose motivation hinges self-fulfillment derived from professional growth and development. Other motivating factor in the library is job design which includes job rotation, job enlargement, and job enrichment. Job design specifies the contents or methods of any job in such a way that various requirements of a job can be effectively satisfied. Adequate motivation of library employees lies with the managers because they generally control work distribution, freedom and autonomy, work challenge Remove, staff development and training, and advancement opportunities. The managers are urged to create enabling environment to ensure improved commitment, satisfaction, and productivity of employees in the library.

Phosphorylation of Eukaryotic Elongation Factor 2 Can Be Regulated by Phosphoinositide 3-Kinase in the Early Stages of Myoblast Differentiation

  • We have previously reported that phosphorylation of eukaryotic elongation factor 2 (eEF2) is related to the differentiation of chick embryonic muscle cells in culture. In the present study, we found that eEF2 phosphorylation declined shortly after induction of differentiation of L6 myoblasts, when the cells prepare for terminal differentiation by withdrawing from the cell cycle. This decrease in phosphorylation was prevented by inhibitors of phosphoinositide 3-kinase (PI3-kinase) that strongly inhibit myoblast differentiation. We hypothesized that PI3-kinase plays an important role in myoblast differentiation by regulating eEF2 phosphorylation in the early stages of differentiation. To test this hypothesis, myoblasts were synchronized at in $G_2/M$ and cultured in fresh differentiation medium (DM) or growth medium (GM). In DM the released cells accumulated in $G_0$/$G_1$ while in GM they progressed to S phase. In addition, cyclin D1 was more rapidly degraded in DM than in GM, and eEF2 phosphorylation decreased more. Inhibitors of PI3-kinase increased eEF2 phosphorylation, but PI3-kinase became more activated when eEF2 phosphorylation declined. These results suggest that the regulation of L6 myoblast differentiation by PI3-kinase is related to eEF2 phosphorylation.

The Effect of AI Development on the Economic Growth: The Case of South Korea (인공지능산업 발전이 경제성장에 미치는 효과 분석)

  • This study examines the impact of the development of the artificial intelligence (AI) industry on the economic growth of South Korea. The study uses variables such as the revenue and patent applications of AI-related companies, as well as industry-specific total factor productivity and GDP, to estimate the effects. The results suggest that the growth of the AI industry has a positive effect on the economic growth with a lag of about one year. Specifically, the effect of government AI revenue on GDP growth appears to be greater than that of private companies or consumer-focused AI revenue. This indicates that government policies aimed at promoting the diffusion of the AI industry have had significant effects. The study notes that the period covered by the AI industry survey data is relatively short, and there is a lack of detailed data for the manufacturing sector. I suggest that further improvements and accumulation of data could lead to more robust results.

EXPRESSIONS OF VASCULAR METASTASIS RELATED FACTORS IN MURINE ORTHOTOPIC TUMOR MODELS OF SALIVARY GLANDS (마우스에서 확립된 타액선 동위종양에서 혈관성 전이관련 인자의 발현)

  • Background and Purpose: Some subtypes of malignant salivary gland tumors such as adenoid cystic carcinoma (ACC) frequently result in distant metastasis of vascular origin, which are main causes of treatment failure. The reasons for the affinity for vascular metastatic potential are unclear. Therefore, molecular characteristics that influence the dissemination of metastatic tumor cells are important for the design of more effective treatment of salivary ACC. Tumor angiogenesis has been known to be essential for the distant metastasis of malignant cells. So, we determined expressions of vascular metastasis related factors in orthotopic (parotid) murine models of parotid ACC and compared with those in ectopic (subcutis) tumors of athymic mice. Experimental Design: Using specimens from murine parotid (orthotopic, experimental group) and subcutaneous (ectopic, control group) tumors, which have developed via transplantation of tumor cells, originated from human parotid ACC, we performed immunohistochemical assays with anti-vascular endothelial growth factor (VEGF), basic fibroblast growth factor (bFGF, FGF2), matrix metalloproteinase (MMP)-9, and interleukin (IL)-8 antibodies. We also performed immunohistochemical assays with VEGF receptor (VEGFR)-1, VEGFR-2, VEGFR-3, and phosphorylated VEGFR-2. Results: Transplantation of human ACC tumor cell $(5{\times}10^5)$ into the parotid and subcutis successfully resulted in orthotopic (parotid) and ectopic (subcutaneous) tumors in athymic mice. Immunohistochemical staining demonstrated higher expression of major angiogenic factors (VEGF, bFGF, MMP-9) in the orthotopic tumors than in ectopic tumors (P<0.05). But the expression level of angiogenic receptors were same in orthotopic and ectopic tumors of parotid ACC. Conclusion: VEGF, bFGF, and MMP-9 could be a good candidates for antiangiogenic therapy for the contol of vascular metastatic lesions of salivary ACC.

Effects on Inhibition of Angiogenesis in MCF-7 Cells by the Aqueous Root Extract of Ailanthus Altissima (저근백피 추출물이 유선암 세포에서의 신생혈관 미치는 영향)

  • The cellular response to hypoxia is controlled to a large degree by the transcription factor Hypoxia-inducible factor-1(HIF-1). HIF-1 is a transcription factor that is activated by hypoxia and plays a critical role in the development of the cancer phenotype. HIF-1 regulates transcription of a number of genes crucial for tumor survival under hypoxic conditions, including vascular endothelial growth factor(VEGF), erythropoietin(Epo) and several glycolytic enzymes. Tumors in which hypoxia can not induce HIF-1 transcriptional activity remain small and fail to metastasize. In this study, we examined whether aqueous root extract of Ailanthus altissima (REA) downregulate HIF-1, VEGF and p53, and raise the possibility that depletion of these proteins and the anti proliferative activities of REA have any effects on inhibition of angiogenesis in MCF-7 cells. Pharmacologic targeting of specific signal transduction pathways related to oncogenic transformation is a promising approach in cancer treatment. Therefore, REA could be a candidate drug for further clinical development.

The Relationship between Job Satisfaction and Quality of Life in Dental Technicians (치과기공사에 있어서 직무만족도와 삶의 질의 관련성)

  • This study was carried out to measure job satisfaction and quality of life and to clarify the factors in job satisfaction which influence on quality of life of dental technicians, using developed tools for job satisfaction and WHOQOL-BREF for quality of life. The subjects for this study were 238 people who worked the in the Daegu-Kyungbuk area. The results of this study were as follows; the general characteristics that influenced job satisfaction, were especially education, hobbies, salary level, job career and work hours. The factors that influenced the quality of life, were especially Married status, hobbies, job position, and office hours. All subfactors in job satisfaction were related to each other. Growth-development factor was intensely related with recognition factor. Also, all subfactors in quality of life were in positive relation to each other, especially psychological domain was highly related with social domain. Factors of safety, recognition and accomplishment in job satisfaction intensely affected quality of life. Therefore, It is suggested that social recognition for dental technician should be promoted and it might be needed to adjust office hours and support hobby life.

The Relationship between Children's Picky Eating Behavior, Physical Growth and Mother's Child Feeding Practice (까다롭게 먹는 유아의 식습관과 성장발육 및 어머니의 식사지도와의 관련성)

  • The purpose of this study was to determine the correlation between young children's picky eating behavior and mother's attitudes, beliefs and practices regarding the eating habits of children. Using a sample of 304 mothers of four or five year-old children, confirmatory factor analysis was conducted to test a 7 factor model, which included mother's beliefs and practices related to the feeding of their children. Following this, a t-test was conducted to examine the differences between the feeding behaviors of mothers of both of picky and non-oicky eaters, respectively. The results indicated that picky eater's eating behaviors were positively related to the pressures exerted by mothers and negatively related to the mother's concerns about their child's weight and child's BMI. These findings suggest that the pressures mothers exert upon their children to eat, their child's weight and picky eating behaviors are correlated to each other.
