Inhibitory effects of environment-friendly materials and defense response signaling chemicals against anthracnose occurrence in Jujube (Zizyphus jujuba Miller)

  • Anthracnose caused by Colletotrichum gloeosporioides, which is one of the major diseases of red dates, causes severe damages in jujube (Zizyphus jujuba Miller) production in Korea. This study was done to evaluate the inhibition of anthracnose occurrence and pathogen growth by the treatment of environment-friendly materials such as a Bordeaux mixture and loess-sulfur mixture and by defense-response signaling in jujube. The in vitro test of the environment-friendly materials and signaling molecules that were routinely applied did not exhibit any antifungal activities against the pathogen for jujube anthracnose. The Bordeaux mixture and loess-sulfur mixture at a two-fold concentration showed inhibition zones that were 16.0 and 20.3 mm in diameter, respectively. In the pathogen inoculation test with detached jujube tree leaves, while treatment with the environment-friendly materials diluted by half showed no inhibition of lesion development, they did show inhibition of lesion development when they were routinely applied to the leaves. In detached jujube fruits inoculated with the pathogen, better suppressive effects by the treatment of the environment-friendly materials were seen in the fruits at a young stage rather than in the ripening stage. The in vivo test with jujube trees in pots showed that the treatment of salicylic acid (1 mM) resulted in the best suppressive effects against lesion development. The results suggest that it is possible to manage the incidence of anthracnose by the treatment of environment-friendly materials such as the Bordeaux and loess-sulfur mixtures and signaling chemicals such as ethephon, hydrogen peroxide, methyl jasmonate, and salicylic acid in jujube trees and fruits. Consequently, these findings suggest that environment-friendly materials and defense response signaling molecules could be used as suitable candidates for sustainable agrochemicals to manage anthracnose in jujube production.

Increased Preservative and Antimutagenic Activities of Kimchi with Addition of Green Tea Leaves

  • Preservative and antimutagenic effects of green tea leaves added Chinese cabbage kimchi (GK1, GK2, GK3, and GK4 : 1, 2, 3 and 4 of green tea leaves (GTL) in proportion of 100 of salted Chinese cabbage were added to kimchi) were compared to those of the Chinese cabbage kimchi without GTL (control kimchi, CK). Fermentation period of GKs was further delayed than that of CK. The initial pH and acidity between GKs an CK were similar, but the time reach optimally ripened status of kimchi (pH 4.3) was different. CK took 6 days, while GK1, GK2, GK3 and GK4 took 6, 10, 12 and 14 days at 10℃, respectively. The growth of Leuconostoc sp. and Lactobacilus sp. in GKs delayed comparing to those in FCK. Among GKs, as the added amount of green tea leaves increased, the growth of lactic acid bacteria was retarded. The antimutagenic effects of juices from GKs and CK were studied against aflatoxin B₁(AFB₁) in the Ames test on Salmonella typehimurium TA100 and N-methyl-N'-nitro-N-nitrosoguanidine (MNNG) in the SOS chromotest using E. coli PQ37. Juices from optimally ripened GKs (pH 4.3) showed 52∼76% inhibition rates against the indirect mutagen, aflatoxin B₁ induced mutagenicity while 49% inhibition rate by CK in the Ames test. Juices from GKs and CK showed 44∼67% and 36% inhibition rate against direct mutagen, MNNG (70 ng/assay) induce mutagenicity in the SOS chromotest. Thus GKs delayed fermentation period of kimchi and exhibited higher antimutagenic activity than CK.

Studies on the Allelopathy of some Poaceae Plants (수종 화본과식물의 Allelopathy에 관한 연구)

  • This investigation includes the sick soil phenomenon caused by the self-poisoning of Setaria italica, Sorghum nervosum, Zea mays and Miscanthus sinensis among Poaceae. It elucidates whether the poison is directly excreted from the root or the secondary product resulting from the decomposition in the soil; the effect of Miscantus sinensis on the germination and growth of other plants, and the effect of Zea mays grown between furrows to shade Angelica gigas on its growth. The results obtained are as follows; Supplied with the leakage water from the pots, in which the same plants as the test ones were grown, in anticipation of the poison to be directly excreted from their roots, Sataria itlaica and Zea mays exhibited the growth inhibition more than 30%, whereas Sorghum nervosum and Miscanthus sisnensis were not effected in growth at all. When cultivated in the soils mixed with the roots of the some plants as the test ones, in anticipation of the poison to be the secondary product resulting from the decomposition in soil, Setaria italica and Zea mays showd growth inhibition of more than 50%, which is greater than that of the case of the leakage water, and Miscanthus sinensis exhibited no inhibition either, whereas Sorghum nervosum in the 50% plot showed heavy growth inhibition of more than 80% to the case of the leakage water. The common or uncommon plants found easily in the group of Misscanthus scinenis were not affected by the extracts of the steam and leaves of Miscanthus sinensis in germination and growth. Supplied with the leakage water from the pots in which Miscanthus sinensis was grown, among Lespedeza crytobotrya, Oenothera odorata, Raphanus sativus val'. acarlthiformis, Zoysia japonica, Patrinia scabiosaefolia. which are easily found in the group of Miscanthus sinensis, only Patrinia scabiosaefolia was slightly inhibited in growth in the 100% plot, whereas the others did not show any inhibition at all. Mean while, Amaranthus patulus. Solanum nigrum, Capsella bursa-pastoris val'. triangularis, Alopecurus amurensis, Chenopodium album val'. centrorubrum, which could not be found in the group of Miscanthus sinensis, were all distinctly inhibited. In the experiment on the effect of Zea mays on the growth of Angelica gigas, its growth was severely inhibited by one-half to two thirds with the increased concentration in both the cases of growing in the mixture of the soil and the powdered root of Zea mays and being supplied with the leakage water from the pot in which Zea mays was grown.

In Fluence Chemicals From Artemisis argyi on the Growth of Selected Species of Plants and Microorganisms (황해쑥에 함유된 화학물질이 다른 식물과 미생물의 생장에 미치는 영향)

  • To investigate phytotoxic substances in Artemisia argyi, the donor plant, and their biological activities, seed germination and seedling growth of receptor plants such as Arundinella hirta, Echinochloa crus-galli, Rumex crispus and Lactuca sativa were examined at different concentrations of aqueous extracts of the donor plant. Germination of four receptor species was inhibited by the extracts, while seedling growth was decreased to a lesser degree than in the germintion test. Germination, seedling growth and dry weight growth of Achyranthes japonica grown in pot were proportionally inhibited by the extracts. Volatile substances emitted from A, argi plant caused slight inhibition in the germination and seedling growth of the receptor species. Essential oil of the plant extracted by Karlsruker's apparatus inhibited growth of microorganisms and callus growth of Pinellia ternata and Oryza sativa. The GC /MS method was employed for analysis and identification of allelochemicals from A. argyi leaves. Sixty-one chemical substances such as a-pinene, camphene, 1. 8-cineol, etc. were identified from essential oil of A. argyi. The results of this experiment on seed germination, seedling growth, microorganism culture and tissue culture indicated that naturally occurring chemical substances from A. argyi would be responsible for the growth inhibition of plants studied.

  • Sulfonamides, a therapeutically important group of antimicrobial drugs, are widely used to treat and prevent the acute systemic and skin infections in dairy cattle. They also pose an economic hazard through inhibition of growth of dairy starter cultures. This study was carried out to compare four screening methods for detection of sulfamethazine in milk and determine the positive milk sample by HPLC method. Sulfamethazine was used to spike at five levels of sulfamethazine. The Lac-Tek test and CharmII test were also consistent better than TTCII test and Delvo SP test in sulfamethazine detection. Analysis probabilities of obtaining a positive response with TTCII test and Delvo SP test assay at 50 ppb sulfamethazine level in milk samples were only 14%, 42% each. Whereas using the Lac-Tek test and CharmII test would have resulted in 100% identification of the five levels. Determination of sufamethazine using the HPLC method in the spiked milk were 10.64, 19.30, 30.76,38.83 and 50.23 ppb, respectively.

Biosafety of the New Soft Contact Lens Materials in the Fibroblast L-929 Cell Line (흰쥐의 섬유아세포 L-929를 이용한 새로운 Soft Contact Lens 소재의 생물안전성 검증)

  • In this study, we polymerized new materials for soft contact lens using HEMA (2-hydroxyethyl methacrylate) which is the based-monomer of soft contact lens, EGDMA (ethylene glycol dimethacrylate) as cross linkage agent, and the new additives of monoester or di-ester derived from itaconic acid commercially produced by the fermentation of Asp. itaconicus. New polymer materials for soft contact lens were synthesized with the mixture of HEMA and mono- or diester at different ratios and presented to a good water content and oxygen transmissibility (Dk/L) values. In case of polymerization with HEMA and mono-ester (15%), the water content and oxygen transmissibility of contact lens were found to be good values at 57.6% and 28.5 Dk respectively. The mixture of HEMA and mono-ester is more excellent than HEMA/di-ester in the water content and oxygen transmissibility. The toxicity of new contact lens materials were confirmed in the fibroblast L-929 cell line using a agar overlay test and a growth inhibition test with the extract solution of contact lens.

  • For the purpose of antibacterial and photocatalystic deodorization functions, the papermaking inorganic fillers and pigments were surface-modified with Ag nano-colloidal solution and $TiO_2$ by using the hybridization technique. The functional specialty sheets and coating papers were produced with the surface-modified fillers and pigments, and evaluated by halo test and inhibition growth test in their antibacterial and photocatalystic characteristics. For the application of specially produced antibacterial handsheet to the wallpaper usages, the photocatalyst efficiency test of benzene in volatile organic compound dissolution experiment of antibacterial and deodorization function wallpaper showed that the efficiency was 45~50% for 80 min of reaction time and 90% of attained resolution was reached at approximately 30 min of response time.

  • During the screening for inhibitors of melanin biosynthesis from plant extract, Angelica polymorpha MAXIM which showed a high level of inhibition was selected. The inhibiting substances were purified form methanol extract of Angelica polymorpha MAXIM followed by silica gel column chromatography and HPLC. The inhibitors were identified as heraclenin, isosaxalin and heraclenol 3'-Me ether, by spectrescopic methods of ESI-MS, H-NMR, C-NMR, DEPT, HMQC and HMBC. These compounds did not have mushroom tyrosinase inhibitory activity, but showed a highly potent melanin biosynthesis inhibition zone in the plate culture of Streptomyces bikiniensis, a bacterium used as an indicator organism in this work. These compounds did not show any growth inhibition against S. bikiniensis at the same concentration of melanin biosynthesis test.

Antioxidant, Anticancer and Antibacterial Activities of Naesohwangryntang and its Ingredients (내소황련탕(內疎黃連湯) 및 구성약재의 항산화효과 검증과 항암 및 항균효과)

  • Objectives : Antioxidant, Anticancer and Antibacterial Activities of Naesohwangryntang and its composition oriental medicines. Methods : We were experimented anti-oxidation effect and growth inhibition ability on cancer cells and antibacterial activity on various kinds of bacteria of skin. Results : The results were obtained as follows : Electron donating ability(EDA) of water extract Naesohwangryntang and ethanol extract Naesohwangryntang was 60% and 70% at 1000 ppm concentration. In the test of SOD-like activity, ethanol extract showed more activity with 27.4% in 700 ppm, while water extract was low in 19.6%. Clear zones formed by sample against the human skin-resident microflora indicated that anti-microbial activity of ethanol extract Naesohwangryntang was higher than that of water extract Naesohwangryntang. The growth inhibition rates of each sample on lung-cancer(A549), at 1000 ppm cancer cell was over 40%. The growth inhibition rate of the each sample melanoma-cancer(B16F10, G361), at 1000 ppm was over 80%. Conclusions : The results indicated that, ethanol extract which is superior in its anti-oxidation and antibacterial effect is useful to be applied in cosmetic industry.

The Evaluation of Allelopathic Potential Barley and Sorghum Residues on Germination and Early Growth of Some Weeds (보리, 수수의 식물체 추출물이 잡초의 발아와 초기생육에 미치는 영향)

  • This experiment was conducted to test the allelopatic activity on germination and early seedling growth of weed species by barley and sorghum plants residues. The fresh barley extraction inhibited the germination and early seedling growth of weeds, Echinochloa crus-galli, Sataria viridis, and P. oleracea. As the extract concentration increased, the germination and early seedling growth of weeds was significantly inhibited. The water extraction of dried barley and sorghum residues also ehibited the strong inhibition effect on germination and Barley seedling growth of weeds. Digitaria sanguinalis, Siegesbechia pubescens, Sectaria viridis, P. oleracea, E. crus-galli. In the dried barley and sorghum residues mixture into the vermiculite, as the dried residue concentration ncreased, emergence percentage, length of shoot and root of weeds, D. saguinalis, S. viridis, S. pubescens, Ammaranthus lividus, and Solanum nigrum, was significantly inhibited. More than 10% concentration of dired residue caused 80% emergence percentage and growth inhibition. From this study, we conclude that barley and sorghym weeds. These results suggest that barley and sorghum has some possibility to control some weed species like natural herbicide.
