Antitumor and Immunomodulatory Activity of Lycoperdon pedicellatum (긴꼬리말불버섯 (Lycoperdon pedicellatum)의 항암 면역활성)

  • Protein-polysaccharide fractions separated from nine Korean wild mushrooms were subjected to an in vitro screening test for lymphoblastogenic activity. Of these, PPLP, the protein-polysaccharide fraction of Lycoperdon pedicellatum, showed the most potent activity and were further investigated for its antitumor activity. When intraperitoneally injected into ICR mice once daily for six days at a dose of 30 mg/kg, PPLP strongly inhibited the growth of sarcoma 180 tumor cells, showing the inhibition ratio of 97.6%. PPLP also showed in vitro inhibitory activity on sarcoma 180 or leukemia L1210 at the concentration of 500 $\mu\textrm{g}$/$m\ell$ or higher. These results strongly suggest that PPLP might exert its antitumor activity through immunostimulation as well as inhibitory activity on the tumor cells.

Effects of Butachlor on Growth of Four Freshwater Algae (Butachlor의 4종 담수조류(freshwater algae)에 대한 생장영향)

  • Algae are vital in the primary production of the aquatic ecosystem, having been considered as good indicators of the bioactivity of pesticides. Algae have short life cycle, respond quickly to environmental change and their diversity and density can indicate the quality of their habitat. The purpose of the study was to determine the growth inhibition effects of butachlor (Tech. 93.4%) and $K_2Cr_2O_7$ (Tech. 99.5%) in Selenastrum capriconutum, Scenedesmus subspicatus, Chlorella vulgaris and Nitzschia palea during and exposure period of 72 hours. The toxicological responses of S. capriconutum, S. subspicatus, C. vulgaris and N. Palea to butachlor, expressed in individual $ErC_{50}$ values were 0.0022, 0.019, 8.67 and $4.94\;mg\;L^{-1}$, respectively. NOEC values were 0.0008, 0.0016, 5.34 and $2.92\;mg\;L^{-1}$, respectively. S. capriconutum was more sensitive than the other algae species. The toxicological responses of S. capriconutum, S. subspicatus, C. vulgaris and N. palea to $K_2Cr_2O_7$ expressed as $ErC_{50}$ values were 0.91, 0.78, 0.85 and $0.57\;mg\;L^{-1}$, respectively. NOEC values were 0.2, 0.2, 0.2 and $0.18\;mg\;L^{-1}$, respectively. Growth inhibition of S. capriconutum, S. subspicatus, C. vulgaris and N. palea from PEC of butachlor were 100, 75, 0 and 0%, respectively.

Inhibition of in Vitro Growth of Three Soil-borne Turfgrass Diseases by Antagonistic Bacteria from Composted Liquid Manure (가축분뇨액비의 길항미생물에 의한 토양전염성 병원균의 생육억제 효과)

  • This study was conducted to test in vitro the antagonistic effect of composted liquid manure (CLM) against soil-borne turfgrass pathogenic fungi, Rhizoctonia solani AG-2-2 (IIIB) (brown patch), R. solani AG-2-2 (IV) (large patch), and Sclerotinia homoeocarpa (dollar spot) for environmentally friendly turfgrass management. CLMs were collected from 9 livestock excretion treatment facilities around the country including Gunwi (GW), Hapcheon (HC), Hoengseong (HS), Icheon (IC), Iksan (IS), Muan (MA), Nonsan (NS), and Yeoju (YJ). CLMs of IC, GW, and IS showed s ignificant (p < 0.05) mycelium growth inhibition that was 17.8%, 20.4%, and 48.0% against R. solani AG-2-2 (IIIB), R. solani AG-2-2 (IV), and S. homoeocarpa, respectively. A t otal of 110 bacterial isolates were obtained from the CLMs that showed antagonistic effects. Among them, 5, 4, and 10 microbe isolates showed promising antifungal activity against mycelium growth of R. solani AG-2-2 (IIIB), R. solani AG-2-2 (IV), and S. homoeocarpa, respectively. The bacterial isolates ICIIIB60, GWIV70, and ISSH20 effectively inhibited the mycelial growth of three soil-borne turfgrass pathogens. Selected bacterial isolates were identified as Alcaligenes sp., Bacillus licheniformis Ab2, and B. subtilis C7-3 through 16s rDNA gene sequence analysis. Among 5 fungicides, the most compatible fungicide with ICIIIB60, GWIV70, and ISSH20 was tebuconazol, toclofos-methyl and toclofos-methyl, respectively. These findings suggested that CLMs could be effectively used not only as organic liquid fertilizer sources but also as biological control agents for soil-borne turfgrass diseases such as brown patch, large patch, and dollar spot.

Antimicrobial Activity of Pectin hydrolysate and its Preservative Effect (펙틴분해물의 항균특성과 식품보존효과)

  • Pectin hydrolysate prepared from citrus pectin by enzymatic hydrolysis has antimicrobial activity. The antimicrobial activity was increased by its hydrolyzing rate and it was rapidly increased after 70% hydrolysis of the pectin. The antibacterial activity of pectin hydrolysate against Escherichia coli ATCC 11229 was the strongest at pH 4.9~5.5, but it diminished slightly at neutral pH values. The antibacterial activity of pectin hydrolysate was stronger than those of against molds and yeasts. The growth of bacteria submitted to this test except lactics was completely inhibited for 48 hrs at $35^{\circ}C$ by adding 2.0~3.0% pectin hydrolysate. While the growth of Lactococcus lactis ATCC 19435 and Lactobacillus bulgaricus and Penicillium funiculosum ATCC 11797 were reached about 60~70% compared with those of the controls in the same condition. But there was no significant effect on the growth of the yeasts. The antibacterial effect of pectin hydrolysate was significantly stimulated by addition of glycine, ethanol, sodium ascorbate, sodium chloride and sodium acetate. The shelf life of Kimchi containing 1.0% pectin hydrolysate was prolonged above 15 days at $4^{\circ}C$ than that of its control. In case of whitish bean jam viable cell counts were inhibited about 2 log cycles by 10 days at $25^{\circ}C$. According to these results, author can sincerely suggest that pectin hydrolysate will be used as a natural food preservative for inhibition of common bacterial growth without inhibition of lactics and yeasts.

Biological Activities of Seven Melania Snails in Korea (국내산 7종 다슬기 추출물의 생리활성 특성 비교)

  • This study was conducted to compare the biological activities of 7 melania snails from the family Pleuroceridae (Semisulcospira coreana, Koreanomelania nodifila, Semisulcospira forticosta, Koreoleptoxis globus ovalis, Semisulcospira libertina, Semisulcospira tegulata and Semisulcospira gottschei) in Korea. Among the 7 species, S. coreana, Korean. nodifila, S. forticosta and S. gottschei showed over 80% cytotoxicities on three cancer cell lines (SNU-1, A549 and Hep 3B) compared to the non-treatment, whereas S. libertina and S. tegulata showed almost no growth inhibition activities on the same cancer cell lines. In relation to ACE inhibition activity, only S. coreana, Korean. nodifila, and S. forticosta showed over 60% ACE inhibition activities, whereas other melania snails exhibited inhibition activities of lower than 25%. DPPH radical scavenging activities were also determined, and used to categories melania snails into three groups based on Duncan's multiple range test at P<0.05. The amount of TNF-${\alpha}$ produced by in vitro mouse peritoneal macrophage was determined according to bioactivity on L-929 cells. Three melania snails, S. coreana, Korean. nodifila and S. gottschei, exhibited 95.2%, 89.7% and 93.7% cell death(%) on L-929 cells, respectively. Glucose-6-phosphate dehydrogenase inhibitory activity was also obtained in the extract of S. coreana (31.9%) and Korean. nodifila (28.1%), showing that these extracts can be used as supplemental dietary health foods. In conclusion, we believe that the extracts of melania snails should be given due consideration in functional health food development.

Inducing apoptosis by the inhibition of c-myb in oral squamous carcinoma cell line, KB cell

  • Oral squamous cell carcinoma (OSCC) is the most common malignancy and is a major cause of worldwide cancer mortality. The proto-oncogene c-myb plays an important role in regulation of cell growth and differentiation, and it is expressed at high levels in hematopoietic cells and many other types of cancers. However, the function of c-myb is not well known in OSCC. The present study aimed to reveal the function of c-myb and to test the alternation of cell growth and signaling by c-myb in OSCC. In this study, c-myb and dominant-negatibe myb(DNmyb) were expressed in an adenovirus-mediated gene delivery system to KB cells. The over-expressed c-myb brought increased cellular proliferation compared with control cells. However, DN-myb infected KB cells showed significant reduction of cell growth and enhanced induction of apoptosis to activate PARP and caspase 9. c-myb induced increase of IGF-I, -II and IGF-IR expressions while DN-myb down-regulated these expression. Activation of ERK and Akt/PKB pathway was shown only in c-myb transduced cells. These findings suggest that the role of c-myb in cell growth of oral cancer cells is partially mediated through the modulation of IGFs, ERK and Akt/PKB. From this results, DN-myb is strongly recommended as a curable gene for the treatment of c-myb dependent malignancies such as OSCC.

Isolation and Identification of Antagonistic Bacteria for Biological Control of Ginger Rhizome Rot Caused by Pythium zingiberum

  • Sixteen isolates showing relatively strong antagonicity against the ginger rhizome rot pathogen, Pythium zingiberum, were selected among the 155 isolates from ginger rhizome surfaces and rhizospheres of ginger cultivation fields in Wanju, Chonbuk. The isolate, 'HB 26-5'showing the strongest antagonicity was finally selected by testing duration of inhibition effect and pathogenicity to ginger. The isolated antagonistic microorganism, 'HB 26-5' was rod shape, gram positive and formed endospore. The isolate produced acids utilizing glucose, arabinose, xylose and mannitol, and acetoin at VP test, and grew anaerobically. Temperature range for growth was from 10 to 4$0^{\circ}C$ . Reaction to catalase and gelatin, hydrolysis were positive, and casein hydrolysis and indol production were negative. Based on the mycological characters and the fatty acid composition, it was identified as Bacillus polymyxa. The pathogenicity test of isolated Bacillus polymyxa 'HB 26-5'on 22 crop cultivars resulted that only the lettuce was influenced in germination, and the others were not affected.

Detection of tetracyclines in beef and pork (식육중 테트라사이클린계 항생물질 잔류조사)

  • In an effort to Improve the quality of meat and to monitor farms, residual tetracyclines in local beef and pork produced in the province of North Chung-cheong were determined by a EEC 4-plate, Charm II and HPLC, respectively. The results were summarized as follows ; 1. Of the 547 samples, 4 beef and 13 pork samples were judged to be positive by EEC 4 - plate method. 2. Detection rates of tetracyclines by type 1(++--) and type 2(+++ or ++$\pm$-) micro-bial growth Inhibition In EEC 4-plate method were 100% and 71%, respectively. 3. Of 17 positive samples, 6 were positive for tetracyclines, 4 were positive for tetracyclines and sulfonamides, 1 was positive for sulfonamides, and 2 were positive for others by Charm II test. 4. The best eluents were 0.01M methanolic oxalic acid, and the ideal temperature for stable concentration was $40^{\circ}C$ as optimal HPLC analytical conditions for the detection of tetracy-clines. 5. Of the 10 positive samples for tetracyclines by Charm II test, tetracyclines were confirmed in 2 beef and 6 pork samples, using HPLC, at levels ranging from 3.64~4.22 ppm and 0.2~1.20 ppm, respectively.

Suppression of Ripe Rot on 'Zesy002' Kiwifruit with Commercial Agrochemicals

  • Ripe rot caused by Botryosphaeria dothidea is one of the serious diseases of postharvest kiwifruit. In order to control ripe rot on Actinidia chinensis cultivar 'Zesy002', several commercial agrofungicides were selected by an antifungal test on an artificial medium. Furthermore, disease suppression by the selected fungicides was evaluated on the kiwifruit by inoculation with a conidial suspension of B. dothidea. On the artificial media containing boscalid + fludioxonil was shown to be the most effective antifungal activity. However, in the bio-test pyraclostrobin + boscalid and iminoctadinetris were the most effective agrochemicals on the fruit. On the other hand, the infection structures of B. dothidea on kiwifruit treated with pyraclostrobin + boscalid were observed with a fluorescent microscope. Most of the fungal conidia had not germinated on the kiwifruit treated with the agrochemicals whereas on the untreated fruit the fungal conidia had mostly germinated. Electron microscopy of the fine structures showed morphological changes to the conidia and branch of hyphae on the kiwifruit pre-treated with pyraclostrobin + boscalid, indicating its suppression effect on fungal growth. Based on this observation, it is suggested that ripe rot by B. dothidea may be suppressed through the inhibition of conidial germination on the kiwifruit treated with the agrochemicals.

The Antioxidative and Antitumor Activity of Extracts of Agrimonia pilosa (선학초 추출물의 항산화 및 항암활성)

  • This study was carried out to investigate the anti oxidative and antitumor activities of medicinal plants for the purpose of developing a functional food. The methanol extracts of Agrimonia pilosa was fractionated with five solvents and examined anti oxidative activities and enzyme inhibitory activities in addition to growth inhibitory activity of human cancer cell. The contents of total phenol compounds in EtOAc and BuOH fraction were 39.89% and 39.56%, respectively. Strong electron donating abilities(>90%) were shown in these fractions and its abilities were 92.90% ($500\;{\mu}g/ml$), 94.47% ($1000\;{\mu}g/ml$) in EtOAc fraction and 93.77% ($500\;{\mu}g/ml$), 92.90% ($1000\;{\mu}g/ml$) in BuOH fraction, respectively. These fractions exhibited more than 50% nitrite scavenging ability and potent inhibition activities to XOase activity (93.06%, 91.73%) at concentration of $1000\;{\mu}g/ml$. In antitumor activity test, hexane fraction showed the strongest growth inhibition activity against HT-29, SNU-1 and HeLa cells. Inhibition levels were 51.50, 90.09% in HT-29, 88.19, 95.11 % in SNU-1 and 42.66, 96.40% in HeLa at the concentration of 50, $100\;{\mu}g/ml$, respectively.
