Isolation of Growth Inhibition Substance on Food borne Microorganisms from Hypericum ascyron L. and Application to Food Preservation (물레나물(Hypericum ascyron L.)의 식중독 미생물 증식 억제 물질의 분리 및 식품적용)

  • The ethanol extract and n-hexane fraction from Hypericum ascyron L. showed strong growth inhibition at 25 ppm on 5 strains of Listeria monocytogenes for 72 hr at $32^{\circ}C$. The purified substance, H2-5-2 fraction, was isolated by silica gel column and preparative thin layer chromatography from n-hexane fraction of Hypericum ascyron L. The H2-5-2 fraction showed a strong bacteriostatic activity on 5 strains of L. monocytogenes at 10 ppm in tryptic soy broth, and the viable cell was reduced 1 log cycle compared to initial cell number. The n-hexane fraction of Hypericum ascyron L. showed strong growth inhibition at 25 ppm on Bacillus cereus and Staphylococcus aureus, and at 50 ppm on Vibrio parahaemolyticus for 72 hr. The purified antimicrobial substance, the H2-5-2 fraction, was assumed as high unsaturated sterol by $^1H-NMR$ and $^{13}C-NMR$. On application test using minced Alaska pollack and ground beef, the n-hexane fraction of Hypericum ascyron L. at the level of 250 ppm was applied at $32^{\circ}C$ and $5^{\circ}C$. At $32^{\circ}C$ storage condition, the antimicrobial substances did not reduced L. monocytogenes ATCC 19113, meanwhile at $5^{\circ}C$ storage condition, L. monocytogenes ATCC 19113 was reduced in viable number.

Antioxidative and Antitumor Activity of Extracts from Saussurea lappa (목향 추출물의 항산화 및 항암활성)

  • This study was carried out to investigate the antioxidative and antitumor activities of Saussurea lappa for the purpose of developing a functional food. The methanol extract of Saussurea lappa was fractionated with five solvents (hexane, chloroform, EtOAc, BuOH, water) and examined for antioxidative activities and xanthine oxidase inhibitory activity in addition to growth inhibitory activity of human cancer cell (HT-29, SNU-1, HeLa). Total phenol compound contents were higher in EtOAC fraction than other fractions. Also, electron donating ability was over 90% at $500{\mu}g/ml$ (93.1 %) and $1000{\mu}g/ml$ (94.3%). The hexane fraction revealed stronger nitrite scavenging ability than the positive control (ascorbic acid) and its abilities were 22.4% and 42.8% at $500{\mu}g/ml$ and $1000{\mu}g/ml$, respectively. Xanthine oxidase inhibitory activity had similar tendency to electron donating ability. The EtOAc fraction showed high inhibition to xanthien oxidase activities at $500{\mu}g/ml$(81.9%) and $1000{\mu}g/ml$(90.4%). In the antitumor activity test, the hexane fraction exhibited potent growth inhibition activity against HT-29, SNU-1 and HeLa cells. Therefore, we believe that Saussurea lappa can be developed into a functional food with antioxidant activity. Additional studies are required for the hexane and chloroform fractions of Saussurea lappa to develop them into therapeutic supplements for stomach cancer, colon cancer, and cervical cancer.

Screening of Natural Antimicrobial Edible Plant Extract for Dooboo, Fish paste, Makkoli Spoilage Microorganism (식용 식물로부터 얻은 추출물의 두부, 어묵, 막걸리 변질균에 대한 항균성 검색)

  • To extend the self-life of Dooboo (tofu), fish paste and Makkoli (Korean traditional rice wine) for which severe heat treatment are not expectable, main putrefactive microorganisms were isolated from each product and tested their growth inhibition by ethanol and water extracts of several edible plants. The ethanol extract of Phellodandron amurense Ruprs had the strong effect of growth inhibiting to all three isolates from Dooboo and Makkoli, and Eugenia caryophyllus, Pinus rigia Mill, Bletilla striata (Thunb) Reichb. Fill and Paeonia albiflora Pall were also same effect to isolates from fish paste. The ethanol extract was more effective inhibiting than water extract in all test microorganisms. The evident inhibition level of each extract was 2000 ppm of Pa ethanol for fish paste and 1000 ppm for Makkoli.

Trichostatin A-induced Apoptosis is Mediated by Krüppel-like Factor 4 in Ovarian and Lung Cancer

  • Background: The istone deacetylase (HDAC) inhibitor trichostatin A (TSA) is known to mediate the regulation of gene expression and antiproliferation activity in cancer cells. Kr$\ddot{u}$ppel-like factor 4 (klf4) is a zinc finger-containing transcription factor of the SP/KLF family, that is expressed in a variety of tissues and regulates cell proliferation, differentiation, tumorigenesis, and apoptosis. It may either either function as a tumor suppressor or an oncogene depending on genetic context of tumors. Aims: In this study, we tested the possibility that TSA may increase klf4 expression and cancer cell growth inhibition and apoptosis in SKOV-3 and A549 cells. Materials and Methods: The cytotoxicity of TSA was determined using the MTT assay test, while klf4 gene expression was assessed by real time PCR andto ability of TSA to induce apoptosis using a Vybrant Apoptosis Assay kit. Results: Our results showed that TSA exerted dose and time dependent cytotoxicity effect on SKOV-3 and A549 cells. Moreover TSA up-regulated klf4 expression. Flow cytometric analysis demonstrated that apoptosis was increased after TSA treatment. Conclusions: Taken together, this study showed that TSA increased klf4 expression in SKOV3 and A549 cell lines, consequently, klf4 may played a tumor-suppressor role by increasing both cell growth inhibition and apoptosis. This study sheds light on the details of molecular mechanisms of HDACI-induced cell cycle arrest and apoptosis.

Inhibition of Growth and Microcystin Toxicity, and Characterization of Algicidal Substances from Lactobacillus graminis against Microcystis aeruginosa (Microcystis aeruginosa에 대한 Lactobacillus graminis의 성장 억제능, microcystin 분해 및 살조 물질의 특성)

  • For several decades, lactic acid bacterium (Lactobacillus graminis: LAB) has been generally recognized as safe. To develop the pan-environmental bio-control agent, algicidal activity of the live LAB cell and its culture filtrate (CF) was examined against Microcystis aeruginosa. LAB cells perfectly lysed M. aeruginosa within 3 days, while the CF had a less effect than the live cells, approximately 78% inhibition of algal growth during a same culture period. The concentration of microcystin in alone culture of M. aeruginosa was $7.1{\mu}gL^{-1}$, but gradually increased and leach $158.5{\mu}gL^{-1}$ on 10 days. However, LAB cells clearly decreased the microcystin by $10.3{\mu}gL^{-1}$ in the same period, approximately 93.5%. CF of LAB showed a strong algicidal activity over 75% between pH 2-7, 91.3% by the treatment of proteinase K, 87.8% by below 3 kDa in particle size, and 75.3% by heat treatment, respectively. Of five solvents, fractions of CF passed through solvents diethyl ether and ethyl acetate showed an obvious algicidal activity in the algal-lawn test. Among 5 fractions purified by silica-gel TLC plate, two spots showed a most strong removal activity on M. aeruginosa. Another analysis of GC indicate that CF contained six representative fatty acids. Even though most of these substance have been known as an anti-algal substance against M. aeruginosa, oleic acid is the most effective. These results suggested that the culture filtrate or specific substances, like a fatty acids, in comparison with live L. graminis can be a successful and eco-friendly agent to control Microcystis bloom.

Variation in seedling growth inhibition due to Maleic Hydrazide treatment of rice(Oryza sativa) and ragi(Eleusine coracana) genotypes and its relationship with yield and adaptability

  • Multilocation trials on 36 rice(Oryza sativa) genotypes of 3 different maturity groups were conducted at four different locations of Orissa for 3 years and 30 ragi(Eleusine coracana) genotypes of 2 different maturity groups were evaluated in three environmental conditions for 3 years. Grain yield data were subjected to stability analysis following linear regression model to estimate adaptability and stability parameters, i.e. b, and $S^2d$ Stability of performance of genotypes was also estimated by two other stability parameters viz., ecovalence W and AMMI stability value ASV. The rice and ragi genotypes of different duration groups showed wide variation in their mean yield, b, $S^2d$, W and ASV parameters. Seeds of the 36 rice and 30 ragi genotypes were treated with 500 and 100 ppm aqueous solution of maleic hydrazide(MH) for 24 hours, respectively to study MH-sensitivity. Sensitivity of genotypes to MH treatment was estimated in terms of seedling growth inhibition index(SGI). The rice and ragi genotypes showed wide differences in their MH-sensitivity in terms of SGI. Relationship of MH-sensitivity of genotypes with their yielding ability, adaptability and stability of performance was tested by contingency $x^2$ test. Low sensitivity of rice and ragi genotypes to MH in terms of SGI appeared to be good indicators of high yielding ability of genotypes. Also, low and high MH-sensitivity of genotypes would be a good indicator of better adaptability to rich and poor environments, respectively, in ragi but not in rice. Low MH-sensitivity of genotypes could be the good indicator of stability of yield performance in rice but not in ragi.

Studies on the Anti-bacterial, Anti-inflammatory and Anti-oxidant Effect of BPH (비피(鼻皮) 증류액의 항균, 소염, 항산화 작용에 대한 연구)

  • Objectives : The purpose of the study is to test the antibacterial, anti-inflammatory and antioxidant effects of BPH, which is composed of Pini Densiflorae Nodi Lignum and Querci Acutissimae Fructus, Angelicae Gigantis Radix, Cnidii Rhizoma, Angelicae Dahuricae Radix, Angelicae Tenuissimae Radix. Method : Antibacterial and anti-inflammatory effects of BPH on Propionibacterium acnes, one of anaerobic bacteria species were evaluated by measuring the levels of 2,2-diphenyl-1-picrylhydrazyl (DPPH) elimination and lipid peroxidation. Result : When BPH was applied to CCD-986sk (Human normal fibroblast) to confirm the level of cytokine(tumor necrosis factor-alpha, interleukin-8), its level increased in proportion to that of BPH's concentration, which indicated dose-dependent relationship. Using the Disk diffusion to measure the bacterial growth inhibition zone varying BPH concentration, it was found that the antibacterial effect of BPH was less than that of erythromycin, the control group, but was higher than that of saline, and it increased with higher concentrations. In a liquid culture medium containing BPH, the growth rate of Propionibacterium acnes was decreased by more than 10% at 25% BPH. After adding P. acnes to THP-1 monocyte, and treated it with BPH, and measuring the concentration of TNF-a and IL-8, it was observed that the amount of TNF-alpha and IL-8 significantly decreased depending on the level of BPH concentration. The ability to eliminate DPPH increased with higher BPH concentration. The inhibition of lipid peroxidation was increased by BHT treatment in a dose-dependent manner. Conclusion : Using Propionibacterium acnes, an anaerobic bacteria, we confirmed that BPH has antibacterial, anti-inflammatory and antioxidant effects.

Effect of Water Treatment Sludge Application on the Growth of Korean Local Corn(Zeas Mays L.) (옥수수에 대한 정수 슬러지(Alum sludge)의 시용효과와 Aluminum이 옥수수의 유식물 생장에 미치는 영향)

  • Effect of water treatment sludge(alum sludge) application on the growth of Korea local corn(Zeas Mays L.) was evaluated and phytotoxicity of alum sludge on the growth of corn roots and plant height was tested. Alum sludge itself and composted alum sludge compost were applicated by 1500kg/ha in the plot respectively. For the fertility test, yield, chlorophyll content, plant height of corn treated by alum sludge and composted alum sludge were better than that of control. No clear toxixity of alum sludge was found. At the food chemical analyses of seed corn, no obvious component variation were evaluated either. Test of pH effect on phytotoxicity of alum sludge on corns showed that plant height and root growth of corns were inhibited at low pH 4 solution. Phytotoxicity test of alum sludge extract which was extracted at various pH solutions, showed that growth and cations uptake inhibition were found the more serious the lower the pH. Plant height and root growth injury were serious at higher than 100ppm Al content solution.

A comparison of bioresorbable membranes alone or in combination with platelet-derived growth factors and insulin-like growth factors on the periodontal healing of the dehiscence defects in dogs. (성견의 열개형 골 결손부에서 흡수성 차단막과 PDGF-BB 및 IGF-I의 혼합 사용시 치주조직의 치유에 미치는 영향)

  • The purpose of present study is to compare the effect of treatment using $Guidor^{(R)}$ as a barrier membrane in conjunction with platelet-derived growth factor and insulin like growth factors on experimental dehiscence defects. Following the resection of premolar crowns, roots were submerged. After 12 weeks of healing period, experimental dehiscence defects of 4mm in height and 4mm in width were surgically created on the mid-facial aspect of the lower premolar roots in each of 4 adult dogs. After root planning and demineralization of the root surface with citric acid, the control groups received 4% methylcellulose gel only, the test group I received 4% methylcellulose gel and were covered by $Guidor^{(R)}$ and the test group II were treated with PDGF and IGF and 4% methylcellulose gel with $Guidor^{(R)}$ coverage. Histological and histomorphometric analysis following 8 weeks of healing revealed the following results. 1. The new bone formation showed no statistically significant difference in all groups with $0.59{\pm}0.82mm$($14.03{\pm}19.60%$) for control, $0.70{\pm}0.39mm$($16.30{\pm}9.01%$) for group I, $0.87{\pm}0.76mm$($18.74{\pm}16.03%$) for group II. 2. The new cementum formation showed no statistically significant difference in all groups with $0.54{\pm}0.48mm$($l6.38{\pm}14.57%$) for control, $0.95{\pm}0.38mm$($23.43{\pm}9.30%$) for group I, $1.01{\pm}0.75mm$($22.10{\pm}16.ll%$) for gorup II. 3. The root resorption showed statistically significant differences betweenthe control group and all test groups(p<0.05) with $2.11{\pm}0.53mm$($52.93{\pm}12.32%$) for control, $0.63{\pm}0.27mm$($15.32{\pm}7.05%$) for group I, $0.89{\pm}0.33mm$ ($19.26{\pm}7.11%$) for group II. On the bases of these results, there were no statistically difference between treatment using resorbable membrane and resorbable membrane in conjunction with PDGF and IGF in the dehiscence defects, where it was difficult to maintain space. The use of membrane seemed to be more effective in the inhibition of root resorption.

Bacterial Reverse Mutation Test of CP pharmacopunture (세균을 이용한 CP 약침의 복귀돌연변이 시험)

  • Objectives: This study aimed to evaluate the toxicity of CP pharmacopunture using bacterial reverse mutation test. Methods: To determine the mutagenic potential of CP pharmacopunture, histidine requiring Salmonella typhimurium (TA98, TA100, TA1535, and TA1537) and tryptophan requiring Escherichia coli (WP2uvrA, pKM101) strains were used. The negative (normal saline solution) and positive (Sodium azide, 2-Nitrofluorene, 2-Aminoanthracene, 9-Aminoacridine, and 4-Nitroquinoline N-oxide) control groups were used. To determine the dose levels of the main study, a dose range-finding study was conducted. Results: As a results of the dose range-finding study, the growth inhibition by CP pharmacopunture was not evident at any dose levels in the absence and presence of metabolic activation. As a results of the main study, the mean number of revertant colonies was less than twice when compared to the negative control values at all dose levels of the CP pharmacopuncture in the presence and absence of metabolic activation, showing no dose-related increase. In the positive control group, the number of revertant colonies was markedly increased by more than twice when compared to the negative control group. Conclusion: According to the results of this study, CP pharmacopunture did not show any signs of mutagenic potential.
