This study aims to investigate how ovulation induction of sevenband grouper, Epinephelus septemfasciatus varies when treated with different hormonal substances such as ovaprim, pimozide, LHRHa, and HCG. As for LHRHa, we injected it in different concentrations: $50{\mu}g/kg$, $100{\mu}g/kg$, $150{\mu}g/kg$, and $200{\mu}g/kg$. All hormonal substances including LHRHa were injected into back muscles. As a result, ovulation occurred in all sample groups. The sample group injected with HCG released the largest amount of eggs, and the dimeter of eggs and oil globules were also significantly bigger than those of others. Fertilization rate, embryonic survival rate, and hatching rate were the highest in a LHRHa ($100{\mu}g/kg$) +pimozide ($1,000{\mu}g/kg$) group and buoyant rate in a LHRHa group. Ovulation rate was 100% when LHRHa was injected in different concentrations. The largest amount of eggs was released in $200{\mu}g/kg$. The diameter of eggs and oil globules were the biggest in $200{\mu}g/kg$ and the smallest in $50{\mu}g/kg$. Fertilization rate showed no significant difference through all different levels of concentration: 94.6-95.3%. Buoyant rate was the highest in $100{\mu}g/kg$ and embryonic survival rate and hatching rate in $150{\mu}g/kg$. Based on these experimental results, we concluded that LHRHa in $100{\mu}g/kg$ is the most efficient for the ovulation induction of sevenband grouper.

This study aims to investigate how ovulation induction of red spotted grouper, Epinephelus akaara varies when treated with different hormonal substances such as ovaprim, pimozide, LHRHa, and HCG. As for LHRHa, we injected it at four different concentrations: 50 ㎍kg-1, 100 ㎍kg-1, 150 ㎍kg-1, and 200 ㎍kg-1. All hormonal substances including LHRHa were injected into the back muscles. Buoyant, embryonic survival, fertilization, and hatching rate were the highest in LHRHa and LHRHa + pimozide group. When LHRHa was injected at different concentrations, buoyancy and fertilization rate were the highest in 100 ㎍kg-1 group. Embryonic survival and hatching rate were the highest in 150 ㎍kg-1. Based on these experimental results, we concluded that LHRHa at the concentration of 100 ㎍kg-1 is the most efficient for the ovulation induction in this fish species.

The effects of dietary mannan oligosaccharides, Lactobacillus plantarum, Bacillus subtilis, and Bacillus licheniformis supplementation on hybrid grouper Epinephelus akaara ♀×Epinephelus lanceolatus ♂ were evaluated. The fish were fed a basal diet and five other diets consisting of 0.6% mannan oligosaccharides, L. plantarum, B. subtilis, and B. licheniformis and mixture of each 0.15% prebiotic and all the probiotics (designated as MOS, LP, BS, BL, and SYN) for 56 days. Growth performance and feed utilization showed no significant differences among all experimental groups. Lipid level of whole-body was significantly high in MOS and BL groups. Plasma aspartate aminotransferase was significantly low in BL and SYN groups. Nitro-blue tetrazolium, lysozyme and anti-protease, and glutathione peroxidase in BS, SYN, and all probiotic groups, respectively, were significantly high. Intestinal Vibrio bacteria was significantly low in all probiotic and SYN groups. Gene expression of interleukin-1β and interleukin-10 in SYN group; transforming growth factor β2 in MOS and BS groups, toll-like receptor 2-2 in BS and BL groups; and C-type lectin in MOS, LP and SYN groups were significantly upregulated. Our findings indicate that mannan oligosaccharides, L. plantarum, B. subtilis, and B. licheniformis could improve innate immunity, antioxidant capacity, anti-inflammation, and intestinal microbiota of hybrid grouper.

The Serranidae is high-quality fish species with good meat quality and is traded at high price, and is attracting attention in South Korea as a cultured species that creates high added value. However, the high-density fish farming for mass production increases the risk of mass mortality due to infectious diseases, leading to enormous economic losses. Therefore, in order to safely prevent and protect farmed fish from serious infectious diseases, it is necessary to conduct disease monitoring on a regular basis. In this study, Hyporthodus septemfasciatus, Epinephelus moara, and the hybrid longtooth grouper (E. moara ♀×E. lanceolatus ♂) were collected once a month from fish farm of Garorim and Aquabiotech Co., Ltd for a total of six months, from July to December 2023. We investigated infections of five species of bacterial diseases, including Flavobacterium columnare, six species of viral diseases, including LCDV (lymphocystis disease virus), and parasitic pathogens in grouper farms. As the result, Vibrio vulnificus and V. harveyi were detected in H. septemfasciatus in August, in the case of viral diseases, NNV was detected in H. septemfasciatus from July to August using RT-PCR or PCR. Finally, In the case of parasitic diseases, Tricodina sp. was detected in E. moara and the hybrid longtooth grouper from August to December.

The physiological response of the longtooth grouper, Epinephelus bruneus to low water temperature (LWT) stress while on three different concentrations of dietary vitamin E (1 mg/diet g, 5 mg/diet g, and 10 mg /diet g) were investigated. Plasma cortisol, glucose, and aspartate aminotransferase (AST) levels were higher in the control and sham control compared with the vitamin E supplemented groups. The most addition with vitamin E in the diet (10 mg/diet g) showed the lowest levels of cortisol, glucose, and AST. However, plasma alanine aminotransferase (ALT) showed no significant differences across all experimental conditions (P>0.05). Also the more disposing vitamin E into feeding the better resistance against stress on the LWT was shown through cortisol, glucose, ALT, and AST.
To investigate the properties of mucosubstances of the epidermis in various teleostean species, conventional histochemical stainings were used on the skin in five species of order Perciformes, i. e., yellowtail, Seriota quinqueradiat, striped beakperch, Oplegnathus fasciatus, brown spotted grouper, Epinephelus chlorostigma, sea chub, Ditrema temmincki and multicolorfin rainbowfish, Halichoeres poecilopterus. The following methods were used: periodic acid Schiff (PAS), alcian blue (AB) pH at 2.5, AB pH at 1.0, AB pH at 2.5-PAS, AB pH at 1.0-PAS, aldehyde fuchsin (AF) pH at 1.7-AB pH at 2.5 and high iron diamine (HID)-AB pH at 2.5. The epidermis of all five species consisted of three layers: superficial, middle, and basal layer. The superficial layer was comprised of rather flattened cells. In particular, the outermost layer of striped beakperch and middle layer of sea chub consisted of mucus-secreting cells. Mucous cells, the unicellular glands, were found in epidermis but varied in number in different body regions and species. Although there was a slight difference in the amount in various species and body regions, the secretory contents of the mucous cells in the five teleostean species contained acidic mucopolysaccharides. In yellowtail, striped beakperch, and multicolorfin rainbowfish, the property of mucosubstances was identified as sialomucin, while it was sulphomucin in brown spotted grouper and sea chub.
Isolation and cloning of seven-band grouper (Epinephelus septemfasciatus) growth hormone cDNA from pituitary gland revealed an open reading frame of 612 bp coding for a pre-growth hormone of 204 amino acids with a 17 amino acid putative signal peptide. Deduced amino acid sequence showed that there was one possible N-glycosylation site at $Asn^{l84}$ and four cysteine residues $(Cys^{52},\;Cys^{160},\;Cys^{177},\;Cys^{185})$ on t e same positions as in some other species where they were involved in the stabilization of the tertiary structure. The seven-band grouper growth hormone (sbgGH) presented a $99.5\%$ amino acid sequence identity with the growth hormone of Epinephelus coioides and contained the conserved hormone domain region. Comparison of growth hormone sequences from evolutionarily diverse species revealed 25 amino acid residues conserved in jawless fishes to modern mammals. It also revealed an evolutionary trend to retain the same polypeptide sequence even in the distantly related animals while allowing alterations to occur in polypeptides of the closely related species. In order to create a recombinant system to produce high levels of the growth hormone, it was expressed in Escherichia coli (BL21) cells. The gel analysis revealed theoretically expected molecular weights for both mature and pre-sbgGHs.

Red spotted grouper, Epinephelus akaara, is a popular aquaculture species and a protogynous hermaphrodite. Induction of artificial sex change at the time of primary sex differentiation is of interest but has not been successful due to the lack of necessary basic information. To find out the potential neuroendocrine influence on the primary sex differentiation, the expression of key genes in the brain was investigated during the formation of ovarian cavity. Expression of cyp19a1b, esr1, gnrhr1, fsh, lh and cga in the brain was positively associated with the formation of ovarian cavity, showing gradual increase as the formation proceeds. However, the expression of gnrh1 was suppressed during the early part of the ovarian cavity formation, signifying potential hypothalamic influence on the primary sex differentiation in this species.
Higher thermal exposure can influence the blood cell morphology and count. Hence, based on the previous results (Rahman et al., 2019), the present study evaluated the morphometric indices of erythrocytes and their nucleus under different water temperatures (25℃, 28℃, 31℃, and 34℃) to investigate their use as an indicator of thermal stress in red spotted grouper, Epinephelus akaara. 180 healthy specimens of E. akaara were exposed to four temperature conditions (25℃ as control, 28℃, 31℃, and 34℃) for 42 days, following 2 weeks of acclimation at 25℃. Erythrocyte major axis (EL), erythrocyte minor axis (EW), nucleus major axis (NL), and nucleus minor axis (NW) were examined from the blood smears on each sampling day (i.e., 2, 7, and 42 days of thermal exposure). EL and NL were significantly decreased, whereas EW and NW were increased at higher water temperature (31℃ and 34℃). The major-minor axis proportions of erythrocytes and their nucleus (EL/EW; NL/NW) were decreased with increasing water temperature (31℃ and 34℃). The strong relationships were observed among the morphometric indices of erythrocytes and their nucleus, especially in EL vs. NL and EW vs. NW. This study reveals that elevated water temperature (31℃ and 34℃) can influence the major and minor axis morphometry of erythrocytes and their nucleus in red spotted grouper. These indices may be used as stress indicators to monitor the health status of E. akaara and probably for other fish species.
We studied oocyte steroidogenesis in blacktip grouper Epinephelus fasciatus ovarian follicles during vitellogenesis. Vitellogenic oocytes with average diameters of 0.45, 0.48 and 0.50 mm were incubated in vitro in the presence of $[^3H]17{\alpha}$-hydroxyprogesterone as a precursor. The steroid metabolites were analyzed using thin layer chromatography (TLC), high performance liquid chromatography (HPLC), and gas chromatography-mass spectrometry (GC/MS). The major metabolites in the vitellogenic oocytes were androstenedione ($A_4$), testosterone (T), estradiol-$17{\beta}$ ($E_2$), and estrone ($E_1$). The metabolites of androgen ($A_4$ and T) were higher in the 0.50-mm oocytes than in the 0.45- and 0.48-mm oocytes, while the estrogen metabolites (E2 and E1) were lower in the 0.50-mm oocytes. These results suggest that 0.50-mm oocytes are fully vitellogenic following initiation of the maturation process.
