Affinity chromatography and capillary electrophoresis for analysis of the yeast ribosomal proteins

  • We present a top down separation platform for yeast ribosomal proteins using affinity chromatography and capillary electrophoresis which is designed to allow deposition of proteins onto a substrate. FLAG tagged ribosomes were affinity purified, and rRNA acid precipitation was performed on the ribosomes followed by capillary electrophoresis to separate the ribosomal proteins. Over 26 peaks were detected with excellent reproducibility (<0.5% RSD migration time). This is the first reported separation of eukaryotic ribosomal proteins using capillary electrophoresis. The two stages in this workflow, affinity chromatography and capillary electrophoresis, share the advantages that they are fast, flexible and have small sample requirements in comparison to more commonly used techniques. This method is a remarkably quick route from cell to separation that has the potential to be coupled to high throughput readout platforms for studies of the ribosomal proteome.

Gas Permeation Properties of Hydroxyl-Group Containing Polyimide Membranes

  • A series of hydroxyl-group containing polyimides (HPIs) were prepared in order to investigate the structure-gas permeation property relationship. Each polymer membrane had structural characteristics that varied according to the dianhydride monomers. The imidization processes were monitored using spectroscopic and thermog-ravimetric analyses. The single gas permeability of He, $H_2$, $CO_2$, $O_2$, $N_2$ and $CH_4$ were measured and compared in order to determine the effect of the polymer structure and functional -OH groups on the gas transport properties. Surprisingly, the ideal selectivity of $CO_2/CH_4$ and $H_2/CH_4$ increased with increasing level of -OH incorporation, which affected the diffusion of $H_2$ or the solubility of $CO_2$ in HPIs. For $H_2/CH_4$ separation, the difference in the diffusion coefficients of $H_2$ and $CH_4$ was the main factor for improving the performance without showing any changes in the solubility coefficients. However, the solubility coefficient of $CO_2$ in the HPIs increased at least four fold compared with the conventional polyimide membranes depending on the polymer structures. Based on these results, the polymer membranes modified with -OH groups in the polymer backbone showed favorable gas permeation and separation performance.

  • PURPOSE. The aims of this study were to suggest a method of fabrication of the record base using a light-polymerized resin by applying the two-phase fabrication method for the improvement of the fit of the record base and to compare the degree of fit according to the separation site. MATERIALS AND METHODS. In the edentulous cast of maxilla, four test groups were considered. In the first, second, third, and fourth test groups (n = 12 in each group) the separation was done at 0, 5, 10, and 15 mm, respectively below the alveolar crest along the palatal plane. For the control group, the record base was made without separating the two sections. The light-body silicone material was injected into the fitting surface of the record base. It was then placed onto the cast and finger pressure was applied to stabilize it in a seated position followed by immediate placement onto the universal test device. Finally, the mass of the impression material was measured after it was removed. ANOVA was performed using the SAS program. For the post-hoc test, the Wilcoxon Rank-Sum test and the Tukey-Kramer HSD test were performed ($\alpha$ = 0.05). RESULTS. The control group and Group 3, 4 showed significant differences. The Group 3 and 4 showed significantly smaller inside gaps than the control group which was not made with the two-phase fabrication method. CONCLUSION. The two-stage polymerized technique can improve the fit of the denture base particularly when the separation was made at 10 to 15 mm from the alveolar crest.

Extraction & Separation Behavior of Light Rare Earth Elements from the Mixed Solutions by Cyanex 572 (경희토류(La, Ce, Pr, Nd, Sm) 혼합용액에서 Cyanex 572에 의한 추출·분리 거동 고찰)

  • Extraction and separation behaviors of La, Ce, Pr, Nd and Sm from light rare earth multi - component mixed solutions by Cyanex 572 were studied. As extractant concentration increased, the $pH_{50}$ values of all the five components decreased. When extractant concentration was larger than 0.6 M, the separation factor of La and Ce, Nd and Sm was higher than 10, while the separation factor between Ce and Pr, Pr and Nd was as low as 0.5~2.2. Addition of TBP to the 0.6 M Cyanex 572 had little synergistic effect on the phase separation rate and separation factor. From the analysis of experiment results, group separation of [La]/[Pr, Nd, Sm] and [Pr, Nd]/[Sm] could be possible, but in case of the group separation between [La, Ce] and [Pr, Nd] was not available because of the low separation factor between Ce and Pr.

  • We investigated the pore properties of inorganic membranes applied for hydrogen separation industry. Inorganic membranes were derived from polysilazanes. The thermal reactions involved were studied using thermogravimetry(TG) and IR spectroscopy(FTIR) of the solids. To determine the thermal effect of pore properties, polysilazanes were pyrolysed in inert atmosphere. Pore volume and BET surface area showed the maximum value at a pyrolysis temperature of $500^{\circ}C$. For amorphous SiCN membrane derived from polysilazanes, selectivity of $H_2/N_2$ was 4.81 at $600^{\circ}C$.

  • An object of this study is to confirm the opening amount of the throttle valve that is begun the temperature separation of vortex tube for various engine speed and load condition in a common rail diesel engine. The vortex tube located at downstream of the exhaust manifold is a device separating the incoming exhaust gas to hot and cold stream. To find optimum separation efficiency of vortex tube, the opening amount of throttle valve has been investigated for various engine speed and load conditions. Engine speed was found that the influence of engine speed was dominant compared with that of engine load. As engine speed was increased, the throttle opening amount starting temperature separation was reduced.

  • Solvent extraction experiments of Pt(IV), Pd(II) and Rh(III) by Alamine336 were performed from the mixed chloride solutions. In the HCl concentration range from 1 to 5 M, most of Pt and Pd were extracted from the mixed solutions. However, the extraction percentage of Rh was much smaller than that of Pt and Pd. Lower concentration of Alamine336 in strong HCl solution led to higher separation factor of Rh from Pt and Pd. Adding $SnCl_2$ to the mixed solutions increased the extraction percentage of Rh, while the extraction percentage of Pt and Pd was little affected. Our results showed that selective separation of Rh or coextraction of the three platinum group metals from the mixed solution would be possible by adjusting the extraction conditions.

Enhanced Separation Technique of Heavy Metal (Pb, Zn) in Contaminated Agricultural Soils near Abandoned Metal Mine (폐금속 광산지역 농경지 납, 아연 오염 토양의 중금속 고도선별)

  • The study is to propose the optimal separation technique of heavy metals (Pb and Zn) contaminated in soil for improving the removal efficiency by various applicable techniques. The heavy metal contaminated soil samples near abandoned mine X-1 and X-2 were used for the study. Firstly, the wet classification process was shown more than 80% of removal efficiency for lead and zinc. Meanwhile, the magnetic separation process was shown low removal efficiency for lead and zincs because those heavy metals were non-magnetic materials. For the next step, the flotation separation process was shown approximately 24.4% of removal efficiency for zinc, while the gravity concentration process was shown approximately 57% of removal efficiency for lead, and 19.9% of removal efficiency for zinc, respectively. Therefore, zinc contaminated in soil would be effectively treated by the combination technique of the wet classification and the flotation technique. Meanwhile, lead contaminated in soil would be effectively treated by the combination technique of the wet classification process and the flotation process. Furthermore, the extraction of organic matter was shown more effective with aeration, 3% of hydrogen peroxide and 3% of lime such as calcium hydroxide.

  • Asymmetric polyetherimide membranes were prepared by phase inversion method. In the modification of the skin layers of polyetherimide membranes, the effects of NaOH concentration on the morphology and pervaporation separation of water-isopropanol mixtures were investigated. With increasing concentration of NaOH solution, polyamicacid structure was formed by the hydrolysis of imide group of polyetherimide, and the thickness of dense layer of the asymmetric membrane increased. In the pervaporation separation of water-isopropanol mixtures the overall permeation rate decreased and the separation factor increased with increasing concentration of NaOH solution. However, when the concentration of NaOH solution was very high, the permeation rate increased but separation factor decreased. From these results, it was found that the permeation behaviors of asymmetric polyetherimide membranes depended upon the concentration of NaOH solution. These modified membranes showed that both the permeation rate and separation factor increased as the operating temperature increased.

  • Cognitive radio (CR) is proposed as a key solution to improve spectral efficiency and overcome the spectrum scarcity. Spectrum sensing is an important task in each CR system with the aim of identifying the spectrum holes and using them for secondary user's (SU) communications. Several conventional methods for spectrum sensing have been proposed such as energy detection, matched filter detection, etc. However, the main limitation of these classical methods is that the CR network is not able to communicate with its own base station during the spectrum sensing period and thus a fraction of the available primary frame cannot be exploited for data transmission. The other limitation in conventional methods is that the SU data frames should be synchronized with the primary network data frames. To overcome the above limitations, here, we propose a spectrum sensing technique based on blind source separation (BSS) that does not need time synchronization between the primary network and the CR. Moreover, by using the proposed technique, the SU can maintain its transmission with the base station even during spectrum sensing and thus higher rates are achieved by the CR network. Simulation results indicate that the proposed method outperforms the accuracy of conventional BSS-based spectrum sensing techniques.
