주요 오염물질로 오염된 지하수에서 미생물의 무배양식 군집분석방법과 미생물상에 대한 조사방법 연구 (Culture-Independent Methods of Microbial Community Structure Analysis and Microbial Diversity in Contaminated Groundwater with Major Pollutants)

  • 최근에 적용된 지하수 미생물의 군집구조를 밝히는 분자생물학적 및 생화학적 방법들에 대해서 알아보았고 그 결과로서 지하수의 주요 오염물질에 따른 활성화된 미생물군집들이 무엇인지를 밝힌 연구논문들을 종합하여 정리하였다. PCR에 의한 유전자 증폭기술의 발달로 배양 없이 미생물 종류와 개체군을 파악할 수 있게 되었고 각종 finger-printing 방법 (DGGE, SSCP, RISA, microarray) 과 지방산분석법 (PLFA/FAME)을 이용하여 활성화 된 미생물군집구조를 분석하였으며 FISH 등의 방법으로 특정균의 활성도를 알아본 사례들을 조사하였다. 대표적인 지하수오염물질인 유류성분 (n-alkanes, BTEX, MTBE, ethanol)과 염소계 용매 (TCE, PCE, PCB, CE, carbon tetrachloride, chloro-benzene) 등으로 오염되었을 때 우점하는 지하수 미생물상에 대해 보고된 내용을 포함하였다.

  • 비음용 목적의 지하수에 대한 국내 수질기준은 기준항목 및 기준치설정에 근거가 명시되지 않아 체계적인 기준설정체계 마련이 시급한 실정이다. 본 연구에서는 비음용 목적의 특정유해물질에 대한 위해성기반 한국 지하수 수질기준 설정체계를 마련하기 위해 흡입 및 피부접촉 노출경로를 고려한 위해성을 반영하고 있는 선진국의 지하수기준 설정근거를 분석하였으며, 국내 타 수질 관련 매체의 기준 설정 체계를 바탕으로 향후 한국 지하수기준의 설정체계를 제안하였다 모니터링 우선항목 선정, 모니터링 수행, 위해성평가, 지하수 수질기준 후보물질 선정, 특정유해물질 지하수 수질기준 신규항목 선정 및 항목별 기준치 설정 단계로 제안하였다.

Assessment of Groundwater Quality for Irrigation and Agro-based Industrial Usage in Selected Aquifers of Bangladesh

  • Groundwater sampled from 24 tube wells of three districts namely Sherpur, Gaibandha and Naogaon in Bangladesh was appraised for their water quality for irrigation and agro-based industrial usage. All waters under test were slightly alkaline to alkaline (pH = 7.2 to 8.4) in nature and were not problematic for crop production. As total dissolved solid (TDS), all groundwater samples were classified as fresh water (TDS<1,000 mg/L) in quality. Electrical conductivity (EC) and sodium adsorption ratio (SAR) values reflected that waters under test were under medium salinity (C2), high salinity (C3) and also low alkalinity (S1) hazard classes expressed as C2S1 and C3S1. As regards to EC and soluble sodium percentage (SSP), groundwater samples were graded as good and permissible in category based on soil properties and crop growth. All water samples were free from residual sodium carbonate (RSC) and belonged to suitable in category. Water samples were under soft moderately hard, hard and very hard classes. Manganese, bicarbonate and nitrate ions were considered as major pollutants in some water samples and might pose threat in soil ecosystem for long-term irrigation. For most of the agro-based industrial usage, Fe and Cl were considered as troublesome ions. On the basis of TDS and hardness, groundwater samples were not suitable for specific industry. Some water samples were found suitable for specific industry but none of these waters were suitable for all industries. The relationship between water quality parameters and major ions was established. The correlation between major ionic constituents like Ca, Mg, K, Na, $HCO_3$ and Cl differed significantly. Dominant synergistic relationships were observed between EC-TDS, SAR-SSP, EC-Hardness, TDS-Hardness and RSC-Hardness.

  • The groundwater has been a major source of water supply throughout the ages. Around 50% of the rural as well as urban population in the developing countries like India depends on groundwater for drinking. The groundwater is also an important source in the agriculture and industrial sector. In many parts of the world, groundwater resources are under increasing threat from growing demands, wasteful use and contamination. A good planning and management practices are needed to face this challenge. A key to the management of groundwater is the ability to model the movement of fluids and contaminants in the subsurface environment. It is obvious that the contaminant source activities cannot be completely eliminated and perhaps our water bodies will continue to serve as receptors of vast quantities of waste. In such a scenario, the goal of water quality protection efforts must necessarily be the control and management of these sources to ensure that released pollutants will be sufficiently attenuated within the region of interest and the quality of water at points of withdrawal is not impaired. In order to understand the behaviour of contaminant transport through different types of media, several researchers are carrying out experimental investigations through laboratory and field studies. Many of them are working on the analytical and numerical studies to simulate the movement of contaminants in soil and groundwater of the contaminant transport. With the advent of high power computers especially, a numerical modelling has gained popularity and is indeed of particular relevance in this regard. This paper provides the state of the art of contaminant transport and reviews the allied research works carried out through experimental investigation or using the analytical solution and numerical method. The review involves the investigation in respect of both, saturated and unsaturated, porous media.

Estimating anaerobic reductive dechlorination of chlorinated compounds in groundwater by indigenous microorganisms

  • Tetrachloroethylene (PCE) and trichloroethylene (TCE), critical pollutants to human health and groundwater ecosystems, are managed by groundwater quality standards (GQS) in South Korea. However, there are no GQSs for their by-products, such as cis-dichloroethylene (DCE) and vinyl chloride (VC) produced through the dechlorination process of PCE and TCE. Therefore, in this study, we monitored PCE, TCE, cis-DCE, and VC in 111 national groundwater wells for three years (2016 to 2018) to evaluate their distributions, a biological dechlorination possibility, and human risk assessment. The detection frequency of them was 30.2% for PCE, 45.1% for TCE, 43.9% for cis-DCE and 13.4% for VC. The four chlorinated compounds were commonly detected in 21 out of 111 wells. In the results of statistical analysis with 21 wells data, DO and ORP also had a negative correlation with four organic chlorinated compounds, while EC and sulfate has a positive correlation with the compounds. This indicates that the 21 wells were relatively met with suitable environments for a biological dechlorination reaction compared to the other wells. Finally, cis-DCE had a non-carcinogenic risk of 10-1 and the carcinogenic risk of VC was 10-6 or higher. Through this study, the distribution status of the four chlorinated compounds in groundwater in South Korea and the necessity of preparing plans to manage cis-DCE and VC were confirmed.

울산지역의 지하수 수질에 관한 통계학적 연구

  • One hundred and thirty two ground water samples from the Usan area were divided into urban and non-urban groupings and were assessed between 1993 and 1996. The results of statistical analysis were as follows: There were significant differences between the two groupings in the average value of the following water quality parameters: total hardness, nitrate , pH, iron, ammonia and chloride ion in the order of Z-score . Because total hardness, nitrate, and pH were also significant in ANOVA test, these three parameters can be regarded as the most sensitive parameters of artirial pollutants. By the comprehensive com- parison of Ulsan water Quality to that of Kangwon-do, all of the major parameters(pH, KMnO4 consumption, sulfate, chloride and hardness except nitrated were confirmed as being significantly Increased levels.

  • Acoustically enhanced soil flushing method is a newly developed in-situ remediation technique. However, there has not been an analytical method that can be used to evaluate the effectiveness of ultrasonic wave under different conditions. This study was undertaken to investigate the degree of enhancement in contaminant removal due to ultrasonic energy on the soil flushing method. The test conditions included different levels of ultrasonic power and hydraulic gradient. The test soils were Ottawa sand, a fine aggregate, and a natural soil, and the surrogate contaminant was a Crisco Vegetable Oil. The test results showed that sonication could increase contaminant removal significantly. Increasing sonication power increased pollutant removal. The faster the flow is, the smaller the degree of enhancement will be. The pollutants in dense soils are more difficult to be removed than in loose soils.

  • This paper presents preliminary laboratory investigations on the electrokinetic (EK) remediation coupled with permeable reactive barrier (PRB) system. Atomizing slag was adopted as a PRB reactive material for remediation of groundwater contaminated with inorganic and/or organic substances. A series of laboratory experiments were performed with variable conditions such as (i) type of contaminant, (ii) applied electric field strength, (iii) processing time, and (iv) the application of PRB system. From the preliminary investigations, the coupled technology of EK with PRB system would be effective to remediate contaminated grounds without the extraction of pollutants from subsurface due to the reactions between the reactive materials and contaminants.

  • Humic acid, fulvic acid and humin present in volcanic ash soil were isolated by IHSS standard procedure and their characteristics were analyzed as a basic study to evaluate the effect of humic substances on the behaviour of pollutants in contaminated surface soil. The volcanic ash soil contained 42.1 % of total organic matter based on the oven-dried soil, and humin, humic and fulvic acids corresponded to 67.5 %, 15.2 %, 7.6 % of TOM respectively. Structural informations of the humic fractions were obtained from their elemental analysis and IR, CPMAS C-13 NMR spectral analysis and the differences among them are discussed with their C/H, O/C ratios and distributions of carbon types in the molecules.

  • In this study, the combined electrokinetic and ultrasonic remediation technique onto simple soil flushing was studied for the enhancement of water and liquid flows and the removal of contaminants in contaminated soils. The ultrasonic technique has been used to increase liquid flow and remove pollutants in contaminated soil. The laboratory soil flushing tests combined electrokinetic and ultrasonic technique were conducted using specially designed and fabricated devices to determine the effect of these both techniques. A series of laboratory permeability experiments involving the simple, electrokinetic, ultrasonic, and electrokinetic & ultrasonic flushing test were carried out. A soil admixed with sand and kaolin was used as a test specimen, and Pb and ethylene glycol were used as contaminants. An increase in out flow, permeability and contaminant removal rate was observed in electrokinetic and ultrasonic flushing tests.
