Comparison of Cold Hardiness in Canes and Buds of Kiwifruit Cultivars (품종에 따른 키위나무 눈과 가지의 내한성 비교)

  • In Korea kiwifruit growing area is limited to southern coastal region and Jeju island, partly due to the lack of information on their cold hardiness in winter. This study was carried out to investigate cold hardiness of Korean kiwifruit cultivars in a period of dormancy for using it as preliminary data to expand the cultivation area of kiwifruit in Korea. A total of five kiwifruit cultivars in two species and hybrid, Actinidia deliciosa ('Hayward' and 'Garmrok'), A. chinensis ('Goldone') and A. arguta hybrid ('Bangwoori' and 'Skinny Green') were subjected to five freezing treatments of -12℃, -15℃, -18℃, -21℃ and -24℃. Cell membrane damage in all cultivars initiated in -18℃/32h and cell membrane stability was lost in -24℃ in most cultivars, except for 'Skinny Green'. Cold hardiness was estimated by 50% lethal temperature (LT50) which was determined by triphenyl tetrazolium chloride (TTC) reduction. In branches, LT50 was -15℃ in 'Hayward' and 'Garmrok', -18℃ in 'Bangwoori' and -21℃ in 'Goldone.' The LT50 of buds on 'Hayward' and 'Garmrok' was 56 and 42 hours in -15℃ and 4 and 11 hours in -18℃, respectively; however, LT50 of buds on 'Goldone' was 51 hours in -18℃ and that on 'Bangwoori' was 3 hours in -24℃. Cold hardiness results imply that it may be difficult for cultivars in A. deliciosa such as 'Hayward' and 'Garmrok' to be grown in the north of southern coastal region in Korea; however, it can be possible for several cultivars in A. chinensis and A. arguta hybrid to be grown in the northern part of Korean kiwifruit belt if cold tolerance in the thaw is confirmed.

The Effect of Root Zone Cooling at Night on Substrate Temperature and Physiological Response of Paprika in Hot Climate (고온기 야간시간 근권냉방이 파프리카 배지온도와 생리적 반응에 미치는 영향)

  • This study examined a technique for cooling root zone aimed at lowering substrate temperature for sweet pepper (Capsicum annum L. 'Orange glory') cultivation in coir substrate hydroponics during hot season, from the $16^{th}$ of July to $15^{th}$ of October in 2012. The root zone cooling technique was applied by using an air duct (${\varnothing}12$ cm, hole size 0.1 mm) to blow cool air between two slabs during night (5p.m. to 3a.m.). Between the $23^{rd}$ of July and $31^{st}$ of August (hot temperature period), average daily substrate temperature was $24.7^{\circ}C$ under the root zone cooling, whereas it was $28.2^{\circ}C$ under condition of no cooling (control). In sunny day (600~700 W $m^{-2}{\cdot}s^{-1}$), average substrate temperatures during the day (6a.m. to 8p.m.) and night (8p.m. to 6a.m.) were lower about $1.7^{\circ}C$ and $3.3^{\circ}C$, respectively, under the cooling treatment, compared to that of control. The degree of temperature reduction in the substrate was averagely $0.5^{\circ}C$ per hour under the cooling treatment during 6p.m. to 8p.m.; however, there was no decrease in the temperature under the control. The temperature difference between the cooling and control treatments was $1.3^{\circ}C$ and $0.6^{\circ}C$ in the upper and lower part of the slab, respectively. During the hot temperature period, about 32.5% reduction in the substrate temperature was observed under the cooling treatment, compared to the control. Photosynthesis, transpiration rate, and leaf water potential of plants grown under the cooling treatment were significantly higher than those under the control. The first flowering date in the cooling was faster about 4 days than in the control. Also, the number of fruits was significantly higher than that in the control. No differences in plant height, stem thickness, number of internode, and leaf width were found between the plants grown under the cooling and control, except for the leaf length with a shorter length under the cooling treatment. However, root zone cooling influenced negligibly on eliminating delay in fruiting caused by excessively higher air temperature (> $28^{\circ}C$), although the substrate temperature was reduced by $3^{\circ}C$ to $5.6^{\circ}C$. These results suggest that the technique of lowering substrate temperature by using air-duct blow needs to be incorporated into the lowering growing temperature system for growth and fruit set of health paprika.

FABRICATION OF MYOMUCOSAL FLAP USING CULTURED ORAL EPITHELIUM IN RABBIT MODEL (가토모델에서 배양 구강상피를 이용한 근-점막 피판의 형성에 관한 연구)

  • Purpose : Extensive defect of oral and maxillofacial area is usually reconstructed with composite flap including skin paddle. However, if the defects are lined with only skin components, the mucosa's role in mastication and texture are not restored. Furthermore, stiffness and hair-growing prevent denture rehabilitation and good oral hygiene. This study was performed to overcome the disadvantages of composite soft tissue flaps including the skin and to make a model for myo-mucosal flaps. Materials and methods : Buccal mucosa sized $0.5\times1.0\;cm^2$ from New Zealand rabbit (around 1.5kg) was harvested and cultivated by the modification of Rheinwald and Green's keratinocyte culture method. Cultured mucosa was grafted on the fascia of latismus dorsi as form of mucosal sheet. After 7, 10, 14 days, the myomucosal flap was excised and evaluated under light microscope with H & E and immunohistochemical staining. As control group, harvested buccal mucosa from rabbit was transplanted to gracilis muscle(n=6). Results : From 7 days after prelamination, the basal layer of the grafted mucosa resembled that of normal mucosa. As control group, transplanted mucosa had original shape but there's slight inflammatory reaction. Prelaminated mucosa has 19.8$\pm$4.59 cell layers and some samples have more than 20 layers. The expression rate of PCNA was relatively strong (42.9%$\pm$14.1) at the basal layer of grafted mucosa and the laminin was found at the basal layer. On the contrary, prelaminated mucosa at 10 days showed moderate expression rate of PCNA(32.4%$\pm$4.62). We found the mucosal layer was somehow disappeared and there is strong inflammatory reaction. After 14 days prelamination, the grafted oral keratinocytes were almost disappeared and expression of PCNA was not observed. Conclusion : We can make 75 fold large mucosal($3850mm^2$) sheet from small samples of mucosa $(50mm^2)$. Epithelial sheet that grafted on the fascia of muscle underwent differentiation and proliferation. But after 10, 14 days, there was strong inflammatory reaction and the grafted mucosa was destroyed from surface layer. In rabbit model, transfer of fascio-mucosal flap should be done from 7 to 10 days after prelamination.

  • Mutual growth limitation was observation when the two antagonistic fungi was come in contact with each other. Brown line was formed 2day after contact with Trichoderma spp., and then, green spores formed overnight. The laccase activity of L. edodes was stimulated when this fungus wsa co-incubated with Trichoderma spp. for a few days in liquid media. In sawdust-rice bran nixtures, outstanding broun line developed when the two antagonistic fungi co-cultured. The pH of the substrates changed from 5.5 to 4.5 after overgrowth, suggesting a difference in the degradation ability and the preference of the two fungi for the lignocellulose material.

Forage Productivity of Two Sorghum and a Sorghum-Sudangrass Hybrids harvested at Different Growth Stages (수확기에 따른 수수와 수수-수단그라스 교잡종의 사료생산성)

  • Forage productivity of two sorghum hybrids, Pioneer(P) 931 and P 956 and a sorghum-sudangrass hybrid, P 988, was studied. The 1st-cut of sorghun hybrids was made for silage 6 times from heading stage at a week-interval and regrowth (2nd-cut) was harvested on Oct. 13 although plants were not reached the same growth stage of 1st-cut. However, the 1st- and 2nd-cut of sorghum-sudangrass were made for green fodder 6 times from 4 weeks before heading stage at a week-interval. 1. Percent dry matter(DM) of 1st-cut of sorghum and 1st- and 2nd-cut of sorghum-sudangrass increased as growth stage advanced. Percent DM of 2nd-cut of sorghum decreased as the 1st-cut date delayed due to insufficient growing period on Oct. 13. 2. In the 1st-cut of sorghum the proportion of leaf blade (LB) or culm+leaf sheath (LS) to total DM decreased, but that of panicle increased as growth stage advanced. The panicle proportion of P 956 increased and culm+LS decreased more rapidly compared with those of P 931. IIowever, in sorghum-sudangrass hybrid the proportion of LB decreased and culmf LS increased as growth stage advanced through heading stage. 3. In the 1st-cut of sorghum hybrids crude(C) protein and C. fiber decreased, but nitrogen free extract (NFE) increased as growth stage advanced although C. fat and C. ash were similar among the growth stages. In the 2nd-cut of sorghum C. protein and C. ash increased, but NFE decreased as harvesting date delayed due to plants were younger although C. fat and C. fiber were similar among the harvesting dates. In the sorghum-sudangrass C. protein of 1st-cut decreased, but C. fiber of 1st-cut and NFE of the 1st-and 2nd-cut increased as growth stage advanced. 4. DM yield of 1st-cut of sorghum increased from heading stage to 3 weeks after heading and then levelled off, but total DM including regrowth was similar among the harvesting time. In the sorghum-sudangrass hybrid DM yield of 1st-cut and total DM yield including 2nd-and 3rd-cut increased as the harvestingtime delayed.

Evaluation of Green House Gases Emissions According to Changes of Soil Water Content, Soil Temperature and Mineral N with Different Soil Texture in Pepper Cultivation (고추재배에서 토성별 토양수분, 토양온도, 무기태 질소 변화에 따른 온실가스배출 평가)

  • Importance of climate change and its impact on agriculture and environment has increased with a rise of greenhouse gases (GHGs) concentration in Earth's atmosphere, which had caused an increase of temperature in Earth. Greenhouse gas emissions such as methane($CH_4$) and nitrous oxide($N_2O$) in the field need to be assessed. GHGs fluxes using chamber systems in the fields(2004~2005) with pepper cultivation were monitored at the experimental plots of National Academy of Agricultural Science(NAAS), Rural Development Administration(RDA) located in Suwon city. $N_2O$ emission during pepper growing period was reduced to 74.0~82.1% in sandy loam soil compared with those in clay loam soil. Evaluating $N_2O$ emission at different levels of soil water conditions, $N_2O$ emission at -50 kPa were lowered to 13.2% in clay loam soil and 40.2% in sandy loam soil compared with those at -30 kPa. $CH_4$ emission was reduced to 45.7~61.6% in sandy loam soil compared with those in clay loam soil. Evaluating $CH_4$ at different levels of soil water conditions, $CH_4$ emission at -50 kPa was lowered to 69.6% in clay loam soil and 55.8% in sandy loam soil compared with those at -30 kPa. It implied that -50 kPa of soil water potential was effective for saving water and reducing GHG emissions. From the path analysis as to contribution factors for $N_2O$ emission, it appeared that contribution rate was in the order of mineral N(51.2%), soil temperature (25.8%), and soil moisture content(23.0%) in clay loam soil and soil moisture content(39.3%), soil temperature (36.4%), and mineral N(24.3%) in sandy loam soil.

Application of Molecular Biological Technique for Development of Stability Indicator in Uncontrolled Landfill (불량매립지 안정화 지표 개발을 위한 분자생물학적 기술의 적용)

  • This study was conducted for developing the stability parameter in uncontrolled landfill by using a biomolecular investigation on the microbial community growing through leachate plume. Landfill J(which is in Cheonan) and landfill T(which is in Wonju) were chosen for this study among a total of 244 closed uncontrolled landfills. It addressed the genetic diversity of the microbial community in the leachate by 165 rDNA gene cloning using PCR and compared quantitative analysis of denitrifiers and methanotrophs with the conventional water quality parameters. From the BLAST search, genes of 47.6% in landfill J, and 32.5% in landfill T, respectively, showed more than 97% of the similarity where Proteobacteria phylum was most significantly observed. It showed that the numbers of denitrification genes, i.e. nirS gene and cnorB gene in the J site are 7 and 4 times higher than those in T site, which is well reflecting from a difference of site closure showing 7 and 13 years after being closed, respectively. In addition, the quantitative analysis on methane formation gene showed that J1 spot immediately bordering with the sources has the greatest number of methane formation bacteria, and it was decreased rapidly according to distribute toward the outer boundary of landfill. The comparative investigation between the number of genes, i.e. nirS gene, cnorB gene and MCR gene, md the conventional monitoring parameters, i.e. TOC, $NH_3-N,\;NO_3-N,\;NO_2-N,\;Cl^-$, alkalinity, addressed that more than 99% of the correlation was observed except for the $NO_3-N$. It was concluded that biomolecular investigation was well consistent with the conventional monitoring parameters to interpret their influences and stability made by leachate plume formed in downgradient around the uncontrolled sites.

Variations in Soil-to-Red Pepper Transfer Factors of Radionuclides with Time of Their Application and Fruit Harvest (고추 재배시 방사성 핵종 처리 및 열매수확 시기에 따른 토양-작물체간 전이계수의 변이)

  • A mixed solution of $^{54}Mn$, $^{60}Co$, $^{85}Sr$ and $^{137}Cs$ was applied to the soil of culture boxes in a greenhouse 2 days before transplanting red pepper and at 3 different times during its growth for investigating transfer factors ($m^2/kg-dry$) for its green and red fruits. Transfer factors varied with radionuclide, application time and harvest time by factors of about $20{\sim}100$. They decreased mostly in the order of $^{85}Sr>^{54}Min>^{60}Co>^{137}Cs$ while $^{54}Mn$ and $^{60}Co$ was higher than $^{85}Sr$ when time lapse between application and harvest was short. Transfer factors of $^{85}Sr$ and $^{137}Cs$ at the last application were lower than those at the previous one by factors of $3{\sim}20$ depending on harvest time. Variations in $^{54}Mn$ and $^{60}Co$ transfer factors with application time after transplanting were comparatively low. Transfer factors of $^{54}Mn$, $^{60}Co$ and $^{85}Sr$ mixed with topsoil before transplanting were up to $3{\sim}9$ times higher than those for the application onto soil surface 2 days after transplanting while there was no difference in $^{137}Cs$. The present results can be referred to in estimating root-uptake concentrations of the radionuclides in red pepper fruit and taking proper measures for its harvest and consumption at the event of an accidental release during the growing season of red pepper.

Measuring Water Content Characteristics by Using Frequency Domain Reflectometry Sensor in Coconut Coir Substrate (FDR(Frequency Domain Reflectometry)센서를 이용한 코코넛 코이어 배지내 수분특성 측정)

  • This experiment has investigated suitable methods to improve precision water content monitoring of coconut coir substrates to control irrigation by frequency domain reflectometry(FDR) sensors. Specifically, water content changes and variations were observed at different sensing distances and positions from the irrigation dripper location, and different spaces between the FDR sensors with or without noise filters. Commercial coconut coir substrates containing different ratios of dust and chips(10:0, 7:3, 5:5, 3:7) were used. On the upper side and the side of the substrates, a FDR sensor was used at 5, 10, 20, 30cm distances respectively from the irrigation dripper point, and water content was measured by time after the irrigation. In the glass beads, sensors were installed with or without noise filtering. Closer sensing distance had a higher water content increasing rate, regardless of different coir substrate ratios. There were no differencies of water content increasing rates in 10:0 and 3:7 substrates between the upper side and the side. Whereas, 7:3 and 5:5 substrates showed higher increasing rates on the upper side measurements. Substrates with higher ratios of chip(3:7) had lower increasing rates than others. And, with noise filters, the exatitude of measurement was improved because the variation and deviation were reduced. Therefore, in coconut coir with FDR sensors, an efficient water content measurment to control irrigations can be achieved by installing sensors closer to an irrigation point and upper side of substrates with noise filters.

Seasonal Survival Characteristics of Conifer Seedlings and Their Suitable Planting Season (침엽수(針葉樹) 식재시기별(植栽時期別) 활착특성(滑着特性)과 식재적기(植栽適期)에 관한 연구(硏究))

  • Main consideration of this trial is to know whether the planting work should be possible to do not only in the early spring but also in the summer or autumn, for giving the guide to get the work plan and to broaden the employing season of the skilled forest worker. Seedling of Pinus koraiensis, Larix leptolepsis, Pinus rigida, Pinus rigida${\times}$ P. taeda(wind) and Chamaecyparia obtusa as the test species had been planted in 15 days interval from the middle of March to the end of November. The seedling survival was investigated in the spring time of coming year because the winter damage could be problems. At the same time the climate data was measured daily and the shoot growth of test species were also measured in other near plantation at 15 days interval to know the influence to survival. From these results the spring and autumn planting is showing the good survival and the summer planting seems to give the difficulties. The spring planting in the southern temperate zone could be stared earlier as the end of February or beginning of March because the soil temperature are increasing up more $5^{\circ}C$ from this time. But the summer planting from the beginning of May until the end of August in better to avoid with excluding specially the good season of rainfall distribution because of the shoot growth of green confer seedling and the leave sprouting of Larix leptolepsis are so vigorously growing up from the begining of May and its wood structure is too weak to compensate the water loss. But among the test species Pinus koraiensis and Chamaecyparis obtusa have more possibility to plant in the summer season. The autumn planting seems to be very reasonable to accept newly in the trial region. This may be the reasons of still high soil temperature to grow the seedling root and of hardened school to resist from the dry winter wind. But it will be carefully that the strongly exposured site could be to avoid for the autumn planting in case of specially Pinus rigida${\times}$P. taeda and Chamaecyparis obtusa. From these discussion the guide table 1 for planting season with the test species is proposed and can be used for planing and employing in the trial zone.
