Insertional mutations exhibiting high cell-culture density HCD phenotypes are enriched through continuous subcultures in Chlamydomonas reinhardtii

  • Low efficiency in microalgal biomass production was largely attributed to the low density of algal cell cultures. Though mutations that reduced the level of chlorophyll or pigment content increased efficiency of photon usage and thus the cell-culture density under high-illumination growth conditions (e.g., >$500{\mu}mol\;photon\;m^{-2}\;s^{-1}$), it was unclear whether algae could increase cell-culture density under low-illumination conditions (e.g., ${\sim}50{\mu}mol\;photon\;m^{-2}\;s^{-1}$). To address this question, we performed forward genetic screening in Chlamydomonas reinhardtii. A pool of >1,000 insertional mutants was constructed and subjected to continuous subcultures in shaking flasks under low-illumination conditions. Complexity of restriction fragment length polymorphism (RFLP) pattern in cultures indicated the degree of heterogeneity of mutant populations. We showed that the levels of RFLP complexity decreased when cycles of subculture increased, suggesting that cultures were gradually populated by high cell-culture density (HCD) strains. Analysis of the 3 isolated HCD mutants after 30 cycles of subcultures confirmed that their maximal biomass production was 50-100% higher than that of wild type under low-illumination. Furthermore, levels of chlorophyll content in HCD mutant strains were similar to that of wild type. Inverse polymerase chain reaction analysis identified the locus of insertion in two of three HCD strains. Molecular and transcriptomic analyses suggested that two HCD mutants were a result of the gain-of-function phenotype, both linking to the abnormality of mitochondrial functions. Taken together, our results demonstrate that HCD strains can be obtained through continuous subcultures under low illumination conditions.

  • In this paper, radiation pattern of rectangular microstrip patch antenna with anisotropy substrates and superstrate is studied by using a rigorous full-wave approach and a moment method calculation. Dyadic Green's function is derived for selected anisotropy material by constitutive relation. From these results, integral equations of electric fields are formulated. The electric field integral equations are discretized into the matrix form by applying Galerkin's moment method and then the current coefficients are obtained.. After solving the current coefficients, the far-zone electric field in spherical coordinates can be obtained by using the stationary phase method. To verify the validity of numerical result, we compare our result with existing one and get a good agreement between them. From the numerical results, the radiation patterns for variation of uniaxial superstrate thickness, anisotropy ratio of substrate and superstrate layer are presented and analyzed.

Inhibitory Activity of Advanced Glycation Endproducts (AGE) Formation of Edible Plants for Development of Anti-Wrinkle Ingredients (피부 주름개선 소재개발을 위한 식용작물의 최종당화산물 생성 억제활성)

  • Skin wrinkles typically appear as a result of aging processes. One of causes may be the nonenzymatic glycation followed formation of browning products called advanced glycation endproducts (AGEs), an irreversible cross-linked protein. The accumulation of glycated collagen cross-linked in skin inhibits the formation and function of skin tightening agents such as collagen and elastin. To development for anti-wrinkle ingredients from edible plants, MeOH and hot-water extracts were prepared and evaluated for their inhibitory effects of AGEs formation. The activities of both extracts from bay laurel (Laurus nobilis), cinnamon (Cinnamomum loureirii), clove (Eugenia caryophyllate), oregano (Origanum vulgare), rosemary (Rosemarinus officinalis), savory (Satureja hortensis) and star anis (Illicium verum) of western spices, and blackberry (Rubus coreanus), dayflower (Commelina communis), Epimedium koreamun (whole), termunalia frutus (Terminalia chebula) and turkestan rose (Rosa rugosa) of medicinal plants were higher than the others. Of Korean vegetables, however, MeOH and hot-water extract from only Asters caber and green tea showed higher activities, and no activity in Korean marine plants (seaweeds).

  • Originally, ancestral rites is a ceremony to pay filial piety to ancestors continuously even though they died, and this is a startingpoint based on the thought of paying respect to ancestors and 'root consciousness' which Korean people have. In the earliest days, Korean people worshipped nature and the sacrificial rituals were performed mainly for the fods of the skies and the earth. After the end-Koryeo Era introduced [Juja-Garye]ancestrial rites of forefather in home generalized. By the way, the foods for ancestrial rites on the original literature centering the [Sa-Rye-Pyon-Ram]are as follows: raw meat(saeng: 牲), rice and broth(ban, gaeng: 飯羹), fruits(果), slice meats(po: 脯), salted dry fish(jaban(佐飯), rice punch(sikhye: 食醯), soup(tang: 湯), fish and meat, roasts beef (jeok: 炙), roast vegetable(sookchae: 熟菜), rice cake(pyon: 餠), noodles(麵), soy(醬), Kimchi(沈菜), drink(酒), green tea(cha: 茶) etc. Today, ancestral rites, basically with this conception, has a function to bind the family and relatives as one. But as double income family are increasing, most housewives have heavy burden mentally and financially to prepare the food for the ancestral. The foods for ancestral rites can be said a way to express the internal true heart, and a basic medium to practice the filial piety. Many documents let us know that a few days before the ancestral rites, we should prepare the food for it with careful and pious attitude. And, they stress that our sincere attitude in preparing food is important rather than its quantity. In this industrial society, we have lots of difficulty preparing and observing the same service as it in the traditional socity. But I think that housewives can be freed from the burden they have to some degree when they realize what the true meaning of ancestral rites is and that the food for it plays a role an external expression.

  • This study is the visually identification lighting for easy to distinguish using colors LED on AtoN facility in harbor gate, which have realized a controller for certainly express the gate of harbor by red light and green light of both sides a harbor and for synchronization at a time of right and left or sequential the harbor guidance light through synchronizer or timer by GPS. There is expectation effect that is prevent a confusion about distinguish of facility by ship's operator and to beautify a night scene of harbor, which is expressed to identification lighting differ from great many lighting of harbor with variable color lighting the lighthouse body and vertical layer color lighting using LED. Especially the function of AtoN is displayed for harbor safety message by CW lighting, and this system is the power consumption greatly reduce by candle alternated high light LED.

Proposal of Urban Agricultural Park Management and Operation Plan Using the Public Service Design Process

  • Background and objective: With the revision of the Act on Urban Parks, Green Areas, Etc. in 2013, the "urban agricultural park" was newly established under the subcategory of "themed park," thereby establishing the institutional basis for the creation of urban agricultural parks. However, urban agricultural parks are still in the early stages of their introduction. There is a lack of research on direction setting and specific operation management that considers urban residents' needs and the city's physical infrastructure. Methods: We utilized the public service design process suggested by the Ministry of the Interior and Safety of the Republic of Korea in 2019 to identify problems and develop directions for urban agricultural parks. The process consisted of the following four steps: Understanding, Discovering people's needs, Defining real problems, and Developing ideas. Results: As four types of ideas for revitalizing urban agricultural parks, 'information users want to know,' 'user participation in design,' 'venue for local communities,' and 'urban agricultural parks as health and rest areas' were derived. This means that urban agricultural parks must provide the information users want; users must plan, decide, and implement such information by directly participating in the creation and efficient management and operation of urban agricultural parks; and urban agricultural parks must be used as a venue for local communities. Urban agricultural parks should also be spaces for health and relaxation. Conclusion: Urban agricultural parks should avoid the unified space and passive participation patterns of existing urban parks, and become real spaces for resident participation that can satisfy all the production, leisure, landscape, ecology, and psycho-social needs of the users of urban agricultural parks. Furthermore, it is necessary to introduce a more systematic and diverse operating system so that it can work to revitalize the local community and connect organically with the function of the city.

A Study on the Image Evaluation for the Improvement of the Landscape of Horticultural Complex in Rural Area (농촌지역 시설원예단지의 경관 개선을 위한 이미지평가)

  • Humans are provided with a wide range of public benefits from ecosystems and agricultural ecosystems, but the establishment of the horticulture complex could be a space that hampers the public function of the agricultural ecosystem. In this study, we sought to focus the function of landscape creation of agricultural landscape and tried to analyze the landscape of the horticulture complex. Therefore, This study aims to suggest ways to build a greenhouse complex which is being indiscreetly introduced in the agriculture landscape through environmentally-friendly manner and minimize the function of the ecosystem service. We divided the greenhouse complex into two categories of Plastic Greenhouse(v) and Glass Greenhouse(g), and compared them to the Netherland and Japan counterparts. Each image of research areas was selected by 3 pics and polled by a total of 101 people. The results of the Evaluation of Landscape Image are as shown in the figure. Netherland Glass Greenhouse scored 1.80 in terms of 'Neat' which is one of the given 15 adjectives. Study results shows that Korean Plastic Greenhouse landscapes need to endeavor Japanese vinly greenhouses and Dutch glasshouses. Consequently, an analysis on the elements of landscapes including green area, variant elements, separation distance is essential in order to improve our country's greenhouse complex landscapes. In this regard, continuous research is required to improve rural landscapes and harmonize large-scale horticultural facilities into the existing agricultural ecosystem.

  • With the advent of the time when the competitiveness of a nation or a city can be evaluated by the extent of cultural reception, the art exhibitions, like other genres of art, are actively held in various ways by different institutions. The spaces of exhibition offered not only by public museums and galleries but also by private galleries, alternative spaces and open-air exhibition spots are being increased. Likewise, the number of exhibitions organized on a large scale by Biennales and local governments is on the increase. In accordance with actual tendency, the Ministry of Culture and Tourism has planned to increase the number of museums and galleries up to 500 by the year of 2011. Under these circumstances, the actual situation shows that the central and local governments are adopting beneficial measures to support the private galleries such as permission of construction on green belt areas and tax deduction. That is, our society has come to realize that the necessity and importance of public as well as private galleries to play their roles as cultural and educational institution. Now, the number of galleries is growing. Exhibitions are held everywhere. Therefore, spectators have more chance to visit them than before. With these conditions and the growing number of amateurs, social interest in curator organizing exhibitions is getting popular. Because of the rise of curatorial popularity on the social level, the influence of curators which is exercised within the gallery seems to become gradually enlarged. In reality, it is true that the curatorial influence cannot be ignored more than ever for appear in various fields their new features made from their professional cometence. The function of gallery is more strengthened and its position is more heightened than those in 80s. For these reasons, the curators whose playground is the gallery, they may be regarded as invisible power to the artists. Futhermore, their influence as such seems to be more likely because our world of art does not still have any reliable system of art criticism. Nevertheless, considering the actual situation in which national and public galleries as cultural institution would rather exist on the local sentiments or political dynamics than on autonomy, there must be some restrictions for the galleries and the curators to obtain the power. In this context, the actual moment is looked upon as transitional period to make the curatorship settled down. Before discussing the power relevant to curatorship, we are going to examine first the role and function of curator and then the new curatorial role and function at the age of information. Finally, we will see in sequence the development and problems of curatorship, the power and dilemmas of curator.

  • This study is on perfume case that is one of a great number of ornaments which was designed by out ancestors. We had developed the tradition and the history of perfume case bag fan-weight etc. These have the same function as the present perfume. case. There were basket-shaped perfume cases which were the smellest one among the personal girdle ornaments in the silla era. The various patterned perfume case were made of gold metal coral platinum or green jadeite etc. In the single-crop trinkets a embroidered perfume bag with the gold and silver thread whose forms perfectly match their functions appeared during the Yi Dynasty. There was also a perfume bag which was one of the daily necessities. A precious "jul perfume" was carried by noble women. A fan-weight perfume bag was attached to the fan to emphasize the intrinsic beauty of utility and function. It is necessary to know the function of prefume case. As perfume case is weared on the clothes it was given more decorative effects as well as the function of medicinal amulet with a sweet smell. Therefore it is very important for us to study perfume case that has various function as an ornament. So The purpose of this study is to investigate the practical and decorative side of perfume case with the general examines of perfume finding how to practice use through our life. The results were as follows; 1. The first use of perfume is perfume through smoke which is for ceremony of religion It removes human body odor by degrees and spreads a sweet odor. Also the materials for making perfume of early age is aromatic plants which will be used flour-made flowers stems. As the materials for making perfume use is increasing today we can invent alcohol perfume today 2. Our country the custom of perfume-used is wide. Among them men's perfume-used was very special phenomenon. For example men were wearing perfume bag in the Silla era. Because perfume represented wealth and noble in those days. They shew off social position personality through perfume-used. 3. One of early religion ceremony article there was the perfume. And perfume case was means for containing perfume. Gradually the perfume case was used widly as increasing needs of perfume in human life. 4. In the middle period of 'Koryo' Dynasty perfume cases had a close relationship with clothes but after Mongolian has been attacked 'Koryo' there were changes in wearing clothes therefore the position of perfume cases were transfered to coat string that was the origin of decoration style that they began. That is to say the perfume case has been influenced the position of perfume case shapes with changing of fashion. 5. The perfume case has been made manifest various function as an ornament. In the practical side First medical-perfume in perfume case has been played an important role in first-aid medicine in critical condition. Second it was amulet for self protection.
