Evaluations on Antioxidant Effect of Water Extract from Graviola Leaves (그라비올라잎 열수추출물의 항산화 효능 평가)

  • This study examined the antioxidant activity of the water extract from graviola leaves to develop a harmless and highly stable natural antioxidant. The total polyphenol content, total flavonoid content, DPPH radical scavenging activity, and MTT assay activity were measured. As a result, 62.3 g of the water extract from graviola leaves was obtained at $98^{\circ}C$ using 300 g of graviola leaf powder. The total polyphenol content was $291.97+2.39{\mu}g/mL$ and the total flavnoid content was $161{\pm}7.85{\mu}g/mL$ in a 1 mg/mL water extract from graviola leaves. The DPPH radical scavenging activity showed 51.6%, 67.8%, 79%, 82.4% and 83.9% at concentrations of 1, 2.5, 5, 10 and 15 mg/mL. This shows concentration-dependent scavenging activity and significant antioxidant activity. As a result of measuring the toxicity about HDF cells, a HDF cell survival rate of 100% was observed at a 150 mg /mL concentration, which was the same as that of the control group and a higher cell survival rate at a lower concentration. In conclusion, the graviola leaf extract can be developed as a material of food or cosmetics containing natural antioxidants.

Antibacterial Activity of Graviola extract to inhibit the Staphylococcus epidermidis (Staphylococcus epidermidis를 억제하는 Graviola 추출물의 항균활성)

  • This study was carried out to verify the antimicrobial effect of Graviola leaves against pathogenic bacteria for the purpose of developing an antibacterial material. The extraction conditions of graviola leaves were $60^{\circ}C$, $80^{\circ}C$ and $98^{\circ}C$, and graviloa leaves werewater extracted at $60^{\circ}C$, $80^{\circ}C$ and $98^{\circ}C$.The extraction yields and extraction conditions were determined. The antimicrobial activity against S. epidermidis, S. aureus and E. coli TOP10 was evaluated by agar diffusion method. The extraction yields were 3.02%, 14.73% and 20.76% at $60^{\circ}C$, $80^{\circ}C$ and $98^{\circ}C$, respectively. The antimicrobial activity against S. epidermidis, S. aureus and E. coli TOP10 was found in S. epidermidis.In the samples extracted at $98^{\circ}C$, a clear zone of 13 mm was observed at 200 mg/mL and of 20 mm at 500 mg/mL. The MIC was 100 mg/mL. The higher the extraction temperature and concentration, the better was the growth inhibition effect. As a result, the natural antimicrobial activity contained in natural materials can solve the problem of resistance to antibiotics. It is considered that antimicrobial activity against S. epidermidis in skin is highly applicable to basic cosmetics and cosmetic materials.

Effect of Graviola Leaves Extracts on Antioxidant Activity and Melanin Production (멜라닌 생성 및 항산화 활성에 미치는 그라비올라 잎 추출물의 영향)

  • The purpose of this study was to investigate the effect of methanolic extracts of graviola, Annona muricate leaves (AMME) on antioxidant activity and melanin production. First of all, DPPH radical and reducing power were performed to determine the antioxidant effect of AMME and organic solvent fractions. AMME and organic solvent fractions showed antioxidative activity in a concentration dependent manner. The ethyl acetate fraction of AMME among organic solvent fractions showed the highest antioxidant activity. Moreover, tyrosinase activity was performed to confirm the effect of organic fractions on melanin production. AMME, ethyl acetate, and hexane fractions increased tyrosinase activity a dose dependent manner. Next, the hexane fraction with the best effect on melanin synthesis in AMME organic solvent fraction was divided into 12 fractions by silica column chromatography. Among them, the fraction 7 and 8 showed the highest DPPH radical scavenging activity and reducing power. In addition, the fraction 7 and 8 at $64{\mu}g/ml$ showed melanin synthesis by 260% and 184%, respectively. Finally, the fraction 8 at $4{\mu}g/ml$ showed melanin synthesis by 34% in B16F1 cells. LC-MS analysis showed that fraction 7 and fraction 8 have a molecular weight of 617 and 619, respectively. FT-IR analysis showed that fractions 7 and 8 is similar to bis(2-hydroxyethly)dimerate. Above results suggest that graviola leaves extracts could be applicable to the development of natural antioxidants or hair cosmetics which are related to the promoting effect of melanin production.
