Plasma G-CSF and GM-CSF Concentrations and Expression of their Receptors on the Granulocyte in Children with Leukocytosis (백혈구 증가증 환아의 혈장내 G-CSF와 GM-CSF의 농도 및 과립구에서의 이들 수용체의 발현)

  • Purpose : Granulocyte-colony stimulating factor(G-CSF) and granulocyte macrophage-colony stimulating factor(GM-CSF) are principal cytokines in granulopoiesis and their physiologic effects are mediated through binding to specific cell surface receptors. Although it is known that the level of serum G-CSF and GM-CSF, and presentation of the receptors are increased in infectious diseases, there have been no studies to find the correlation between the granulopoiesis and leukocytosis. This study was designed to measure G-CSF and GM-CSF in leukocytosis and in control and to demonstrate the possible pathogenesis of granulopoiesis in leukocytosis using quantitative analysis of G-CSF, GM-CSF and their CSFr. Methods : The plasma levels of G-CSF, GM-CSF of 13 children without leukocytosis and 14 children with leukocytosis were measured. Counts of cell surface G-CSFr and GM-CSFr were measured by combining anti G-CSFr and anti GM-CSFr monoclonal antibodies to their respective receptors by using quantitative flow cytometric assay. Results : There was no significant difference betweeen the plasma concentration of G-CSF and GM-CSF in acute leukocytosis and in the control group. However, levels of G-CSFr in acute leukocytosis decreased significantly compared to the control(P=0.012) and the levels of GM-CSFr in both groups revealed no significant difference. Conclusion : Increase in the number of leukocyte in leukocytosis was mediated by increasing the number of neutrophil, and increased plasma concentration of G-CSF may be the cause of neutrophilia. But GM-CSF did not have any influence on leukocytosis.

Effects of Curcuma longa Rhizoma on Asthma induced intra-nasal instillation of Ovalbumin in Mice (강황이 난황의 비강내 점적을 통하여 유발된 생쥐의 천식에 미치는 영향)

  • Objective : This study was designed to investigate the effects of Curcuma longa Rhizoma(CLR) on asthma. Methods : Detecting splenocyte proliferation rates, cytokines and antigen specific antibody isotypes in bronchoalveolar lavage fluid (BALF). In addition, the present author I also investigated changes in spleen and histopathological changes of lung tissues. Results : Oral administration of CLR lowered spleen weight and splenocyte proliferation rates. In addition, levels of IL-4, IL-17A and Granulocyte/Macrophage Colony-Stimulating Factor(GM-CSF), Th2 driven cytokines, were lowered respectively and IFN-g, and Th1 driven cytokine, were elevated by CLR. Levels of Ovalbumin(OVA) specific IgE and IgGl in BALF were also lowered by oral administration of CLR too. Conclusion : CLR is useful to treat patients with asthma and the mechanisms are related to the in suppression of Th2 skewing reactions.

  • The fine structure of the hemocytes in Wolf spider, Pardosa astrigera, is discribed and compared with that of similar cells in other spider species and insects. Five hemocyte types are identified in the hemolymph: prohemocyte, plasmatocyte, granulocyte, spherulocyte and adipohemocyte. Prohemocytes are small with a relatively undifferentiated cytoplasm. The nucleus is comparatively large and has a perinuclear space. Plasmatocytes and granulocytes are pinocytotic function in the hemolymph of the body. The plasmatocytes have some coated pits on the plasma membrane and well developed Golgi complex, The granulocytes appear sequence of events in the formation of coated vesicle from a coated pit on its plasma membrane. Golgi complex become well expressed and give rise to small secretory vesicles which fuse to large bodies. The spherulocytes are larger in cell size than other hemocytes. Their cytoplasm is filled with spherules. The spherules contain the floccurent materials and the helical structured materials, which are 220nm in length and 80nm in width. The adipohemocytes are oval shaped and have a number of lipid droplets.

Increased production of human granulocyte-macrophage colony stimulating factor (hGM -CSF) by the addition of stabilizing polymer in plant suspension cultures

  • The effect of stabilizing polymer on hGM-CSF production was investigated in suspension cell cultures of transgenic tobacco. Secreted human GM -CSF from cell suspension cultures was detected in the medium at a maximum concentration of 180 ${\mu}g/L$ by ELISA. However, the secreted hGM -CSF was unstable in the medium, and rapidly degraded after day 5. In order to stabilize the secreted hGM-CSF, three stabilizing polymers were tested, polyethylene glycol, polyvinylpyrrolidone and gelatin. Gelatin was the most effective in stabilizing the secreted GM-CSF. Following the addition of 5% (w/v) gelatin, the maximum GM -CSF concentration reached 783 ${\mu}g/L$, a 4.6-fold increase over control.

The Expression of Galectin-3, a Beta-Galactoside Binding Protein, in Dendritic Cells

  • Background: Dendritic cells (DCs) are the most potent APCs (antigen-presenting cells) and playa critical role in immune responses. Galectin-3 is a biological lectin with a beta-galactoside binding affinity. Recently, proteomic analysis revealed the presence of galectin-3 in the exosome of mature DCs. However, the expression and function of galectin-3 in DCs remains unclear yet. Methods: We used bone marrow-derived DCs of mouse and showed the expression of galectin-3 in DCs by using flow cytometry analysis and Western blot analysis. Results: Galectin-3 was determined as single band of 35 kDa in Western blot analysis. Flow cytometry analysis showed the major growth factor for DCs, granulocyte-macrophage colony stimulating factor (GM-CSF) and maturing agents, anti-CD40 monoclonal antibody (mAb) and lipopolysaccharide (LPS) consistently increased the intracellular expression of galectin-3 in DCs compared to medium alone. In addition, DCs treated with maturing agents did marginally express galectin-3 on their surface. Conclusion: This study suggests that galectin-3 in DCs may be regulated by critical factors for DC function.

The effects of immune response on maximal aerobic and anaerobic exercise (급성적인 최대 유·무산소 운동이 면역반응에 미치는 영향)

  • The purpose of this study is to examine the effect of the acute aerobic and anaerobic exercise on immune response. Fourteen university soccer players are sampled and divided into two groups -- one group of seven for aerobic exercise, the other for anaerobic exercise. The one group of aerobic exercise went through the Graded Exercise Test of Bruce protocol, using Treadmill, and were driven to be all-out ; the other of anaerobic exercise took the Wingate Test. Blood the respective periods of rest, post exercise, and recovery for ten minutes. The results are as follows. (1) The acute aerobic and anaerobic exercise don't affect monocyte and granulocyte. (2) The change in lymphocyte and NK cell resulting from the acute aerobic and anaerobic exercise shows a significant differences in the three different periods, but not significant differences between the two groups.

General Pharmacology of Recombinant Human Granulocyte-Macrophage Colony Stimulating Factor Expressed in Saccharomyces cerevisiae (효모에서 발현된 유전자 재조합 인간 GM-CSF의 일반 약리작용)

  • The general pharmacological tests with rhGM-CSF indicated that it had no influences on rotarod and locomotor activity tests, but shortened hexobarbital-sleeping time at the large dose of 3 mg/kg s.c. in mice. It elicited no hypothermic, analgesic and antiepileptic action. No influences on blood pressure and respiration in rabbits were observed at the dose of 1 mg/kg, i.v. and it did neither affect the receptors of adrenaline, acetylcholine, serotonin, histamine, kinin and oxytocin, nor antagonize the actions of histamine, serotonin and oxytocin at its concentrations of 1$\times$$10^{-6}$g/ml. However, this substance was demonstrated to stimulate the formation of leucocytes in rats.

  • Tobacco cells transformed with hGM-CSF gene produced $162\;{\mu}g/L$ of hGM-CSF, a valuable therapeutic protein in seven days after inoculation. The protein concentration decreased after the maximum point. It is evidenced that the environment of cell culture was inappropriate for the stability of the protein(e.g., low osmolarity which can cause cell lysis).

Histone tail cleavage as a novel epigenetic regulatory mechanism for gene expression

  • Chromatin is an intelligent building block that can express either external or internal needs through structural changes. To date, three methods to change chromatin structure and regulate gene expression have been well-documented: histone modification, histone exchange, and ATP-dependent chromatin remodeling. Recently, a growing body of literature has suggested that histone tail cleavage is related to various cellular processes including stem cell differentiation, osteoclast differentiation, granulocyte differentiation, mammary gland differentiation, viral infection, aging, and yeast sporulation. Although the underlying mechanisms suggesting how histone cleavage affects gene expression in view of chromatin structure are only beginning to be understood, it is clear that this process is a novel transcriptional epigenetic mechanism involving chromatin dynamics. In this review, we describe the functional properties of the known histone tail cleavage with its proteolytic enzymes, discuss how histone cleavage impacts gene expression, and present future directions for this area of study.

  • Objective : The aim is to identify the effects of Glycyrrhiza uralensis Fisch on immunocyte and cytokine production in asthmatic laboratory mice. This experiment was designed to investigate the antiallergic and antiinflamatary the effects of Glycyrrhiza uralensis Fisch on asthma. Materials and Methods : We measured the eosinophil, IL-4, IL-5, IL-13, $IFN-{\gamma}$, CD4, CDS, CD69, CCR3, CD11b, Gr-1 in bronchoalveolar lavage fluid of ovalbumin induced asthmatic mice. Results : Glycyrrhiza uralensis Fisch increased the proliferation of eosinophils, IL-4, IL-5, IL-13, IgE, granulocyte, CCR3, CD4, IgE, CD69. Glycyrrhiza uralensis Fisch increased the proliferation of $IFN-{\gamma}$. Conclusion : Results suggest that Glycyrrhiza uralensis Fisch extract is useful in treatment and prevention allergic asthma.
