This study was performed to develop jumbo granules floated and spreaded on the water surface after application, having low production and formulation cost, and safe to environment for paddy herbicides and to establish the formulation recipe. The jumbo granules of azimsulfuron with molinate(0.075+7.5%) was formulated by KCl as water soluble carrier and paraffin oil as solvent to impose the floating and spreading force to granules. That showed 100% of total granules to be floated on and spreaded upto the water surface within 25 minutes after application. Change in carriers, surfactants and pH did not affected to improve the time-course degradation of azimsulfuron in jumbo granules, but salt formation of azimsulfuron added by 1.15M solution of NaOH a little. Addition of N-methyl acrylate and modification of formulation process affected decrease in degradation of azimsulfuron upto 1.2, 2.1, and 7.2% after 2, 6 and 12 weeks under storage at $40{\pm}2^{\circ}C$, respectively, which showed the establishment of formulation recipe of the jumbo granules.
For the propose of increasing the value added, zeolite powder was granulated by adding Polyvinyl Alcohol (PVA) as cementing material. The optimal concentration of PVA for granulation and the physical properties of the prepared granule were investigated. Four percent PVA stock solution was made by adding distilled water at $90^{\circ}C$ water bath and incorporated with zeolite. The contents of water stable granule above 2.0mm in diameter was 98.0% in the case of 0.3% PVA addition and almost 100% in the case of 0.6% PVA addition. Increasing PVA concentration of granules increased the hardness and decreased the infiltration rate and the maximum water holding capacity of granules, However, the drying method hardly affected the physical properties of granules. With the increase in the mixing ratios of kaolinite or bentonite to zeolite, the hardness of granules increased but high water contents of granules resulted in rapid decrease in the hardness of granules. However, when smectite or perlite was mixed with zeolite, the increase in mixing ratios to zeolite resulted in the decrease in hardness, the increase in the infilteration rate and maximum water holding capacity of granules.
Starch was isolated from turnip (Brassica rapa L.), and to elucidate the structure-function relationship its structural and physical properties were characterized. Morphological structure of the starch was analyzed by SEM (Scanning Electron Microscopy). Most of the starch granules were spherical in shape with diameter ranging from 0.5-10mm. Apart from larger granules ($<10\;{\mu}m$) which dominated the population size of turnip starch, significant amount of small ($0.5-2\;{\mu}m$) and mid-size granules (${\sim}\;{\mu}m$) were also detected. It was revealed that presumably, erosion damages occurred due to the attack of amylase-type enzymes on the surface of some granules. Branch chain-length distribution was analyzed by HPAEC (High-Performance Anion-Exchange Chromatography). The chain-length distribution of turnip starch revealed a peak at DP12 with obvious shoulder at DP18-21. The weight-average chain length ($CL_{avg}$) was 16.6, and a large proportion (11.8%) of very short chains (DP6-9) was also observed. The melting properties of starch were determined by DSC (Differential Scanning Calorimetry). The onset temperature ($T_o$) and the enthalpy change (${\Delta}H$) of starch gelatinization were $50.5^{\circ}C$ and 12.5 J/g, respectively. The ${\Delta}H$ of the retrograded turnip starch was 3.5 J/g, which indicates 28.2% of recrystallization. Larger proportion of short chains as well as smaller average chain-length can very well explain relatively lower degree of retrogradation in turnip starch.
The ultrastructure of the sac and tail portion of the large ampullate gland and production of the silk materials in the orb web spider, Nephila clavata L. Koch, are studied with electron microscope. Large ampullate glands, the largest glands among the seven kinds of silk glands in this species, are composed of three parts which are the excretory duct, the storage sac and the convoluted tail. The wall of the sac is composed of a single layer of columnar epithelial cells. In the cytoplasm of these cells several kinds of the secretory granules, which are commonly associated with the rough endoplasmic reticula and had characteristic crystalloid, are seen. According to the morphology and internal textures of these granules, the sac portion is subdivided into proximal(connected with the convoluted tail) and distal(connected with excretory duct) portion. Between these two portions, the proximal portion is longer than the distal by four times. Silk materials, being synthesized to the shape of secretory granules, within the glandular epithelial cell of the tail portion are released to the inner cavity by the mechanism of the eccrine secretion. These secretory granules are originated from the rough endoplasmic reticula of the glandular epithelial cells, whereas no Golgi complexes has been found in any of the cells which have been examined.
The stimulation-secretion coupling in the pancreatic acinar cell have been studied by electron microscope. Morphological changes in the cells exhibited the cellular response induced by acetylcholine and MNNG. MNNG, a guanylate cyclase activator, induced the formation of numerous secretory granules in a period after the agent administration. This result suggest that guanylate cyclase potentiated the early sustained response in pancreatic acinar cells stimulated by acetylcholine. Cycloheximide and dibucaine reduced the secretory granules in number during sustained period. In pancreatic acinar cells, the secretion granules were considered to be directly packaged from cisternal space of endoplasmic reticulum.
Ultrastructure of the secretory portion of the small ampullate gland in the orb web spider, Nephila clavata, has been investigated using the electron microscope. The secretory portion of this gland comprise two different regions which are a S-shaped storage sac and a long, convoluted tail. By the internal textures of the secretory granules, the sac is subdivided into two regions ; the proximal region near the tail and the distal region near the duct. Commonly single layered connectives cover the basal portion of the sac epithelium, and apical portion of the epithelial cells is occupied by the thick cuticles. Within the epithelial cells of both the proximal and distal region, several types of the secretory granules surrounded by a limiting membrane and had characteristic crystalloid are scattered throughout the cytoplasm. The granular size and its electron densities are not coincide with each other according to the maturation level of the granules. The wall of the tail is composed of single layer of columnar epithelial cells, and their nuclei are found at the basal portion of the cells. Dissimilar to the epithelial cells of the sac, apical cuticles are not found at this portion. In the cytoplasm of these cells, numerous secretory granules, synthesized from the rough endoplasmic reticula commonly and had fine fibrous materials, are found. At the cell surface bordering the lumen, microvilli are seen, arid along the cellular boundaries specialized septate junctions and desmosomes appeared.
Pancreatic acinar cells exhibit a polarity that is characterized by the localization of secretory granules at the apical membrane. However, the factors that regulate cellular polarity in these cells are not well understood. In this study, we investigated the effect of Mist1, a basic helix-loop-helix transcription factor, on the cellular architecture of pancreatic acinar cells. Mist1-null mice displayed secretory granules that were diffuse throughout the pancreatic acinar cells, from the apical to basolateral membranes, whereas Mist1 heterozygote mice showed apical localization of secretory granules. Deletion of the Mist1 gene decreased the expression of type 3 inositol 1,4,5-triphosphate receptors ($IP_3R$) but did not affect apical localization and expression of $IP_3R2$. Mist1-null mice also displayed an increase in luminal areas and an increase in the expression of zymogen granules in pancreatic acinar cells. These results suggest that Mist1 plays a critical role in polar localization of cellular organelles and in maintaining cellular architecture in mouse pancreatic acinar cells.
The morphological characteristics and changes of the foregut in the grasshopper (Oxya sinuosa Mistshenko) have been studied with the special reference to the developmental stage. In light microscope level, the epithelia are seen the cuboidal shape in the 5th instar nymph and the adult. A number of pigment granules are appeared in the cuboidal epithelium of the 5th instar nymph, however the pigment granules were a few in the adult. Indistinct or undifferentiated folds of the epithelial layer were appeared in the 5th instar nymph, whereas the well-developed folds were in the adult. The well-developed muscular layers are seen in the 5th instar nymph, however in the adult the muscular layers are appeared thin or a few layer except the crop. In electron microscope level, in the foregut epithelium, a few round or oval shaped mitochondria, well-developed rER, Golgi complex, lysosome and a number of vacuoles were appeared in the cytoplasm. Two types of pigment granules, electron dense or homogenous and low electron dense or lamellar profiles, are seen in the esophageal epithelium. The crystalline profiles and plasma membrane infoldings were also appeared. In the foregut epithelium of the 5th instar nymph, however the cell organelles were generally undifferentiated and in the crop two types of pigment granules as the adult esophageal epithelium were also found.
In order to know the histological changes of rat preputial gland during normal sexual cycle and sex hormone treatment, matured Wistar rats (B.W.about 200g) were used for the experiment. Rats were subcutaneously given $2{\mu}g$ 17-${\beta}$-estradiol (Sigma) and 2.5g progesterone (Nakarai Chem., Japan) daily in 0.5ml propylen glycol for ten days respectively. The results obtained are as follow: At the stage of estrus and metestrus, the eosinophilic crystalloid granules and large vacuoles in the acinar cells appeared numerously, and the excretory ducts were severely extended. The developed connective tissue between the acinus were also found. At the stage of proestrus and diestrus, however, the small vacuoles and a few eosinophilic crystalloid granules appeared in the acinar cell. In the estradiol treatment, on the other hand, severely extended excretory ducts and a small number of the eosinophilic crystalloid granules compared with the progesterone treatment were found. The cyst-like structure was found, and in that the acinus disappeared completely. In the progesterone treatment, remarkably extended excretory ducts and nomerous appearance of oval formed eosinophilic granules in the acinar cells were found. As these findings, it could be suggested that the secretion of rat preputial gland was active at the estrus and metestrus stage, and estrogen may concerned in the secretion, land progesterone in the formation of secretory products.
To study the effects of graphite shape and the composite fabricating method on the mechanical properties of graphite/copper (Gr/Cu) composites, a copper composite using graphite flakes or graphite granules as reinforcing phases is fabricated using mechanical mixing or electroless plating method. The mechanical properties of the Gr/Cu composites are evaluated by compression tests, and the compressive strength and elongation of the Gr/Cu composites using graphite granules as a reinforcing phase are compared with those of Cu composites with graphite flakes as a reinforcing phase. The compressive yield strength or maximum strength of the Gr/Cu composites with graphite granules as a reinforcing phase is higher than that of the composites using graphite flakes as a reinforcing phase regardless of the alignment of graphite. The strength of the composite produced by the electroless plating method is higher than that of the composite material produced by the conventional mechanical mixing method regardless of the shape of the graphite. Using graphite granules as a reinforcing phase instead of graphite flakes improves the strength and elongation of the Gr/Cu composites in all directions, and reduces the difference in strength or elongation according to the direction.
